Effect of Three Different Pastes on Remineralization of Initial Enamel Lesion: An in Vitro Study

Objective: Dental caries in enamel is unique as enamel is both acellular and avascular. Fluoride is one of the most effective remineralizing agents in caries prevention. The purpose of the study was to assess whether nano-hydroxyapatite and CPP-ACP can be used as an alternative to fluoride for remineralization of early carious lesions. Study design: Initial artificial carious lesions were prepared in enamel with demineralization solution. The treatment materials used as remineralizing agents were Nano-hydroxyapatite (10%), CPP – ACP (10%), NaF (1000 ppm) and Deionized water (negative control). Surface microhardness (SMH) measurements were performed before/after demineralization and after 3, 6, 9 and 12 days of pH cycling and the percentage surface microhardness recovery (%SMHR) was calculated. The specimens were then examined by scanning electron microscope. Results: Percentage surface microhardness recovery of nano-hydroxyapatite and fluoride was significantly greater than CPP – ACP and negative control. There was no significant difference of %SMHR between n-HA and fluoride at different time periods in the pH cycling. SEM analysis revealed n-HA particles were deposited on the demineralized enamel surface which formed a new surface layer. When observed under SEM CPP – ACP failed to show any significant surface remineralization while surface globular crystal depositions with multiple discrete porosities were evident with fluoride. Conclusion: It was concluded that nano-hydroxyapatite and fluoride had the potential to remineralize initial enamel lesions. CPP – ACP can be used as an effective adjunct to fluoride therapy but cannot be used as an alternative to fluoride.

This study aimed to evaluate microhardness of a dentifrice containing fluoride and arginine compared to a positive control (fluoride only) and a negative control (no fluoride) on sound and demineralized bovine enamel surfaces. Specimens were randomly assigned to different treatments that included daily pH cycling and brushing three times a day with one of the following dentifrices (n = 8): Neutraçucar (arginine and fluoride), Colgate Total 12 (fluoride) and My First Colgate (no fluoride). Enamel carious lesions were artificially created one week before the beginning of these treatments (demineralized bovine enamel (DE) groups). The same groups were also tested in sound enamel (sound bovine enamel (SE) groups). Microhardness was measured at baseline and after one, two, and five weeks of treatment using a Knoop indenter. Statistical analysis involved two-way Analysis of Variance (ANOVA) and Tukey’s test. After five weeks, both Total 12 and Neutraçucar had increased the microhardness of DE specimens (p < 0.05). Only Neutraçucar had increased the microhardness of the sound enamel after five weeks of treatment. Thus, it could be concluded that arginine-based dentifrices increase the microhardness of sound and demineralized bovine enamel surfaces.

ABSTRACT Objective: This in vitro study was designed to assess shear bond strength (SBS) of ormocer flowable (OF) resin as a luting agent, ormocer as an indirect veneer material with portrayal of modes of failures using scanning electron microscope (SEM). Materials and Methods: Sixty maxillary central incisors were divided into Group I, II, and III with 20 samples each based on luting cement used. They were OF, self-adhesive (SA) cement, and total etch (TE) cement. These groups were subdivided into “a” and “b” of ten each based on the type of veneering materials used. Veneer discs were fabricated using Ormocer restorative (O) and pressable ceramic (C). Specimens were thermocycled and loaded under universal testing machine for SBS. The statistical analysis was done using one-way ANOVA post hoc Tukey honest significant difference method. Results: A significant difference was observed between the Groups I and II (P < 0.05). The highest mean bond strength when using ormocer veneer was obtained with the Group Ia (19.11 ± 1.92 Mpa) and lowest by Group IIa (8.1 ± 1.04 Mpa), whereas the highest mean bond strength while using ceramic veneer was of similar range for Group Ib (18.04 ± 4.08 Mpa) and Group IIIb (18.07 ± 1.40 Mpa). SEM analysis revealed OF and TE presented mixed type of failure when compared with SA where failure mode was totally adhesive. Conclusion: OF was found equally efficient like TE. Bond strength of ormocer as a veneer was not inferior to ceramic making it one of the promising additions in the field of dentistry.

SUMMARY This in vitro study evaluated the effect of six surface treatments on the shear bond strength of three resin-modified glass ionomers (RMGIs) to dentin. Occlusal surfaces of caries-free third molars were reduced to expose only dentin. Surface treatments were smear layer intact (negative control), Cavity Conditioner, EDTA, Ketac Primer, Self Conditioner, and etching with 35% phosphoric acid followed by the application of Optibond Solo Plus. Filtek Z250 composite resin bonded with Optibond Solo Plus served as a positive control. Conditioning agents were used according to the manufacturers' instructions. After surface treatments, Fuji II LC, Riva LC, Ketac Nano, and Filtek Z250 were placed in copper-band matrices 5 mm in diameter and 2 mm in height and were light-cured for 20 seconds. Specimens were stored in 100% humidity for 24 hours, after which they were placed in deionized water for 24 hours at 37°C. They were then tested under shear forces in an Instron Universal Testing Machine at a crosshead speed of 0.5 mm/min. A two-way analysis of variance and Tukey honestly significant difference statistical analyses (p&lt;0.05) indicated significant interaction between RMGIs and conditioning agents. Acid etching followed by Optibond Solo Plus provided highest bond strengths for all three RMGIs, which were not statistically different from the positive control.

