In vitro Propagation of Mentha pulegium L. and Testing, The Antifungal Activity of Stem Nodes Callus and Shoots Extracts Against Acremonium strictum L

In this study, we evaluate the potential for in vitro propagation of Mentha spp. for mass production. The two Mentha spp. (Mentha x piperita L., M. pulegium L.) were propagated with four successive 60-day subcultures in MS medium supplemented with for 100µL/L NAA (Naphthylacetic Acid) and 600µl/L IBA( Indole Butyric Acid). The shoots were rooted in the same media. The rooted plantlets were finally acclimatized in a growth room. Callus induction was carried out in MS (Murashige and Skoog) media supplemented with 100µL/L NAA and 250µL/L BAP (Benzylaminopurine). Callus was successfully induced from nodes of Mentha pulegium L. Through micropropagation, both Mentha spp increased in multiplication rates around 6-fold per month compared to the traditional propagation method. Mentha x piperita and Mentha pulegium showed the most significant potential for plantlet production through the micropropagation method.

The aim of this work was to prepare and evaluate the topical carbopol gel formulation containing ketoconazole encapsulated liposomes. Ketoconazole loaded liposomes were prepared by thin film hydration technique. The prepared liposomes were incorporated into 1% carbopol gel, and the systems were evaluated for in-vitro drug release, drug retention into skin and in-vitro antifungal activity. The in-vitro permeation of ketoconazole using wistar albino rat skin from liposomal gel was compared with that of plain drug gel and also with plain drug cream containing 2% w/w of ketoconazole. The release of ketoconazole from liposomal gel was much slower than from non liposomal formulations. Gel containing liposomal ketoconazole showed maximum antifungal activity after 30 hours over plain ketoconazole gel and cream formulations.

Shallot (Allium ascalonicum L.) has been known as traditional medicine. Shallot which has same genus with garlic(Allium sativum L.) contains allicin that is also found in garlic and has been suspected has fungicidal activity toCandida albicans. It is supported by several researches. Therefore, shallot is suspected has antifungal activity too.The aim of this research was to know antifungal activity of shallot’s water extortion againsts Candida albicans invitro. This was a laboratory experimental research which used completely randomized design, with diffusion method.Shallot’s water extortion was devided into three concentrations, there were 50%, 100% and 200%. Ketoconazole 2%was positive control and aquadest was negative control. The result of this research based on analysis of varians(Anova), there was significant difference between several treatments and was confirmed with Duncan New MultipleRange Test (DNMRT) p<0,05, there was significant difference between 100% shallot’s water extortion with othertreatments, but there was no significant difference between 50% shallot’s water extortion with 200% shallot’s. Theconclusion was shallot’s water extortion had antifungal activity againsts Candida albicans with the best concentration100%, but it was lower than ketoconazole 2%.
