A raw extract and four extractive fractions were obtained from Cedrus brevifolia (Cyprus cedar) bark. They were all studied regarding the phenolic content and profile using spectrophotometry and HPLC-DAD-ESI-MS. The antioxidant activity was investigated using in vitro assays: DPPH and ABTS radicals scavenging and reducing power assays. The ethyl acetate fraction had the highest total phenolic and proanthocyanidin contents; a taxifolin-O-hexoside, catechin, epicatechin and procyanidin oligomers (three dimers, two trimers) were identified in this fraction. The ethyl acetate fraction was found to possess the highest DPPH and ABTS radicals scavenging effects (EC50=13.9 ± 0.3 and 2.3 ± 0.0 μg/mL, respectively) and reducing capacity (EC50=9.1 ±0.1 μg/mL). Antioxidant effects were highly correlated with total phenolic and proanthocyanidin contents (r=0.89-0.99). These results suggest that Cedrus brevifolia bark is a new source of antioxidants.

Exudates (DE) secreted from two shiitake mushroom mycelia (strains 1358 and L5458) were evaluated for their antioxidative properties and phenolic content. 1358DE and L5458DE showed distinct antioxidant activity in different in vitro assays, including scavenging activity on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals, hydroxyl radical, superoxide anions and hydrogen peroxide; the ability to chelate ferrous ions; reducing power; hemolysis inhibition activity in rat erythrocyte; and lipid peroxidation inhibition (IC50 values of 1358DE and L5458DE were 3.3 and 132.6; 44.5 and > 1000; 26.9 and 53.7; 153.6 and >175.0; 176.0 and 521.0; 26.7 and 746.4; 47.8 and 736.9; and 3.1 and > 1000 μg/mL, respectively). Their total phenolic content was 237.33 and 24.08 mg gallic acid equivalent (GAE)/g of dry DE, respectively. Overall, these results show that 1358DE generally possesses better antioxidant properties than L5458DE, possibly due to its larger total phenolic content. Shiitake mushroom mycelial exudates, particularly of 1358DE, could be a good source of natural antioxidants.

The present study was designed to confirm the traditional use of the fruits of Ficus hispida Linn. (Moraceae) as an antioxidant agent. Fruits of the plant extracted with methanol and crude methanol extract (CME) were further fractionated with n-hexane, chloroform, and ethyl acetate. All the fractions, n-hexane (NHF), chloroform (CHF), ethyl acetate (EAF), aqueous (AQF) and CME were preliminary screened for in vitro antioxidant activity and total phenolic and total flavonoid content. In DPPH radical scavenging assay, CME exhibited highest scavenging activity (IC50 = 11.20 µg/mL) as compared to other fractions. In this assay, IC50 of reference standard BHT was 5.10 µg/mL. The reducing power of the samples was in the order as AQF > CME > CHF > EAF > NHF. The results for hydrogen peroxide scavenging activity indicated that CME, EAF and AQF had almost the same scavenging activity except NHF. Total antioxidant capacity of CME and other fractions were ranked as CHF > AQF > CME > EAF > NHF.  In the assay of antioxidant constituents (total phenol and total flavonoids content), the CME had highest phenolic and flavonoids content. The results indicate that Ficus hispida fruits could be considered as a potential source of natural antioxidant.

The aim of this study is to investigate antioxidant activity and total phenolic content of ethyl acetate fraction of Sembung leaves. Sembung (Blumea balsamifera L.) is a plant that has the potential as an antioxidant because it contains secondary metabolites of alkaloids, steroids, flavonoids, saponins, and phenolic compounds. In this study, antioxidant activity was tested by using 1,1-diphenyl-2-picrylhydrazyl (DPPH) for free radical scavenging activity assay. Evaluation of total phenolic content was tested by using the Folin-Ciocalteau method. The results showed that the ethyl acetate fraction of Sembung leaves possess showed medium antioxidant activity with IC50 valueof 221.821 compare with reference standard ascorbic acid with IC50 value 68.25 ppm. The ethyl acetate fraction of Sembung leaves has a total phenolic content of 161.101 mg GAE/g. This study provided that Blumea balsamifera leaves possess antioxidant.

