The CRISPR gene-editing (GE) breeding method is used to increase the resilience of high-yielding tomato cultivars against pests and diseases, reducing crop protection requirements. This study investigated consumers’ willingness to buy CRISPR GE tomatoes in a repeated discrete-choice experiment. We observed a strong positive effect of providing information on the CRISPR breeding technology, while the sensory experience of the CRISPR GE tomatoes in a visit to a greenhouse had a rather weak, predominantly negative effect on the participants’ willingness to buy CRISPR GE tomatoes. We found that roughly half of the 32 participants demonstrated constant CRISPR GE tomato choices during the experiments, and these participants were mainly employed as scientists. However, the rest of the participants changed their CRISPR GE tomato choices, with the majority showing an increase in their willingness to buy CRISPR GE tomatoes; these “changers” were dominated by non-scientists. Science communication on CRISPR GE breeding technology should target people with little knowledge about the technology, and consumers of organic tomatoes seem to have more specified, stable preferences regarding the technology. Further, scientific information about the CRISPR GE methodology should preferentially be provided when new technology and information about it are not yet widespread and people have not yet formed a strong opinion about the technology.

<p class="042abstractstekst">Plant height, straw mass and flag leaf area are recognized by physiologists as morphological markers of drought stress tolerance. Developing varieties intended for arid and semi-arid zones need to select for these traits. Understanding the genetic control of a given trait helps breeder to handle the segregating populations under study in a more efficient and consistent manner by choosing the best breeding method available to realize significant genetic advance. For this purpose, six generations: parents, F1, F2, BC1, BC2, derived from MBB x ‘Gaviota’ durum wheat (<em>Triticum durum </em>Desf.) cross were grown to investigate the nature of gene action involved in the inheritance pattern of the three traits. The results indicated that the six-parameter model fitted the best the data related to the variability present in the generation means of the studied traits. Generation mean analysis indicated that non-allelic interactions were important factors controlling the expression of these characters with complementary type of gene action governing FLA and STW inheritance. High heritability estimates, moderate to high expected responses to selection, significant genetic correlations with grain yield and greater role of non-additive effects in controlling the inheritance of the three studied traits suggested that breeding methods exploiting both fixable and non-fixable components be applied to break unfavorable linkage and to accumulate useful genes in the base population, followed by mono-trait or index based selection in late advanced generations.</p>

The protection of Dalmatian lamb ("Dalmatinska janjetina") with a protected designation of origin label (PDO) is based on the specific quality of meat of Dalmatian pramenka lamb and the traditional lamb breeding method in the geographic area that coincides with the breeding area of Dalmatian pramenka (indigenous Croatian sheep breed). Under the name of "Dalmatinska janjetina", only the meat of lambs of Dalmatian pramenka, bred exclusively in Dalmatia, can be placed on the market. In the process of obtaining a PDO label, which has recently been completed at the Croatian national level and is currently in the process at the EU level, a survey among Croatian consumers (a sample of 1,034 consumers from all over Croatia) was conducted to determine the recognition of this product on the Croatian market, as well as consumer preferences considering similar products from other Croatian regions. The research has shown that Croatian consumers recognize Dalmatian lamb as a traditional, high-quality product. Thus, almost 75 % of the respondents recognize it as a particular type of lamb of better quality than others on the Croatian market. Almost 85 % of the respondents use lamb regardless of type in their diet, but more than 58 % always on special occasions (folk festivals and family celebrations), especially if prepared in the traditional way (lamb on a spit) by which Dalmatia as a region is recognized by more than 85 % of the respondents. The market reputation and connection with the geographic production area are why Croatian consumers rather buy Dalmatian lamb over others.

