SUMMARY In dairy farm animals, one the most important goal of the selection is the improvement of milk yield and composition. Several studies have demonstrated that the candidate genes of the kappa-casein (CSN3) and β - lactoglobulin (β-LG) are associated with milk yield, milk quality and health traits in dairy animals. Therefore the aim of this study was to detect polymorphisms in CSN3 and β-LG genes and its association with milk yield in up to 305 days (305MY) and predicted transmission capacity (PTA) for 305MY in Girolando cattle. Totally, 138 bulls and 729 cows (n=867) were sampled. The genotypes of both genes were obtained by the PCR-RFLP method using HinfI and HaeIII enzymes for CSN3 and β-LG genes, respectively. Statistical results revealed two alleles A and B for both genes. The genotypes and alleles more frequents for CSN3 and β-LG genes were respectively: AA (0.7324) and A (0.8558), and AB (0.4827) and A (0.5017). The x2 test revealed that the two loci were at Hardy-Weinberg equilibrium (p<0.001). The allele substitution effects for the variants were not significant on 305MY and PTA for 305MY (p>0.05). The allele variants of β-LG and CSN3 might be more investigated before include them into future breeding schemes designed for Girolando dairy cattle with objective of improving milk traits as milk yield in up to 305 days (305MY) and predicted transmission capacity (PTA) for 305MY..

Abstract The animal selection with favourable phenotypes of the past has been, currently, replaced by the genotype selection on quantitative traits, assisted by the expanding molecular techniques in the context of livestock improvement. In this study, the c.112T>C polymorphism in exon II of β-lactoglobulin (β-LG) gene was investigated in Karagouniko and Chios sheep breeds by using polymerase chain reaction - restriction fragment length polymorphism (PCR-RFLP), and possible associations with milk traits were examined. In total, 125 blood DNA samples were isolated for PCR-RFLP analysis and the respective 217 milk samples′ composition profile was obtained. The goodness of fit test to Hardy-Weinberg equilibrium (HWE) for β-LG genotypes was estimated and associations found between β-LG genotypes and raw milk composition. Two alleles and three genotypes were observed (AA, AB and BB) in both breeds, and Chios breed significantly deviated (P≤0.05) from Hardy-Weinberg equilibrium (HWE). Conclusively, linear mixed model analysis on samples, from both breeds collectively, showed significant effects of β-LG genotype on lactose percentage and somatic cell count (SCC), lactation stage on daily milk yield and protein, while the breed effect was significant only on daily milk yield.

Objective of this study was to analyze the effect of the dairy farm on milk traits of cows in Vojvodina. The research was carried out on small farms with 10 to 20 cows, medium farms with 20 to 50 cows, and large farms with over 50 dairy cows. The study included registered animals of Simmental (SM) and Holstein-Friesian breed (HF; including Red Holstein) in the first lactation for traits of milk yield and yield and content of milk fat. Total of 1323 first lactations were analyzed. The average milk yield (both breeds) in the first lactation of 305 days was 6295 kg of milk with 234,3 kg of milk fat and average milk fat content of 3,74%. Milk performance of cows varied significantly (CV=22,9% and SD=1447,8), as well as milk fat yield (CV=21,6% and SD=50,8). Large farms produced in average 6534 kg of milk, medium farms 6347kg and small farms 4717kg. Size of the farm exhibited significantly high effect on all observed traits, and the tendency was that farms with higher number of animals realize also higher average of production. Farm management and various breeding-zootechnical conditions present on farms had significant effect on milk performance of cows.

