AbstractPurple waxy corn is a good source of antioxidant compounds such as anthocyanins and polyphenols. Promotion of its use requires an appropriate assay to determine antioxidant activity. The aim of this study is to verify compatibility of the antioxidant activity assays by comparing five different assays in daily kernel samples of the fresh purple waxy corn during grain filling. The levels of antioxidants measured by 2,2-diphenyl-1-picrylhydrazyl, ferric reducing antioxidant power, and chemiluminescence showed significant positive correlations with the levels measured by the other assays (r = 0.761–0.893; p < 0.01) and with anthocyanin content (r = 0.798–0.924; p < 0.01). Reducing capacity of 2,2ʹ-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) and total phenolic content also showed significant correlations (r = 0.764; p < 0.01). However, during late grain filling, the levels of antioxidants measured by all methods showed strong correlations with each other (r > 0.699; p < 0.05). The inconsistencies among the measurements are largely dependent on the developmental stage of the corn kernel. The combination of at least two assays is required to ensure reliable antioxidant activity estimates, especially for early grain-filling stages. These results will inform efforts to promote fresh purple corn as a source of antioxidants.

Phytophthora capsici Leon. is the main pathogen that limits the production of peppers. In this study, the effects of 1 and 10 mM proline (Pro), prior to exposure of resistant (CM-334) and susceptible (SD-8) pepper seedlings to P. capsici, on some physiological parameters were investigated. A lower Pro concentration (1 mM) was found to be more effective than 10 mM Pro in increasing the stress tolerance of the CM-334 cultivar. Namely, in CM-334 cultivar, the highest chlorophyll a, chlorophyll b, carotenoid, glucose and fructose content and 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging activity percentage were detected on the seventh day after application of 1 mM Pro + P. capsici, while the lowest malondialdehyde (MDA) amount was measured on the third day in the same treatment. The highest ferric reducing antioxidant power (FRAP) increase was determined on the seventh day in the 10 mM Pro + P. capsici application. The effects of the same Pro treatments on the SD-8 cultivar somewhat differed; the highest amounts of chlorophyll a, chlorophyll b, anthocyanins, fructose, total protein and endogenous Pro were detected on the seventh day in the 1 mM Pro + P. capsici application, while the lowest MDA amount was measured on the third day after the 10 mM Pro + P. capsici application, the highest DPPH % and FRAP values were detected on the seventh day with 10 mM Pro + P. capsici application. Although some differences were detected between the cultivars, Pro application against the P. capsici stress in general resulted in a positive effect on photosynthetic pigments, soluble carbohydrates and antioxidant capacity in pepper. The exogenous application of Pro helped the non-resistant cultivar to overcome the stress.

Various beneficial biological activities have been reported for passion fruit (Passiflora edulis), grown in tropical and subtropical regions. However, no study has yet demonstrated its action against the Zika virus (ZIKV) infection. The present study aimed to investigate the anti-ZIKV of the ethanolic passion fruit seed extract (PFSE), from which, liquid chromatography tandem mass spectrometry (LC-MS/MS) analysis allowed to putatively annotate the occurrence of piceatannol, astringin, scirpusin A, scirpusin B, isookanin-7-O-glucoside and naringenin-7-O-glucoside, two quadranguloside derivatives, tyrosine, and phenylalanine. PFSE is well tolerated by cell lines (Vero E6 and HTR-8/SVneo) and tissue explants from the human placenta. With 1- and 24-h treatments, PFSE consistently reduced the viral load and NS1 protein expression of the two strains of ZIKV (MR766 and PE243) in placental explants and cell lines. Thus, PFSE has a promising potential for developing anti-ZIKV treatments, protecting the placenta against infection caused by ZIKV, along with relevant antioxidant potential, represented by ferric reducing antioxidant power (FRAP) of 390.5 Trolox eq per g dry extract and half maximal inhibitory concentration (IC50) for 2,2-diphenyl-1-picrylhydrazyl radical (DPPH•) of 12.1 μg mL–1, and more than 90% protection from lipid peroxidation. Although further studies need to be conducted, PFSE treatment was safe and effective in trophoblast cells and placental explants, thus representing a promising class of all-accessible products based on the reuse of industrial by-products.

