The total phenolics, flavonoids and in vitro antioxidant activity of petroleum ether, benzene, ethyl acetate, methanol and ethanol extracts of stem of Drypetes sepiaria were determined using various antioxidant model systems viz, DPPH, hydroxyl, superoxide, ABTS and reducing power. Total phenolic content was estimated by Folin-Ciocalteau method. Flavonoids were determined by Aluminium chloride method. The total phenolics and flavonoids contents were found to be 0.81 g 100 g-1 and 1.12g 100 g-1 respectively in the methanol extract. Among the solvent tested, methanol and ethanol extracts of D. sepiaria stem showed potent in vitro antioxidant activities. This investigation explored that D. sepiaria stem is a potential source of natural antioxidant.

The objective of the current study was to evaluate the antioxidant activity and enzyme inhibitory effect of different parts of medlar including fruit, leaf and flower bud by using various in vitro methods, and also determination of total phenolic and flavonoid content in the samples. Ethanol extracts of medlar parts were prepared and their antioxidant activities were determined using 1,1-diphenyl-2-picryl-hydrazil (DPPH•) scavenging and β-carotene bleaching methods. The leaf extract showed the strongest antioxidant activity. DPPHradical scavenging activity was in the order of BHA > leaf > bud > fruit. This ordering was the same for β-carotene bleaching activity, tocopherol > leaf > bud > fruit. The highest total phenolic (60.3 ± 1.69 mg GAE g-1 extract) and flavonoid (14.77 ± 1.15 mg QE g-1 extract) content were determined in leaf extract. For possible antidiabetic effects of extracts, α-amylase and α-glucosidase inhibitory activities were investigated, the bud extract showed the highest inhibition activities among the all extracts.

Background:Propolis contains compounds with antibacterial and antioxidant capacities, such as phenolic compounds and flavonoids. The aim of this study was to investigate the antibacterial and antioxidant effects of six propolis samples from northern Morocco and to determine their content of polyphenols and flavonoids.Methods:The total polyphenols and flavonoids of Ethanol Extracts (EEP) were evaluated by colorimetric assays. To test the antibacterial activity, the well diffusion and micro-dilution methods were adopted. The kinetic of bacterial growth in the presence of EEP was tested. The antioxidant activity was evaluated using three techniques: reducing power, β-carotene bleaching test, and DPPH scavenging capacity assay.Results:The EEPs were found to have high total phenolic content (65.09 - 122.00 mg GAE/g) and flavonoids (41.95 - 50.44 mg QE/g). The screening of the antibacterial activity showed that all tested bacteria were sensitive to EEP except Escherichia coli. The inhibitory zones varied from 11.50 ± 0.70 to 32.00 ± 0.70 mm, and MIC and MBC ranged from 0.07 to 5 mg/mL and 0.07 to >5 mg/mL, respectively. The antioxidant activity was proportional to the phenolic content of propolis. The IC50 varied from 48.70 to 156.00 mg/mL and the RAA% ranged from 75.07 to 96.68 %.

Objective: The aim of the present study was to evaluate the alpha (α)-amylase and alpha (α)-glucosidase inhibitory activities and in vitro antioxidant activities of the 80 % aqueous ethanol extracts of Tinosporasinensis Lour (Merr.).Methods: The 80% aq. ethanol extract of the plant was prepared. The plant extract was examined for its antioxidant activity by using free radical 1,1-diphenyl-2-picryl hydrazyl (DPPH) scavenging method, ABTS radical scavenging ability, reducing power capacity, estimation of total phenolic content, flavonoid content and flavonol content. Different concentrations (2, 4, 8,10and 15 μg/ml) of the extract was subjected to α-amylase inhibitory and α-glucosidase inhibitory activities and IC50were calculated.Results: The study revealed that the different concentrations of the plant extract possessed a very good amount of total phenolics, flavonoid and flavonol and exhibited potent radical scavenging activity using DPPH and ABTS as a substrate. The ethanol extracts exhibited significant α-amylase and α-glucosidase inhibitory activities with an IC50 value1.093µg and 1.04µg dry extract respectively and well compared with standard acarbose drug.Conclusion: Thus, it could be concluded that due to the presence of antioxidant components the plant extracts could be used for the treatment of hyperglycemia, diabetes and the related condition of oxidative stress. This knowledge will be useful in finding more potent components from the natural resources for the clinical development of antidiabetic therapeutics.