Free radical is any molecular species that have unpaired free electrons in their outer orbital shell that make radicals highly reactive, resulting in pathogenesis conditions such as cellular injury, premature aging, cancer, hepatic disorders, neurodegenerative diseases, cardiovascular disease, and kidney disease. One source of natural antioxidant is jackfruit. The purpose of this research was to determine the phenolic and flavonoid contents in the extracts and fractions of jackfruit peel and their potential as antioxidants. Jackfruit peel powder was extracted from maceration. The total phenolic content was determined by the Folin-Ciocalteu method. Meanwhile, flavonoid content was determined using the aluminium chloride complex colorimetric method. Measurements of antioxidant activity were conducted using the 2,2-diphenyl-1-picrylhydrazyl (DPPH). The ethyl acetate fraction had high phenolic and flavonoid contents, which were 49.667±1.508 g GAE/100 g of sample and 70.199±0.374 g of quercertin equivalent/100 g. The ethyl acetate fraction had the strongest antioxidant activity with IC50 value of 4.539±0.201 µg/ mL and correlation value (R2 ) of 0.5881 for phenols and R2 of 0.7241 for flavonoids. Ethyl acetate fraction of jackfruit peel is very potential to be developed as a natural antioxidant and functional food.

Glutinous rice-based foods have a long history are consumed worldwide. They are also in great demand for the pursuit of novel sensory and natural health benefits. In this study, we developed a novel fermented glutinous rice product with the supplementation of Fu brick tea. Using in vitro antioxidant evaluation and phenolic compounds analysis, fermentation with Fu brick tea increased the total phenolic content and enhanced the antioxidant activity of glutinous rice, including scavenging of 1,1-Diphenyl-2-picryl-hydrazyl (DPPH) radical, 2,2′-azino-bis-3-ethylbenzthiazoline-6-sulphonic acid (ABTS) radical, and hydroxyl radical, ferric-reducing antioxidant power, and ferric ion reducing power and iron chelating capability. Besides, compared with traditional fermented glutinous rice, this novel functional food exhibited a stronger activity for protecting DNA against hydroxyl radical-induced oxidation damage. Quantitative analysis by HPLC identified 14 compounds covering catechins and phenolic acids, which were considered to be positively related to the enhanced antioxidant capability. Furthermore, we found that 80% ethanol was a suitable extract solvent compared with water, because of its higher extraction efficiency and stronger functional activities. Our results suggested that this novel fermented glutinous rice could serve as a nutraceutical food/ingredient with special sensory and functional activities.

n-Hexane, ethyl acetate, and butanol fractions from methanol extract of Vietnamese Phellinus baumii fruiting body consisted of phenolics, flavonoids, and terpenoids. Ethyl acetate and butanol fractions showed a high level of total phenolics. Ethyl acetate fractions possessed the highest phenolic content (532.96 ± 19.95 mg GAE\g), accounting for approximately 53% fraction dry weight. Free radical DPPH (1,1-diphenyl-2-picrylhydrazyl) scavenging capacity of all fractions correlated with their total phenolic content. Ethyl acetate fraction exhibited the strongest capacity with an IC50 value of 0.059 mg\mL, equivalent to nearly 50% of ascorbic acid’s capacity. Its ferric reducing power was off more than half of quercetin and acid ascorbic at the concentration of 0.2 mg\mL. Ethyl acetate also had remarkable α-amylase inhibitory activity (IC50= 0.38 mg\mL). The results suggested P. baumii fruiting body as a potent source for antioxidative and α-amylase inhibitory compounds.

Objective: This study was to investigate the phytochemical screening, total phenolic content (TPC), and antioxidant activity of fractions of water, ethyl acetate, and n-hexane from Dendrophthoe pentandra (L.) Miq. and mistletoe of Moringa oleifera Lam.Methods: Simplicia extracted with 96% ethanol using maceration method. Liquid-liquid extraction was used for fractionation with water, ethyl acetate, and n-hexane as solvent. Phytochemical screening, determination of TPC, and antioxidant activity were performed against these fractions. Antioxidant activity was determined by 2,2-diphenyl-1-picrylhydrazyl method. The TPC was measured by Folin–Ciocalteu reagent and use gallic acid (GA) as a standard.Results: Water fraction contains saponin and polyphenols. Ethyl acetate fraction contains alkaloids, flavonoids, saponins, terpenoids, and polyphenols. N-hexane fraction only contains alkaloids and terpenoids. The TPC and antioxidant activity of fractions of water, ethyl acetate, and n-hexane were 9.42±1.16%, 21.35±1.86%, and 67.40±0.82% mg GA equivalent/g fraction and 29.46 μg/ml±0.99%, 7.08 μg/ml±0.39%, and 10.90 μg/ml±1.05%, respectively.Conclusion: Fraction of ethyl acetate exhibited the highest antioxidant activity and TPC. This study showed that there were significant correlations between TPC and antioxidant activity at significance level p<0.05.

