Background:Withania somnifera (family solanaceae) is a well-investigated medicinal plant which is also called Indian ginseng due to its wide spectrum of medicinal properties. The contents and activity of the plant may vary depending on the habitat and part of the plant and the solvent used for extraction. The present study deals with the comparative chemical analysis and in vitro antioxidant activity of methanolic fruits extracts and its subfractions (in ethyl acetate, butanol and water) of W. somnifera collected from two different geographical locations.Methods:In the present study, Withania somnifera fruits were collected from two different geographical locations (Uttarakhand and Rajasthan). The different fruit extracts were prepared and studied for total phenolic contents and total flavone contents. The in vitro antioxidant activity was assessed by DPPH free radical scavenging assay and peroxide scavenging assay.Results:Methanol extract of W. somnifera Uttarakhand and ethyl acetate subfraction of W. somnifera Rajasthan showed the highest amount of Total Phenolic Contents (TPC). In W. somnifera Uttarakhand, ethyl acetate extract showed the highest amount of Total flavonoids while in W. somnifera Rajasthan, methanol extract was found to be the richest in flavonoids. Methanolic extract of W. somnifera Uttarakhand showed the highest free radical scavenging activity while in W. somnifera Rajasthan, the highest antioxidant activity was shown by the methanolic extract followed by butanolic extract, water extract and then ethyl acetate. In the peroxide scavenging assay of antioxidant activity, water extract of W. somnifera Uttarakhand showed the highest activity, while in W. somnifera Rajasthan, ethyl acetate extract showed highest scavenging activity.Conclusion:It was concluded that the geographical location exerts a vital effect on the presence of active constituents and also on the antioxidant potential of W. somnifera.

The present study was designed to confirm the traditional use of the fruits of Ficus hispida Linn. (Moraceae) as an antioxidant agent. Fruits of the plant extracted with methanol and crude methanol extract (CME) were further fractionated with n-hexane, chloroform, and ethyl acetate. All the fractions, n-hexane (NHF), chloroform (CHF), ethyl acetate (EAF), aqueous (AQF) and CME were preliminary screened for in vitro antioxidant activity and total phenolic and total flavonoid content. In DPPH radical scavenging assay, CME exhibited highest scavenging activity (IC50 = 11.20 µg/mL) as compared to other fractions. In this assay, IC50 of reference standard BHT was 5.10 µg/mL. The reducing power of the samples was in the order as AQF > CME > CHF > EAF > NHF. The results for hydrogen peroxide scavenging activity indicated that CME, EAF and AQF had almost the same scavenging activity except NHF. Total antioxidant capacity of CME and other fractions were ranked as CHF > AQF > CME > EAF > NHF.  In the assay of antioxidant constituents (total phenol and total flavonoids content), the CME had highest phenolic and flavonoids content. The results indicate that Ficus hispida fruits could be considered as a potential source of natural antioxidant.

The aim of this study is to evaluate in vitro antioxidant activities of Ruta montana L.  extracts. This activity was evaluated by three methods : DPPH (2, 2'-diphenyl- 1- picrylhydrazy), bleaching of β-carotene and chelation of ferrous iron. Results showed that ethyl acetate extract (EAE) represents the highest amount of total polyphenols, tannins and flavonoids with 257,1 ± 0,703µg gallic acid equivalent/mg of extract,  251 ± 1.41 µg tannic acid equivalent /mg of extract,117,4 ± 3,451 µg quercetin equivalents/mg of extract, 139,5 ± 4,107 µg rutin equivalents/mg of extract, respectively. In the DPPH assay, ethyl acetate extract showed the higher scavenging capacity (IC50 = 0.044 ± 0.001 mg/ml) followed by methanol, aqueous and chloroform extract. Whereas, AqE showed the best chelating effect and the best inhibitory capacity of the coupled oxidation of linoleic acid/ β-carotene. Background:: Algerian propolis has gained interest in the last recent years. Many researches concerning both its biological effects and chemical composition were performed. Objective:: The present study was designed to investigate the chemical composition in particular phenolic compounds and the antioxidant activity of five Algerian propolis collected from different geographical parts of Algerian north namely: Constantine (CN), Boumerdes (BN), Mila (MN), Tebessa (TN) and Guelma (GN). Method:: Propolis extracts were obtained using solvents of varying polarity: petroleum ether, chloroform, ethyl acetate and finally methanol. Phenolic compounds were determined by liquid chromatography-tandem mass spectrometry (LCMS/ MS). In addition, total flavonoid and phenolic contents were also determined. Antioxidant activity was investigated using five complementary tests namely: DPPH., ABTS.+ assays for radical-scavenging activity, β-carotene-linoleic acid assay for lipid peroxidation activity, CUPRAC and FRAP assays for reduction capacity. Results:: The main phenolic compounds detected in the present study were: caffeic, p-coumaric, cinnamic and chlorogenic acids as well as naringenin and kampferol. Among the tested extracts, ethyl acetate and methanol extracts exhibited the highest phenolic and flavonoid contents and the strongest antioxidant activity. Propolis of Constantine was the most active one. Conclusion:: Our results suggest a potential use of Algerian propolis as a natural source of bioactive compounds with antioxidant activity.

