STUDY OF THE ANXIOLYTIC ACTIVITY OF ETHANOLIC EXTRACT OF ROOT OF ACORUS CALAMUS IN ALBINO MICE

Objective: Anxiety is classified as a form of sub-acute or chronic fear. Root of Acorus calamus has been traditionally used as an anxiolytic. The aim of the study is to assess the anxiolytic activity of ethanolic extract of A. calamus (EEAC) by elevated plus-maze test on Albino mice. Methods: Albino mice of either sex were taken and divided into five groups, each consisting of 5 mice. One group was used as control, one as standard (diazepam), and three as test groups treated with 100, 200, and 400 mg/kg of EEAC. The drugs, that is, 10 ml/kg of normal saline for control, diazepam 2 mg/kg (standard), and 100, 200, and 400 mg/kg EEAC (test groups) were injected intraperitoneally (i.p.), 30 min before placing them in the center of the maze. The preferences of the animal to open/enclosed arm, average time spent in open arm, and numbers of entries in open arm were compared in each group. Data were statistically analyzed by one-way analysis of variance followed by multiple Dunnett’s test. Results: The number of entries in open arm and the average time spent in the open arm by the mice is increased by EEAC in a dose-dependent manner. Conclusion: EEAC has anxiolytic activity.

Latency to enter the chamber, number of entries in five minutes and total time spent in the chamber during 5 minutes test period were compared in each group. Results: EEAC showed significant increase in all the parameters for the doses of 100, 200 and 400 mg/kg in a dose dependent manner. Conclusion: EEAC has anxiolytic activity.

AbstractThe current study investigates the anorectic interaction and safety of the mazindol-metformin combination in rats. Isobologram and interaction index were used to determine anorectic interaction between mazindol and metformin in the sweetened milk model. The safety profile of the mazindol-metformin combination was determined by measuring anxiety, blood pressure, hematic biometry and blood chemistry. An acute dose of mazindol and metformin administered per os, individually or as a mixture, has reduced the milk consumption in rats in a dose-dependent manner. Theoretical effective dose 40 (ED40t) did not differ from the experimental effective dose 40 (ED40e) obtained with the mazindol-metformin mixture in the anorexia experiments, by Student′s t-test. In addition, the interaction index confirmed the additive anorectic effect between both drugs. A single oral dose of ED40e mazindol-metformin mixture induced anxiolysis in the elevated plus-maze test. Moreover, oral administration of mazindol-metformin combination for 3 months significantly decreased glycemia, but not blood pressure nor other parameters of hematic biometry and blood chemistry. Results suggest that mazindol-metformin combination exerts an additive anorectic effect, as well as anxiolytic and hypoglycemic properties. Mazindol-metformin combination might be useful in obese patients with anxiety disorders or diabetes risk factors.

The present study investigates the anxiolytic activity of NR-ANX-C, a standardized polyherbal formulation containing the extracts ofWithania somnifera, Ocimum sanctum, Camellia sinensis, Triphala, and Shilajit in ethanol withdrawal- (EW-) induced anxiety behavior in rats. Ethanol dependence in rats was produced by substitution of drinking water with 7.5% v/v alcohol for 10 days. Then, ethanol withdrawal was induced by replacing alcohol with drinking water, 12 hours prior to experimentation. After confirming induction of withdrawal symptoms in the alcohol deprived animals, the anxiolytic activity of the test compound in graded doses (10, 20, and 40 mg/kg) was compared to the standard drug alprazolam (0.08 mg/kg) in the elevated plus maze and bright and dark arena paradigms. In our study, single and repeated dose administration of NR-ANX-C reduced EW-induced anxiety in a dose-dependent manner. Even though the anxiolytic activity was not significant at lower doses, NR-ANX-C at the highest dose tested (40 mg/kg) produced significant anxiolytic activity that was comparable to the standard drug alprazolam. Based on our findings we believe that NR-ANX-C has the potential to be used as an alternative to benzodiazepines in the treatment of EW-induced anxiety.

Objectives: The objectives of the study were to study the effect of Morus alba leaves extract (MAE) on sleep by phenobarbitone-induced sleeping time and the antianxiety effect by elevated plus maze apparatus model in rats. Methods: In this study, the effect of MAE on sleep was evaluated by the phenobarbitone-induced sleeping time of rats. The onset and the duration of sleep were recorded in minutes. The antianxiety effect was evaluated by the elevated plus maze apparatus model in rats. During 5 min test period, the number of entries into the open arm and closed arm and time spent in the open arm and closed arm were recorded in seconds. Results: MAE at the dose 200 and 400 mg/kg, highly significantly (p<0.001) decreased the onset of phenobarbitone-induced sleeping time. The duration of sleeping time was increased significantly (p<0.01) for 200 mg/kg and highly significantly (p<0.001) for 400 mg/kg as compared to the control group. M. alba has significant antianxiety activity in comparison with control in a dose-dependent manner. M. alba in a dose of 200 mg showed significant (p<0.01) and 400 mg/kg treated groups showed highly significant (p<0.001) anxiolytic activity by increasing the mean time spent in open arms as compared to control but less significant with standard (diazepam). Conclusion: Results indicate that the MAE has a significant dose-dependent effect on phenobarbitone- induced sleeping time and antianxiety effect in the elevated plus maze test.

