Effect of Different Intermediary Bases on Microleakage of a Restorative Material in Class II Box Cavities of Primary Teeth

Introduction The aim of this in vitro study was to assess and compare the effect of different intermediary bases on microleakage between tooth and a nanocomposite interface in Class II box cavities in primary teeth. Methods Standard Class II box cavities were prepared in 52 primary molars and randomly divided into 9 groups according to the intermediary base used (Multicore Flow, Fuji II LC, SDR, Smart Dentin Replacement, and Biodentine). All specimens were subjected to thermocycling and prepared for microleakage testing and evaluation. Results There was significant difference in the mean ranks of microleakage between the 9 groups, which was observed in the gingival side (p<0.0001) and the occlusal side (p<0.0001). The mean ranks microleakage was significantly higher with experimental SDR, experimental Multicore Flow, and positive control materials when compared with the other 6 groups. The microleakage mean ranks were statistically significantly lower in experimental Fuji II LC, experimental Biodentine, and all negative control groups when compared with the other 3 groups. Conclusions Microleakage is affected by the application of intermediate material. Experimental Biodentine and Fuji II LC showed the lowest microleakage while experimental SDR and experimental Multicore Flow showed the highest microleakage.

ABSTRACT Objective: The purpose of this study was to evaluate the effect of different concentrations of platelet-rich plasma (PRP) on the proliferation of undifferentiated periodontal ligament (PDL) fibroblasts. Materials and Methods: The undifferentiated PDL fibroblasts were obtained from two healthy human premolar teeth and cultured in Dulbecco's modified Eagle's medium. Cell wells were divided into five groups. Experimental groups received 0.1%, 5%, or 50% PRP; the positive and negative control groups were cultured in fetal bovine serum (FBS) 12% and in a medium without FBS 12%, respectively. The plates were incubated at 37°C for 1, 2, 3, 4, and 7 days. PDL cell viability was assessed by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide proliferation assay. Statistical analysis of the data was accomplished using repeated measure ANOVA and Tukey's test. P < 0.05 was considered statistically significant. Results: The 5% PRP had the greatest effect on undifferentiated fibroblast proliferation, which was significant on the 3rd day. There was no significant difference between 0.1% PRP and positive control during the first 3 days. The group with 50% PRP presented significantly lower proliferation, compared to other experimental and control groups. Conclusions: It may be concluded that the growth-stimulating effect of PRP is dose dependent with the best results in low concentrations.

SUMMARY This in vitro study evaluated the effect of six surface treatments on the shear bond strength of three resin-modified glass ionomers (RMGIs) to dentin. Occlusal surfaces of caries-free third molars were reduced to expose only dentin. Surface treatments were smear layer intact (negative control), Cavity Conditioner, EDTA, Ketac Primer, Self Conditioner, and etching with 35% phosphoric acid followed by the application of Optibond Solo Plus. Filtek Z250 composite resin bonded with Optibond Solo Plus served as a positive control. Conditioning agents were used according to the manufacturers' instructions. After surface treatments, Fuji II LC, Riva LC, Ketac Nano, and Filtek Z250 were placed in copper-band matrices 5 mm in diameter and 2 mm in height and were light-cured for 20 seconds. Specimens were stored in 100% humidity for 24 hours, after which they were placed in deionized water for 24 hours at 37°C. They were then tested under shear forces in an Instron Universal Testing Machine at a crosshead speed of 0.5 mm/min. A two-way analysis of variance and Tukey honestly significant difference statistical analyses (p&lt;0.05) indicated significant interaction between RMGIs and conditioning agents. Acid etching followed by Optibond Solo Plus provided highest bond strengths for all three RMGIs, which were not statistically different from the positive control.