Molineria latifolia is a perennial-monocotyle-donous herb and it is well-known in the treatment of melasma, freckle and black spot in Thai traditional medicine. The purpose of this study was to determine the phytochemicals and antioxidant activity in the aerial parts and the underground parts of M. latifolia by water, ethanolic and ethyl acetate extracts. The phytochemicals were analyzed by HPTLC. The antioxidant activity was measured by using the DPPH radical scavenging activity assay, Ferric reducing antioxidant power assay and Total phenolic content was measured by Folin‑Ciocalteu. The result showed the highest antioxidant activity of the underground parts (RW) were extracted by water at EC50=245.03±6.59 µg/mL(DPPH) and 640.03±0.36 Fe2+/ mg (FRAP). Consequently, the highest phenolics content was found in the water extract of the aerial parts (LW) (191.70±1.64 µg FA/mg). M. latifolia extracts contain a bioactive compound of Tannin, Flavonoids and Steroid . Tannin and Flavonoid were found in both of parts. In the aerial parts were found steroid extracting by ethyl acetate and ethanolic, as the underground parts were seen steroid extracting by water. It was found that M. latifolia has high bioactive compounds and moderate antioxidant activity. These results for the cosmetic product development in the future.

Bersama abyssinica, belonging to the Melianthaceae family, is distributed across Sub Saharan Africa. Decoctions of the bark, leaves, and roots of B. abyssinica have been extensively used in traditional medicine to manage many stomach complications such as colic, diarrhea, dysentery, and intestinal worms. In this study, we examined three extracts (ethyl acetate, methanol and water) obtained from B. abyssinica stem barks in terms of antioxidant properties. The antioxidant abilities were investigated by different chemical methods, including free radical scavenging (DPPH and ABTS), reducing power (CUPRAC and FRAP), metal chelating and phosphomolybdenum assay. In addition, total phenolic and flavonoid contents in the extracts were calculated. The highest level of phenolics was determined in water extract (230.83 mg GAE/g extract), followed by methanol (216.79 mg GAE/g extract) and ethyl acetate (100.57 mg GAE/g extract). In same line with total phenolic content, the best antioxidant properties were noted for water and methanol extracts. Our findings suggested that B. abyssinica stem bark extracts could be considered as promising sources of natural antioxidants.

Background: Tylophora hirsuta Wall. has long been used as traditional medicine for the treatment of diabetes. Current study is designed to evaluate the antidiabetic and anti-inflammatory activity of different extracts of aerial parts of Tylophora hirsuta. Methods: Sequential maceration was conducted to obtain extracts. Total phenolic contents were determined by FolinCiocalteau method. The antioxidant activity was assessed by DPPH free radical scavenging assay. The extracts were tested for its inhibitory activity against α-amylase in-vitro. In-vivo anti diabetic assay was conducted using alloxan induced diabetic model and OGTT was conducted on normal rats. ELISA was used to determine the proinflammatory cytokines (TNF-α and IL-6). Polyphenolic composition of the extract was analyzed using a HPLC system. Results: Aqueous extract exhibited highest total phenolic contents (985.24± 3.82 mg GAE/100 g DW), antioxidant activity (IC50 = 786.70 ± 5.23 Conclusion: These results showed that Tylophora hirsuta possess strong antidiabetic and anti-inflammatory potentials and justify its folklore use for the management of diabetes.

Objectives: The objective of this study was to estimate the total phenolic and flavonoid contents and to explore the antioxidant potential of various leaf extracts (chloroform, ethyl acetate, and ethanol) of Canthium coromandelicum, which is considered traditionally as an important medicinal plant.Methods: Antioxidant properties of the extracts were assessed using 1, 1- diphenyl -2- picrylhydrazyl and hydrogen peroxide radical scavenging assays for ascorbic acid equivalents. The total phenolic and flavonoid contents were also investigated to determine their correlation with the antioxidant activity of the leaf extracts and expressed in Gallic acid and quercetin equivalents, respectively.Results: The results showed that the content of total phenols and flavonoids was found to be high in ethyl acetate extract which was recorded as 61.02±1.30 mg Gallic acid equivalent (GAE/g) and 81.72±0.61 QE/g, respectively. Compared to other extracts, ethyl acetate leaf extract was found to possess high antioxidant activity at p<0.05 level, with high percentage of inhibition at 100 μg/ml concentration (82.70%) toward hydrogen peroxide radical scavenging with IC50 value 62.94 μg/ml. Statistically, two-tailed Pearson’s correlation showed strong positive correlations between hydrogenperoxide radical scavenging activity and total phenolic contents (TPC) (r=1.000) at p<0.05 level.Conclusion: The results obtained in this study clearly signifies that the ethyl acetate leaf extract of C. coromandelicum has high content of total phenols which are correlated to its antioxidant activity and thus has the potential to use as a source of natural antioxidants and can be explored as a therapeutic agent in free radical induced diseases.Keywords: Canthium coromandelicum, In-vitro antioxidant activity, Ethyl acetate extract, Pearson’s correlation, 1, 1- diphenyl -2- picrylhydrazyl, Hydrogen peroxide, Nitric oxide, Superoxide.

