Golden (Mesocricetus auratus) and Siberian (Phodopus sungorus) hamsters are widely used as animal models for seasonal reproduction; butM. auratusshows no developmental delay in short days until after sexual maturity, whereasP. sungorusjuveniles delay development in short days. As the photoperiodic response ofPhodopus campbelliis not well established, litters of the twoPhodopusspecies were gestated and reared under long days (14 h light:10 h darkness) or short days (10 h light:14 h darkness) until 70 days of age. As expected, under short photoperiodP. sungorusshowed reduced body, testes, epididymides, uterus, and ovary weight; antral follicles and corpora lutea were absent and vaginae remained closed. Animals moulted to winter pelage, and low concentrations of each of leptin, testosterone, and prolactin were present in male serum.Phodopus campbellijuveniles also responded to the short photoperiod as measured by reduced body, testes, epididymides, and ovary weight. The summer pelage persisted. However, both sexes ofP. campbellideveloped functional reproduction under 10 h light:14 h darkness. All females had a patent vagina by 10 weeks; ovaries contained antral follicles and corpora lutea, and uteri were not reduced in weight. In males, the concentrations of testosterone, leptin, and prolactin were not reduced by short photoperiod. Developmental patterns in the three species of hamster, therefore, differ and are not predicted by relatedness or latitude of origin. Other ecological traits, such as predictability of summer rainfall, ambient temperature, and differential responses to social cues might be important.

OBJECTIVE: In contrast to the elaborate information available on the effects of the photoperiod on the testes of hamsters, little is known about the influence on their ovaries. This study aimed to describe the ovarian follicular development and steroid hormone production in Djungarian hamsters kept from birth under a short daylight regime. DESIGN AND METHODS: Female Djungarian hamsters (Phodopus sungorus) were kept under two different light regimes: (i) 16 h light:8 h darkness (long daylight; LD) and (ii) 4 h light:20 h darkness (short daylight; SD). They were killed at 28, 56 and 80 days after birth; blood and ovaries were collected. Ovaries were either fixed in Bouin's solution or frozen. Fixed material was dehydrated, embedded in paraffin, serially sectioned at 5 microm and stained with haematoxylin and eosin, whereafter all healthy and atretic follicles were classified and counted. 3beta-Hydroxysteroid dehydrogenase (3beta-HSD) was histochemically demonstrated in 10 microm sections of frozen ovaries. Serum oestradiol-17beta and progesterone levels were determined by RIA. RESULTS: The numbers of healthy preantral and antral follicles were higher in LD than in SD hamsters. Antral follicles did not significantly differ in number during development in LD hamsters, but they were completely absent from 80-day-old SD animals. In LD animals the number of apoptotic preantral follicles dramatically increased with age. In SD animals the numbers of apoptotic antral follicles strongly decreased with age, whereas numerous non-apoptotic follicles with luteinized granulosa cells and a degenerated oocyte appeared, and in increasing numbers with age. During development, moderate 3beta-HSD activity was present in interstitial cells, theca cells of healthy follicles, and in both theca and granulosa cells of degenerating follicles. Strong enzyme activity was found in the hypertrophied granulosa cells of luteinized atretic follicles. Mean serum progesterone values varied from 2 to 6 nmol/l and were not different in LD and SD hamsters. Mean serum oestradiol levels varied from 132 to 542 and 325 to 2353 pmol/l in LD and SD hamsters respectively. The highest oestradiol levels were found in SD animals at day 28 of development. CONCLUSIONS: Folliculogenesis was dramatically disturbed in Djungarian hamsters raised under a short photoperiod. These animals developed high serum oestradiol levels and numerous luteinized atretic follicles with highly steroidogenic granulosa cells, which appear to be the source of the increased serum oestradiol levels.

