Introduction: A successful in vitro propagation system was developed for Sandra and Rocky cultivars of tomato plants (Lycopersicon esculentum Mill.) commonly grown in Kurdistan Region of Iraq by in vitro culture of shoot tips and node explants. Methods: Shoot tip and node explants were excised and cultured on basal MS medium containing several concentrations of BAP and Zeatin. Results: Multiple shoots formation of up to 2 shoots were obtained on MS medium supplemented with 2.0 mgl-1 Zeatin in node and shoot explants for the two cultivars. Microshoots were tested for root initiation on full MS medium supplemented with different concentrations of IBA, NAA and IAA. The best root formation was observed on a medium containing 1.0 mgl-1 IAA in Sandra cultivar and 0.5 mgl-1 of NAA in rocky cultivar. Conclusion: The regenerated plants were successfully acclimatized and transplanted to the open field conditions.

Agrobacterium-mediated genetic transformation system has been developed for two tomato (Lycopersicon esculentum Mill.) varieties, namely Pusa Ruby (PR) and BARI Tomato-3 (BT-3). Prior to the establishment of transformation protocol cotyledonary leaf explants from the two varieties were cultured to obtain genotype independent in vitro regeneration. Healthy multiple shoot regeneration was obtained from the cut ends of cotyledonary leaf segments for both the varieties on MS containing 1.0 mg/l BAP and 0.1 mg/l IAA. The maximum root induction from the regenerated shoots was achieved on half the strength of MS medium supplemented with 0.2 mg/l IAA. The in vitro grown plantlets were successfully transplanted into soil where they flowered and produced fruits identical to those developed by control plants. Transformation ability of cotyledonary leaf explants was tested with Agrobacterium tumefaciens strain LBA4404 harboring binary plasmid pBI121, containing GUS and npt II genes. Transformed cotyledonary leaf explants were found to produce multiple shoots on MS containing 1.0 mg/l BAP and 0.1 mg/l IAA. Selection of the transformed shoots was carried out by gradually increasing the concentration of kanamycin to 200 mg/l since kanamycin resistant gene was used for transformation experiments. Shoots that survived under selection pressure were subjected to rooting. Transformed rooted plantlets were transferred to soil. A rapid and efficient protocol is outlined for in vitro propagation of muskmelon(Cucumis melo L.) Shoot tips, nodal segments and cotyledonary nodes from invitro grown seedlings were used as explants. The explants were inoculated on MS medium fortified with different combinations and concentrations of growthregulators viz., BAP, NAA, GA3 and IBA for multiple shoot regeneration.Effective result was found on MS medium supplemented with 2.0 mg/l BAP, inwhich 90% and 70% cultures induced multiple shoots from nodal segments andshoot tip explants, respectively. Whereas, 70% cultures of cotyledonary nodeswere found to induced shoots on MS medium with 1.5 mg/l BAP + 0.1 mg/l GA3. In vitro regenerated shoots were subcultured on half strength MS mediumsupplemented with different concentrations of IBA and NAA for successful rootinduction and the effective result (up to 70%) was found in medium with 1 mg/lIBA. Well rooted in vitro grown plantlets were acclimatized in sandy soil, whereas 70% plantlets survived

This experiment was conducted in faculty of Science labs, Kufa University, carried out during 2015 to applied methods for extraction, purification and Quantitative of Lycopene red pigments, from callus tissue and tomato fruits mother plant (Lycopersicon esculentum Mill).This study include of three parts, Firstly; Tomato seeds(Supper queen) hybrid were germinated in free MS medium and callus induction from shoot tip (3cmpieces) by using MS medium supplemented with Dichlorophenoxiactic acid (2,4-D) at different concentration (0.5,1, 1.5mg/l)with benzyl adenine (BA) at concentration of (0.3 mg/l). Secondly; identically callus fresh weight re-cultured in the same MS medium supplemented with high molecular weight polyethylene glycol (PEG) was used as selective agent at level of (5,10,15 and 25%). Thirdly; comparisons study were made between in vitro and in vivo grown plant. Powder of control lycopene used as standard solution. The content of lycopene was done by using high performance liquid chromatography (HPLC), and compare of the quantitatively of lycopene with these content in fruits of mother plant, and callus tissue. Also, include alcohol extraction of Lycopene from tomato fruit by using acetone and hexane mixture. The result showed significant increased (P< 0.05) of lycopene production and the superiority of lycopene content in callus than the content in fruits of mother plant. Antioxidant enzymes activity like Catalase (CAT),Guaiacol peroxidise (POX) and Superoxide dismutase(SOD) were high in callus under drought stress than in fruit of mother plant. However, Proline and total sugar content were at higher levels in callus under drought stress than in fruit of mother plant.

In vitro rooting of micropropagated shoots were carried out with the aim of evaluating the root induction responses of two tomatoes (Lycopersicon esculentum MILL) varieties (Gelilema and Chali) using Indole -3- butyric acid (IBA). Seven levels of IBA (0.0, 0.25, 0.5, 0.75, 1.0, 1.25 and 1.5 mg L-1) were used in a completely randomized design (CRD) in factorial combinations (seven level of IBA*two varieties) with three replications.  After the plantlets were kept in the rooting media for three weeks, data on rooting percentage, number of roots/shoots and root length in cm were collected. The analysis of variance showed that the interaction of IBA*Var was highly significantly different for rooting percentage, a number of roots/shoot and root length at p<0.01. The highest rooting percentage (100.00±0.00), number of roots/shoot (14.20±0.35) and root length (10.7±0.29) were received from Chali on free Murashige and Skoog medium (MS). At the same time, the lowest percentage of rooting (11.11±0.00), number of roots/shoot (0.887±0.19) and root length (1.00±0.00 cm) were obtained from Gelilema on MS+1.5mg/l IBA. For acclimatization, the in vitro rooted shoots were transplanted into plastic pots containing a mixture of oven sterilized soil and sand at a ratio of 2:1. After three weeks, a survival rate of 67.7% for Chali and 58.1% for Gelilema was obtained. From the above result, it can be concluded that free MS medium was the best for in vitro rooting of the two tomato varieties. The optimized protocol will be useful for rapid in vitro multiplication of the two tomato varieties.

