Hill-Activity and P700 Concentration of Chloroplasts Isolated from Radish Seedlings Treated with -Indoleacetic Acid, Kinetin or Gibberellic Acid

Abstract The Hill-activity (reduction of DCPIP or methylviologen) and the concentration of P700 were studied in chloroplasts isolated from cotyledons of radish seedlings (R aphanus sativu s L. Saxa Treib), which had been grown with the addition of β-indoleacetic acid (IAA), kinetin, or gibberellic acid.1) The photosynthetic activity of young chloroplasts from 3 day old Raphanus seedlings is very high (c. 180 μmol O2/mol chlorophyll × h) and decreases continuously thereafter with increasing age. The steady state Hill-activity is readied after 8 to 10 days (values of 55 to 50 μmol 02/mg chlorophyll × h).2) Chloroplasts from plants treated with IAA or kinetin not only exhibit higher plastoquinone levels 1,2, but also a higher P700-content and a higher Hill-activity. The promotion effect is more pronounced with kinetin (+ 36 to 40%) than with IAA (+ 12 to 17%).3) Gibberellic acid has a different effect on composition and activity of chloroplasts. In younger seedlings the Hill-activity appears to be somewhat stimulated, without promotion effect on plasto­ quinone 2 or P700 concentration. After 10 days GA3-treated plants show signs of chlorosis combined with a strong decrease in photosynthetic activity.4) The data clearly demonstrate that the composition and activity of the photosynthetic ap­ paratus are under phytohormone control. IAA and even better kinetin promote the light induced formation of pigment systems and electrontransport chains. GA3 seems to block the rebuilding of the photosynthetic apparatus under steady state conditions.

Abstract Barley seedlings were grown for 7, 10 or 13 days under continuous white light (Fluora lamps) on a nutrient solution containing simazine (2-chloro-4,6-bis-(ethylamino)-s-triazine, 10, 100 μᴍ) . Accumulation of chlorophylls and in part of carotenoids was increasingly enhanced depending on age and concentrations applied. The ratio chlorophyll a/b decreased on this line in 10 and 13 day old plants, the ratio xanthophylls/β-carotene and the ratio chlorophyll a/prenylquinones (plasto-quinone-90X. + red. , α-tocopherol, α-tocoquinone) increased. The way how these prenyllipid ratios are changed in 10 and 13 day old plants is characteristic of a shade type adaptation, as it was shown earlier for other herbicides inhibiting photosystem II.In 7 day old plants the ratio chlorophyll a/prenylquinones decreased. Photosynthetic activity (Hill-reaction) was enhanced in the simazine plants. The ratio chlorophyll a/b was higher, the ratio xanthophylls/β-carotene was lower than in the older seedlings.Similar changes of prenyllipid ratios like in 7 day seedlings and a higher Hill activity were also found in plants grown under blue light (sun type adaptation) as compared to red light (shade type adaptation). This points to similar metabolic changes in the chloroplasts which could be related to a common site of regulation, perhaps the endogenuous cytokinins. The Hill activity, increasing with age in the 10 and 13 day plants, indicates that the mode of action of simazine may be a multiple process resulting to a parallel formation of shade type and sun type characteristics.

ABSTRACT Transit searches have uncovered Earth-size planets orbiting so close to their host star that their surface should be molten, so-called lava planets. We present idealized simulations of the atmosphere of lava planet K2-141b and calculate the return flow of material via circulation in the magma ocean. We then compare how pure Na, SiO, or SiO2 atmospheres would impact future observations. The more volatile Na atmosphere is thickest followed by SiO and SiO2, as expected. Despite its low vapour pressure, we find that a SiO2 atmosphere is easier to observe via transit spectroscopy due to its greater scale height near the day–night terminator and the planetary radial velocity and acceleration are very high, facilitating high dispersion spectroscopy. The special geometry that arises from very small orbits allows for a wide range of limb observations for K2-141b. After determining the magma ocean depth, we infer that the ocean circulation required for SiO steady-state flow is only 10−4 m s−1, while the equivalent return flow for Na is several orders of magnitude greater. This suggests that a steady-state Na atmosphere cannot be sustained and that the surface will evolve over time.