ABSTRACT In mink, termination of the delayed implantation period, following reactivation of the corpora lutea, and onset of the spring moult are associated with a rise in prolactin secretion triggered by increasing daylength, while decreasing daylength induces the autumn moult. To establish whether suppression of the function of the pineal rendered the mink unresponsive to daylength changes, the superior cervical ganglion was removed bilaterally 2–4 weeks before mating. Intact and operated females were then left outdoors or were put under a lighting regime of either 15 h light: 9 h darkness (15L: 9D) or 8L: 16D. In July, at the end of the spring moult, the 15L: 9D lighting regime was changed to one of 8L: 16D. Under artificial photoperiods ganglionectomy suppressed the stimulatory role of long days and the inhibitory role of short days on prolactin secretion, and consequently on progesterone secretion and spring moult. Neither was the autumn moult, induced early in intact females by the change to a short photoperiod, advanced in ganglionectomized females, showing that the latter were unresponsive to the artificial modification of the photoperiod. However, in animals kept outdoors, prolactin and progesterone secretion and spring moult were not changed by ganglionectomy. Increase in body weight and autumn moult were only slightly delayed by the operation suggesting that other environmental factors had replaced the synchronizing effect of the daylength changes. Alternatively the desynchronization between intact females responsive to photoperiodism and those rendered unresponsive may be too slow to be observed soon after ganglionectomy. J. Endocr. SUMMARYThe vernalization and photoperiodic response of six locally adapted bread wheat cultivars grown under natural daylength conditions during the summer or winter months was examined in glasshouse experiments. The wheat was vernalized by chilling imbibed grains at 2 ± 1°C for 0, 15 or 45 days. Vernalization for 45 days followed by long summer days led to floral initiation in all cultivars within 28 days but vernalization for 0 or 15 days only led to floral initiation in one cultivar. Vernalization followed by long days reduced the time from transplanting to anthesis, resulting in early ear emergence. Vernalization followed by short days accelerated the development of all the cultivars, but normal development could also occur without vernalization at this time of year. Apical differentiation of the primary shoot and its length and development gave the most reliable information on the period of vernalization required.

Most of the previous studies on ovarian hyaluronan (HA) have focused on mature antral follicles or corpora lutea, but scarcely on small preantral follicles. Moreover, the origin of follicular HA is unknown. To clarify the localization of HA and its synthases in small growing follicles, involvement of HA in follicle growth, and gonadotropin regulation of HA synthase (Has) gene expression, in this study, perinatal, immature, and adult ovaries of Wistar-Imamichi rats were examined histologically and biochemically and byin vitrofollicle culture. HA was detected in the extracellular matrix of granulosa and theca cell layers of primary follicles and more advanced follicles. Ovarian HA accumulation ontogenetically started in the sex cords of perinatal rats, and its primary site shifted to the intrafollicular region of primary follicles within 5 days of birth. TheHas1–3mRNAs were expressed in the ovaries of perinatal, prepubertal, and adult rats, and the expression levels ofHas1andHas2genes were modulated during the estrous cycle in adult rats and following administration of exogenous gonadotropins in immature acyclic rats. TheHas1andHas2mRNAs were predominantly localized in the theca and granulosa cell layers of growing follicles respectively. Treatments with chemicals known to reduce ovarian HA synthesis induced follicular atresia. More directly, the addition ofStreptomyceshyaluronidase, which specifically degrades HA, induced the arrest of follicle growth in anin vitroculture system. These results indicate that gonadotropin-regulated HA synthesis is involved in normal follicle growth.

Abstract A cooperative study evaluated effects of supplemental dietary OmniGen AF (OG; Phibro Animal Health Corporation) on reproductive characteristics of gilts. Crossbred gilts (n = 56; initial body weight = 152.3 ± 9.7 kg) were assigned to treatments in a 2 x 2 factorial design [diet (OG or control) and PG600 (Merck Animal Health, El Dora, KS) injection (Yes or No)]. Dietary treatments were: 1) corn-SBM basal diet (control) that met NRC (2012) requirement estimates, or 2) control plus 0.75% OG and were fed 35 to 40 d prior to breeding. A common diet formulation was used by both stations. Gilts were estrous-synchronized by 14-d feeding of Matrix (Merck Animal Health). PG600 injections occurred 24 hours after Matrix withdrawal. Gilts were weighed at breeding and preslaughter (at d 39 to 48 postbreeding) and were euthanized to evaluate reproductive characteristics. There were no OG x PG600 interactions on any response measures (P > 0.15). Mean responses did not differ (P > 0.20) when gilts were fed control or OG diets, respectively, for total uterus weight (6,065 vs. 5,883 g), uterine horn length (129.2 vs. 123.9 cm), ovary weight (9.66 vs. 9.94 g), total corpora lutea (CL) number (25.0 vs. 25.3), individual CL weight (0.41 vs. 0.43 g), total follicular fluid weight (3.17 vs. 3.03 g), individual placenta weight (86.3 vs. 86.4 g), total fetuses (14.2 vs. 13.0), and fetus crown-rump length (7.97 vs. 7.66 cm). Supplementation with OG reduced (P = 0.04) individual placenta fluid volume of gilts (150.6 vs. 115.4 mL). PG600 injection increased (P < 0.001) ovary weight (9.03 vs. 10.57 g), total CL (18.09 vs 32.27) and reduced CL weight (0.46 vs. 0.38 g). The results demonstrated classic PG600 responses on reproductive characteristics but no effects of 0.75% OG supplementation nor an interaction between the two factors.

