Rooting and acclimatization are limiting steps in plant micropropagation, especially in woody plant species. This study aimed to evaluate the IAA and IBA effect on the in vitro rooting and acclimatization of micropropagated shoots of Japanese plum (Prunus salicina Lindl.) cv. América. Shoots from 3 to 4 cm long were inoculated in MS medium with half salt and vitamin concentrations (MS/2) added with IAA and IBA (0, 0.25, 0.5, 0.75 and 1 mg L-1). After a 20-day period in in vitro cultivation, the shoots were evaluated, and then transferred to a greenhouse, and evaluated after 30 days. At the end of the in vitro cultivation period, no significant interactions were observed for number of roots per shoot and rooting percentage, but a significant effect was recorded for auxin type only, for which shoots grown in media added with IBA showed high values - 0.87 and 41.95%, respectively. A linear increase response from 1.45 to 5.75 cm was verified for root length of shoots cultivated in IBA medium; however, no significant effect was observed, and a 0.86 cm average root length per shoot grown in medium added with IAA was found. After 30 days of acclimatization period, the largest survival percentage was obtained from shoots cultivated in medium with 1 mg L-1 of IBA and IAA (88% and 92%, respectively). Although, IBA provided the highest in vitro rooting, most of the surviving shoots were those originated in IAA-added medium, probably because IBA promoted longer fibrous roots, less appropriate for transplant and soil fixation, as they are easily damaged. It was concluded that in vitro rooting with the addition of the highest IAA concentration (1 mg L-1) provided the greatest plant survival during the acclimatization period of the Japanese plum cv. América.

The aim of this study was to determine the concentration and exposure time to NaCl suitable for the micropropagation of banana, through the analysis of growth traits. Banana propagules were inoculated in MS medium with different concentrations of NaCl (0; 50; 75 and 100 mM) for 120 days (multiplication and rooting, 60 days each), with monthly subcultures. These propagules were measured for plant height, number of leaves, sprouting rate, average number of formed propagules, rooting rate, root length and survival rate. After 30 days, NaCl reduced sprouting rate at multiplication; the number of leaves, rooting rate and root length in rooting; and the height and propagules number in both phases. After 60 days, the NaCl affected the sprouting rate and propagules number in the multiplication; length of root in rooting; and the height and number of leaves in both phases. After 120 days, the reduction in the survival rate was proportional to the increase of NaCl in the medium. Thus, it is concluded that NaCl reduces most of the growth traits and the treatments with 75 and 100 mM NaCl affected multiplication and in vitro rooting more intensely.

In vitro cultivation is a highly important biotechnological method widely used for the production of orchid seedlings, but it is necessary to study the suitability of the nutrients used in different kinds of formulation, as the nutritional requirement varies according to the species. The objective was to evaluate different concentrations of iron in the in vitro cultivation of Schomburgkia crispa Lindl seedlings. Seedlings were obtained from seeds germinated in vitro. Modified MS culture medium was used with half of the macronutrient concentration. The micronutrients were added according to the original formulation, except for the iron which was added from a stock solution of FeEDTA (FeSO4.7H2O: 5.6 g L-1 and EDTA: 7.48 g L-1) at 0.0; 2.5; 5.0; 7.5; 10.0 and 12.5mL L-1. At 200 days after seedling transplantation, shoot height, root length, number of leaves, shoot number, leaf length, leaf width, aerial and root dry mass, chlorophyll a, b and carotenoids content were evaluated. A completely randomized design was used, with six treatments and ten replicates of five seedlings. Regression analysis was performed at 5% of significance. The increase in iron concentration caused a reduction in root length and an increase in the number of leaves and shoots. The concentration of 4.13 mL L-1 of FeEDTA was the one that provided the best in vitro growth of S. crispa plants. High concentrations of iron caused a reduction of initial development, but stimulated an increase in the number of shoots.

In vitro rooting of micropropagated shoots were carried out with the aim of evaluating the root induction responses of two tomatoes (Lycopersicon esculentum MILL) varieties (Gelilema and Chali) using Indole -3- butyric acid (IBA). Seven levels of IBA (0.0, 0.25, 0.5, 0.75, 1.0, 1.25 and 1.5 mg L-1) were used in a completely randomized design (CRD) in factorial combinations (seven level of IBA*two varieties) with three replications.  After the plantlets were kept in the rooting media for three weeks, data on rooting percentage, number of roots/shoots and root length in cm were collected. The analysis of variance showed that the interaction of IBA*Var was highly significantly different for rooting percentage, a number of roots/shoot and root length at p<0.01. The highest rooting percentage (100.00±0.00), number of roots/shoot (14.20±0.35) and root length (10.7±0.29) were received from Chali on free Murashige and Skoog medium (MS). At the same time, the lowest percentage of rooting (11.11±0.00), number of roots/shoot (0.887±0.19) and root length (1.00±0.00 cm) were obtained from Gelilema on MS+1.5mg/l IBA. For acclimatization, the in vitro rooted shoots were transplanted into plastic pots containing a mixture of oven sterilized soil and sand at a ratio of 2:1. After three weeks, a survival rate of 67.7% for Chali and 58.1% for Gelilema was obtained. From the above result, it can be concluded that free MS medium was the best for in vitro rooting of the two tomato varieties. The optimized protocol will be useful for rapid in vitro multiplication of the two tomato varieties.

