Chrysophyllum albidum G. is generally regarded as a plant with many ethno-medicinal uses. The study was aimed at evaluating the in vitro antioxidant, diuretic, and hypoglycemic properties of the ethanol extract of leaves of C. albidum Male Wistar albino rats weighing between 150g and 200g were used for the experiment. The DPPH scavenging activity was determined using DPPH assay, inhibition of lipid peroxidation was carried out using the reactions of peroxides with thiobarbituric acid, while nitric oxide was conducted based on the Greiss reaction. Diuretic activity was carried out using the method of Lipschitz with slight modifications. Alloxan (140 mg/kg) was used to induce diabetes in the rats. The extract showed a concentration-dependent increase in DPPH radical scavenging activity, anti-lipid peroxidation activity, and nitric oxide inhibition activity maximal at 200 mg/ml (84.99±7.97, 79.82±6.10, 69.99±2.22 respectively) and this was significant (p<0.05) when compared with that of 100 mg/ml of vitamin C (97.68±0.84, 99.05±0.24 and 98.15±0.29 respectively) which served as the control. The extract also significantly (p<0.05) increased; urine volume, urinary sodium, and urinary chloride particularly at 2000 mg/kg (1.55±0.07, 109.60±0.57, and 106.00±1.41 respectively) when compared with the standard drug (1.85±0.07, 115.20±0.57, and 112.85±0.49 respectively). The increase in potassium excretion was also significant (p<0.05) particularly at 2000 mg/kg (13.70±0.28) but less than those of sodium (109.60±0.57) and chloride (106.00±1.41) at the same concentration. There was no significant (p<0.05) increase in urinary bicarbonate the maximum being at 250 mg/kg (22.55±0.78). The extract caused a non-significant (p<0.05) decrease in the fasting blood glucose level of the rats. It was concluded that the leaves of C. albidum could be used as a source of natural antioxidant boosters and also a diuretic.

Objective. Mallotus oppositifolius(Geiseler) Müll. Arg. (Euphorbiaceae) is folklorically used to “treat” diabetic conditions in some parts of Nigeria therefore the study, to investigate the extract of the leaves for activities on hyperglycaemia, lipid peroxidation, and increased cholesterol levelsin vivoin alloxan diabetic rats as well as its potential antioxidant activityin vitro.Methods. Albino rats (240–280 g) were given an injection of 120 mg/kg body weight, i.p. of alloxan monohydrate. After 8 days, diabetic animals with elevated fasting blood glucose levels (>9 mmol/L) were considered and selected for the study.Results. Oral treatment with the extract administered every 12 h by gavage at doses of 100, 200, and 400 mg/kg of the extract to the test rats, for 14 days, resulted in a significant dose-dependent decrease in blood glucose levels from 12.82 ± 1.02 mmol/dL to 4.92 ± 2.01 mmol/dL at the highest dose of 400 mg/kg compared to the control drug and glibenclamide as well as attendant significant decline in diabetic rats employed in the study.Conclusion. The extract also showedin vitroconcentration-dependent antioxidant activity following the 1,1-diphenyl-2-picryl-hydrazyl (DPPH) and ferric reducing assays. Findings further suggest the presence of active antidiabetic and antioxidant principles inM. oppositifoliusleaves.

Background: Syzygium cumini L., commonly known as Jamun, black-plum, and Indian blackberry, is one of the most widely distributed trees in India with booming medical benefits and possesses antioxidant, anticancer and anti-diabetic properties. It belongs to the family Myrtaceae. Despite countless phytochemicals, seeds are not consumed and are the waste part of Jamun fruit. Objective: The objective of this study was to evaluate the antioxidant capacity of phenolics from Jamun seeds against a bundle of oxidant moieties. Methods: The 50% acetone extract of Jamun seeds was investigated for in-vitro antioxidant profiling. Assays include free radical scavenging activity, metal chelation activity, hydroxyl radical scavenging activity, hydrogen peroxide scavenging activity, total antioxidant activity, total reducing power, nitric oxide scavenging activity, and lipid peroxidation inhibition activity. Results: The extract depicted maximum DPPH radical scavenging activity followed by ABTS radical scavenging activity. Hefty metal chelation and nitric oxide scavenging activity were recorded while lipid peroxidation, H2O2, and OH- scavenging activity was intermediate. Conclusion: Jamun seed showed ample antioxidant activity and certifies that it is the right candidate for exploitation as a source of natural antioxidants to counteract autoxidation-induced pathologies or diseases.