Abstract Many Arachis species collections do not produce pegs in North Carolina even though they flower profusely. To investigate reasons for the failure of fruiting, nine wild peanut species of section Arachis and three A. hypogaea cultivars representing spanish, valencia and virginia types were evaluated for response to short and long-day treatments in the North Carolina State Phytotron Unit of the Southeastern Environmental Laboratories. The objective of this investigation was to determine the flowering and fruiting responses of Arachis species to short and long-day photoperiods. Plant collections grown under a 9-hour short-day treatment were generally less vigorous, but produced more pegs than corresponding plants grown in long-day treatments which were produced by 9 hours of light plus a 3-hour interruption of the dark period. Annual species produced significantly more flowers and pegs than perennial species during both long and short days. The total number of flowers produced ranged from 0 during short days for A. correntina to more than 300 for A. cardenasii in long-day treatments. Only one plant of each species A. chacoense and A. villosa, and no plants of A. correntina, flowered in short days. Total numbers of pegs produced in short-day treatments were generally greater than in long-day treatments and the ratio of total number of pegs/total number of flowers was consistently greater during short-day treatments. A general trend was observed for more flowers produced in long-day treatments, but more pegs produced in short days. This study indicated that photoperiod can be manipulated to increase the seed set of some species and the success rate of obtaining certain interspecific hybrids. Furthermore, introgression from wild to cultivated species may possibly alter the reproductive capacity of A. hypogaea to photoperiod.

Reproductive tracts were collected from 532 female caribou (Rangifer tarandus groenlandicus) in northern Canada over a [Formula: see text]-year period. Ovaries were weighed and scrutinized by gross and histological techniques. Ovaries of nulliparous and nongravid, parous cows responded to follicular fluctuations by increasing in weight in summer and declining in winter. Weights of ovaries from calves, yearlings, and 2 year olds overlapped extensively. Formation of corpora lutea of pregnancy more than doubled ovary weight. During gestation ovary weight declined and then partially recovered; it increased with age in pregnant cows. The number and size of follicles [Formula: see text] diameter increased with age until puberty, reaching greatest development just before the autumn rut. After puberty the seasonal incidence of such follicles remained stable except during gestation, when it declined. Corpora lutea of pregnancy regressed to form apparently permanent scars composed of vascular and connective tissue remnants. Secondary corpora lutea developed in 35% of the cows at or before conception and upon regression produced scars which were not permanent. Regressing corpora lutea of estrus also disappeared into the ovarian stroma, apparently within 1 year. In individual cows one ovary, selected at random, dominated in the production of ova and corpora lutea.

Homozygous androgen receptor (AR)-knockout (ARKO) female mice are subfertile due to both intra- and extraovarian (neuroendocrine) defects as defined by ovary transplantation. Using ARKO mice, this study set out to reveal the precise AR-regulated pathways required for optimal androgen-regulated ovulation and fertility. ARKO females exhibit deficient neuroendocrine negative feedback, with a reduced serum luteinizing hormone (LH) response to ovariectomy (OVX) ( P < 0.01). Positive feedback is also altered as intact ARKO females, at late proestrus, exhibit an often mistimed endogenous ovulatory LH surge. Furthermore, at late proestrus, intact ARKO females display diminished preovulatory serum estradiol (E2; P < 0.01) and LH ( P < 0.05) surge levels and reduced Kiss1 mRNA expression in the anteroventral periventricular nucleus ( P < 0.01) compared with controls. However, this reduced ovulatory LH response in intact ARKO females can be rescued by OVX and E2 priming or treatment with endogenous GnRH. These findings reveal that AR regulates the negative feedback response to E2, E2-positive feedback is compromised in ARKO mice, and AR-regulated negative and positive steroidal feedback pathways impact on intrahypothalamic control of the kisspeptin/GnRH/LH cascade. In addition, intraovarian AR-regulated pathways controlling antral to preovulatory follicle dynamics are disrupted because adult ARKO ovaries collected at proestrus have small antral follicles with reduced oocyte/follicle diameter ratios ( P < 0.01) and increased proportions of unhealthy large antral follicles ( P < 0.05) compared with controls. As a consequence of aberrant follicular growth patterns, proestrus ARKO ovaries also exhibit fewer preovulatory follicle ( P < 0.05) and corpora lutea numbers ( P < 0.01). However, embryo development to the blastocyst stage is unchanged in ARKO females, and hence, the subfertility is a consequence of reduced ovulations and not altered embryo quality. These findings reveal that the AR has a functional role in neuroendocrine regulation and timing of the ovulatory LH surge as well as antral/preovulatory follicle development.