Brown planthopper, blast, and bacterial blight are the main biotic stressors of rice and can cause a massive loss in rice production. Aroma is an important character of rice quality. It is of far-reaching significance to breed resistant and high-quality varieties using germplasms with objective genes. In this study, the introgression and pyramiding of brown planthopper (BPH), blast, and bacterial leaf blight (BLB) resistance genes and aroma genes into elite rice maintainers and restorers were conducted through conventional cross-breeding coupled with the marker-assisted selection (MAS) breeding method. Single-plant selection was performed from F2 onwards to select desirable recombinants possessing alleles of interest with suitable phenotypes. Respective linked markers were used in each generation from intercrossing to the F7 generation for tracking the presence of targeted genes. A total of 74 improved lines (ILs) have been developed which possess a combination of 1 to 4 genes for BPH, blast, and BLB resistance and aroma. These ILs showed moderate to high resistance to multiple biotic stresses (BPH, blast and BLB) or aromatic fragrance without obvious negative effects on agronomic traits. As multiple resistance and aromatic traits have become significant objectives in rice breeding, these resistance and/or aroma gene introgressed or pyramided lines have important application prospects. Core ideas: (1) marker-assisted breeding was used to pyramid multiple genes for an elite breeding line; (2) improved lines with the introgression of 1–4 genes were developed to achieve high resistance against various biotic stresses and aroma; (3) new lines were used as donor parents to introgress multiple genes in other genetic backgrounds.

Abstract A new breeding method of F1 hybrid using male sterility would open an exciting frontier in lettuce breeding, a self-pollinating crop. Male sterility is a crucial trait in F1 hybrid breeding. It is essential to map the causative gene for using male sterility. The ms-S, male-sterile gene of ‘CGN17397’, was mapped to LG8 by double-digest restriction site-associated DNA sequencing (ddRAD-seq) and narrowed down between two markers using two F2 populations. This region spans approximately 10.16 Mb, where 94 genes were annotated according to the lettuce reference genome sequence (version8 from crisphead cultivar ‘Salinas’). The whole-genome sequencing of the male-sterile and fertile lines of ‘CGN17397’ revealed that only one gene differed in the area of Lsat_1_v5_gn_8_148221.1, a homolog of Arabidopsis acyl-CoA synthetase5 (AtACOS5), and was deleted in the male-sterile lines. It was reported that AtACOS5 was needed for pollen wall formation and that the null mutants of AtACOS5 were entirely male sterility. Thus, I concluded that Lsat_1_v5_gn_8_148221.1 designated as LsACOS5 was a biologically plausible candidate gene for the ms-S locus. By using the structural polymorphism of LsACOS5, an insertion/deletion (InDel) marker was developed to select the male-sterile trait. The results obtained here provide valuable information for the genic male-sterility in lettuce.

AbstractIncluding Indirect Genetic Effects (IGE) in breeding programs to reduce aggression in group housed animals has been proposed. However, the effect of selection for IGE for growth on animal metabolism and physiology is unknown. The purpose of this study was twofold: (1) To investigate the effects of this new breeding method along with two housing (barren and straw), coping style (high and low resisters) and sex (female and castrated males) options on the metabolome profile of pigs. (2) To identify and map biological processes associated with a regrouping test at 9 weeks of age. We used Nuclear Magnetic Resonance to quantify 49 serum metabolites at week 8, 9 and 22. Also, we quantified 3 catecholamines (tyramine, epinephrine, phenylethylamine) and serotonin and three water soluble vitamins (B2, B5 and B7). Overall, no significant differences were observed between negative and positive IGE animals. The magnitude of change (delta) of many metabolites as a response to the regrouping test was significantly affected by IGE, especially that of the amino acids (P < 0.05), being greater in positive IGE pigs. The regrouping test was associated with alteration in glycine, serine and threonine metabolism. In conclusion positive and negative IGE animals respond differently to the regrouping test.