Introduction The aim of this in vitro study was to assess and compare the effect of different intermediary bases on microleakage between tooth and a nanocomposite interface in Class II box cavities in primary teeth. Methods Standard Class II box cavities were prepared in 52 primary molars and randomly divided into 9 groups according to the intermediary base used (Multicore Flow, Fuji II LC, SDR, Smart Dentin Replacement, and Biodentine). All specimens were subjected to thermocycling and prepared for microleakage testing and evaluation. Results There was significant difference in the mean ranks of microleakage between the 9 groups, which was observed in the gingival side (p<0.0001) and the occlusal side (p<0.0001). The mean ranks microleakage was significantly higher with experimental SDR, experimental Multicore Flow, and positive control materials when compared with the other 6 groups. The microleakage mean ranks were statistically significantly lower in experimental Fuji II LC, experimental Biodentine, and all negative control groups when compared with the other 3 groups. Conclusions Microleakage is affected by the application of intermediate material. Experimental Biodentine and Fuji II LC showed the lowest microleakage while experimental SDR and experimental Multicore Flow showed the highest microleakage.

Objective: Proper apical seal plays an important role in the success of root canal treatment. The most common cause of failure of root canal therapy is known as the lack of adequate apical seal. The aim of this in vitro study was to compare the apical microleakage of MTA Fillapex, , and Endofill sealers using dye penetration method. Material and Methods: In this in vitro study, 72 single-rooted extracted human teeth were selected. The teeth were randomly divided into three experimental groups of 20 and two positive and negative control groups of 6. The canals were prepared by step-back technique and then filled with gutta-percha and one of the sealers mentioned. In the positive control group, the canals were filled with gutta-percha without sealer, and in the negative control group, the canals were prepared but not filled. The teeth were immersed in 2% methylene blue dye for 72 hours. The teeth were then cut longitudinally and the level of dye penetration was measured under a stereomicroscope. Data were analyzed by SPSS ver. 19 software, ANOVA and Bonferroni post-hoc tests. Results: The mean level of dye penetration in the Endofill test group was significantly higher than that in the  and MTA Fillapex test groups. While, the observed difference between  and MTA Fillapex groups was not statistically significant (p<0.05). Conclusion: The results of this study showed that  and MTA Fillapex sealers did not show any significant difference in apical seal properties. <p><strong>Objective: </strong>Early carious lesions in bovine and humanenamel developed in vitro using a pH cycling regimenwere compared. <strong>Material and Methods: </strong>Fifteencentral bovine incisors and fifteen recently extractedhuman third molars were randomly divided into twogroups: ten for the cross-sectional microhardness test(MT) and five for polarized light microscopy (PLM)analysis. Enamel blocks measuring 5 x 5 mm weremade from the buccal face of the teeth. The blocksused for the MT were sliced into two halves: “A” and“B”. “A” slices were embedded in acrylic resin, withthe face of the dentin-enamel junction left exposedfor the MT prior to pH cycling. “B” slices and wholeblocks were coated with acid-resistant varnish,except a 3 x 3 mm central window, and submitted tothe pH cycling regimen (demineralizing solution for3 h and remineralizing solution for 21 h) over fiveconsecutive days. The “B” slices were then submittedto the MT and the whole blocks were processed forthe PLM study. <strong>Results: </strong>The PLM analysis revealedshallow, extensive lesions in the bovine enamel,hardly showing the superficial, dark and translucentzones, as well as deep cavity lesions in the humanenamel, with the body of the lesion and the darkzone evident. Background. Internal bleaching is a choice of treatment in discolored endodontically treated teeth. Cervical root resorption is one of the important complications of this treatment. A suggested procedure to prevent this type of resorption is using a coronal barrier under the bleaching materials. The aim of the study was to compare the microleakage of mineral trioxide aggregate (MTA), calcium-enriched mixture (CEM) cement, and Biodentine. Materials and Methods. In this in vitro study, a total of 60 single canal incisors were included. They were randomly divided into three experimental groups (n = 16), one positive control group (n = 6), and one negative control group (n = 6). Coronal portion of the canals in the experimental groups was sealed with 3 mm of MTA, CEM cement, or Biodentine as a coronal barrier. After 3 days, specimens were bleached. A fresh Enterococcus faecalis suspension was added to the samples. The culture tubes were observed for 45 days, and the daily turbidity was recorded. Statistical analysis was accomplished by the Kaplan–Meier test and SPSS 22. Results. All positive samples showed turbidity, whereas none of the negative samples allowed bacterial leakage. Results showed no significant difference between MTA, CEM cement, and Biodentine groups. ( P value = 0.304, 0.695, and 0.217). The bacterial microleakage for the two groups also did not show significant differences. Conclusions. CEM cement and Biodentine showed promising results as coronal plug, and clinical studies are needed to test these materials with MTA for avoiding microleakage in internal bleaching treatment.

