AbstractKindlin-2 is known to play important roles in the development of mesoderm-derived tissues including myocardium, smooth muscle, cartilage and blood vessels. However, nothing is known for the role of Kindlin-2 in mesoderm-derived reproductive organs. Here, we report that loss of Kindlin-2 in Sertoli cells caused severe testis hypoplasia, abnormal germ cell development and complete infertility in male mice. Functionally, loss of Kindlin-2 inhibits proliferation, increases apoptosis, impairs phagocytosis in Sertoli cells and destroyed the integration of blood-testis barrier structure in testes. Mechanistically, Kindlin-2 interacts with LATS1 and YAP, the key components of Hippo pathway. Kindlin-2 impedes LATS1 interaction with YAP, and depletion of Kindlin-2 enhances LATS1 interaction with YAP, increases YAP phosphorylation and decreases its nuclear translocation. For clinical relevance, lower Kindlin-2 expression and decreased nucleus localization of YAP was found in SCOS patients. Collectively, we demonstrated that Kindlin-2 in Sertoli cells is essential for sperm development and male reproduction.

The activeness of students in learning is of course very necessary because by activating their five senses, they will learn more meaningfully. One of the teaching methods that can activate students' five senses is the inquiry method. The purpose of this study was to describe the increase in science learning outcomes at MIS Gampong Mutia. The design of this study used a control group posttest design. The instrument used is a test of learning outcomes for the material structure and function of plant parts. The results showed that there was an increase in science learning outcomes in classes that applied the inquiry method. So it can be said that this guided inquiry method is appropriate for use on the material structure and function of plant parts

The ultrasonic vocalizations of mice are attracting increasing attention, because they have been recognized as an informative readout in genetically modified strains. In addition, the observation that male mice produce elaborate sequences of ultrasonic vocalizations (‘song’) when exposed to female mice or their scents has sparked a debate as to whether these sounds are—in terms of their structure and function—analogous to bird song. We conducted playback experiments with cycling female mice to explore the function of male mouse songs. Using a place preference design, we show that these vocalizations elicited approach behaviour in females. In contrast, the playback of whistle-like artificial control sounds did not evoke approach responses. Surprisingly, the females also did not respond to pup isolation calls. In addition, female responses did not vary in relation to reproductive cycle, i.e. whether they were in oestrus or not. Furthermore, our data revealed a rapid habituation of subjects to the experimental situation, which stands in stark contrast to other species' responses to courtship vocalizations. Nevertheless, our results clearly demonstrate that male mouse songs elicit females' interest.

The paper presents the results of research on the development and implementation of environmentally friendly practices of veterinary preparations, namely placental able to normalize metabolic processes in tissues and restore the structure and function of organs and body systems. We established that the yard at multiple input of «Horiotsen» increased protein levels on 1,5 % compared to the index of the first days after farrowing and 5,3 % – rate of animals in the control group at 28th day. Glucose increase after the injection in all groups – p<0,001, but in the experimental groups the figure was higher than in the control one by 8,7, 7,0 and 6,1 %. Against the background of hyperphosphatemia content of inorganic phosphorus in the blood sows of 3rd research group at the 28th day was 17 Im% lower than in the control group of sows.

