AbstractThe strain SARS-CoV-2, newly emerged in late 2019, has been identified as the cause of COVID-19 and the pandemic declared by WHO in early 2020. Although lipids have been shown to possess antiviral efficacy, little is currently known about lipid compounds with anti-SARS-CoV-2 binding and entry properties. To address this issue, we screened, overall, 17 polyunsaturated fatty acids, monounsaturated fatty acids and saturated fatty acids, as wells as lipid-soluble vitamins. In performing target-based ligand screening utilizing the RBD-SARS-CoV-2 sequence, we observed that polyunsaturated fatty acids most effectively interfere with binding to hACE2, the receptor for SARS-CoV-2. Using a spike protein pseudo-virus, we also found that linolenic acid and eicosapentaenoic acid significantly block the entry of SARS-CoV-2. In addition, eicosapentaenoic acid showed higher efficacy than linolenic acid in reducing activity of TMPRSS2 and cathepsin L proteases, but neither of the fatty acids affected their expression at the protein level. Also, neither reduction of hACE2 activity nor binding to the hACE2 receptor upon treatment with these two fatty acids was observed. Although further in vivo experiments are warranted to validate the current findings, our study provides a new insight into the role of lipids as antiviral compounds against the SARS-CoV-2 strain.

If the otoliths of fish are to be used as an ageing tool, they must fulfil three criteria: they must display an internal structure of increments, this structure must be relatable to a regular time scale, and the otoliths must grow throughout the lives of the fish at a perceptible rate. These criteria were assessed for two species of damselfish: Pomacentrus moluccensis and P. wardi from One Tree Reef on the southern Great Barrier Reef. Transverse sections of sagittae from both species contained an alternating sequence of opaque and translucent zones that formed an interpretable pattern of increments. Treatment of fish with tetracycline showed that the opaque zones were formed on an annual basis, generally between spring and early summer. The assessment of growth of otoliths from fish treated with tetracycline indicated that those as old as 10 years of age were still laying down increments of regular thickness. Consequently, the otoliths from both species successfully fulfilled the three criteria required for the direct determination of age. Furthermore, this study helped to identify two levels of variation in otolith structure, manifested in the clarity and interpretability of the increments; there was an interspecies difference and a significant intraspecies latitudinal difference.

Abstract. Determining reliable proxies for the ionospheric signature of the open-closed field line boundary (OCB) is crucial for making accurate ionospheric measurements of many magnetospheric processes (e.g. magnetic reconnection). This study compares the latitudes of Spectral Width Boundaries (SWBs), identified in the morning sector ionosphere using the Super Dual Auroral Radar Network (SuperDARN), with Particle Precipitation Boundaries (PPBs) determined using the low-altitude Defense Meteorological Satellite Program (DMSP) spacecraft, in order to determine whether the SWB represents a good proxy for the ionospheric projection of the OCB. The latitudes of SWBs and PPBs were identified using automated algorithms applied to 5 years (1997-2001) of data measured in the 00:00-12:00 Magnetic Local Time (MLT) range. A latitudinal difference was measured between each PPB and the nearest SWB within a ±10min Universal Time (UT) window and within a ±1h MLT window. The results show that the SWB represents a good proxy for the OCB close to midnight (~00:00-02:00 MLT) and noon (~08:00-12:00 MLT), but is located some distance (~2°-4°) equatorward of the OCB across much of the morning sector ionosphere (~02:00-08:00 MLT). On the basis of this and other studies we deduce that the SWB is correlated with the poleward boundary of auroral emissions in the Lyman-Birge-Hopfield ``Long" (LBHL) UV emission range and hence, that spectral width is inversely correlated with the energy flux of precipitating electrons. We further conclude that the combination of two factors may explain the spatial distribution of spectral width values in the polar ionospheres. The small-scale structure of the convection electric field leads to an enhancement in spectral width in regions close to the OCB, whereas increases in ionospheric conductivity (relating to the level of incident electron energy flux) lead to a reduction in spectral width in regions just equatorward of the OCB.

Omics technologies hold great potential as the basis for development of the new diagnostic approaches in obstetrics. Cervicovaginal fluid (CVF) as part of the mother-placenta-fetus system can be used to diagnose obstetric complications. This study aimed to identify the features of lipid composition of the cervical canal secretion peculiar to Intrauterine Growth Restriction (IUGR) and preeclampsia (PE). We took CVF samples from 57 pregnant women and subjected them to an in-depth clinical-anamnestic and mass-spectrometric analysis. Lipid extracts of CVF were analyzed with a liquid chromatography system coupled with a mass analyzer. As a result, we identified 239 lipid compounds. In case of 17 lipids, mathematical analysis revealed significant differences between samples from women with normal pregnancy indicator values (normal group) and patients from the IUGR group (p < 0.05). As for the normal group and PE group patients, there were significant differences identified for 3 lipids (p < 0.05). Comparison of samples from the PE and IUGR groups yielded statistically significant differences in levels of two lipids (p < 0.05). Mainly, the lipids were oxylipins, sphingomyelins, triglycerides, and cardiolipins. The developed diagnostic model had the sensitivity of 0.81 and specificity of 0.91 (cut-off level — 0.50; AUC — 0.85). The data obtained are valuable in the context of development of the new methods of diagnosing placentaassociated complications of pregnancy and for understanding new mechanisms of pathogenesis of these complications.
