Biodiversitas 18: 1377-1384. Fusarium oxysporum is a soil borne pathogenic fungus that causes wilt disease in members of the family Solanaceae including the eggplant (Solanum melongena L.). One approach to resolving the problem of wilt disease in eggplant is to find endophytic microbes with antagonistic activity against F. oxysporum. The study reported here aimed to isolate such endophytic fungal antagonists from growing eggplants, to determine their antagonistic mechanisms, and to identify them. Samples of pathogenic fungi from diseased plants, assumed to be F. oxysporum, were obtained from the Laboratory of Plant Pests and Diseases of the Faculty of Agriculture, Universitas Sebelas Maret Surakarta. These were used to evaluate the antagonistic potential of endophytic fungi obtained from healthy eggplants in Dawung Village, Matesih, Karanganyar, Central Java. Specimens of various plant parts were collected from the healthy eggplants. The surfaces of these samples were sterilized for four minutes to remove contaminants, and then crushed excisions were cultured on a potato dextrose agar (PDA) medium. Antagonistic tests between endophytic and pathogenic fungi used the agar plug diffusion technique. Identification of fungi isolates was carried out on the basis of morphological characteristics. Six endophytic fungi isolated had antagonist activity against F. oxysporum. The antagonistic mechanism of FEB1, FEB2, FEB5 and FED1 was competition; FED2 was antibiosis, and FED3 was parasitism. Based on their morphological characteristics, FEB2, FEB5 and FED3 were identified as Helicomyces spp.; FEB1 was a Rhizopus sp.; FED1 was a Mucor sp.; and FED2 was a species of Penicillium.

Aims: The present study was carried out to determine the diversity of endophytic fungi that colonize the leaves of Psidium guajava, and to evaluate their antagonistic activity against Fusarium oxysporum f.sp. cubense and Mycosphaerella fijiensis which are the two main phytopathogens of banana plants. Place and Duration of Study: The research was carried out at Microbiology Laboratory, Faculty of Sciences, University of Yaoundé I and Microbiology Laboratory, Faculty of Biotechnologies, University of Agronomic Sciences and Veterinary Medicine Bucharest, between April 2018 and February 2020. Methodology: Fragments of surface sterilized leaves of Psidium guajava were inoculated on Potato Dextrose Agar supplemented with chloramphenicol. The isolated and purified endophytic fungi were identified based on their macroscopic and microscopic characters using a mycological atlas as guide. The non-sporulating isolates were identified by comparing the ITS regions of their DNA to those of known fungi registered in the GenBank database. The antagonistic activity of the endophytic fungi isolated against Fusarium oxysporum and Mycosphaerella fijiensis was screened using dual culture method. Results: A total of 28 endophytic fungal were isolated from the leaves of Psidium guajava corresponding to a colonization frequency of 33.33%. These isolates were identified as: Aspergillus sp., Botryosphaeria sp., Fusarium sp., Neoscytalidium sp., Xylaria sp., Phyllosticta capitalensis, Cercospora apii, Xylaria longipes, Phomopsis sp., Phomopsis asparagi, Aspergillus versicolor, Pallidocercospora thailandica, and Xylaria grammica that belonged to the Deuteromycota and Ascomycota divisions. These endophytic fungi inhibited the growth of Fusarium oxysporum f.sp. cubense and Mycosphaerella fijiensis with the percentage inhibition varying respectively from 23.25% to 73.52% and from 21.36% to 100%. The species Botryosphaeria sp., Phomopsis sp., Phomopsis asparagi, and Xylaria longipes exhibited the greatest activity. Conclusion: The leaves of Psidium guajava have a fairly varied diversity of endophytic fungi. These endophytic fungi can serve as potential biological control agents against Panama and Sigatoka diseases of banana and also would produce secondary metabolites with antifungal properties.

Watermelon is an important summer crop in Palestine, for several decades filling the needs of local market and some Arab countries. The yield of watermelon decreased dramatically in recent years due to severe infections with the soil borne fungal pathogen Fusarium oxysporum f. sp. niveum (FON). Soil fumigation with methyl bromide was commonly applied by Palestinian farmers until it was recently legally banned. Different control mechanisms were not feasible to overcome problems caused by the disease resulting in decreased watermelon cultivation in Palestine for the past 30 years. In this work, we have experimentally shown that Pseudomonas fluorescens was efficient in controlling FON infection and allowing normal seedling growth of both the root and shoot systems. Field experiments are necessary to further confirm the efficacy of biocontrol application.