This in vitro study examined the margin integrity of sculptable and flowable bulk-fill resin composites in Class II cavities of primary molars. Standardized Class II cavities were prepared in human primary molars and restored with the following resin composite materials after application of a universal adhesive: a sculptable bulk-fill composite (Tetric EvoCeram Bulk Fill (TEC) or Admira Fusion x-tra (AFX)), a flowable bulk-fill composite (Venus Bulk Fill (VBF) or SDR), or a conventional composite (Filtek Supreme XTE (FS)). The bulk-fill materials were applied in 4 mm layers, while the conventional composite was applied in either 2 mm (FS2, positive control) or 4 mm layers (FS4, negative control). The specimens were exposed to thermo-mechanical loading (TML) in a computer-controlled masticator. A quantitative margin analysis was performed both before and after TML using scanning electron microscopy, and the percentage of continuous margins (margin integrity) was statistically analyzed (α = 0.05). All composites showed a significant decline in margin integrity after TML. AFX exhibited the significantly highest margin integrity of all materials after TML (97.5 ± 2.3%), followed by FS2 (79.2 ± 10.8%), TEC (73.0 ± 9.1%), and FS4 (71.3 ± 14.6%). SDR (43.6 ± 22.3%) and VBF (25.0 ± 8.5%) revealed the lowest margin integrity. In conclusion, the tested sculptable bulk-fill materials show similar or better margin integrity in primary molars than the conventional resin composite placed in 2 mm increments.

Aim: To analyze the remineralization efficacy of casein phosphopeptide–amorphous calcium phosphate (CPP–ACP) and 8% arginine on artificial early enamel lesions on primary teeth enamel samples. Materials and Methods: In this in vitro study, artificial enamel lesions were created in 120 primary teeth. Teeth were randomly divided into 5 groups: group C: CPP–ACP paste; group CF: CPP–ACPF paste with 900 ppm flour; group A: paste with 8% arginine; group K+: 500 ppm NaF as a positive control; and group K–: deionized water as a negative control. After 4 weeks of the remineralization process, the effect of remineralization on samples of agents used the microhardness, atomic force microscope, and scanning electron microscope analyses. Statistical analysis was done using Statistical Package for the Social Sciences (SPSS version 20.0, SPSS Inc., Chicago, IL, USA). ANOVA and Tukey Post-Hoc multiple comparison test were applied ( p < .05). Results: Increasing percentage values of the microhardness at groups C, CF, and A were significantly better than group K– ( p < .05), whereas there is no statistically significant difference with group K+ ( p > .05). The average surface roughness values of groups C and CF were similar with group K+ in AFM analysis, whereas average surface roughness values of group A were significantly higher than K+ group. Conclusion: The CPP–ACP, CPP–ACPF, and arginine are effective agents for remineralization of early childhood caries lesions.

Introduction: Thorough, chemo-mechanical preparation of the root canal is required to completely eliminate microorganisms from the dentinal tubules. Faulty obturation in the apical area is the pathway from which bacteria and their toxins pass on to the periapical area and this leakage can be studied by sealability studies. Aim: To evaluate the apical sealing ability of three sealers i.e., AH Plus, Roekoseal and Adseal using passive dye penetration test and a dye extraction test with Methylene blue dye to evaluate the apical leakage. Materials and Methods: The present study was an experimental in-vitro study, carried out at MGM Dental College and Hospital, Navi Mumbai, Maharashtra, India over a period of 10 months (from August 2013 to May 2014). Eighty freshly extracted human permanent maxillary central incisor teeth were obtained. The crowns of the teeth were sectioned at/below cemento-enamel junction and divided into five groups: Groups I, II and III were obturated with gutta-percha and three different sealers, and groups IV and V were the two control groups. Passive dye penetration test was performed to evaluate penetration of the dye by means of capillarity followed by dye extraction test. At the end of dye extraction procedure, the supernatant solution obtained was analysed using an Ultraviolet spectrophotometer. Significance was set at 5%. Statistical analysis was performed by using IBM Statistical Package for the Social Sciences (SPSS) statistical software version 20 (Armonk, New York, United States). The normality of data in each group was tested using Shapiro-Wilk test. The mean absorbance values of the five groups were compared using one-way ANOVA. Further to compare the level of absorbance of dye among various groups, Tukey’s post-hoc test was applied. Results: The mean values of dye absorbance of all groups were as follows, AH Plus (0.02638), Roekoseal (0.06713), Adseal (0.03575), positive control group (1.36250) and negative control group (0.01894). One-way ANOVA showed p<0.001, thus indicating there was a significant difference between the dye absorbance values of all the groups. Tukey’s post-hoc test showed statistically significant difference in dye absorbance of the experimental groups when compared to positive control group. There was no significant difference in dye absorbance when experimental groups were compared to each other. When AH Plus was compared with Roekoseal, the p-value was 0.568 and 0.997 with Adseal whereas when Roekoseal was compared with Adseal the p-value was 0.778. Conclusion: The mean dye absorbance value of AH Plus was found to be least and that of Roekoseal was highest in the experimental groups. AH Plus gave the best results, as least dye was extracted from this group, indicating least dye penetration and minimum microleakage followed by Adseal and Roekoseal.