AbstractAim: Pirfenidone is a novel anti-fibrotic agent utilized in the treatment of idiopathic pulmonary fibrosis (IPF). It has been implicated in mitigating myocardial fibrosis and left ventricular (LV) systolic and diastolic dysfunction in animal models. However, its impact on LV mechanics in humans remains unknown. The aim of this study was to retrospectively evaluate the effects of pirfenidone on echocardiographic parameters of LV function and structure in patients with IPF.Methods: A total of 124 patients with IPF were included in this study: 64 patients treated with pirfenidone (treatment group) and 60 patients not taking pirfenidone (control group), who had serial pretreatment/baseline and posttreatment/follow-up echocardiograms done within a time frame of four years. Changes in the means of parameters of LV function (systolic, diastolic, and global longitudinal strain) and LV structure (mass and volume indices) were compared between the treatment and control groups. This was followed by a subgroup analysis that included only 88 patients (47 treated, 41 controls) with echocardiographic evidence of myocardial dysfunction at baseline (defined as an ejection fraction of ≤45, or diastolic dysfunction stage 1 or more) in addition to a known clinical diagnosis of congestive heart failure. To account for potential confounders, a secondary adjusted analysis by way of 1:1 propensity score matching (PSM) was carried out. This yielded a sample consisting of 62 patients with 56 patients in the subgroup cohort. Results: Patients in the treatment group were significantly younger (69.4 vs. 77 years, p<0.001) and had relatively lower forced vital capacity (69.9% vs. 80.6%, p = 0.005) in comparison to the control group. However, after PSM, the age demographics were comparable between both groups (72.18 vs. 72.15, p = 0.9). In the primary unadjusted analysis, there was no statistically significant change in any of the mean parameters of LV function and structure after pirfenidone administration when compared to the control group. Furthermore, no significant differences were noted in the subgroup cohort. Such findings were re-demonstrated after a secondary analysis with PSM.Conclusion: From an echocardiographic perspective, pirfenidone had no significant effects on LV structure and function in patients with IPF, even in patients with more overt cardiac dysfunction.

Abstract Background The purpose of this study was to evaluate left ventricular systolic function in patients with different types of ischemic heart disease using two-dimensional speckle tracking imaging (2D-STI). Methods We retrospectively studied patients who were admitted to Peking University Third Hospital from January 2011 to December 2017 due to chest tightness and chest pain. Two hundred forty-two patients were divided into control group, CMD group and obstructive CAD group. The main coronary artery stenosis was confirmed by coronary angiography or coronary computed tomography and coronary flow reserve (CFR) in patients was measured by transthoracic Doppler echocardiography. Left ventricular strain and strain rate (SR) measured by 2D-STI. Cardiac structure and function were measured by conventional echocardiography. Results Conventional echocardiography showed that there was no significant difference in cardiac structure and function among the three groups (P > 0.05). Moreover, the longitudinal strain (LS) of each ventricular wall in CMD group was notably lower than that in control group (P < 0.01). In addition, global longitudinal SR and longitudinal SR in CMD group and obstructive CAD group were obviously lower than those in control group (P < 0.01). GLS, endocardial LS and epicardial LS were negatively correlated with CFR (P < 0.01). Conclusions Early left ventricular systolic dysfunction was found in patients with CMD and patients with obstructive CAD, with similar degree. CFR is an independent influencing factor of GLS. GLS and stratified LS have certain diagnostic value for CMD.

In the conditions of experi mental chronic gastritis and duodenitis reproduction, carried out among Wistar line 24 rats, was investigated effect of the nitrogen oxide stable metabolites content in the gums homogenates on a lamina propria structure and function. Results of our research have been shown, that level of nitrogen oxide stable metabolites in the rats’ gums tissues homogenates with gastritis and duodenitis decreased in three times, compared with a same period at the intact animals, which indicated about depletion of a nitrogen oxide depot at the given disease. It was revealed a significant decrease typical for the protein and glycoprotein content in the animals gums tissues at the experimental group, which indicated about the catabolic processes predominance. Amount of hexosamines in the gums was significantly increased, which was correlated with a morphological picture (gums lamina propria impregnation with the blood plasma proteins), testified about a change in the vascular wall permeability and microcirculation disruption. In the animals from experimental group was reduced a height of gums papillae connective tissue to 27.18±1.86 x 10-6 m against 56.93±2.64 x 10-6 m in the rats from control group. In the gums lamina propria defines phenomenon of the papillary and reticular layer fibrosis. After medicamentous correction, carried out with an antioxidant and the nitrogen oxide donor was observed repairing structure of the gums lamina propria on a background of increasing microvasculature area. It was determined significantly increased protein and glycoprotein content in the gums tissues with increased levels of nitrogen oxide metabolites.