Axenic shoot culture of Mentha spicata L. was established from young shoots of the plant naturally growing in the field. Large number of multiple shoots were observed on MS media supplemented with NAA (0.5ppm) and BAP (1ppm) within eight weeks of culture. Extensive root formation was observed on MS medium supplemented with 1 ppm NAA. Essential oils from both in vitro and in vivo samples showed l- carvone as a major constituent, however, the in vitro samples showed higher concentration of menthol than in vivo samples. <i>Nepal Journal of Science and Technology</i> Vol. 7, 2006

Plant regeneration of Anubias barteri var. Nana was achieved through organogenesis in shoot tip cultures. Multiple shoots were induced from cultured shoot tips on a modified MS (Murashige and Skoog, 1962) medium supplemented with BA and kinetin. The maximum green shoot numbers were best obtained on MS medium containing 3 mg/L BA with 5 shoots. Rooting in all regenerated shoots was promoted on MS medium devoid of plant growth regulators or kinetin singly. Acclimatization and survival when transferred to field conditions were shown to be 100% in the regenerated plants. Cytological and flow cytometric analyses of the mother plants and in vitro grown plants derived from 5 years old cultures showed no differences in ploidy level, they were all diploid (2n = 2x = 48) with a 2C peak indicating that ploidy alteration did not occur.

An efficient and rapid protocol for the micropropagation of Globba schomburgkii Hook. f. via bulbil explants was investigated. The long divided and undivided bubils of G. schomburgkii Hook. f. were cultured on MS medium (Murashige and Skoog) that had either 3 mg/l benzyladenine (BA) or 0.5 mg/l naphthaleneacetic acid (NAA) added for 8 weeks. The results indicated that the long divided bulbils of G. schomburgkii Hook. f. showed a greater amount of plant regeneration than the undivided bulbils. Callus induction, as well as shoot and root formation, were observed when culturing microshoots of 1 cm in length on media (MS) that had Thidiazuron (TDZ) or NAA plus BA added at a range of concentrations for 8 weeks. The highest percentage of callus induction was 40 % when culturing the microshoots on MS medium supplemented with NAA and BA. The best result for shoot formation was achieved when culturing the microshoots on MS medium with TDZ added. The highest number of roots was obtained when culturing the microshoots on MS medium with NAA and BA added. The in vitro-derived plantlets of G. schomburgkii Hook. f. were transplanted to pots containing different types of potting mixture in a greenhouse. The survival rates were 80 % when G. schomburgkii Hook. f. was transplanted to sand.

An efficient protocol for in vitro propagation of Adhatoda vasica Nees was established using shoot tip and nodal explants from field grown mature plant. Proliferation of multiple shoots was achieved on MS medium supplemented with different concentrations and combinations of cytokinins (0.5-4.0 mg/l) and auxins (0.1-1.0 mg/l). Maximum number of shoots per explant (13.0) was obtained on MS medium supplemented with 2.0 mg/l BAP + 0.2 mg/l NAA. Among two types of explants used in this study, nodal explants showed better response in respect of multiple shoot production. The elongated shoots were excised and subcultured for rooting on MS medium supplemented with different concentrations of auxins (IBA and NAA). Highest 80% rooting was achieved; and three to four roots per shoot were recorded in medium with 1.0 mg/l IBA within 4 weeks of culture. The in vitro raised plantlets were acclimatized and successfully transferred to natural condition in pot. The regenerated plants were healthy, uniform and identical to the donor plants and the survival percentage was 80%. Seeds of four tomato hybrids Ginan, Nirtia, Speedy and Rola were cultured on aseptic germination medium after surface sterilization with NaOCl. Shoot tips 1cm length were dissected and grown on MS medium supplemented with Benzyl adenine (BA) and Indole acetic acid (IAA) for multiplication. Callus was induced on hypocotyls cultured on MS medium supplemented with BA under light intensity of 1000 lux and 25 0C. Results showed that NaOCl at 4% completely disinfected seeds and resulted in pathogen free seedlings. Results also showed that a concentration of 4 mg/l of BA was superior in increasing callus fresh and dry weights. Shoot multiplication was achieved on the same medium supplemented with 2 and 0.8 mg/l of BA and IAA respectively.

Calluses were induced from four tomato hybrid (Davista, superDavista, Muntcarloo,Hymar). On MS medium supplemented with 2, 4-D Auxin at concentration of 4 mg/L.The Callus were divided into two groups the first one was affected with gammaradiation at the doses (0,2,4,6,8)Gray and re cultured on the same mentioned medium.The second group was also affected with gamma radiation with the same dosesmentioned above and re- cultured on MS media supplemented with 0.5 mg/L Kinetinfor regeneration. Fresh and dry weight was measured after 8 weeks. The number andlength of shoots were measured as indicators after 6 weeks. Results revealed that thedose 2 Gray was superior to give highest rate of fresh and dry weight reaching 238.5and 43.1 mg respectively and the hybrid super Davista was superior in giving thehighest fresh and dry weight reaching 231.6 and 30.2 mg respectively. Moreover theresults revealed that control treatment was superior in giving the highest rate innumber and length of plantlets. The 2 Gray treatment showed a significant effect onthe rate and length of plantlets. In addition, there were significant differences amongthe cultivars in most characters mentioned above.