Background: Coniferous cones are a by-product of forestry and wood logging, used for many possible purposes, e.g., the extraction of polyphenols. Objective: The aim of the present article was the comparison of the antioxidant polyphenol content of the differently matured cones of 17 selected conifers, either common in Hungary or yet uninvestigated. Methods: Total polyphenol content, ferric reducing antioxidant power and 2,2-diphenyl-1-picrylhydrazyl assays were used to determine the antioxidant contents. A scoring system was implemented using the three assay results to evaluate and compare the overall antioxidant power of the samples. Result and Conclusion: Highest antioxidant contents were found in green cones, followed by mature and opened cones. Taxa with the highest scores were Tsuga canadensis, Cryptomeria japonica, Chamaecyparis lawsoniana, Thuja orientalis, Metasequoia glyptostroboides and Picea abies. For the samples with the highest overall antioxidant power the high-performance liquid chromatographic/tandem mass spectrometric polyphenol profiling was carried out (green cones of T. canadensis and P. abies) and 83 compounds have been tentatively identified and described. Results contribute to the future bioactivity testing and evaluation of the cone extracts of T. canadensis and P. abies.

Maoberry (Antidesma bunius L.) is a tropical fruit locally referred to as “Mao-Luang” in Thailand. The fruit contains high amounts of ascorbic acid and phenolic compounds with antioxidative potential, which has demonstrated medicinal value in terms of anti-cancer and anti-diabetic effects. In this term, this research purposed to determine the changes of predominant bioactive phytochemicals, antioxidant capacity and microbiological quality of pasteurized (85ºC/1 min), pressurized (500 and 600 MPa/30ºC/30 min), and ultra-sonicated (20 kHz/60% and 80% amplitude/30 min) maoberry juices during storage at 4°C for 30 days. The results displayed that ascorbic acid, phenolic acids (gallic and vanillic acids), anthocyanins (cyanidin 3-o-glucoside and cyanidin 3-rutinoside), flavonoids [(+)-catechin and (˗)-epicatechin), 2,2-diphenyl-1-picryl hydrazyl hydrate (DPPH) radical inhibition and ferric reducing antioxidant power (FRAP) value in pressurized and ultra-sonicated juices displayed higher reduction rate during storage than those in pasteurized juice. Nevertheless, at the final stage of storage, both juices still contained higher levels of antioxidant compounds and properties than in thermally treated juice. All the treated samples were shown to reduce initial microbial load of fresh maoberry juice to a non-detectable amount, while maintaining their quality during prolonged refrigerated storage.

Although avocado is a superfood rich in phytochemicals with high antioxidant activities, studies on the antibacterial properties of its pulp are limited, except for seed and peel portions. In this study, three types of solvent (acetone, methanol, and diethyl ether) were used to obtain the extracts from “Maluma” avocado pulp powder prepared by infrared drying. The extracts were analyzed for total polyphenols, phytopigments (total chlorophylls and carotenoids), antioxidant activities (ferric-reducing antioxidant power (FRAP), 2,2-Diphenyl-1-picrylhydrazyl (DPPH), and 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) assays), and antibacterial activities against seven pathogens (Shigella sonnei ATCC 9290, Escherichia coli ATCC 8739, Salmonella typhi ATCC 6539, Vibrio parahaemolyticus ATCC 17802, Proteus mirabilis ATCC 25933, Staphylococcus aureus ATCC 6538, and Bacillus cereus ATCC 11778). The results showed that the acetone solvent could extract the highest polyphenols and chlorophylls with the highest antioxidant activity in terms of ABTS and DPPH assays. In contrast, diethyl ether exhibited the most significant content of carotenoids and FRAP values. However, the methanol extract was the best solvent, exerting the strongest antibacterial and meaningful antioxidant activities. For the bacterial activities, Gram-positive pathogens (Bacillus cereus and Staphylococcus aureus) were inhibited more efficiently by avocado extracts than Gram-negative bacteria. Therefore, the extracts from avocado powder showed great potential for applications in food processing and preservation, pharmaceuticals, and cosmetics.

In this study, we explored the technical parameters of tree peony seeds oil (TPSO) after their treatment with radio frequency (RF) at 0 °C–140 °C, and compared the results with microwave (MW) and roasted (RT) pretreatment in terms of their physicochemical properties, bioactivity (fatty acid tocopherols and phytosterols), volatile compounds and antioxidant activity of TPSO. RF (140 °C) pretreatment can effectively destroy the cell structure, substantially increasing oil yield by 15.23%. Tocopherols and phytosterols were enhanced in oil to 51.45 mg/kg and 341.35 mg/kg, respectively. In addition, antioxidant activities for 2,2-diphenyl-1-picrylhydrazyl (DPPH) and ferric-reducing antioxidant power (FRAP) were significantly improved by 33.26 μmol TE/100 g and 65.84 μmol TE/100 g, respectively (p < 0.05). The induction period (IP) value increased by 4.04 times. These results are similar to those of the MW pretreatment. The contents of aromatic compounds were significantly increased, resulting in improved flavors and aromas (roasted, nutty), by RF, MW and RT pretreatments. The three pretreatments significantly enhanced the antioxidant capacities and oxidative stabilities (p < 0.05). The current findings reveal RF to be a potential pretreatment for application in the industrial production of TPSO.