Ficus racemosa is used in traditional system of medicine for various health problems and diseases, and is commonly known as Gular fig. The main objective was to study its effects against streptozotocin induced diabetic neuropathy by structural and functional marker. Investigation of diabetic neuropathy was carried out through functional and structural assessment in streptozotocin induced in diabetic rats. Diabetic rats were treated for 28 days in dose dependent manner of Ficus racemosa aqueous extract (250 mg/kg and 500 mg/kg) and ethanolic extract (200 mg/kg and 400 mg/kg). Study showed marked protection observed by Ficus racemosa in hippocampus region of brain and sciatic nerve tissues. Ficus racemosa treatment showed improvement in functional and structural markers, which strongly suggest its protective role in diabetic neuropathy.

Objective: To study the anti-obesity potential of aqueous rhizome extract of Acoruscalamus Linn. (AREAC)in high fat diet fed obese rats.Methods: Adult strain male Wistar rats used in this study were fed with High Fat Diet (HFD) for 60 days. For the treatment groups,AREAC was administered in a dose levels of100, 200 and 300 mg/kgbw, orally once a day along with HFD. Rats fed with normal pellet chow were served as normal control. The effect of AREAC on physical parameterssuch as body weight, organ weight, fat pad weights and various biochemical parameterslike serum glucose, insulin, leptin,lipid profile, liver markers, kidney markers and oxidative stress markers were analysed.In-vitro pancreatic lipase inhibition assay of AREAC was also studied.Results: Data of in-vivo studies revealedsignificant (p<0.05) reduction in percentage body weight gain, organ weights, fat pad weights and levels of serum glucose, insulin and leptin after treatment with AREAC in a dose dependent manner. Also, administration of AREAC significantly inhibited the increases in the concentrations of triglycerides, total cholesterol, LDL-cholesterol, VLDL-cholesterol, free-fatty acid and phospholipids in a dose dependent manner whereas, the level of HDL-cholesterol was found to be elevated on treatment. Moreover, on treatment with test drug,the elevated levels of serum liver and kidney markerssuch as AST, ALT, ALP, urea, creatinine were also brought back to near normalcy. Antioxidant status was found to be enhanced in liver tissues after treatment.In-vitro studies showed significant inhibition in the activity of pancreatic lipaseby AREAC.Conclusion: The data of the results obtained clearly depicted that AREAC was found to have pronounced anti-obesity activity particularly at the dose levels of 300 mg/kg bw.Key Words: Obesity, High Fat Diet, Leptin, AcoruscalamusLinn., Orlistat.

Using the previously obtained first dipeptide ligand TSPO the N‑carbobenzoxy-L‑tryptophanyl-L‑isoleucine amide (GD‑23) as a basis, the new dipeptide was synthesized — the N‑phenylpropionyl–L‑tryptophanyl-L‑leucine amide (GD‑102). GD‑102 expressed anxiolytic activity in the open field test in BALB/c mice and in the elevated plus maze test in ICR mice. The minimum effective dose of GD‑102 was an order of magnitude lower than that of GD‑23. Preliminary administration of the TSPO selective antagonist, compound PK11195, completely blocked the anxiolytic activity of GD‑102, that indicated the participation of TSPO in the realization of the anxiolytic action GD‑102. The results were confirmed by molecular docking data.

Objective: The aim and objectives of the present work is to determine pharmacological activity upon ethnopharmacological survey. The present study deals with phytochemical screening and analgesic as well as muscle relaxant activity of leaves of Mirabilis jalapa.Methods: The present study is aimed at phytochemical screening and evaluating the analgesic and muscle relaxant activities of ethanolic leaf extract of Mirabilis jalapa by using hot plate method and rota rod method respectively [1, 2]. The Phytochemical screening of the extract was done according to the standard procedures to reveal the presence of the active constituents like Alkaloids, flavonoids, phenols, glycosides, tannins, saponins, steroids, carbohydrates etc [3-5].Results: The analgesic and muscle relaxant activity study were dose dependent. The EEMJ extracts (100 mg/kg, 200 mg/kg, 500 mg/kg) and the standard drug Diclofenac sodium (25 mg/kg) shows significant increase in the reaction time when compared with control at 30 min, 60 min, 90 min and 120 min and the effect of standard was found to be highest during the study. Another study was designed to evaluate the skeletal muscle relaxant properties of ethanolic extract of leaves of Mirabilis jalapa. Linn by taking the EEMJ extracts (100 mg/kg, 200 mg/kg, 500 mg/kg) and standard drug Lorzepam (10 mg/kg). Both the extracts and standard drug show decrease in the fall of time in a dose dependent manner when compared with control at 15 min, 30 min, 45 min respectively. Conclusion: Therefore, from the above study it is revealed that Mirabilis jalapa showing better pharmacological activities (Analgesic and Muscle relaxant) in dose dependent manner.

The present work was carried out to assess the Onosma bracteatum anxiolytic and antidepressant properties. Swiss albino mice (male) were fed orally with hydroalcoholic extract at different doses 50, 100, and 200 mg 1 hour prior to test with the standard diazepam and fluoxetine. Anxiolytic and antidepressant activities were evaluated by using open field, elevated plus maze, force swimming, and tail suspension test. Results of open field test showed an increase in number of line crossing as well as number of rearing in dosage-dependent design. Although results of elevated plus maze test evidently showed antianxiety effect of O bracteatum by increasing the time spent in open arms along with decreasing the time spent in closed arms in dosage-dependent way. For the evaluation of antidepressant effect, O bracteatum diminished the immobility time and expanded mobility time in forced swim model in dosage-dependent way. Likewise, O bracteatum expanded time span of mobility along with diminished immobility time in tail suspension method in dosage-dependent way. Outcome demonstrated that plant at the dose of 200 mg/kg body weight showed significant potential which was similar to that standard diazepam and fluoxetine. Hence, O bracteatum may be used as potent natural psychotherapeutic agent against the mental disorders.