ABSTRACT Objective. Analyze the structure and genetic differentiation of a population of Antioquia Holstein cows from the polymorphisms A192G of INHA and A-320T of FSHR, and explore the association of the genotypic combinations with milk traits. Materials and methods. 1240 lactations of 356 animals from 9 herds in 6 municipalities of Antioquia were analyzed. Genotyping was performed by PCR-RFLP. Structure and genetic diversity parameters were determined using GenAlex software. The association of genotypes combinations with productive and reproductive traits was explored through a linear mixed model. Results. SNP A192G showed a frequency of 0.534 and 0.466 for A and G alleles respectively and SNP A-320T had a frequency of 0.660 far A allele and 0.339 for T allele, this way the population is in HWE. The FST, FIS and FIT values were 0.059, 0.285 and 0.328 respectively indicating a moderate genetic differentiation between subpopulations. The A-320T SNP showed significant effect on milk yield. Fat and protein percentage, calving interval and services per conception were not affected by these polymorphisms or their interaction. Conclusions. Phenotypic selection made on this population has not been strong enough to generate noticeable changes in allele frequencies of these polymorphisms or deviations from Hardy-Weinberg equilibrium. The interaction of these polymorphisms has no significant effect on the characteristics of zootechnical interest, so its use in programs of molecular marker assisted selection is not recommended.

Abstract Background: SLC27A3 and β-Lactoglobulin gene are candidate genes that effects milk traits.Results: This study aimed to determine the effect of polymorphisms of the SLC27A3 gene on Hamdani sheep's milk components. The PCR-RFLP method was used to detect the SLC27A3 polymorphisms at exon 2, exon 3 and exon 4. The allele frequency of SNP1, SNP2 and SNP3 were 0.59(G), 0.41(T); 0.57(G), 0.43(C) and 0.58(A) and 0.42(C) respectively. β-lactoglobulin allele frequency was 0.56(A) and 0.44(B). The statistical analysis showed an association between SLC27A3 and β-lactoglobulin polymorphisms and milk composition. Conclusions: The SLC27A3 and β-lactoglobulin polymorphisms can be used as a genetic marker to select milk component traits in the sheep breeding programs.

Pathogens are always a threat to the livestock and domestic animals due to their exposure to the contaminated environments. The study was conducted to evaluation of the prevalence of Escherichia coli, Shigella spp., Salmonella spp., and S. aureus, in farm animals (cattle and buffalos). A total of 150 (n = 150) samples were collected from cattle and buffaloes, 60 samples from cows’ and buffalo’s teats milk, 30 of water samples, and 60 of fecal samples isolates from dairy farm animals, which may act as reservoir disseminating such pathogens. Farm hygiene, management, and milking procedure were listed through a questionnaire. The most common pathogens detected in this study was E. coli 88 (58%) and S. aureus 81 (54%), followed by Salmonella spp. 32 (21%), and Shigella spp. 44 (29%), respectively. During the antibiogram studies, the results revealed that the highest number of bacterial isolates showed resistance against ampicillin 50 (56.8%), followed by ciprofloxacin 23 (26.1%) and augmentin 22 (25%) of Escherichia coli and ampicillin 49 (60.4%), cefpodoxime 23 (28.3%), and augmentin 20 (24.6%) of S. aureus. In the case of Salmonella spp., the highest resistance was showed by amoxicillin 16 (50%). In Shigella spp., the highest resistance was shown by ampicillin 16 (36.3%), followed by cefpodoxime and ceftazidime 10 (22.7%). The high frequency of isolates in this investigation with multiple antibiotic resistance ranges from 15. MARI % value of S. aureus and E. coli 15 (12.5%), followed by Salmonella and Shigella spp. ranges from 12 (10%), suggesting the presence of various antibiotic-resistant bacteria as well as highly resistant bacteria. The mean ± SD zone areas for the greater resistance are for E. coli and S. aureus, already known to be multiresistant, followed by Salmonella spp. and Shigella spp., when the zone areas are for the low resistance, and the findings determined that there was a little difference between S. aureus and E. coli.

