In vitro Antioxidant Capacities of Two Benzonaphthoxanthenones: Ohioensins F and G, Isolated from the Antarctic Moss Polytrichastrum alpinum

Antioxidant agents against reactive oxygen species can be used for several cosmetic and medicinal applications. This study’s objective was to evaluate the antioxidant activities of Polytrichastrum alpinum (Hedw.) G. L. Sm. (Polytrichaceae), an Antarctic moss species collected from King George Island (Antarctica). The identifi cation of the moss species was performed on the basis of morphological characteristics and molecular sequencing of the 18S rRNA gene. Two benzonaphthoxanthenones: ohioensins F and G, were isolated from the extract after several chromatographic procedures. The various in vitro antioxidant capacities of a methanolic extract of P. alpinum and the isolated compounds were evaluated by analyzing the scavenging capacities of free radicals of 2,2-azino-bis(3-ethylbenzthiazoline- 6-sulfonic acid) (ABTS) and 2,2-diphenyl-1-picrylhydrazyl (DPPH), the total phenol assay with Folin-Ciocalteu reagent, the ferric ion (Fe3+) reducing power and the nitric oxide (NO) scavenging activity and compared to those of commercial standards for each assay. The experimental data showed that even the crude extract of P. alpinum exhibited potent antioxidant activity. The antioxidant activity was increased two- to seven-fold for the purified compounds. The antioxidant activities of both purified compounds were found to be more or less the same in all experiments. However, the obtained data showed that the Fe3+ reducing power of the purified compounds and crude methanolic extract was almost the same suggesting the presence of other stronger reducing agents in the methanolic extract which could not be isolated in the present experiment. Therefore, further work on the isolation of these stronger antioxidant agents from this moss specimen of the extreme environment is warranted. Developments of laboratory mass culture techniques are anticipated to achieve bulk production of the active constituents for commercial application.

Background: Quercus serrata Murray leaves have been used traditionally in the treatment of diabetes, dysmenorrhoea, inflammation and urinary tract infection. So, far no study had been reported on the toxicological profile and antioxidant properties of the plant. Objective: The present study was aimed to investigate the in-vivo toxicological profile and in-vitro antioxidant activities of the methanolic extract of standardized Quercus serrata leaves. Methods: Per-oral sub-acute toxicity study was performed in rats using three dose levels (200, 400 and 800 mg/kg b.w.) of the extract for 28-days. Control group received gum acacia suspended in water. Bodyweight was measured weekly. Biochemical parameters were analysed using the serum, the blood-cell count was done using whole blood. Pathological changes were also checked in highly perfused tissues. Further, in-vitro reducing power assay, nitric oxide scavenging assay, DPPH free-radical scavenging assay were performed to check the antioxidant activity of the extract. Results: There were no significant alterations in the blood-cell count and biochemical parameters analysed in the treatment group when compared with the normal control. Histopathology study of liver, kidney, pancreas, heart and brain revealed normal cellular architecture in the treatment groups alike the control group animals. Quercus serrata also showed a significant reduction of DPPH with IC50 4.48±0.254 µg/mL, in-vitro reducing power activity with IC50121.65±0.320 µg/mL and nitric oxide scavenging activity IC50 106.43±0.338 µg/mL. Conclusion: The above study showed that standardized methanolic extract of Quercus serrata leaves was safe after subacute oral administration in rats and has good antioxidant potential.

Amomum subulatum, commonly known as large or black cardamom, is a commonly used spice in Indian kitchens and is traditionally used to treat various ailments. To add more knowledge about the medicinal values of the fruit, this study was conducted to evaluate the in vitro antioxidant activities of aqueous, methanol, ethanol, hydro alcohol, ethyl acetate, acetone and chloroform extracts of the fruit. Preliminary assessment was done to detect the presence of phytoconstituents using identification tests. The antioxidant activity was measured by employing methods such as diphenylpicrylhydrazyl (DPPH) radical scavenging assay, total antioxidant activity equivalent to ascorbic acid, reducing power assay and superoxide anion scavenging assay. The antioxidant activities were compared with their respective phenol and flavonoid contents. Preliminary assessment revealed that large cardamom fruit is a good source of all the bioactive constituents as well as phenol and flavonoid essential for medicinal values. The extract obtained by polar solvents showed the highest antioxidant efficacy in relation to its phenol content. Also, all the solvent-soluble fractions showed a concentration-dependent antioxidant effect. Results from our study prove that large cardamom can alleviate oxidative stress, suggesting the potential of large cardamom as a functional food