Application of biostimulants instigates many physiological processes that enhance nutrition efficiency, abiotic stress tolerance, and quality traits of crops, regardless of their nutrient content. One of such preparations is Atonik which contains nitrophenol compounds naturally occurring in plant cells. Several studies have confirmed its beneficial effect on the growth, development, and improved metabolic activity of plants. Therefore, it seems advisable to investigate the effect of Atonik preparation on the photosynthetic activity of Moldavian dragonhead (Dracocephalum moldavica L.). The reported study was carried out in 2014 in Perespa, Poland. Over the growing season, Atonik was foliar-applied at a dose of 0.3 L/ha (0.1%) and 0.6 L/ha (0.2%) by single and double spraying of plants. Chlorophyll content and nitrogen status (N) were estimated by a Chlorophyll Meter SPAD-502 Plus. The foliar application of Atonik was found to improve the efficiency of the photosynthetic apparatus and chlorophyll content in the leaves of Dracocephalum moldavica plants, but the results were dependent on biostimulant concentration and number of its applications, and on the date of measurement. To conclude, Atonik is an environmental-friendly preparation which has a positive impact on the metabolic processes of plants.

A new modified Reynolds equation is derived with centrifugal force acting on the hydrodynamic oil film being considered. This equation, together with a cavitation model, is used to obtain the steady-state equilibrium and calculate the rotordynamic coefficients of lightly loaded floating bush journal bearings operating at very high shaft speeds. The bush-to-shaft speed ratio and the linear cross-coupling spring coefficients of the inner oil film is found to decrease with the increase in shaft speed as the axial oil film rupture develops in the inner oil film. The present model can give reasonable explanation to the steady-state behavior and the stability behavior of the bearing observed in actual machines.

1. The kinetics of the inactivation of photosynthesis by 2537 Å in Chlorella pyrenoidosa and Scenedesmus D1 indicate that, while the destruction process is largely a first order effect, higher order effects also occur, which become evident at low exposures. In agreement with previous observations, endogenous respiration is insensitive to exposures which inactivate photosynthesis. 2. In Scenedesmus D1 a solid dose of ultraviolet has no more effect on the photosynthetic apparatus than a dose of equal total duration interrupted by periods of photosynthesis. Nor is any difference noted if the cells are in a different buffer, e.g. 0.05 M KH2PO4, or carbonate-bicarbonate buffer 9. 3. In C. pyrenoidosa, a solid dose and an interrupted dose cause equal effects on photosynthesis when neutral phosphate buffer is used. If the ultraviolet exposure schedules are identical, equal effects are also noted in cells suspended in buffer 9, and in 0.05 M phosphate (pH 6.2). Solid exposures are, however, much more effective than interrupted exposures, when buffer 9 is used. 4. Oxygen evolution (Hill reaction), photosynthesis, and photoreduction in Scenedesmus D1 are equally sensitive to a given dose of ultraviolet. The mechanism responsible for adaptation to hydrogen metabolism is not more sensitive to ultraviolet than is the photosynthetic mechanism. The O2/H2/CO2 reaction in darkness is less sensitive to ultraviolet than any of the above reactions. 5. Glucose oxidation by C. pyrenoidosa, and colony formation in Scenedesmus D1 are far more sensitive to a given dose of ultraviolet than photosynthesis in these organisms. 6. The photosynthetic apparatus of C. pyrenoidosa is more sensitive to ultraviolet than that of Scenedesmus D1. 7. The Hill reaction in chloroplast fragments is also inactivated by 2537 Å by a first order process. Exposures which inactivate this reaction completely have no effect on polyphenol oxidase, cytochrome oxidase, or catalase in the same chloroplast preparation. 8. After irradiation, the survival of photosynthesis in Scenedesmus D1 and of the Hill reaction in chloroplast fragments are independent of the light intensity used to measure these processes. 9. No significant changes occur in the ultraviolet absorption of chloroplasts after an exposure to 2537 Å, which completely inactivates the Hill reaction.