Abstract. This study was conducted to determine the polymorphisms of the POU1F1 gene and their relationships with milk yield and components, litter size, birth weight, and weaning weight in goats. For this purpose, a total of 108 Saanen goats from two different farms (Bornova and Manisa) were used as animal materials. Polymorphisms at the exon 6 and the 3′ flanking region of the POU1F1 gene were determined by using PCR-RFLP with PstI and AluI restriction enzymes and DNA sequencing analyses. Two alleles and three genotypes were identified by AluI or PstI digestions of the POU1F1 gene. The genotypes frequencies of TT, TC, and CC were 64.8 %, 31.5 % and 3.7 % for the PstI locus; 54.6 %, 31.5 % and 13.9 % for the AluI locus, respectively. T allele frequencies (0.56 and 0.88 for the AluI locus, 0.80 and 0.81 for the PstI locus, respectively) were predominant in both loci at the Bornova and Manisa farms. In terms of POU1F1-AluI and POU1F1-PstI loci, two populations were found to be in Hardy–Weinberg equilibrium. In the POU1F1-AluI locus, significant associations were found between genotypes and lactation milk yield and litter size. Similarly, a significant relationship between genotypes and birth weight in the POU1F1-PstI locus (p<0.05) was determined. The TC and CC genotypes were observed to be higher than the TT genotype for lactation milk yield and litter size at the POU1F1-AluI locus. Birth weight was found to be higher in animals that have the CC genotype at the POU1F1-PstI locus. In conclusion, the POU1F1 gene can be used as a molecular marker for economic features like reproduction, growth, milk content and yield in Saanen goats.

Abstract Prolactin is a polypeptide hormone, encoded by the prolactin (PRL) gene, synthesized and secreted by anterior pituitary, and affecting milk yield and composition. β-Lactoglobulin (BLG) is the major whey proteinin the milk of ruminants. This study was conducted to identify the PRL and LGB genes polymorphism in the Indonesian FH cattle. A total of 139 individual cattle blood samples from West Java were used to obtain DNA samples through the DNA extraction process. Identification of the PRL and LGB genes was performed using PCR-RFLP method with RsaI (PRL gene) and HaeIII (BLG gene) restriction enzymes. The PRL gene was amplified using forward primer 5’-ccaaatccactgaattatgctt-3’ and reverse primer 5’-acagaaatcacctctctcattca-3’. The BLG gene was amplified using forward primer 5’-tgtgctggacaccgactacaaaaag-3’ and reverse primer 5’-gctcccggtatatgaccaccctct-3’. The PRL and BLG genes in the Indonesia FH cattle were polymorphic based on the PCR-RFLP analysis but the heterozygosity value was low. There were two alleles (G and A) and three genotypes (GG, GA, and AA) identified in the PRL gene of the Indonesian FH cattle with genotype frequencies were 0.914, 0.079, and 0,007 for GG, GA, and AA genotypes respectively. There were two genotypes (CC and CG) identified in the BLG gene with genotype frequencies were 0.91 (CC), and 0.09 (CG). Information about the PRL and BLG genes polymorphism in this study can be considered for further study to analyse its association with milk yield trait.

