Adult male Wistar rats were enrolled in a study to test the acute hepatic effects of 50 mg/kg fumonisin B1 in feed for 5 days. Fumonisin B1 depressed growth and feed intake, and absolute and relative liver weight showed a significant increase. The proportions of C17:0, C18:3 n3, C22:5 n3 and C22:6 n3 fatty acids decreased in the hepatic phospholipid fraction. All proportional decreases modified the hepatocellular membrane lipids into a more rigid state. The fatty acid profile modifications were partly compensated for by endogenous glutathione (preventing the formation of conjugated dienes and trienes as initial phase lipid peroxidation indicators), while the enzymatic antioxidant defence system (glutathione peroxidase) was unaltered. In contrast, hepatic malondialdehyde, the cytotoxic product of end-phase lipid peroxidation showed a concentration increase even after 5 days of feeding. The results indicate a rather strong and rapid hepatic effect of FB1, immediately impairing membrane phospholipids, even before the enzymatic antioxidant defence is activated.

Background: The aim of this study is to evaluate the potential effect of vitamin C as an antioxidant on liver regeneration after partial hepatectomy and acetaminophen-induced liver injury in Wistar rats.Methods: A total of 24 male Wistar rats were divided into four groups, each group consisted of 6 rats: group A (control, partial hepatectomy/PHx alone), group B (PHx and vitamin C 250 mg/kg BW), group C (acetaminophen 500 mg/kg BW and PHx), and group D (acetaminophen 500 mg/kg BW with PHx and vitamin C 250 mg/kg BW). Subtoxic dose of acetaminophen was given 24 hours before partial hepatectomy. Vitamin C was given orally via oral gavage for 6 consecutive days after partial hepatectomy. POD 7, all animals were terminated and performed laparotomy to obtain liver tissue for measurement of liver weight and regeneration rate, blood samples for malondialdehyde (MDA) as a lipid peroxidation measurement and histopathological investigation.Results: The means of regeneration rate in vitamin C groups were significantly higher compared to non-vitamin C group (p<0.05). Similar result, the means of MDA values in vitamin C groups were significantly lower compared to non-vitamin C group (p<0.05). This result suggests a protective effect of vitamin C against lipid peroxidation. Histopathological changes in liver cells were statistically difference between vitamin C groups and non-vitamin C groups (p<0.05).Conclusions: Our results indicate that vitamin C administration promotes liver regeneration and inhibits lipid peroxidation after partial hepatectomy and acetaminophen-induced liver injury in Wistar rats.

The present study was undertaken to assess the beneficial effects of a daily consumption of 8 % freeze-dried red-pigmented leafy lettuce (Lactuca sativa) on CVD. C57BL/6J mice were fed a high-fat high-cholesterol diet supplemented with or without red-pigmented leafy lettuce for 4 weeks. The present results showed that the red-pigmented leafy lettuce-supplemented diet significantly decreased the level of total and LDL-cholesterol and TAG in the plasma of the mice. The atherosclerotic index was calculated to be 46 % lower in the mice fed with the lettuce diet compared with the control diet. Lipid peroxidation measured by 2-thiobarbituric acid-reactive substances was markedly reduced in the plasma, liver, heart and kidney of the mice fed the lettuce diet. The content of antioxidants (total glutathione and β-carotene) was significantly increased by lettuce supplementation. The antioxidant defence system by antioxidant enzymes including glutathione S-transferase, glutathione peroxidase, glutathione reductase, superoxide dismutase and paraoxanase in blood or liver tissues was also increased, and showed the improved oxidative stress in the mice fed the lettuce diet. The measurement of tail DNA (%), tail extent moment and olive tail moment indicated that the lettuce diet increased the resistance of hepatocyte and lymphocyte DNA to oxidative damage. The present study showed that the supplementation of a high-cholesterol high-fat diet with 8 % red-pigmented leafy lettuce resulted in an improvement of plasma cholesterol and lipid levels, prevention of lipid peroxidation and an increase of the antioxidant defence system and, therefore, could contribute to reduce the risk factors of CVD.