Corncobs (Zea mays) are beneficial to human health as they contain tyrosinase inhibitors and natural antioxidants, but they are not used as they are considered as waste. This research evaluated the inhibition test towards tyrosinase enzyme and antioxidant activity of corncob fraction using in-vitro DPPH method and its correlation to phenolic and flavonoids. Corncob fraction was extracted using the maceration method applying 70% ethanol solvent. The ethanol extract of corncob was suspended by water and then partitioned with chloroform, ethyl acetate, and aquadest to produce three fractions (chloroform, ethyl acetate, and aquadest fractions). These fractions were analyzed through the tyrosinase inhibition test, applying in vitro tyrosinase enzyme inhibition and antioxidant activity using radical scavenging test DPPH (2 2,2-diphenyl-1-picrylhydrazyl). Meanwhile, the total phenolic and flavonoids content tests were determined spectroscopically. The results showed ethyl acetate fraction had the highest tyrosinase activity with IC50 values of 185.76 µg/mL, followed by the aquadest fraction (IC50 676.44µg/ml) and the chloroform fraction (IC50 709.26 µg/mL). The antioxidant activity using DPPH radical scavenging method exhibited that ethyl acetate fraction had the highest antioxidant activity with IC50 of 25.79 µg/mL followed by the chloroform fraction (IC50 of 29.15 µg/mL) and the aquadest fraction (IC50 of 32.41 µg/mL). The total phenolic content of the corncob fraction ranged between 1.73 to 7.43% (w/w) gallic acid equivalents (GAE), while the entire flavonoid content ranged between 0.01 to 1.34% (w/ w) quercetin equivalent (QE). The tyrosinase activity and antioxidants of the corncob fractions correlated with the total phenolic and flavonoid contents.

This study was aimed to evaluate the cytotoxicity of methanolic extract (ME) and aqueous extract (AE) of the rhizomes of Bergenia purpurascens through the Brine-shrimp bioassay, which revealed that ME possessed cytotoxicity with LC50 = 181.97 ìg/ml value. The plant extracts were also evaluated in vitro for antioxidant activity using different redox [Folin-Ciocalteu (FC) and Ferric Reducing Antioxidant Power (FRAP)] and radical scavenging [(2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2’-azinobis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS)] assays. Both ME and AE possessed antioxidant activity and ME displayed comparatively a greater potential. The total phenolic contents in ME and AE was 1.27 and 0.06 g gallic acid equivalent (GAE)/100 g dry mass (DM), respectively. The FRAP values determined for ME and AE were 15.82 and 2.58 ìmol Fe(II)/100 g DM, respectively. The DPPH radical scavenging effect (IC50) was for ME 19.86 ìg/ml and 295.91 ìg/ml for AE. At 100 ìg/ml concentration, up to 94% inhibition of ABTS+ was observed in 6 min incubation. Cordia macleodii, an important medicinal plant traditionally used for the treatment of various disorders, including wound healing, jaundice, gastric ulcer, hepatoprotective activity and antivenom potential, thus the plant showed the high antioxidant properties. Successive extraction of leaf and barks powder with methanol and water was tested for quantitative determination of total phenolics, total flavonoids and various in vitro antioxidant activities. Phytochemical screening of crude plant extracts revealed the presence of sterols, alkaloid, flavonoid, phenolic, Reducing sugar, glycosides, tannins and saponins. All extracts showed the significant total phenolic and flavonoid contents and as well as radical scavenging and iron-chelating activities. Among all, methanol leaf extract was observed to be higher antioxidant activity than that of other extract. The methanol extracts showed greater antioxidant activity by DPPH scavenging the free radical with IC50 values of 7.63 ± 0.38 μg gm-1 along with scavenged free radical in a concentration depended manner, which is compare to the standard ascorbic acid.  Our results revealed that the presence of grater antioxidant activity of C. macleodii extract may be due to the high level of flavonoid and phenolic content in the plant. These results clearly indicated that C. macleodii is an effective natural antioxidant. Key words: Cordia macleodii, antioxidant, radical scavenging, phenolics, flavonoid, phytochemical.
