Abstracts Background Chemotherapy is associated with male infertility. Cisplatin (CP), an antineoplastic agent has been successfully used for the treatment of diverse kinds of malignancies, however, the use of this effective agent could induce oxidative stress injury, nephrotoxicity, hepatotoxicity, and testicular damage. Combined CP chemotherapy with plant extracts can diminish the toxicity and enhance the antitumor efficacy of the drug. The objective of the study was to determine the protective effect Lophira lanceolata leaf extract (LLLE) on CP-induced toxicity on male reproductive organs. Methods The study was carried out with 30 (n = 30) male Wistar rats (Rattus norvegicus). The rats were randomly assigned into 6 groups of 5 rats each. Rats in group 1 (Control) were administered distilled water per os. Rats in group 2 were administered 5 mg/kg of CP intraperitoneally (i.p). Rats in groups 3 and 4 were administered per os LLLE at the doses of 200 and 400 mg/kg body weight and rats in groups 5 and 6 were administered 5 mg/kg body weight of CP + LLLE at the doses of 200 and 400 mg/kg body weight respectively. Results The results showed a significant decrease in the sperm parameters in the group treated with CP alone when compared with the control and there in the sperm parameters in the groups administered CP + LLLE. The body and organ weights of the rats were significantly (p < 0.05) decreased in the CP treated group relative to the control. However, there was an increase in the weight of the organs in the LLLE pretreated groups. The photomicrographs showed degenerative changes in the testicular tissues of the rats administered CP alone whereas the group pretreated with the LLLE showed amelioration induced by the CP. Our study revealed that CP treatment has deleterious effects on sperm parameters and testicular tissues and the accessory sex organs (Epididymis, prostate, seminal vesicles) of the rats. Oral administration of LLLE at 200 and 400 mg/kg bodyweight for 26 days conferred protective effects against testicular damage induced by CP. Conclusion This study revealed that pretreatment with LLLE protected against CP-induced testicular toxicity.

Abstract Background Waltheria indica is a multipurpose medicinal plant with abundance of phytochemical compounds. Antifertility effect of Waltheria indica Linn. root and leaves have been reported. However, the fraction responsible for this antifertility effect needs to be isolated for possible male contraceptive purpose. Therefore, this research was designed to isolate the antifertility fraction of Waltheria indica Linn. root (WILR) in an in vivo model using male Wistar rats. Crude ethanol extract of WILR was sequentially dissolved in hexane, dichloromethane, and ethyl acetate. Rats (n = 5) were administered with 200, 500, or 1000 μg/kg of hexane, dichloromethane, and ethyl acetate soluble extracts of WILR, while control received distilled water, daily for 15 days to determine the soluble extract with most antifertility effect. Thereafter, fractions were separated from dichloromethane soluble WILR extract by column and thin-layer chromatography. Rats (7 groups, n = 5) were administered with each of the fractions (DF1 to DF7; at 1000 μg/kg) to determine the fraction with the highest antifertility. Rats were thereafter sacrificed, and sperm parameters, reproductive hormones, testicular cholesterol, and protein were determined according to standard procedure. Histology of the testis was also done. Data were analyzed using ANOVA at p ≤ 0.05. Results Dichloromethane soluble fraction (500 μg/kg) significantly decreased sperm concentration (137.00 ± 9.85 to 107.00 ± 13.08 × 106 cells/mL), levels of testosterone (2.90 ± 0.65 to 1.50 ± 0.37 ng/mL), and FSH (0.08 ± 0.08 to 0.99 ± 0.08 IU/L). The dichloromethane soluble fraction also caused the loss of testicular interstitium and spermatogenic cells. DF5 significantly reduced sperm motility (92.00 ± 2.74 to 76.00 ± 5.48%) and LH (2.86 ± 0.52 to 1.47 ± 0.18 IU/L). DF5 also significantly increased levels of prolactin (1.22 ± 0.10 to 1.88 ± 0.48 ng/mL), testicular total protein (7.36 ± 0.35 to 8.54 ± 1.06 g/dL), and testicular cholesterol (34.17 ± 3.65 to 55.76 ± 6.08 mg/mL). Conclusion The results indicate that the DF5 is the bioactive fraction of WILR responsible for its antifertility effect. The possible antifertility mechanisms are through the reduction in sperm parameters, reproductive hormones, and histological changes in the testis.

Abstract Background Numerous uses of Waltheria indica plant such as antitrypanosomal, antibacterial and antimalarial effects have been reported. It has however been reported that most plants with antibacterial and antiprotozoal effects have adverse effect on male reproduction. Hence, we evaluated the effect of Waltheria indica root on male reproductive parameters. Methods Twenty adult male Wistar rats were randomly divided into four groups (n=5); A–D. Group A served as control group while groups B, C and D were administered with 200, 400 and 800 mg/Kg body weight of crude ethanolic extract of Waltheria indica root. After 28 days of administration, the rats were sacrificed and sperm parameters, sperm morphology, serum reproductive hormones and lipids were determined. Results There was a significant reduction in sperm count and motility as well as significant increase in percentage abnormal sperm cell (p<0.001) at the 400 and 800 mg/kg BW. The serum levels of testosterone was also significantly reduced while total cholesterol increased significantly (p<0.05) at the highest dose. Conclusion Waltheria indica root has adverse effect on male reproduction through reduction in sperm parameters and male reproductive hormones.