Tea is a popular beverage and shows very strong in vitro antioxidant activity. However, the relationship among in vitro and in vivo antioxidant activities in teas is seldom reported. In this study, in vivo antioxidant and hepatoprotective activities of 32 selected Chinese teas were evaluated on a mouse model with acute alcohol-induced liver injury. The results showed that most teas significantly reduced the levels of alanine transaminase, aspartate transaminase, alkaline phosphatase, triacylglycerol, and total bilirubin in the sera of mice at a dose of 400 mg/kg. In addition, most teas greatly decreased the malondialdehyde level and increased the levels of superoxide dismutase, glutathione peroxidase, and glutathione in the liver of mice, indicating the antioxidant and hepatoprotective activities of teas. Furthermore, the in vivo antioxidant activity of dark tea was stronger than that of green tea, opposite to the results of the in vitro study. Among these 32 teas, Black Fu Brick Tea, Pu-erh Tea, and Qing Brick Tea showed the strongest antioxidant and hepatoprotective activities. Moreover, total phenolic content as well as the contents of epicatechin, gallocatechin gallate, and chlorogenic acid were found to contribute, at least partially, to the antioxidant and hepatoprotective actions of these teas. Overall, teas are good dietary components with antioxidant and hepatoprotective actions.

Objective: The current investigation was contemplated to evaluate the phytochemicals and in vitro antioxidant activities in peel and pulp of some commercially grown citrus fruits of South India, namely, lemon (Citrus aurantifolia), orange (Citrus reticulata), sour orange (Citrus aurantium), pomello (Citrus grandis), and citron (Citrus medica).Methods: The peel and pulp of the fruits were separated and subjected to cold extraction using 70% alcohol. The extracts obtained were screened for the presence of their phytoconstituents using various qualitative and further quantified for major constituents. Further, the in vitro antioxidant activity was assayed by different radical scavenging methods, namely, 2,2-diphenyl-1-picrylhydrazyl, superoxide anion, nitric oxide, lipid peroxidation inhibition, iron chelating activity, and reducing power assay at different concentrations.Results: All the citrus fruits have shown significant in vitro antioxidant activity for the parameters assessed, wherein peel extracts recorded superior antioxidant potential than their corresponding pulps. The broad range of activity of the extracts suggests that multiple mechanisms mediated by the phytoconstituents are responsible for the antioxidant activity.Conclusion: The study thus revealed that peel and pulp of citrus fruits are potential sources of bioactive compounds which are reflected in antioxidant activity and supports their health-promoting claims of plethora of investigations.

Summary Introduction: Chrysopogon aucheri is a species native to the South of Iran. It is used for the treatment of some oxidative-based disorders. Objective: Due to lack of biological research on C. aucheri, we were prompted to investigate the antioxidant activities of leaf total extract and different sub-fractions for the first time. The phenolic and flavonoid contents were also determined in the leaves as the interaction between these components and the antioxidant activity. Methods: The antioxidant effects of total extract and sub-fractions were evaluated by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging method. Total phenolic and flavonoid contents were also determined by Folin-Ciocalteu and aluminium chloride colorimetric methods, respectively. Results: Total extract of C. aucheri leaves was found to possess desirable antioxidant activity in vitro in comparison with standard antioxidant BHT (butylated hydroxytoluene). The highest phenolic and flavonoid contents were observed in the methanol sub-fraction. Results showed weak and moderate correlations with phenolics and flavonoids, respectively, and therefore other metabolites of C. aucheri leaves might be responsible for antioxidant activities. Conclusion: The results suggested that C. aucheri leaves possess desirable antioxidant activity when compared with BHT and support the ethnomedicinal claims of the use of the leaves in the management of some oxidative-based diseases.