This study was done to evaluate the efficiency of non-pathogenic Fusarium oxysporum isolates (141/3, 233, 233/1, 245, 245/1, 251, 251/2, 251/5, and 257) in controlling vascular wilt caused by F. oxysporum f. sp. lycopersici, race 2 (isolates C-21A, TO11, and TO245) in tomato (Lycopersicon esculentum) cv. Viradoro seedlings. In order to determine the effect of non-pathogenic F. oxysporum isolates in tomato plants, the root system of 30-day-old seedlings was immersed in conidial suspensions (10(6) ml-1) of each isolate and the seedlings were transplanted to a cultivation substrate. Thirty-five days after transplanting it was observed that the non-pathogenic F. oxysporum isolates were not pathogenic to the cv. Viradoro nor did they affect seedling development. The efficiency of the non-pathogenic F. oxysporum isolates in controlling Fusarium wilt was determined by immersing the tomato seedling roots in the conidial suspension (10(6) ml-1) of each isolate and then transplanting them into substrates previously infested with isolates of F. oxysporum f.sp. lycopersici, race 2 (10(5) conidia ml-1 of substrate). Evaluations were performed 35 days after transplanting, for severity in scale with 1=healthy plant to 6=dead plant or plant showing vessel browning and wilted leaves up to the leader shoot and seedling height. The non-pathogenic F. oxysporum isolates were efficient in reducing the severity of the disease and maintaining normal plant development. These results provide evidence of the antagonistic activity of non-pathogenic F. oxysporum isolates in controlling vascular wilt caused by F. oxysporum f. sp. lycopersici race 2 in tomato.

To investigate the biocontrol mechanisms by which the antagonistic Fusarium oxysporum strain Fo47 is active against Fusarium wilt, a Fot1 transposon-mediated insertional mutagenesis approach was adopted to generate mutants affected in their antagonistic activity. Ninety strains in which an active Fot1 copy had transposed were identified with a phenotypic assay for excision and tested for their biocontrol activity against F. oxysporum f. sp. lini on flax in greenhouse experiments. Sixteen strains were affected in their capacity to protect flax plants, either positively (more antagonistic than Fo47) or negatively (less antagonistic). The molecular characterization of these mutants confirms the excision of Fot1 and its reinsertion in most of the cases. Moreover, we demonstrate that other transposable elements such as Fot2, impala, and Hop have no transposition activity in the mutant genomes. The phenotypic characterization of these mutants shows that they are affected neither in their in vitro growth habit nor in their competitiveness in soil compared with wild-type strain Fo47. These results show that mutants are not impaired in their saprophytic phase and suggest that the altered biocontrol phenotype should likely be expressed during the interaction with the host plant.

The medicinal plants Centella asiatica, Curcuma xanthorrhiza, Guazuma ulmifolia, and Hydrocotyle verticillata are widely used  in Indonesian traditional medicine, but little is known about their associated endophytic fungi. This research aimed to study the diversity of endophytic fungi derived from functional parts of these plants and to evaluate their potential as antifungal agents against the plant pathogenic fungus Fusarium oxysporum. A total of 17 isolates of endophytic fungi were obtained: nine from leaves of G. ulmifolia, three each from leaves of C. asiatica and H. verticillata, and two from rhizomes of C. xanthorrhiza. The genus Colletotrichum was found in all plants studied, but each plant was associated with different species. Colletotrichum aeschynomenes was associated with C. xanthorrhiza, C. siamense was associated with C. asiatica, and C. tropicale was associated with G. ulmifolia and H. verticillata. The species Curvularia affinis, Diaporthe tectonae, Lasiodiplodia mahajangana, Parengyodontium album, Talaromyces trachyspermus, and Speiropsis pedatospora were found only in G. ulmifolia; while Didymella coffeae-arabicae and Muyocopron laterale were found only in H. verticillata. The endophytic fungi showed inhibition activity against F. oxysporum with inhibition values of 6.0-78.9%, T. trachyspermus JBd10 and C. affinis JBd14 gave the highest inhibition activity.