The aim of this work is to deepen the pharmacological effect of 4-phenyl-1,5-benzodiazepin-2-one derivatives which have a similar structure to nonionic surfactants: 4-phenyl-1,5-benzodiazepin-2-one is the hydrophilic head, and the carbon chain is hydrophobic tail. The antinociceptive activity of 4-phenyl-1,5-benzodiazepin-2-one derivatives was determined using acetic acid-induced writhing and tail immersion tests. In addition, the in vitro antioxidant activities of the tested derivatives were determined by using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging method and ferric reducing power assay. A single oral administration of these compounds at the doses of 50 and 100 mg/kg significantly reduced the number of abdominal writhes induced by acetic acid injection. Acute pretreatment with 4-phenyl-1,5-benzodiazepin-2-one derivatives at the dose of 100 mg/kg caused a significant increase in the tail withdrawal latency in the tail immersion test. Additionally, a significant scavenging activity in DPPH and reducing power was observed in testing antioxidant assays. Finally, we carried out a study of the antioxidant activity of these derivatives. The results of this study reveal that these compounds have a low antioxidant activity compared to the BHT. It decreases with the polarity of the molecule. The present study suggests that 4-phenyl-1,5-benzodiazepin-2-one derivatives possess potent antinociceptive and antioxidant effects, which suggest that the tested compounds may be useful in the treatment of pain and oxidation disorders.

Rosmarinus eriocalyx Jord. & Fourr is a well-known aromatic and medicinal plant whose consumption serves to remedy a number of disorders, evergreen bush endemic to Algeria. The present study aimed it investigating the in-vitro anti-inflammatory and antimicrobial for two extracts of Rosmarinus eriocalyx. The antioxidant activity was evaluated by DPPH and the reducing power essay, anti-inflammatory activity with the Human Red Blood Cell (HRBC) membrane stabilization method. However, the antimicrobial activity was tested with three bacterial strain Pseudomonas aeruginosa ATCC 27853, Staphylococcus aureus ATCC25923 and Bacillus subtilis ATCC6633. The results show an excellent antioxidant and an interesting inflammatory activities but a weak power against the used strains therefore. © 2020 International Formulae Group. All rights reserved. The study was designed to investigate the antioxidant activities of the methanolic extract of leaves of Begonia roxburghii by five different in vitro methods namely Folin-Ciocalteu, total flavonoid content, phosphomolybdenum, total reducing power determination, and DPPH scavenging activity. Phenol content (Gallic acid equivalent) was found as 180.40±0.03 mg/g of the dry sample by the Folin-Ciocalteu method. Flavonoid assay method revealed the content of flavonoids (Quercetin equivalent) as 60.43±0.27 mg/g of the dry sample. Total antioxidant capacity of the extract was observed 243.43±0.11 mg/g ( ascorbic acid equivalent) by phosphomolybdenum method. Free radical scavenging activity (measured by the DPPH test) of the methanolic extract was found 158.36±0.22 μg/mL whereas ascorbic acid, the standard, showed 19.53±0.04 μg/mL. The methanolic extract of Begonia roxburghii leaves exhibited positive and linear dose dependent reducing power activity and its activity was comparable to the standard ascorbic acid for a given dose. Phytochemical screening of the extract showed positive results for alkaloids, carbohydrates, flavonoids, glycosides, saponins, steroids, and tannins, which demonstrate definite pharmacological actions on human body. Ayurveda contains a variety of medicines including the polyherbal fermented traditional medicines namely Arishta and Asava. It remains as a rich source of bioactive phytochemicals including antioxidants. There are scanty publications on the antioxidant activity of these medicines. There is a need to explore the natural antioxidants to replace synthetic counterparts which requires an assessment in terms of antioxidant activity by in vitro analysis. This study employs a range of antioxidant assay systems containing various free radicals (DPPH, hydroxyl, hydrogen peroxide, super oxide, nitric oxide and ABTS), Inhibition of free radical generation (metal chelating) and H+ – donating ability (reducing power) were performed. Ashokarishta, Aswagandharishta and Dasamoolarishta exhibited higher degree of antioxidant activities but less than the corresponding standards. Based on IC50 values, the hydrogen peroxide scavenging ability of Ashokarishta is comparable to the standard L-ascorbic acid. In ABTS assay, Aswagandharishta and Dasamoolarishta are comparable to the standard Butylated hydroxytoluene (BHT). These may be due to the phytochemical heterogeneity of the samples assayed. Thus study can lead to the sourcing of Ayuvedic polyherbal fermented medicines as a novel source of natural antioxidant molecules that might be acting as free radical quencher or scavenger and to treat human ailments.