ABSTRACT Objective: This in vitro study analyzed the effects of a bleaching treatment containing 35% hydrogen peroxide (HP) with or without calcium on bovine enamel, using the Knoop hardness number (KHN), tristimulus colorimetry (TC), and scanning electron microscopy. Materials and Methods: Forty-five specimens were randomly divided into groups (n = 5), which included artificial saliva (negative control [NC]), 35% HP (positive control [PC]), and 35% HP Blue Calcium (HP Blue). The specimens were subjected to three bleaching sessions. During the sessions, the specimens were immersed in artificial saliva at 37°C. Before and after bleaching, KHN tests were conducted using a force of 25 gf for 5 s. TC was performed using the CIE-L*a*b* system and readouts were obtained at the following 4 time points: Before the bleaching treatment; after the first session, the second session, and the third session. The specimens were dehydrated and coated with gold, and the photomicrographs were analyzed in a double-blind manner with a LEO microscope. Results: Using one-way analysis of variance and Tukey's test (P < 0.05), a statistically significant difference was identified between the initial and final mean KHNs of the NC and PC groups, while the initial and final mean KHNs were not significantly different in the HP Blue group. The final mean values of ⋄E, ⋄L, and ⋄b of the PC and HP Blue groups were significantly higher than the initial values (P < 0.01 for both). The photomicrographs revealed no differences among the groups. Conclusions: Therefore, treatment with HP Blue prevented changes in the KHN without reducing the efficacy of bleaching.

Background and Aims: Trichomonas vaginalis is a flagellated protozoa that is associated with vaginitis, cervicitis, urethritis and other vaginal disorders. Current study aimed to evaluate the anti-Trichomonas activity of Medicago sativa and Satureja hortensis, in vitro.Materials and Methods: Ethanolic extract of Medicago sativa and Satureja hortensis were obtained by rotary evaporator. anti-Trichomonas vaginalis activities of the extracts in different concentrations were evaluated after 24, 48 and 72 hr of incubation of the cultured media.Results and Conclusions: The data showed a significant difference between concentration and time regarding the Satureja hortensis and Medicago sativa extracts compared to the negative control (p<0.05). According to the results, the anti- trichomonas activity of the Medicago sativa and Satureja hortensis extracts may make it possible to use them in the treatment of trichomoniasis.

Objective: Studies have focused on use of non-expired composites. Unfortunately some clinicians still use expired composite resins without considering their effects. The objective of this in vitro preliminary research was to investigate cytotoxicity of expired(6-months) and non-expired composite resins. Materials and methods: Expired (E) and non-expired (NE) samples of one bulk-fill (Tetric N-Ceram Bulk-fill [TNB], Ivoclar Vivadent), two nano-hybrid (Tetric N-Ceram [TN], Ivoclar Vivadent; Clearfil Majesty ES-2 [CM], Kuraray) composite resins were tested on L929 fibroblast cells. Medium covering cells was removed then plastic rings (2-mm height) were filled with non-polymerized composite resins, placed in direct contact with cells and polymerized with LED light curing unit (LCU). Three samples were prepared for each group. After polymerization, removed medium was added to the cells. Cells that were left without medium (WOM) and cells that were exposed to LCU were used as positive control groups. Cells without any treatment were used as negative control group (C). Cells were incubated with tested materials for 7-days to evaluate cytotoxicity. Cell viability was calculated by sulforhodamine B test as a percentage (%). One-way ANOVA and post-hoc Tukey tests were used for statistical analyses (p<0.05). Results: Comparison between E and NE groups of same composite resins did not result in statistically significant differences (p>0.05), except between TN NE and TN E (p<0.05). TN E group was significantly more cytotoxic than TN NE group. When NE composite resin groups were compared to each other, statistically significant difference was only obtained between TNB NE and TN NE (p<0.05). Among all tested groups, TN NE group showed the least cytotoxic profile. No statistically significant differences were determined when E composite resin groups were compared to each other (p>0.05). All experimental groups compared with C group showed statistically significant cytotoxicity (p<0.05). A statistically significant difference existed between LCU and C groups (p<0.05). Conclusions: In clinical practice, expired composite resins should never be used. Although a correlation was found between expiration dates of nano-hybrid composite resins and cell viability, opposite data were obtained for bulk-fill composite resin. Researches are still required to evaluate biocompatibility of bulk-fill composite resins at various thicknesses with current LCUs.
