Treatment of inflammatory diseases of the uterine appendages with turpentine. Fuchs (Zentr. T. Gyn., 1920, No. 4), Hinze (ib., No. 28) and Sonnenfeld (Berl. Klin. Woch., 1920, No. 30) observed very good results from the treatment of salpingo-ophoritis with intramuscular injections of a mixture of turpentine with olive oil in a ratio of 1: 4.

Inflammation of the adipose tissue plays an important role in the development of several chronic diseases associated with obesity. Polyphenols of extra virgin olive oil (EVOO), such as the secoiridoids oleocanthal (OC) and oleacein (OA), have many nutraceutical proprieties. However, their roles in obesity-associated adipocyte inflammation, the NF-κB pathway and related sub-networks have not been fully elucidated. Here, we investigated impact of OC and OA on the activation of NF-κB and the expression of molecules associated with inflammatory and dysmetabolic responses. To this aim, fully differentiated Simpson-Golabi-Behmel syndrome (SGBS) adipocytes were pre-treated with OC or OA before stimulation with TNF-α. EVOO polyphenols significantly reduced the expression of genes implicated in adipocyte inflammation (IL-1β, COX-2), angiogenesis (VEGF/KDR, MMP-2), oxidative stress (NADPH oxidase), antioxidant enzymes (SOD and GPX), leukocytes chemotaxis and infiltration (MCP-1, CXCL-10, MCS-F), and improved the expression of the anti-inflammatory/metabolic effector PPARγ. Accordingly, miR-155-5p, miR-34a-5p and let-7c-5p, tightly connected with the NF-κB pathway, were deregulated by TNF-α in both cells and exosomes. The miRNA modulation and NF-κB activation by TNF-α was significantly counteracted by EVOO polyphenols. Computational studies suggested a potential direct interaction between OC and NF-κB at the basis of its activity. This study demonstrates that OC and OA counteract adipocyte inflammation attenuating NF-κB activation. Therefore, these compounds could be novel dietary tools for the prevention of inflammatory diseases associated with obesity.

Diet plays a key role in the maintenance and optimal functioning of immune cells. The Mediterranean dietary pattern is an example of a prudent choice of lifestyle and scientifically accepted to help preserve human health by protecting against major chronic and inflammatory diseases. Mediterranean diets (MedDiets) are characteristically high in the consumption of fruits, vegetables and salad, bread and whole grain cereals, potatoes, legumes/beans, nuts, and seeds. Their common central feature is the usage of olive oil as the main source of fat. The health benefits attributed to olive oil are specifically related to extra virgin olive oil (EVOO) intake with its high nutritional quality and multiple positive effects on health. Overall, MedDiets have direct (mono-unsaturated fatty acids (MUFAs), tocopherols, polyphenols) and indirect (low saturated fats, well-balanced linoleic/alpha linolenic acid) effects on the immune system and inflammatory responses. In the present paper, we summarize the current knowledge on the effect of olive oil per se and MedDiets generally on immune-mediated and inflammatory diseases, such as coronary heart disease (CHD)/cardiovascular diseases (CVD), obesity, type-2 diabetes, cancer, asthma, and allergies.

Curcumin is a traditionally used spice with a potential to treat various inflammatory diseases including arthritis. This study was aimed at observing curcumin’s effects on the histopathological progression and interleukin-1β (IL-1β) levels in collageninduced arthritis (CIA). Thirty male Sprague-Dawley rats (150+50 g) were divided into five random groups. A group was assigned as the normal control (CTRL), while the remaining were subcutaneously immunised with 150 μg of collagen emulsion on day 0. CTRL and CIA-Curcumin-d0 groups were supplemented daily with olive oil (1 ml/kg) and curcumin (110 mg/ml/kg) from day 0, respectively. The CIA-OV (negative control), CIA-Beta and CIA-Curcumin-d14 groups were given daily supplementation of olive oil (1 ml/kg), Betamethasone (0.5 mg/ml/kg), and curcumin (110 mg/ml/kg) from day 14, respectively. The daily oral supplementations continued until day 42. The study showed that CIA-Beta (**P<0.05) and CIACurcumin- d0 (**P=0.01) groups had significantly lower mean histological scores compared to CIA-OV, respectively. Serum IL-1β levels for CIA-Beta and CIACurcumin- d0 were not significantly raised on day 42 as to compared to day 0, and the mean increment of IL-1β levels from day 0 to day 42 were significantly lower (**P≤0.01) for all the CIA groups compared to CIA-OV. There was no significant difference in both mean histological scores and IL-1β levels of CIA-Curcumin-d0 compared to CIA-Beta. Early supplementation of curcumin could potentially minimise disease progression of CIA in rats.

Recent studies have shown that dietary patterns confer protection from certain chronic diseases related to oxidative stress, the immune system and chronic low-grade inflammatory diseases. The aim of this study was to evaluate the anti-inflammatory potential and the capacity to attenuate cartilage degradation using extra-virgin olive oil–derived polyphenols for the treatment of osteoarthritis. Results show that both nutraceuticals ligstroside aglycone and acetylated ligstroside aglycone showed an anti-inflammatory profile. Acetylated ligstroside aglycone significantly reduced the expression of pro-inflammatory genes including NOS2 and MMP13 at both RNA and protein levels; decreased nitric oxide release; and, importantly, reduced proteoglycan loss in human osteoarthritis cartilage explants. Our study demonstrated that a new synthetic acetylated ligstroside aglycone derivative offers enhanced anti-inflammatory profile than the natural nutraceutical compound in osteoarthritis. These results substantiate the role of nutraceuticals in osteoarthritis with implications for therapeutic intervention and our understanding of osteoarthritis pathophysiology.