This study was designed to determine the effect of some environmental factors on milk yield and composition in Saanen goats and the effect of CSN3 and AGPAT6 gene polymorphisms on milk production traits. Saanen goats were genotyped for milk traits, and the samples were collected during the lactation, and the milk yield/compositions of each goat were specified. In terms of the CSN3 and AGPAT6 genes, the genotypes were defined by PCR-RFLP. The milk yield (MY), lactation length (LL), protein, fat, total solid (TS), solid not fat (SNF), casein, lactose rates and yields were as follows; 388.9 ± 17.5 kg, 243 ± 5.81 days, 3.29 ± 0.05%, 3.59 ± 0.07%, 11.60 ± 0.13%, 8.43 ± 0.06%, 2.57 ± 0.04%, 4.31 ± 0.03%, 11.21 ± 0.47 kg, 12.79 ± 0.64 kg, 40.95 ± 1.82 kg, 29.53 ± 1.31 kg, 8.82 ± 0.37 kg, 15.40 ± 0.74 kg respectively. The determined genotype and allele frequencies of CSN3 and AGPAT6 gene were as follows; FF (%99.20), MF (%0.80), F (0.996), M (0.004); GG (%12), GC (%43.20), CC (%44.80), G (0.336), C (0.664). The impacts of lactation length on all milk yield parameters (protein, fat, TS, SNF, casein, lactose, MY) (P<0.001); the age on all milk yield parameters (P<0.001) and the protein, fat, SNF, casein rates (P<0.05), the TS rate and LL (P<0.01) were found to be significant. The birth type was not found to significantly affect any milk yield parameters. Although Saanen goats showed the variation for the AGPAT6 gene (g.9263C>G), the herd was found to be monomorphic (FF) for the CSN3 gene. The effect of the AGPAT6 gene on milk traits in Saanen goats was determined not to be statistically significant. Environmental factors, such as maternal age and lactation length, were found to significantly affect some milk traits in Saanen goats. So consideration of factors such as maternal age or lactation length could be useful for improving breeding strategies for dairy goats.

Attitudes toward practical dairy cow welfare issues were evaluated based on a questionnaire answered by 500 dairy farm workers and 27 veterinary practitioners. Primarily, the effect of demographic characteristics on attitudes toward cattle welfare was tested. Professionally, five themes were identified: effect of welfare awareness on productivity, knowledge of cattle’s senses and social structure, effects of man–animal interactions on milk yield, pain perception and prevention, and knowledge transfer from veterinary practitioners to farm workers. Farms with a higher welfare awareness score also had higher annual milk yield, with an annual mean difference of 1000 L of milk per cow between farms with higher and lower awareness scores. Veterinary practitioners showed high awareness of cows’ social structure, senses, and pain perception. Farm workers were aware of the influence of man–animal interactions during milking and stress effects on milk yield, and the possible effect of man’s behavior on heifers and cows. Practitioners and farm workers had different views regarding pain perception, mostly involving mutilation procedures. All veterinary practitioners advocated the use of pain alleviation in painful procedures, but only some of them instructed the farm workers to administer it. The survey results emphasize the variation in welfare knowledge and practical applications across farms, and the interest of both the animals and their managers to improve applied knowledge of best practice.

BACKGROUND: Obesity, as a medical condition, results from interactions between environmental and genetic factors. The rs17782313 polymorphism, located 188kb downstream of the Melanocortin 4 Receptor (MC4R) gene, is one of the essential candidate genetic markers that has shown the highest association with obesity in different populations. OBJECTIVE: This study aimed to investigate the possible associations of rs17782313 polymorphism near the MC4R gene with obesity/overweight, body mass index (BMI), and hedonic hunger among women from the Iranian Azeri ethnic group. METHODS: Five hundred sixty-three women, composed of 396 patients with obesity/overweight and 167 unrelated healthy controls, were genotyped for the rs17782313 polymorphism by applying the polymerase chain reaction-restriction fragment length polymorphism (PCR–RFLP) method. RESULTS: This population was in Hardy-Weinberg equilibrium (P = 0.878). The study confirmed a significant association of rs17782313 with obesity, where subjects carrying the C/C genotype had higher odds of obesity (OR = 2.681, P = 0.005, 95%CI:1.340–5.365). Also, C allele carriers have statistically significantly higher BMI scores than those carrying the T allele (P = 0.029). However, no significant associations were found among PFS scores and genotypic/allelic groups of rs17782313 polymorphism (P = 0.368). CONCLUSIONS: Our findings suggest that rs17782313 polymorphism is strongly associated with obesity and BMI but not with hedonic hunger among Northwest Iran women. Moreover, the sequencing data analysis in several homozygous and heterozygous carriers of the C allele led to identifying a novel frameshift variant with TCT deletion (rs534212081) in the 166 upstream of rs17782313, which has not been reported so far.