The aim of this study was to evaluate and compare the sealing ability of two calcium silicate-based sealers (TotalFill BC RRM Fast Set Putty and White ProRoot MTA) when used as apical plugs in immature teeth through nuclear medicine. Single-rooted extracted teeth (n = 34) had their crowns and root tip sectioned to obtain 14 mm long root segments to simulate an in vitro apexification model. Were created two experimental groups, namely MTA (n = 12) and BC (n = 12), and two control groups, PG (positive group, n = 5) and NG (negative group, n = 5). On the 4th day after placing the respective apical plug, the apical portions of the teeth were submerged in a solution of sodium pertechnetate (99mTcNaO4) for 3 h. Statistical analysis showed a significant difference between the MTA group and the controls (p < 0.05). The BC group had a significant difference regarding the negative control (p < 0.001) but showed no statistical significance regarding the positive control (p = 0.168). There was a statistically significant difference (p = 0.009) between the BC group (7335.8 ± 2755.5) and the MTA group (4059.1 ± 1231.1), where the last showed less infiltration. Within the limitations of this study, White ProRoot MTA had a significantly better sealing ability than TotalFill BC RRM Fast Set Putty.

Shallot (Allium ascalonicum L.) has been known as traditional medicine. Shallot which has same genus with garlic(Allium sativum L.) contains allicin that is also found in garlic and has been suspected has fungicidal activity toCandida albicans. It is supported by several researches. Therefore, shallot is suspected has antifungal activity too.The aim of this research was to know antifungal activity of shallot’s water extortion againsts Candida albicans invitro. This was a laboratory experimental research which used completely randomized design, with diffusion method.Shallot’s water extortion was devided into three concentrations, there were 50%, 100% and 200%. Ketoconazole 2%was positive control and aquadest was negative control. The result of this research based on analysis of varians(Anova), there was significant difference between several treatments and was confirmed with Duncan New MultipleRange Test (DNMRT) p<0,05, there was significant difference between 100% shallot’s water extortion with othertreatments, but there was no significant difference between 50% shallot’s water extortion with 200% shallot’s. Theconclusion was shallot’s water extortion had antifungal activity againsts Candida albicans with the best concentration100%, but it was lower than ketoconazole 2%.

