Abstract A commercially available (Syva Co.) enzyme-multiplied immunoassay technique (EMIT) for the quantitative determination of procainamide (PA) and N-acetylprocainamide (NAPA) was modified to allow automated quantitative analysis of approximately 100 samples per day, in a working range of 0.1 to 2.0 micrograms/mL. Such a test was needed to evaluate the pharmacokinetic characteristics of controlled-release dosage forms characterized by long half-lives at low plasma concentration. Analytical recovery of PA and NAPA from serum, plasma, and urine was satisfactory, but at extreme ratios for PA:NAPA the accuracy of determining the lower-concentration component became unsatisfactory. In fact, however, we found no such ratios in 5400 clinical samples assayed by this procedure.

Abstract A method is described for the quantitative determination of the ratio of d- and l-amphetamine stereoisomers by GLC. A derivative formed with N-trifluoroacetyl-(l)-prolyl chloride is chromatographed and the isomeric ratio is read from a standard calibration curve. The method is applicable to crystalline salts of amphetamine and to commercial tablet dosage forms. It is recommended that this method be subjected to collaborative study.

Tablet film coating is a common but critical process providing various functionalities to tablets, thereby meeting diverse clinical needs and increasing the value of oral solid dosage forms. Tablet film coating is a technology-driven process and the evolution of coated dosage forms relies on advancements in coating technology, equipment, analytical techniques, and coating materials. Although multiple coating techniques are developed for solvent-based or solvent-free coating processes, each method has advantages and disadvantages that may require continuous technical refinement. In the film coating process, intra- and inter-batch coating uniformity of tablets is critical to ensure the quality of the final product, especially for active film coating containing active pharmaceutical ingredients in the coating layer. In addition to experimental evaluation, computational modeling is also actively pursued to predict the influence of operation parameters on the quality of the final product and optimize process variables of tablet film coating. The concerted efforts of experiments and computational modeling can save time and cost in optimizing the tablet coating process. This review provides a brief overview of tablet film coating technology and modeling approaches with a focus on recent advancements in pharmaceutical applications.

Abstract A simple, accurate, and rapid method for the quantitative determination of ritodrine hydrochloride (RTH) and isoxsuprine hydrochloride (ISH) in both pure and dosage forms, is described. The method is based on the development of pink colored product as a result of the condensation of 4-aminoantipyrine with phenols in the presence of an alkaline oxidizing agent. The resulting products are measured at 510 nm for both drugs, with molar absorptivities of 0.98 × 104 and 1.20 × 104 L/mol·cm for RTH and ISH, respectively. A study of the effect of commonly associated excipients revealed that they did not cause interference.

AbstractCarcinogenic and mutagenic properties of aflatoxin species are known in literature. Their intake over a long time period might be health-dangerous for human even at trace levels. It is well known that different foodstuffs can be contaminated by aflatoxin species through growing and storage. Due to the serious health effects, sensitive determination of aflatoxin species in any matrices related with the human being is very crucial at trace levels. In literature, there are sensitive techniques to analyze the different samples for the contents of their aflatoxin species. Each technique has some advantages and disadvantages over the other techniques. This review aims to summarize the different health effects of aflatoxin species, development of analytical techniques and applications of developed techniques in a variety of matrices.
