Despite intensive efforts put on prevention of environment pollution by nitroaromatic compounds, these xenobiotics have not been eliminated from the biosphere. The physicochemical properties make nitroaromatics extremely recalcitrant to biodegradation. Therefore, microbial degraders of these pollutants are sought after. This paper reports preliminary results of the study on degradation of 3,5-dinitrosalicylic acid (DNS) by a basidiomycetous fungus Phanerochaete chrysosporium under stationary conditions in a culture medium containing 0.05–0.5% v/v of DNS. The results obtained suggest that the fungus degrades DNS through the reductive pathway.

A new bioassay for A.C.T.H. has been devised utilising non-proliferative organ culture of segments of guinea pig adrenal. The cultured adrenal is exposed to standard or unknown A.C.T.H. and sections stained for the presence of reducing groups. The intensity of staining in the zona reticularis is measured by scanning and integrating microdensitometry and is inversely proportional to the logarithm of the concentration of A.C.T.H. in the culture medium over the range 0.0027–27 pg/ml. Preliminary results using the assay, which greatly exceeds others in sensitivity, on human plasma are similar to those obtained with standard techniques.

The felsic volcanic rocks and tuff aged from Late Mesozoic to Kainozoic occur in many places in Viet Nam Territory, such as Tu Le area (in Northwest Region), Binh Gia area, Binh Lieu area (Northest Reigion), Nha Trang area, Dalat area (Central Region). The mafic volcanic ashes aged from Late Neogen to Quarternary distribute in central part of Vietnam such ash Con Co Island, Van Hoa High land. The samples of the tuff rock of Don Duong Stratum collected from field trip in Da Lat area have been treated (burned) at 650oC, 750oC, 900oC. The experimental results show that these materials, after treatment (heating in funace at the temperature 650oC, 750oC and 900oC) expressed absortion capacity up to 90% of selected organic matters (MO, MD) and heavy Metal (Pb2+) in the testing solutions. Among them, the sample heating at 900oC is the best material for Pb2+ absorption. The preliminary results show that the volcalnic felsic rocks and volcanic ashes in Viet Nam, after suitable treatments, can be used as an enviroment treatment materials.

The aim of this study was to analyze the response of bovine embryos fertilized in vitro to a tumor dog cell- conditioned TCM-199 culture medium used in the development stage. Two hundred ninety-five oocytes obtained from slaughterhouse ovaries were cultured in groups of 20 cells at 39°C in TCM-199 medium droplets with 20% estrous cow serum (ECS) for 24h in a humid atmosphere containing 5% CO2. After incubation, these oocytes were fertilized with a 1 × 106 sperm cell dose, previously capacitated by a 1-h incubation in SP-TALP medium under usual conditions, by incubation in small petri dishes with 1 mL of Fert-TALP medium for 24 h under the conditions described previously. Alleged fertilized oocytes were randomly separated into 2 groups. A control group (n = 144) was cultured in TCM-199 medium with 10% fetal calf serum (FCS) and the other (n = 151) was cultured in TCM-199 medium previously conditioned by incubation for 72 h with 1 × 106 cells of a complex adenocarcinoma grade 1 obtained from a 6-year-old boxer dog and sterilized by filtration with 0.2-(im membrane. After 72 h of culture, the zygotes were harvested and prepared for a quick microscope observation. Alleged zygotes were classified as “not fertilized,” “divided,” or “degenerated.” Statistical differences (P < 0.01) were found among both groups with a greater number of normal divided embryos (65.2%) in the conditioned medium group and a lower number (30.4%) in the control group. The number of normal divided embryos in the conditioned medium after 72 h of incubation was similar to that achieved by current methods. These preliminary results are consistent with results obtained by other researchers in which growth factors derived from established cell lines can be used to condition culture medium for in vitro bovine embryo development. Further studies will be necessary to clarify how these factors contribute to in vitro embryo development. Spanish Retinta Breed Asociation, Spain.

SummaryCrosslinked gels of albumin as well as heparinized albumin gels, potential sealants of prosthetic vascular grafts, were studied with regard to in vitro stability, binding of basic fibroblast growth factor (bFGF) and cellular interactions. A small percentage of the heparin present in these gels, was released during storage in SDS solution. During storage in cell culture medium at 37° C, heparin release was 21-25 percent. Release of albumin did not occur.Human umbilical vein endothelial cells (HUVECs) rapidly adhered and subsequently spread on (heparinized) albumin gels, but proliferation was only observed if heparin was present in the gel.Binding of 125I-bFGF to heparinized albumin gel was 35 percent higher than to non-heparinized albumin gel. Growth of HUVECs occurred only on heparinized albumin gel loaded with bFGF and not on bFGF-loaded albumin gel.The number of platelets deposited under stationary conditions onto heparinized albumin gel was about twice the number found on nonheparinized albumin gel. Seeding of HUVECs on heparinized albumin gel, significantly reduced the number of platelets adhering to this surface. Moreover, no spreading of platelets was observed on substrates seeded with HUVECs.It can be concluded that crosslinked gels of albumin to which heparin is immobilized, are candidate sealants for prosthetic vascular grafts and suitable substrates for endothelial cell seeding.

Galaxolide (1,3,4,6,7,8-hexahydro-4,6,6,7,8,8-hexamethyl-cyclopenta-γ-2- benzopyran, HHCB) is recognized as a novel contaminant in water and has potential adverse impacts on aquatic organisms. The toxic effect of HHCB on Phanerochaete chrysosporium was investigated by exposure of the fungus in nitrogen-limited culture medium to various concentrations of HHCB. DNA damage of P. chrysosporium by HHCB was detected. Comparing with that in the control, the percent polymorphism under different concentrations of HHCB increased, from 21.4% to 42.9%. In addition, the result of UPGMA (un-weighted pair group method of arithmetic means) dendrogram showed that the Simple Matching Coefficient (SM) was decreased with an increase in the concentrations of HHCB. Thus, as an environmental pollutant, HHCB has the toxic effect on P. chrysosporium at molecular level.

The article provides information about the way of obtaining chitin-chitosan biosorbents from the fungus Rhizopus oryzae, using mild deacetylation and demineralization conditions. Such physicochemical properties as deacetylation degree, total ash, weight loss on drying etc. were determined. Method of obtaining the chitin-chitosan complex, shown in the current study, allows to obtain a pure biosorbent with a high degree of deacetylation possible to be further used as entero-and hemosorbent.
