Eighteen commercial taxa in genera Liriope and Ophiopogon were evaluated genetically and morphologically. Three hundred and forty-four Amplified Fragment Length Polymorphism (AFLP) markers were identified and used in the analysis. Relatedness among taxa was estimated by analyzing the AFLP data using the Dice coefficient of similarity and UPGMA methods. The molecular analysis revealed relatively no genetic differentiation among the taxa ascribed to their respective genera, Liriope and Ophiopogon, based on morphological assessment of floral structure. Greater than 95% of the total genetic variability present was attributed to within group effects using various grouping strategies among the taxa. Among group effects were correspondingly low and not significant (P ≤ 0.05). Results suggest an indistinguishable genetic relationship may exist in the tribe Ophiopogonae between what is currently ascribed as two distinct genera.

Genetic variability and population structure of Cercospora sorghi from wild and cultivated sorghum were investigated to gain insight into their potential impact on epidemics of gray leaf spot of sorghum in Africa. Population structure was examined using data derived from amplified fragment length polymorphism (AFLP) of C. sorghi by Nei's test for population differentiation, GST, and analysis of molecular variation (AMOVA). Two ecological populations of C. sorghi in Uganda were devoid of population structure (GST = 0.03, фFST = 0.01, P = 0.291). AMOVA revealed that genetic variability was due mainly to variations within (99%) rather than between (0.35%) populations, and Nei's genetic distance between the two populations was 0.014. Phenetic analysis based on AFLP data and polymerase chain reaction-restriction fragment length polymorphism analyses of the internal transcribed spacer regions of rDNA and mitochondrial small subunit rDNA separated Cercospora cereal pathogens from dicot pathogens but did not differentiate among C. sorghi isolates from wild and cultivated sorghum. Our results indicate that Ugandan populations of C. sorghi compose one epidemiological unit and suggest that wild sorghum, while not affecting genetic variability of the pathogen population, provides an alternative host for generating additional inoculum.

The extent and pattern of genetic differentiation between two naturally occurring phenotypes, grey–green leaf (GGL) and yellow–green leaf (YGL), of Leymus chinensis (Trin.) Tzvel., which colonize distinct habitats in the Songnen Prairie in northeast China, were investigated by amplified fragment length polymorphism (AFLP) analysis. Twelve selected AFLP primer pairs amplified 593 reproducible bands, of which 148 (24.96%) were polymorphic among 69 individuals taken from three populations: two natural ones (YGL and GGL1) and one transplanted (GGL2). Cluster analysis based on the AFLP data categorized the plants into distinct groups that are in line with their phenotypes and population origins, thus denoting clear genetic differentiation between the two phenotypes. This, together with their adaptation to contrasting natural habitats, suggests that the two phenotypes probably represent stabilized ecotypes. The grouping was supported by multiple statistical analyses including Mantel’s test, principal coordinate analysis (PCOORDA), and analysis of molecular variance (AMOVA). The GGL phenotype harbors a higher level of within-population genetic diversity than YGL, possibly reflecting selection by habitat heterogeneity. Although GGL2 is largely similar to its original population (GGL1), further diversification since transplantation was evident. Sequence analysis of a subset of phenotype-specific or phenotype-enriched AFLP bands implicated diverse biological functions being involved in ecological adaptation and formation of the two phenotypes.

Analysis of secondary molecular polymorphism at the level of amplified mitochondrial DNA, digested by restriction enzymes (CAPS markers), revealed genetic differentiation among ten Phytophthora infestans accessions. Mitochondrial DNA was amplified with four pairs of primers (H1,H2,H3 and H4) and after that the products of amplification were digested with Hha I (for H1 products of amplification), Msp I (for H2 products of amplification) and Eco R I (for H3 and H4 products of amplification) restriction enzymes. On this basis we have established that the ten Phytophthora infestans accessions belongs to two haplotypes, one accession belongs to Ia haplotyp and the others belong to IIa haplotype.