PENENTUAN TOTAL FENOLIK DAN AKTIVITAS ANTIOKSIDAN FRAKSI ETIL ASETAT DARI TEPUNG PELEPAH AREN (Arenga pinnata) ABSTRAK Tujuan penelitian ini untuk menentukan total fenolik dan aktivitas antioksidan fraksi etil asetat dari tepung pelepah aren (Arenga pinnata). Tepung pelepah aren diekstraksi maserasi menggunakan etanol hasil redestilasi cap tikus kemudian di partisi menggunakan pelarut petroleum eter, etil asetat dan aquades. Penentuan kandungan total fenolik menggunakan metode Folin-Ciocalteu dan aktivitas antioksidan menggunakan metode serapan radikal 1,1-difenil-2-pikrilhidrazil (DPPH). Didapati kandungan total fenolik fraksi etil asetat sebesar 172.857 ± 4.762 mg/kg dan aktivitas penangkal radikal bebas fraksi etil asetat sebesar 90.573 µg/mL. Hasil ini menunjukan bahwa fraksi etil asetat berpotensi sebagai antioksidan alami. The determination of phenolic content was using Folin-Ciocalteu method and the antioxidant activity was done using diphenylpycrylhydrazyl (DPPH) radical scavenging. Total phenolic value of ethyl acetate fraction is 172.857 ± 4.762 and IC50 value of ethyl acetate fraction is 90.573 µg/mL. This result shows ethyl acetate fraction as natural antioxidant potential. Herbal medicines, dietary supplements, or infusions with health promoting effects can note strong consumer demand. Hence, researchers and scientists have been increasingly focusing on the promotion of medicinal plants. In this respect, the present study has the evaluation of the biological properties of different extracts (hexane, chloroform, methanol, ultrasonic, essential oil, and supercritical) obtained from two Centaurea species (C. kroumirensis (Coss.) and C. sicula L. subsp sicula) as purpose. Antioxidative activities were assessed by in vitro assays including total phenolic content, free radical scavenging assay and ferric ion reducing power. Among the fourteen extracts examined, the highest total phenolic content (TPC) and antioxidant abilities were recorded in ultrasonic ethanol extracts of both Centaurea species. Additionally, these extracts exhibited a noticeable strong protective effect of the human skin fibroblast cell line (HS-68) that was exposed to oxidative stress induced by hydrogen peroxide. According to our results, the sonication with ethanol might be an ideal, rapid, and eco-friendly extraction method for obtaining a polyphenol-rich extract that exhibited high antioxidant activity. The results presented herein suggest that Centaurea species could be promoted as an antioxidant resource that could be exploited for the development of nutraceuticals or in pharmacology industry.

This work aimed to evaluate the phenolic content and in vitro antioxidant, antimicrobial and enzyme inhibitory activities of the methanol extracts and their fractions of two edible halophytic Limonium species, L. effusum (LE) and L. sinuatum (LS). The total phenolic content resulted about two-fold higher in the ethyl acetate fraction of LE (522.82 ± 5.67 mg GAE/g extract) than in that of LS (274.87 ± 1.87 mg GAE/g extract). LC-MS/MS analysis indicated that tannic acid was the most abundant phenolic acid in both species (71,439.56 ± 3643.3 µg/g extract in LE and 105,453.5 ± 5328.1 µg/g extract in LS), whereas hyperoside was the most abundant flavonoid (14,006.90 ± 686.1 µg/g extract in LE and 1708.51 ± 83.6 µg/g extract in LS). The antioxidant capacity was evaluated by DPPH and TAC assays, and the stronger antioxidant activity in ethyl acetate fractions was highlighted. Both species were more active against Gram-positive bacteria than Gram negatives and showed considerable growth inhibitions against tested fungi. Interestingly, selective acetylcholinesterase (AChE) activity was observed with LE and LS. Particularly, the water fraction of LS strongly inhibited AChE (IC50 = 0.199 ± 0.009 µg/mL). The ethyl acetate fractions of LE and LS, as well as the n-hexane fraction of LE, exhibited significant antityrosinase activity (IC50 = 245.56 ± 3.6, 295.18 ± 10.57 and 148.27 ± 3.33 µg/mL, respectively). The ethyl acetate fraction and methanol extract of LS also significantly inhibited pancreatic lipase (IC50 = 83.76 ± 4.19 and 162.2 ± 7.29 µg/mL, respectively). Taken together, these findings warrant further investigations to assess the potential of LE and LS as a bioactive source that can be exploited in pharmaceutical, cosmetics and food industries.
