The objective of this study was to verify the genetic diversity between and within seven populations of Moxotó goat (n = 264) from the States of Pernambuco, Paraíba and Rio Grande do Norte, using RAPD (Random Amplified Polymorphic DNA). Moxotó, as well as other naturalized breeds, suffers genetic losses due to the indiscriminate miscegenation with breeds raised in the Northeast Region of Brazil. The genetic characterization of these genetic resources is essential to conservation and breeding programs. DNA was extracted from lymphocytes using a non-organic protocol. The 16 primers used were selected from 120 decamer oligonucleotide primers and generated 56 polymorphic bands. The analysis of molecular variance (AMOVA) showed that the greater part of total genetic variability (71.55%) was due to differences between individuals within populations, while 21.21% was among populations. The analysis of variance among the pairs of populations demonstrated that the populations located in Floresta, PE x Angicos, RN presented a smaller value of intrapopulational differentiation (8.9%), indicating low genetic variability among them. Nei's genetic distances varied between 0.0546 and 0.1868 in the populations. The dendrogram generated showed that the Canindé breed, used as outgroup, clustered with the populations of Moxotó, indicating a possible common origin of the naturalized goat breeds.

Random amplified polymorphic DNA (RAPD) markers were used to study genetic diversity in dog samples belonging to populations of German Shepherd and Japanese Spitz. A total of twelve samples were typed using eight RAPD primers. Out of eight primers, three primers gave result in six individuals of dogs. The phylogenetic tree constructed by the neighbor joining method based on Nei. Original measures revealed highest genetic identity found in German Shepherd as 0.9444 and highest genetic distance as 1.2809. The analysis predicts the number of polymorphic loci as 15 and the percentage of polymorphic loci as 83.3. Genetic analysis with RAPD markers has been extensively used to determine diversity among maize genotypes. The aim of the present study was to estimate genetic relationships among 70 genotypes, provided from SCDA Turda Cluj germplasm collection. RAPD analysis was performed with 14 decamer primers. These primers generated, among the studied genotypes, a number of polymorphic bands comprised between 13 bands (OPA 04) and 7 bands (OPAL 20). The highest numbers of polymorphic bands were obtained with primer OPA 04, respectively 13 bands, following by OPO 12 (12 polymorphic bands), OPAB 11 and OPA 17 (11 polymorphic bands). Lowest number was obtained with primer OPAL 20, respectively 7 polymorphic bands. Genetic distances were established using Nei-Li coefficient and UPGMA dendrogram was constructed with RAPDistance 1.04 software. The built dendrogram shows phylogenetic relationships between genotypes analyzed.

This study aimed to characterize the genetic variability in commercial accessions of passion fruit from the Federal District, Brazil, by RAPD markers. Genetic analyses were done with leaf samples of 30 accessions. DNA samples were amplified by RAPD technique, and respective markers converted into a binary matrix, from which the genetic distances between the accessions were estimated. Clustering analyis based on genetic distances allowed to detect a wide range of genetic variabillity among the accessions of sour passion fruit, and to separate them from the two sweet passion fruit. The graphical positioning of 'BRS Ouro Vermelho' confirms its potential to improve the genetic variability of commercial varieties of sour passion fruit. Dispersal of genetic distances among commercial accessions of sour passion fruit supports evidence for different genetic origins of the materials planted in the Federal District. The verified genetic variability indicates the potential success of future breeding programs for this region.

Our analysis of the genetic variability and phylogenetic relationships among the isolates of mitosporic entomopathogenic fungi Paecilomyces fumosoroseus (Wize) Brown & Smith (10 isolates), Paecilomyces farinosus (Holm ex S.F. Gray) Brown & Smith (3 isolates), Paecilomyces lilacinus (Thom) Samson (1 isolate), and Verticillium lecanii (Zimmerman) Viegas (6 isolates) was based on the use of random amplified polymorphic DNA (RAPD) markers. Neighbor-joining analysis characterized P. fumosoroseus and V. lecanii as monophyletic taxa. The analysis also supported monophyly of the genus Paecilomyces. We have used the distribution of genetic distances to demonstrate the genetic variability within studied species, in Paecilomyces spp. (Hyphomycetes) and Aschersonia spp. (Coelomycetes) in particular. We conclude that the RAPD-based analysis is useful for the reconstruction of intraspecific and interspecific relationships of the entomopathogenic fungi. Moreover, it can be employed to study the intergeneric relationships, especially in the case of closely related genera of the hyphomycetous entomopathogenic fungi. We propose that RAPD-based phylogeny and genetic variability reflect their life strategy. In our opinion, the mobility of spores represents a factor with major influence on the evolution of these entomopathogenic fungi.Key words: Paecilomyces, Verticillium, Aschersonia, RAPD, phylogeny, genetic variability.