SummaryThe camel population in India is facing a severe decline which demands that immediate steps are taken to ensure its conservation. Characterisation is an integral part of the conservation program. The Polymerase Chain Reaction-Randomly Amplified Polymorphic DNA profile of unrelated camels of the Bikaneri (29), Jaisalmeri (30) and Kachchhi (18) breeds were analyzed. Reproducible polymorphic bands with varying frequencies among the three breeds of camel were obtained with five oligonucleotide primers. A total of 75 bands were amplified, of which 27 (36%) were polymorphic. The probability of obtaining identical fingerprints was observed to be the lowest in primer GC-10 (5.7%) followed by OP-08 (8.7%), GT-10 (11.3%), G-2 (15.5%) and G-1 (80%). Breed informative bands were amplified. The maximum genetic variability was observed in the Bikaneri (0.80±0.05) followed by the Kachchhi (0.84±0.06) and the Jaisalmeri (0.87±0.05) breeds. The inter-breed genetic distance estimates indicated a closer relationship in the Bikaneri-Kachchhi camels, (0.075), followed by the Jaisalmeri-Kachchhi (0.106) and Bikaneri-Jaisalmeri (0.132) breeds. A similar genetic relationship was observed when the degree of population subdivision was measured between the Bikaneri-Kachchhi (0.529), Jaisalmeri-Kachchhi (0.558) and Bikaneri-Jaisalmeri (0.566) breeds.

Amplified fragment length polymorphism (AFLP) using three primer sets was used to characterize 211 Colletotrichum coccodes isolates from North America, 112 of which were assigned to six vegetative compatibility groups (VCGs) using nitrate nonutilizing (nit) mutants. These isolates clustered into five corresponding groups by unweighted pairgroup method with arithmetic means-based cluster analysis of AFLP banding patterns. Isolates of C. coccodes belonging to NA-VCG1 and NA-VCG3 were closely related, as were isolates belonging to NA-VCG2 and NA-VCG5. Based on bootstrap analysis of AFLP data, the two isolates originally assigned to NA-VCG4 clustered with isolates belonging to NA-VCG2 and NA-VCG5. C. coccodes isolates that clustered with two isolates belonging to NA-VCG6 were the most diverged from other groups, including seven isolates collected from hosts other than potato. As opposed to the bootstrap analysis, a quadratic discriminant analysis (QDA) of AFLP data correctly categorized the two isolates of NA-VCG4. Furthermore, in isolates where VCG determinations had been made, this model correctly classified isolates of all VCGs. QDA classifications were identical to those made by the bootstrap analysis, with the exception of VCG4. Overall, classifications made by the QDA model were strongly correlated (r = 0.970, P < 0.001) to the VCGs assigned by traditional methods. All 99 C. coccodes isolates evaluated only by AFLP also were subjected to QDA, leading to the assignment of a presumptive VCG for each isolate. No isolates of VCG4 or VCG6 were identified by QDA within this population. Symptoms of black dot developed in plants inoculated with isolates collected from both potato and non-potato hosts. However, total yield was not significantly reduced by infection with non-potato isolates. The lack of any additional groups identified by AFLP analysis may be an indicator of a limited level of genetic variation among North American C. coccodes isolates. AFLP is a much more efficient technique for subspecific characterization in C. coccodes than VCG analysis utilizing nit mutants and will provide an effective means by which the population biology of this pathogen can be further investigated worldwide.

AbstractAnthyllis vulneraria L. (Fabaceae) s. lato includes many cryptic taxa, ranging from 25 to 60 subspecies according to different authors. The delimitation of intraspecific taxa of A. vulneraria s. lato has always been complicated and inconsistent. Different data sets (multivariate analyses of morphological variation, allozymes, chloroplast SSRs and ITS) have not resolved the existing problem with distinguishing some subspecies. We used the amplified fragment length polymorphism (AFLP) analysis to describe the differentiation in this species complex and to characterize variation on a geographic scale. Some correlation was found between genetic variability and geographic distribution (western-eastern directional variation), but AFLP data analysis did not reveal clear intraspecific structure of the seven analysed taxa. The analysed specimens did not comprise groups correlated with the subspecies.

Germplasm accessions of wildLactucaspecies are maintained worldwide inex situcollections as gene reservoirs for quality and disease resistance traits for cultivated lettuce. Accessions of 12Lactucaspecies from 6 genebanks were compared via morphological characterization and AFLP (Amplified Fragment Length Polymorphism)-based profiling to estimate the extent of duplication. A method of assessing redundancy within very similar, but not identical accessions, is proposed, based on 352 polymorphic AFLP products. Seven duplication groups showed a high level of AFLP similarity, and one pair ofLactuca salignaaccessions displayed identical AFLP profiles. In several cases, the morphological assessment indicated that a taxonomic reclassification of accessions was necessary. Candidate duplicates were identified using population parameters and inter- and intra-accession variability. The implications of these findings on the conservation of wild species are discussed.