Abstract Anthropogenic activities, such as agriculture and industrial activities, are a main source of pollution contributing for the degradation of water quality and thus affecting the living organisms of aquatic systems. Copper is widely used at these practices being often released into the aquatic systems and may cause negative effects in its communities. This study proposes to determine the effects of copper in the antioxidant defence system of two size classes (big and small sizes) of Scrobicularia plana and Cerastoderma edule, two marine bivalve species with commercial interest. It was observed the behaviour activity of the organisms during the exposure to copper sulphate (CS) and was determined the enzymatic activities of glutathione-S-transferases (GST), glutathione reductase (GR) and glutathione peroxidase (GPx) (both selenium-dependent (SeGPx) and total (tGPx)) in the muscle tissue (foot). Lipid peroxidation (LPO) was evaluated through thiobarbituric acid reactive substances (TBARS) measurement in the foot. Changes in the behaviour and enzymatic activity were observed. Lipid peroxidation was observed at C. edule and S. plana big and small size classes, respectively, according to TBARS levels. The foot showed to be a good tissue to be used in biochemical analysis to detect the presence of toxicants.

The aim of the current study was to asses the effect of goat or cow milk-based diets, either normal or Fe-overloaded and folic acid supplement on some aspects of hepatic physiology, enzymatic antioxidant defence and lipid peroxidation in liver, brain and erythrocyte of control and anaemic rats after chronic Fe repletion. 160 male Wistar rats were placed on 40 d in two groups, a control group receiving normal-Fe diet and the Fe-deficient group receiving low Fe diet. Lately, the rats were fed with goat and cow milk-based diets during 30 d, with normal-Fe content or Fe-overload and either with normal folic or folic acid supplemented. Fe-overload increased plasma alanine transaminase and aspartate transaminase levels when cow milk was supplied. Dietary folate supplementation reduced plasma transaminases levels in animals fed goat milk with chronic Fe overload. A remarkable increase in the superoxide dismutase activity was observed in the animals fed cow milk. Dietary folate supplement lead to a decrease on the activity of this enzyme in all the tissues studied with both milk-based diets. A concomitant increment in catalase was also observed. The increase in lipid peroxidation products levels in rats fed cow milk with Fe-overload, suggest an imbalance in the functioning of the enzymatic antioxidant defence. In conclusion, dietary folate-supplemented goat milk reduces both plasma transaminases levels, suggesting a hepatoprotective effect and has beneficial effects in situation of Fe-overload, improving the antioxidant enzymes activities and reducing lipid peroxidation.

The paper analyzes the impact of chemotherapy in patients with chronic lymphocytic leukemia in the functional state of the liver.The formation of differentiated types of drug- induced liver damage, depending on the characteristics of cytostatic therapy has been established. The intensity of lipid peroxidation and antioxidant defence stress in patients with drug- induced liver damage on a background of chemotherapy and the presence of correlation relationships between the intensity of cytolysis and severity of lipid peroxidation have been proved.

Selenium, as part of selenocysteine, is the active centre of selenoenzymes. Excess amount of selenium generates oxygen free radicals and reacts with thiol compounds such as reduced glutathione, composing seleno-diglutathione. These reactions impair the amount and/or activity of the biological antioxidant defence system. In the present experiment the effects of two inorganic selenium sources (Na2SeO3, Na2SeO4) on lipid peroxidation and on the content and activity of some components of the antioxidant defence mechanism were studied in Ross 308 hybrid cockerels. In the tissues examined, the glutathione redox system was altered in different ways as an effect of excess selenium. The amount of glutathione and, consequently, glutathione peroxidase activity declined in the blood plasma and in the red blood cell haemolysate, while in the liver malondialdehyde concentration increased only at the end of the experimental period as a result of the boosted enzyme activity. The authors suppose that the cause of increased lipid peroxidation was the potential toxic effect of selenium accumulation above the actual demand.