Acmella oleracea is a tropical plant, typical of the northern region of Brazil. The species belongs to the Asteraceae family and has great therapeutic, pharmacological and industrial potential. A limiting factor for the production of this species on a large scale is the short life cycle. The tissue culture programs use synthetic hormones based on cytokinins, such as kinetin and benzylaminopurine (BAP) and auxins such as naphthalene acetic acid (ANA). The objective of this research was to evaluate the effect of growth regulators on the production of Acmella oleracea "in vitro". The experimental test was carried out with control (C), without the addition of growth regulators and five treatments, composed of: (T1) 0.1; (T2) 0.3; (T3) 0.5 mg L-1 kinetin; (T4) 0.1 mg L-1 of BAP and ANA; (T5) 0.5 mg L-1 of BAP and ANA. The experimental design was a completely randomized block in a factorial arrangement with six treatments, three blocks and twenty-five repetitions per block. The evaluated parameters were: germination, root formation, aerial part length, root length, aerial part fresh mass and root fresh mass, aerial part dry mass and root dry mass. The data obtained were subjected to analysis of variance (p <0.05) and compared using the Tukey test. The results showed that kinetin positively contributed to seed germination and aerial part dry mass development. Treatment 1 had the best results for the parameters root length, shoot length and root dry mass.

The experiment was conducted to investigate the combined effect of different plant growth regulators with and without charcoal supplementation for root formation and plantlet development from protocorm like bodies (PLBS) of orchid. The combination of BAP + NAA, BAP + IAA, BAP + IBA, and IAA + IBA at different concentrations were studied. It revealed that the highest number of roots was obtained from 1.0 mg/L each of IAA + IBA combination (6.667) and the highest root length was recorded from 2.0 mg/L BAP + 1.0 mg/L IBA with charcoal supplementation. The treatment combinations, 1.0 mg/L each of BAP + NAA, BAP + IAA, BAP + IBA, and IAA + IBA were found best for producing more rooted plantlets with charcoal supplementation. Nodal explants of difficult-to-root Gordal olive were cultured on half-strength OM medium after surface sterilization with 15% NaOCl for 20 minutes. The in vitro-proliferated shoots were then submitted for 25 days to rooting trials including various concentrations of NAA and IBA and different half-strength rooting media as well.Among the media tested during rooting stage, highest rooting percentage 62% found in both OM and MS half-strength media when both the growth substances were used, and 87.5 and 90% gained when IBA and NAA were added to the rooting media at 2 Mg/L each, respectively.Highest number of roots 1.85 and highest root length 3.09 mm obtained with half-strength MS and OM media respectively, when IBA was used with these media. And IBA at 2 Mg/L in the rooting media gave the highest number of roots 2.5 with 4.13 mm length.However, 2 Mg/L NAAin the rooting media gave 1.94 number of roots with length of 3.88 mm, while number of roots 1.38 with 2.79 length obtained with half- strength OM media when NAA was used in the rooting media .It is therefore concluded that best medium for in vitro rooting of Gordal microcuttings is half- strength OM supplemented with 2 mg/L of IBA.

Abstract: The objective of this work was to evaluate the effects of the solidifying agents agar and phytagel and of activated charcoal on the in vitro cultivation of two maná cubiu (Solanum sessiliflorum) varieties: Thaís and Santa Luzia. The phytotechnical characteristics analyzed included number of leaves, number of roots, shoot and root length, and fresh matter of shoot and root. Regardless of the variety, phytagel was superior to agar as a culture medium. A greater number of leaves and longer shoots were observed in the Santa Luzia variety, in the absence of charcoal. The Thaís variety showed longer shoots and roots in the presence of charcoal.

Abstract Lavender, an aromatic and medicinal plant, used in the extraction of essential oil, as an ornamental and meliferous plant, can be successfully propagated as long as the light, the nutrient medium and the growth regulators are adequate. The aim of the study was to evaluate the effect of different concentrations of indolebutyric acid (IBA) associated to distinct light spectra, on the in vitro rooting of Lavandula angustifolia. The experiment consisted in the combination of two concentrations of indolebutyric acid in the growth medium (0 and 0.1 mg L-1) and two light spectra, using cellulose acetate filters (blue and red), besides the control (without filter), totaling six treatments with four repetitions. For each repetition five explants were used. After 30 days, the variables evaluated were: survival percentage, number of buds and leaves, shoot length, number of roots, length of the longest root, shoot fresh and dry matter weight. There was a significant interaction among the indolebutyric acid concentrations and the light filters for the variables number of buds, leaves and roots and shoot length. Except for leaf number, promising results were obtained when the explants were held under red filters and with a concentration of 0 mg L-1 of indolebutyric acid in the growth medium. From these results, it can be concluded that the best in vitro rooting of Lavandula angustifolia is obtained when there is no indolebutyric acid in the growth medium associated with the use of the red filter.

The aim of this study was to evaluate the effect of aqueous ora-pro-nobis (Pereskia aculeata Mill.) extract on in vitro growth, survival rate, and acclimatization of Miltonia flavescens. Crude ora-pro-nobis extract was produced by infusing 10 g of dry leaves fractionated in 100 mL of distilled water. The treatments consisted of the crude extract (100%) and dilutions of 75%, 50%, and 25% added to Murashige and Skoog ½ culture medium (control treatment: absence of extract) where seedlings germinated in vitro were cultivated. After 270 days of cultivation, growth data were collected, and the plants acclimated. The survival rate was evaluated every 30 days, and the growth of the aerial part and root system were measured after 120 days of acclimatization. The data were submitted to analysis of variance and a test of the separation of means (Skott-Knott). For the number of leaves, the 100% treatment had the highest average value, differing statistically from the other treatments. For the length of the root system and fresh mass, the highest averages occurred in the control and 100% treatments. For the number of roots and shoots, there were no differences between treatments. After 120 days of acclimatization, plants from the 100% treatment had the lowest survival percentage, and plants from the 25% and 50% treatments had the greatest increase in the growth of the aerial part and root systems, demonstrating these treatments would be beneficial for the cultivation of this species.