Cendana merupakan spesies asli Indonesia bagian timur, yang menjadi ras lahan di Gunung Sewu. Sembilan ras lahan di Gunung Sewu, seluruhnya tersusun atas tiga varian yang berbeda struktur dan warna bunganya (MK, merah kecil; MB, merah besar; dan KB, kuning besar). Penelitian ini bertujuan untuk menganalisis karakter pembungaan ketiga varian cendana di Gunung Sewu pada periode pembungaan tahun 2015; di tiga ras lahan yang mewakili tiap zona yaitu Nglanggeran (Zona Utara), Bleberan (Zona Tengah), dan Petir (Zona Selatan). Observasi meliputi fenologi, kemasakan organ reproduksi, display, dan struktur pembungaan. Struktur pembungaan dan longevity organ reproduksi berbeda antar varian. MB dan KB didominasi warna merah dan marun, perigonium lebih besar, organ reproduksi lebih panjang, posisi putik sejajar/lebih rendah dari benang sari, dan longevity lebih singkat. MK didominasi kuning hingga oranye, lebih kecil, posisi putik sejajar/lebih tinggi, dan longevity lebih lama. Display pembungaan berbeda antar ras lahan, namun tidak berbeda antar varian. Petir, dengan ukuran populasi terbesar, memproduksi paling banyak pohon berbunga dan total bunga untuk seluruh varian. Ras lahan dan varian berbunga dua kali setahun, namun dengan inisiasi dan durasi yang berbeda. KB berbunga lebih awal di seluruh ras lahan, sedangkan MB paling akhir. MK memiliki durasi pembungaan terpanjang. Perbedaan antar ras lahan lebih dipengaruhi oleh faktor lingkungan seperti ketinggian, jenis tanah, dan iklim. Ras lahan yang lebih rendah, kering, dan panas berbunga lebih awal dan singkat. Peningkatan temperatur dan turunnya status air tanah menyebabkan keguguran massal yang diikuti sequential replacement, mengakibatkan perubahan frekuensi pembungaan. Strategi konservasi cendana di Gunung Sewu perlu dirancang berbasis keragaman genetik, sinkroni pembungaan, dan kompatibilitas antar varian, dengan tetap mempertimbangkan keberagaman tempat tumbuhnya.Kata kunci: cendana; Gunung Sewu; pembungaan; ras lahan; varian bunga The Flowering Characters Variation Among Floral Variants and Landraces Along Geographical Gradients in Gunung SewuAbstractSandalwood was origin to the south-eastern islands of Indonesia, but is recently occured as new land-races in Gunung Sewu, Java island. All of land-races consisted of three floral variants (MK, refers to “merah kecil”; MB, “merah besar”; and KB, “kuning besar”, respectively). This research aimed to determine flowering of these variants among three of land-races which were representing geographical zone in Gunung Sewu: Nglanggeran, Blebera, and Petir. Observation on phenology, sexual organs maturity, floral structure, and display was conducted in 2015. Floral structures varied among variants. MB and KB dominated by red and maroon colors, possessed bigger perigonium, longer sexual organs but with shorter longevity, and similar/lower position of stylus to stamens. MK dominated by yellow to orange colors, smaller, similar/higher position of stylus, and longer longevity. Floral display varied among land-races but were similar among variants. Petir, with biggest population size, produced the most abundant flowers and flowering trees at all variants. Sandalwood flowered twice a year in all of land-races and variants, however, the onset and duration varied. KB flowered earliest while MB was the latest. MK possessed longest flowering period. Flowering varied among land-races due to the altitude, soils, and climatic differences. Lower altitude, drier, and warmer sites flowered earlier and shorter. Mass abortion and sequential replacement, resulting in the alteration on flowering frequency, occured due to the extreme temperature increment. Conservation strategy should be arranged based on genetic differentiation, flowering differences, and crossing abilities among variants. Differences of landscapes, which may contribute to the differences of environmental conditions and flowering processes, should also be considered.

Several subspecies of cutthroat trout, Salmo clarki, including the Snake River, Yellowstone, and Colorado cutthroat, are native to Wyoming. Because these fish have evolved in independent drainages their morphological and ecological characteristics have diverged significantly. Based upon these kinds of differences Behnke (1971, 1972) has identified two subspecies of cutthroat trout in the Snake River drainage in Wyoming. Recent fish taxonomic studies, however, also employ electrophoretic techniques to measure the extent of genetic divergence between suspected species or subspecies populations. The amount and kind of genetic variability and the extent of genetic divergence between Wyoming's cutthroat trout populations is unknown.