Background: Protocatechuic acid (PCA) is a natural phenolic acid widely distributed in plants and is considered as an active component of some traditional Chinese herbal medicines such as Cibotium barometz (L.) J.Sm, Stenoloma chusanum (L.) Ching, Ilex chinensis Sims. PCA was reported to possess various pharmacological effects which may be closely correlated with its antioxidant activities. However, the antioxidant of PCA has not been investigated systematically yet. Methods: In the study, the antioxidant activities of protocatechuic acid were measured in vitro using various antioxidant assays including 1,1-diphenyl-2-picryl-hydrazyl (DPPH•), 2,2’-azino-bis (3-ethylbenzthiazoline-6-sulfonic acid) (ABTS+•), superoxide anion radicals (•O2-) and hydroxyl radical (•OH) scavenging activity, ferric ions (Fe3+) and cupric ions (Cu2+) reducing power, ferrous ions (Fe2+) and cupric ions (Cu2+) chelating activity, compared with the positive controls Trolox or BHT.Results: In all assays, PCA along with positive controls exhibited dose-dependently antioxidant ability. Comparing to a standard antioxidant Trolox, the relative antioxidant activity of PCA (i.e. the ratio of IC50(Trolox)/IC50(PCA) ) was calculated as 2.8, 2.3, 3.7, 6.1, 4.2, 1.0, 2.7, 1.5, respectively, for DPPH, ABTS, reducing power (Fe3+), reducing power (Cu2+), superoxide anion radical-scavenging, hydroxyl radical-scavenging, chelating ability (Fe2+) and chelating ability (Cu2+). Conclusion: Comparing to Trolox, PCA shows much more effective antioxidant activity in vitro in both lipid and aqueous media. Hence, it could therefore be used in pharmacological or food industry as a natural antioxidant. It may exhibit antioxidant activity by both chelating metal transition ions as well as by scavenging free radicals via donating hydrogen atom (H•) or electron (e).Keywords: Protocatechuic acid, antioxidant, reducing power, free radical-scavenging, chelating ability.

Water extract of Acacia seyal bark is used traditionally by the population in Djibouti for its anti-infectious activity. The evaluation of in vitro antibacterial, antioxidant activities and cytotoxicity as well as chemical characterization of Acacia seyal bark water and methanolic extracts were presented. The water extract has a toxicity against the MRC-5 cells at 256 μg/mL while the methanolic extract has a weak toxicity at the same concentration. The methanolic extract has a strong antioxidant activity with half maximal inhibitory concentration (IC50) of 150 ± 2.2 μg/mL using 1-diphenyl-2-picrylhydrazyl (DPPH) and IC50 of 27 ± 1.3 μg/mL using 2,2′-azino-bis 3-ethylbenzthiazoline-6-sulphonic acid (ABTS) radical methods. For ferric reducing/antioxidant power (FRAP) assay, the result is 45.74 ± 5.96 μg Vitamin C Equivalent (VCE)/g of dry weight (DW). The precipitation of tannins from methanol crude extract decreases the MIC from 64 µg/mL to 32 µg/mL against Staphylococcus aureus and Corynebacterium urealyticum. However, the antioxidant activity is higher before tannins precipitation than after (IC50 = 150 µg/mL for methanolic crude extract and 250 µg/mL after tannins precipitation determined by DPPH method). By matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) analysis, the results showed that the condensed tannins consist of two types of catechin and gallocatechin-based oligomers. The fractionation led to the identification of three pure compounds: two flavanols catechin and epicatechin; one triterpene as lupeol; and a mixture of three steroids and one fatty acid: campesterol, stigmasterol, clionasterol, and oleamide.

The total phenolics, flavonoids and in vitro antioxidant activity of petroleum ether, benzene, ethyl acetate, methanol and ethanol extracts of stem of Drypetes sepiaria were determined using various antioxidant model systems viz, DPPH, hydroxyl, superoxide, ABTS and reducing power. Total phenolic content was estimated by Folin-Ciocalteau method. Flavonoids were determined by Aluminium chloride method. The total phenolics and flavonoids contents were found to be 0.81 g 100 g-1 and 1.12g 100 g-1 respectively in the methanol extract. Among the solvent tested, methanol and ethanol extracts of D. sepiaria stem showed potent in vitro antioxidant activities. This investigation explored that D. sepiaria stem is a potential source of natural antioxidant.

Objective: To standardise the protocol for rapid callogenesis in Mussaenda frondosa L. using leaf explants. Qualitative and quantitative phytochemical analysis of leaf, stem and callus cultures.Methods: The leaf explants were inoculated onto MS medium supplemented with varying concentrations of growth regulators such as 2, 4 - D, NAA, BAP, Kn for the induction of callus. Qualitative and quantitative analysis of total phenol, flavonoids and alkaloids contents of leaf, stem and callus were tested by standard methods.  The antioxidant activities were investigated using DPPH radical scavenging method and reducing power assay. The anti - inflammatory activity was evaluated by membrane stabilizing activity.Results: Pale green, healthy, friable and fast growing callus was obtained on the medium enriched with NAA (2mg/l) + Kn (4mg/l). Quantitative determination showed the highest concentration of total phenolics in the methanolic extract of in vitro grown callus (10 ± 1.1 mg of GA/g of extract), flavonoids in methanolic stem extract (137±1.6 mg of Quercitin/g of extract) and alkaloids in methanolic extract of leaf (118.3±1.5 mg/10g of extract). The methanolic leaf extract exhibited highest free radical scavenging activity with IC50 value of 40.6±10.06 μg/ml. The highest membrane stabilizing activity was shown by chloroform extract of the leaf (66.02%).Conclusion: The present preliminary phytochemical and pharmacological analysis may form the basis for drug development in future using callus cultures of M. frondosa.