Aims: This study was designed to assess antiplasmodial and antioxidant activities in relation to phytochemical contents and toxicological profile of crude extracts of Cola millenii leaves. Place and Duration of Study: The study was carried out from April 2015 to November 2018 at University of Abomey-Calavi, Laboratory of Biochemistry and Bioactives Natural Products and Laboratory of Infectious Vector-borne Diseases. Methodology: Phytochemical screening of aqueous and ethanolic crude extracts was performed using standard methods. Estimation of total phenolics contains (TPC) and total flavonoids contains (TFC) was done using colorimetric methods. Antioxidant activity was assessed in vitro by 2,2-diphenyl-1-picrylhydrazyl (DPPH) Radical-Scavenging, Reducing power, superoxide radical scavenging and Hydrogen peroxide scavenging assay. Antiplasmodial activity was investigated in vitro using both field isolate and laboratory 3D7 strains of P. falciparum using Plasmodium lactate dehydrogenase (pLDH) assay. Hemoglobin release was measured spectrophotometrically to assess hemolytic power. Acute oral toxicity of extracts was evaluated on rats according to OECD 423 guideline. Results: Flavonoids, anthraquinones, coumarins, and triterpenes were recorded in both extracts while tannins were recorded only in ethanolic extract. High TPC and TFC were recorded with ethanolic extract with significate difference (P <0.01) when compared to aqueous extract. Ethanolic extract exhibited the highest parasite growth inhibition against both field isolate (41.62±1.78%) and 3D7 (45.89±1.66%) strains at 100 µg/mL. Both extracts exhibited strong antioxidant activity according to different methods used. Toxicological profile revealed no hemolytic effect on human red blood cells as well as acute toxicity signs in rats. Conclusion: This study demonstrated strong antioxidant and moderate antiplasmodial activities of C. millenii extracts without toxicity effect on rats and human erythrocytes. It would play an important role in malaria and oxidative damage control.

Objective: The objective of the study was to evaluate and compare the antioxidant activity of (EA and E) ethyl acetate and ethanol extracts of (CM) Callicarpa macrophylla. Methods: The physiochemical parameters were assessed according to guidelines given by the world health organization. The total content of phenols and flavonoids was assessed by Folin–Ciocalteu and aluminum chloride methods. In vitro, antioxidant activity was screened by (DPPH) 1, 1-diphenyl-2- picrylhydrazyl and(H2O2) hydrogen peroxide scavenging and reducing power assay. Results: The physicochemical parameters fulfilled the standards of WHO guidelines. Total phenol and flavonoid content were more in ethanol extract as compared to ethyl acetate extract of CM. The antioxidant activity of ethanol extract was further high as compared to ethyl acetate extract of Callicarpa macrophylla. The IC50 of Callicarpa macrophylla ethanol extract was less than the ethyl acetate extract. So, more antioxidant activity of ethanol extract compared to ethyl acetate extract of CM. Conclusion: Overall, both the extracts showed antioxidant activity and can be used further for diseases that can be managed using antioxidants. Ethanol extract possessed significant antioxidant effects than the ethyl acetate extract.

Objectives: The objective of the study is to evaluate in vitro antioxidant activity of aqueous- and freeze-dried fruits extracts of Rhus chinensis and Citrus macroptera and their comparative studies. Methods: The sumac fruit extracts were prepared by macerating in distilled water for 36 h, strained, and concentrated. Hatkora juice was used as it is for further concentration. Both the sample were dried using two methods, that is, evaporation in the water bath at 50°C and freeze-dried in a lyophilizer. The total phenolic content and total flavonoid content were determined using Folin–Ciocalteu and aluminum chloride method. The extracts were evaluated for antioxidant activity by 2, 2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging assay and reducing power assay. Results: Both sumac extract and Hatkora juice show potent antioxidant activity. When compared, the freeze-dried sample shows better activity as compared to the aqueous dried sample in both DPPH and reducing power assay. Conclusions: This study result shows high phenolic and flavonoid content which is responsible for its high antioxidant activity. This suggests that the fruits can be viewed as a potential source of natural antioxidants that can provide valuable functional ingredients useful for the prevention of diseases related to oxidative stress, dietary supplements, functional food, nutraceuticals, etc.

Different ageing technology of wine spirits (WSs) has been investigated, but little has been published on the chemical evolution of aged WS during storage in bottle. The purpose of this study was to examine how 12 months of storage in bottle affected the evolution of antioxidant activity (DPPH, FRAP and ABTS assays), total phenolic index (TPI) and low molecular weight (LMW) compounds content of the WSs aged through alternative technology using three micro-oxygenation levels (MOX) and nitrogen control (N). Results revealed the ability of phenolic compounds from aged WSs to scavenge free radicals during storage in bottle. Among the in vitro antioxidant-activity methods, FRAP assay was the more effective to differentiate WSs according to the ageing technology. Concerning the overall influence of storage in bottle on antioxidant activity, and TPI and LMW compounds content, the higher results were obtained for the MOX modalities (O15, O30 and O60), which showed a similar evolution. In summary, this study provides innovative information, demonstrating that the differences between the aged WSs imparted throughout the ageing process (resulting from different MOX levels) were mostly retained, and only slight modifications during storage in bottle were found.