Abstract Objective This study evaluated four types of pH adjustment of the coconut water (CW) on viability of human fibroblasts (HFF). Material and method Natural and industrialized CW were adjusted to pH 7.0 using: (1) Sodium Hidroxide (NaOH), (2) Sodium bicarbonate (NaHCO3), (3) Triethanolamine (C6H15NO3), (4) 2-Amino-2-Methil-1-Propanol (C4H11NO). Fibroblasts were plated at 2×104/ well in 96 well plates and maintained in the CW solutions for 2 h and 4 h. Positive control was represented by HFF maintained in DMEM and the negative control by tap water. Cell viability was analyzed by MTT formazan method. Data were analyzed by 3-way ANOVA followed by Tukey’s and Dunnet’s test. Result There are no significant effect on the cell viability regarding type of CW, period of evaluation, and the interactions between CW and period of evaluation, CW and pH adjustment method, pH adjustment method and period of evaluation (p>0.05). Conclusion The product used for CW pH adjustment did not influenced HFF viability, thought there are a tendency of better performance in natural CW.

<p><strong>ABSTRACT</strong></p><p><strong>Background</strong>; In Indonesia, most diarrhea disease in 1995 to 2001 are caused by Shigella spp. Shigella spp infection can cause various symptom dan complication. Generally, the treatment by using antibiotic can cause antibiotic resistance. Sea cucumber (Holoturia scabra) is an herb that known, available, and easy to consume by society and has an antibacterial effect. Therefore, further research to study the effect of Holoturia Scabra on <em>Shigella Dysentriae</em> growth in vitro is needed.</p><p><strong>Objectives</strong>: The goal of this research is demonstrate the effect of sea cucumber (Holoturia scabra) to the growth of the <em>Shigella dysentriae</em> bacteria in vitro.</p><p><strong>Method</strong>: The method in this research is Posttest Only Control Group. There are 6 groups, 4 types of and 2 control groups. The concentration of the treatment group is 100%,50%, 25%, and, 12.5% while for positive control tests using chloramphenicol and aquadest as a negative control.</p><p><strong>Result</strong>: The result showed there is an influence on the intake of sand cucumber to the growth of the Shigella dysenteriae.</p><p><strong>Conclusion</strong>: Sea cucumber (<em>Holoturia scabra</em>) inhibit the growth of <em>Shigella dysenteriae</em>.</p><p><strong>Key words</strong>: <em>Shigella dysenteriae</em>, sea cucumber (<em>Holoturia scabra</em>), antibacterial</p>

Recurrent caries is still considered the main reason restorations need to be replaced. There are different materials available now that promise to reduce the possibility of recurrent caries by releasing fluoride and inhibiting restoration marginal caries. The purpose of this in vitro study was to evaluate the demineralization inhibition potential of a non-fluoride-releasing resin (Z100TM 3M, St. Paul, MN, USA) and a glass containing resin-based composite (Tetric EvoCeram Bulk Fill, Ivoclar/Vivadent AG, Schaan, Liechtenstein), which contains fluoride. Class V preparations were placed on 22 premolars; the gingival margin was below the cementoenamel junction and the occlusal margin was placed above the cemento-enamel junction. Ten teeth were randomly selected to be restored with Z100 while the other 10 were restored with Tetric EvoCeram Bulk Fill. Both groups were restored following manufacturer’s instructions. All teeth had an acid resistant varnish placed within one millimeter of the preparation margins. Both groups were placed in artificial caries challenge solution (pH 4.4). At the end of the 4 days; 100 µm buccolingual sections were obtained for each tooth; these were photographed under polarized light microscopy and the demineralized areas adjacent to the restorations were measured and quantified. The mean (±S.D.) area (µm2) of demineralization from the occlusal margin (enamel) and dentin margin were: Z100 2781.889 ± 1045.213; 3960.455 ± 705.964 and for Tetric EvoCeram Bulk Fill 1541.545 ± 1167.027; 3027.600 ± 512.078. Student’s t-test indicated that there was significantly less enamel and dentin demineralization adjacent to Tetric EvoCeram Bulk Fill compared to Z100; there was significantly less demineralization in enamel compared to dentin in both Tetric EvoCeral Bulk Fill and Z100. Tetric EvoCeram Bulk Fill performed better inhibiting demineralization at restoration margins when compared to Z100 and provided better demineralization inhibition in enamel than cementum/dentin.