The objective of this study was to examine the molecular and phenotypic variability of 'Prata Anã' banana clones cultivated in northern Minas Gerais State. For the phenotypic and molecular characterization, the clones were collected from five properties. The morphological characterization of 20 clones was accomplished through a qualitative evaluation of 74 descriptors. For the molecular evaluations, 45 clones were collected, and 14 Random Amplified Polymorphic DNA primers were used. Among the 74 morphological descriptors evaluated in the 20 'Prata Anã' banana tree clones, 53 descriptors presented in the same homogeneous class for all clones. The presence of anthocyanin in the pseudostem (ANT) and the shape (BUS) and flexion (FLX) of the bunch clearly differentiated the 'Prata Anã' banana clones from the commercial cultivar. A total of 176 bands were produced from the molecular analysis using the 14 RAPD primers; there were 116 monomorphic and 60 polymorphic bands, with an average polymorphism percentage of 52.2%. The amplification using the OPP 14 primer resulted in the greatest percentage of polymorphic bands at 88%. The dendrogram indicated that each of the 45 'Prata Anã' clones that were evaluated using RAPD markers clustered with the commercial 'Prata Anã' cultivar.

Identified germplasm is an important component for efficient and effective management of plant genetic resources. Traditionally, cultivars or species identification has relied on morphological characters like growth habit or floral morphology like flower colour and other characteristics of the plant. Studies were undertaken for identification and analysis of genetic variation within 34 rose cultivars through random amplified polymorphic DNA (RAPD) markers. Analysis was made by using twenty five decamer primers. Out of twenty five, ten primers were selected and used for identification and analysis of genetic relationships among 34 rose cultivars. A total of 162 distinct DNA fragments ranging from 0.1 to 3.4 kb was amplified by using 10 selected random decamer primers. The genetic similarity was evaluated on the basis of presence or absence of bands. The cluster analysis indicated that the 34 rose cultivars form 9 clusters. The first cluster consists of eight hybrid cultivars, three clusters having five cultivars each, one cluster having four cultivars, two clusters having three cultivars each and two clusters having one cultivar each. The genetic distance was very close within the cultivars. Thus, these RAPD markers have the potential for identification of clusters and characterization of genetic variation within the cultivars. This is also helpful in rose breeding programs and provides a major input into conservation biology.

Elephantgrass pastures are limited by yield variations and reductions in forage quality and availability, thus making the search for genotypes with reduced seasonality a major concern. In order to verify the extent of genetic variability among contrasting cultivars, ten elephantgrass accessions were analyzed through DNA amplification by RAPD technique. A total of 160 DNA bands were generated with the use of 44 random primers and 23% of these bands were monomorphic for all accessions. Gel-obtained binary data (1 for presence and 0 for absence) were used for generating a genetic distance matrix, which was utilized in a UPGMA grouping analysis. Elephantgrass cultivars Cameroon and Vruckwona were the accessions mostly divergent from the others, with an average genetic distance of 0.34. The accessions with the lowest average genetic distances from the others were Pioneiro and CNPGL 27-5, both with a distance of 0.25. Overall, genetic distances ranged from 0.06 to 0.43, indicating little genetic variability for the set of accessions, despite the contrasting morphology of the studied genotypes.

Abstract. The use of molecular markers can support the management of endangered populations and should be combined with appropriate breeding strategies to improve productive traits avoiding the decline of the breed. The genetic variability at 10 microsatellite loci were investigated in a sample of 100 unrelated Markhoz goats (77 females and 23 males). The investigated population was reared at the Sanandaj Markhoz goat Performance Testing Station in Kurdistan, Iran. Markhoz goat, a multipurpose breed, is one of the most valuable genetic resources in Iran. All the studied loci were found to be polymorphic and a total number of 52 alleles were identified with an average number of alleles of 5.2. Moreover, some population genetic indices, such as observed and expected heterozygosity, observed and expected number of alleles, Shannon's index, Nei's expected heterozygosity, and polymorphism information content were also calculated. Despite the decreasing population size, Markhoz goat genetic diversity is still conserved. The breed seems to have a good level of genetic variability and, as a consequence, a potential margin of adaptability to environment and for future genetic improvement.

Eight species and 57 selections/cultivars of Penstemon were compared for genetic variability using Random Amplified Polymorphic DNAs (RAPDs). The RAPD technique was used to help understand the genetic relationships in species and cultivars in the genus Penstemon. Ten RAPD primers (from Operon) were screened to identify polymorphisms among these eight species and 57 selections. More than 100 RAPD polymorphic bands were obtained. A principle component analysis was used to study genetic relationships. Variation among species was greater than variation among selections/cultivars within species. RAPD markers distinguished differences between most cultivars tested. DNA fingerprints generated by RAPDs should be useful to distinguish cultivars of Penstemon, as well as to assist in determining genetic relationships between species.

Camellia sinensis is a beverage tree crop native to Southeast Asia and introductions have been made into several nonindigenous countries. No systematic assessment of genetic variability in tea has been done anywhere. In this study, random amplified polymorphic DNA (RAPD) analysis was used to estimate genetic diversity and taxonomic relationships in 38 clones belonging to the three tea varieties, assamica, sinensis, and assamica ssp. lasiocalyx. Extensive genetic variability was detected between species, which was partitioned into between and within population components. Seventy percent of the variation was detected within populations. Analyses based on band sharing separated the three populations in a manner consistent with both the present taxonomy of tea and with the known pedigrees of some clones. RAPD analysis also discriminated all of the 38 commercial clones, even those which cannot be distinguished on the basis of morphological and phenotypic traits.Key words: genetic diversity, RAPDs, Camellia sinensis.