Introduction: The purpose of this experiment is to examine the effect of different oligosaccharides with proven prebiotic effects on enzymes of carbohydrate metabolism and the antioxidant protection of erythrocytes in vitro under conditions of hyperglycemia. Materials and methods: This experiment included 10 healthy men (27±3 years of age). The isolated erythrocytes were treated with 1% and 5% solutions of the following oligosaccharides: lactulose, inulin, galactooligosaccharide and fructooligosaccharide in the presence of 5mM, 50mM and 100mM glucose. After incubation, for 2 hours at 37 °C, the erythrocytes were lysed, and the supernatant was used for analyses of lactate dehydrogenase, hexokinase and glutathione reductase. FRAP (Ferric reducing antioxidant power) method was used for determining the total antioxidant activity of erythrocytes. Results: Lactate dehydrogenase was decreased in the presence of 5% lactulose in groups with 50mM and 100 mM Glc. An increase in the activity of glutathione reductase under severe hyperglycemia (100mM glucose) was observed after treatment with: 1% lactulose, 1% inulin, 1% galactooligosaccharide, 1% and 5% fructooligosaccharides (p<0.005). A significant difference in the enzymatic activity of hexokinase was found in all groups (p<0.05) and of glutathione reductase only in the control group as well as in the groups treated with 1% lactulose, 1% galactooligosaccharide, 1% and 5% fructooligosaccharides Conclusions: Galactooligosaccharides 1% and fructooligosaccharides 1% and 5% cause a statistically significant increase of the enzymatic activities of hexokinase and glutathione reductase in in vitro hyperglycemia induced by 100 mM glucose, as well as an increase in FRAP.

While Aurantiochytrium sp. is an increasingly popular source of polyunsaturated fatty acids (PUFAs), its extraction generates high amounts of waste, including the spent, defatted residue. The composition and bioactivities of this by-product could prove to be a major part of the sustainable valorisation of this organism within the framework of a circular economy. In this study, the defatted biomass of commercial Aurantiochytrium sp. was nutritionally characterised, and its amino acid profile was detailed. Additionally, the antioxidant and prebiotic potentials of an enzymatically digested sample of defatted Aurantiochytrium sp. were evaluated under a set of miniaturised in vitro assays. The nutritional profile of the spent Aurantiochytrium biomass revealed a protein and dietary-fibre rich product, with values reaching 26.7% and 31.0% for each, respectively. It also held high concentrations of glutamic and aspartic acid, as well as a favourable lysine/arginine ratio of 3.73. The digested samples demonstrated significant Weissela cibaria and Bifidobacterium bifidum growth-enhancing potential. Residual ferric reducing antioxidant power (FRAP) activity was likely attributed to antioxidant amino acids or peptides. The study demonstrated that some of the nutritional and functional potential that reside in the defatted Aurantiochytrium sp. waste encourages additional studies and the development of food supplements employing this resource’s by-products under a biorefinery framework.

Currently, oxidative stress is one of the major problems that threatens human health. It is at the root of many diseases such as cancer. Despite the enormous efforts provided to combat this scourge, oxidative stress is still relevant and hence comes the need for research of new remedies especially from natural origin. For this purpose, the study of the antioxidant activity of extracts of Cistus monspeliensis from Morocco is a principal research objective. The phenolic extracts were obtained by maceration of the plant in a water/acetone mixture and then separated by liquid/liquid extraction with solvents of increasing polarity. The first phytochemical tests carried out on these extracts showed the existence of different families of phenolic compounds, such as flavonoids, tannins, and others. Assays for total polyphenols, flavonoids, hydrolysable, and condensed tannins were carried out by known colorimetric methods. The results of these assays have shown that the studied extracts are rich in phenolic compounds present in the plant in the form of flavonoids (69.81 ± 0.22 mg EQ/g DM), hydrolysable tannins (61.86 ± 0.89 mg ETA/g DM), and condensed tannins (70.05 ± 1.61 mg EC/g DM). The evaluation of the antioxidant activity is carried out by two different methods: the DPPH test (2,2-DiPhenyl-1-Picryl-Hydrazyl) and the FRAP test (Ferric Reducing Antioxidant Power). The results obtained show that the extracts of Cistus monspeliensis are active and have interesting antioxidant powers. In particular, the water/acetone (WAE) (IC50 = 0.079 mg/mL) and butanolic (BUE) (IC0.5 = 0.099 mg/mL) extracts are the most active with values comparable to that of ascorbic acid. The interesting results obtained in this study clearly show that Cistus monspeliensis originating from Morocco can be considered as a source of natural antioxidants. Therefore, the extracts of this plant deserve to be tested in the medicinal field, against cancer and cardiovascular diseases, and in food field as an additive and preservative.