The Bornean Spotted Barb is endemic fish species and widely distributed throughout Borneo in various freshwater bodies and can be considered as a potential aquaculture candidate for food security and the ornamental trade. However there has yet to be scientific studies on the early life history and developmental stages of this species. Thus the present study reveals the embryonic development of Puntius sealei embryo, from fertilization to post-hatching using the induced breeding method. The induced spawning on P. sealei using Ovaprim hormone at a dose of 0.5 ml/kg for females and 0.25ml/kg for males respectively was successfully conducted and the embryonic development of the fertilized eggs were observed. The zygote, cleavage, blastula, gastrula, segmentation, and larval phases were the six major stages of embryogenesis that investigated in this experiment. This study contributed major knowledge to the early developmental biology of this particular species. The information will be useful for accessing the potentiality of this species for aquaculture as well as the management, and production of the species. Asian Australas. J. Food Saf. Abstract Shuttle breeding is a breeding system where generations undergo sequential evaluations at different locations, developed by N. Borlaug. Our study aims to demonstrate an opportunity of application shuttle breeding method in sunflower lines development and also to show its advantages and disadvantages. Experiments were mainly conducted at the Central Station (Krasnodar) of All-Russia Research Institute of Oil Crops (VNIIMK). Krasnodar region is situated in the Southern part of Russia near the Black Sea. Part of our work was made at Seed and Plant Improvement Institute (Karaj, Iran) and Experimental Station near Borozdjan (Busher Province of Iran). Released and experimental sunflower hybrids and lines of VNIIMK breeding were used as a material. The experimental design was randomized blocks with three replications. Each replication had four rows and two central rows were analyzed only to exclude the border effect. It is shown that the application of the shuttle breeding technique to sunflower was highly successful and results in elite line development.

Abstract ObjectiveThis study was conducted to evaluate the relationship between market weight, slaughter age, yield grade, and primal cut yield in Hanwoo. MethodsA total of 403 Hanwoo (Korean native cattle) was assessed for carcass traits such as carcass cold weight, backfat thickness, ribeye area, dressing percentage, yield index, and marbling score. The production yield of the individual major primal cuts of Hanwoo beef was also measured. FindingsCarcass cold weight, ribeye area, and backfat thickness, which affect meat quality increased with increased market weight (p<0.05). The production yield of the ten major primal cuts also increased with increased market weight (p<0.05). In terms of slaughter age, carcass cold weight, ribeye area, and backfat thickness all increased from 25 months to 28-29 months, and the production yield of all prime cuts also increased with increasing slaughter age. According to the meat yield grade, carcass cold weight and backfat thickness increased from grade A to grade C, although the ribeye area was not affected. The combined findings of the study suggest that slaughtering Hanwoo at the weight of 651-700 kg and 701-750 and age of 28.23 and 29.83 months might be desirable to achieve the best quality and quantity grade of Hanwoo beef. However, the positive correlation of carcass cold weight and backfat thickness, and the negative correlation of the yield index according to primal cuts yield indicated that it is necessary to couple the slaughtering management of cattle with improved genetic and breeding method of Hanwoo to increase the production yield of the major prime cuts of Hanwoo beef.

Parthenogenesis activation (PA), as an important artificial breeding method, can stably preserve the dominant genotype of a species. However, the delayed development of PA embryos is still overly severe and largely leads to pre-implantation failure in pigs. The mechanisms underlying the deficiencies of PA embryos have not been completely understood. For further understanding of the molecular mechanism behind PA embryo failure, we performed transcriptome analysis among pig oocytes (meiosis II, MII) and early embryos at three developmental stages (zygote, morula, and blastocyst) in vitro fertilization (IVF) and PA group. Totally, 11,110 differentially expressed genes (DEGs), 4694 differentially expressed lincRNAs (DELs) were identified, and most DEGs enriched the regulation of apoptotic processes. Through cis- and trans-manner functional prediction, we found that hub lincRNAs were mostly involved in abnormal parthenogenesis embryonic development. In addition, twenty DE imprinted genes showed that some paternally imprinted genes in IVF displayed higher expression than that in PA. Notably, we identified that three DELs of imprinted genes (MEST, PLAGL1, and DIRAS3) were up regulated in IVF, and there was no significant change in PA group. Disordered expression of key genes for embryonic development might play key roles in abnormal parthenogenesis embryonic development. Our study indicates that embryos derived from different production techniques have varied in vitro development to the blastocyst stage, and they also affect the transcription level of corresponding genes, such as imprinted genes. This work will help future research on these genes and molecular-assisted breeding for pig parthenotes.