The study assessed the in vivo antioxidant and hepatoprotective activity of an ethanol (70%) extract of Momordica tuberosa Cogn. (Cucurbitaceae) (TMT) tubers in experimentally induced liver damage by paracetamol (2 g/kg, po.) in albino rats. The degree of protection was ascertained by estimating the levels of biochemical markers like SGPT, SGOT, bilirubin (total and direct), ALP, and triglycerides. Tissue GSH and lipid peroxidation were also determined. The ethanol (70%) extract of tubers in an oral administration of 20 and 40 mg/kg doses produced significant protection by decreasing the activity of serum enzymes, bilirubin, cholesterol, triglycerides and tissue lipid peroxidation, while it increased tissue GSH at 40 mg/kg dose. The effects of the extract were comparable to the standard drug silymarin (100 mg/kg). Results suggested that an ethanol (70%) extract of the tubers of the plant at 40 mg/kg possesses potential hepatoprotective activity against paracetamol-induced hepatic damage and significant antioxidant activity in rats.

Background: Fagopyrum esculentum Moench. is a herb consumed as food and has medicinal value. It is a rich source of bioactive nutrients which cure and prevent many ailments. Traditionally, it is used to treat hypertension, diabetes, constipation, cancer etc. Methods and Objective: Present work illustrates morphological, microscopic and physicochemical parameters of Fagopyrum esculentum seeds as per WHO guidelines, in vitro antioxidant activity; assessed by DPPH scavenging method, hydrogen peroxide scavenging assay and β-carotene linoleic acid bleaching method and study of lipid lowering potential of the ethyl acetate and ethanol extract of seeds on normal diet fed Wistar rats. Results: Morphological studies delineated the triangular shape, dark brown colour, 8 mm length and 6 mm width of the seed. The microscopic examination of the transverse section of seed depicted features like testa or pericarp (seed coat), the endosperm, embryo and sclerenchyma cells. Study of physiochemical parameters exhibited 0.3±0.02% of foreign matter and 1.44±0.51% crude fibre content. Total ash, acid insoluble ash and water soluble ash value were 6.7±1.7%, 1.9±0.23% and 3.9± 0.31% respectively. Alcohol soluble and water soluble extractive value came out to be 65.02± 3.21 mg/g and 12.7±1.24 mg/g respectively. Foaming index was less than 100, swelling index was found to be 0.5±0.01 ml/g. Loss on drying was 4.02±1.27%. Phytochemical screening of ethyl acetate and ethanol extract revealed the presence of alkaloids, carbohydrates, phenolic compounds, phytosterols and flavonoids. Trace amount of heavy metals (arsenic, cadmium, lead, mercury) were determined by atomic absorption spectrophotometer. Pesticide residue analysis confirmed the presence of nontoxic pesticides like dimethipin, hymexazol, phenothrin-2, methoprene, triadimenol, prohydrojasmon- 1, jasmolin ii, triademinol, jasmolin i, prohydrojasmone i, cyromazine in both the extracts by gc-ms spectrometer. The ethyl acetate and ethanol extract has shown significant in-vitro antioxidant activities demonstrated by the DPPH method (IC50 = 94.37±2.51 and 216.04±4.39 μg/ml respectively), hydrogen peroxide scavenging assay (IC50 = 83.72±3.72 and 193.47±5.05 µg/ml respectively) and β-carotene bleaching method (IC50 = 100.67±4.01 and 205.39±2.89 µg/ml respectively). Lipid lowering study performed on Wistar rats demonstrated a significant (p<0.001) decrease in serum Total Cholesterol (TC), Triglyceride (TG) and increase in High Density Lipoprotein (HDL) level as compared to normal group. Both the extracts have shown a non significant difference in the level of TG as compared to standard drug atorvastatin, depicting that the efficacy of extracts is at par with that of standard drug atorvastatin. Conclusion: Pharmacognostical study of the plant can be a very good tool for identification as well as authentication of a herb. Moreover, these parameters may be helpful in the development of monograph of the plant. Pharmacological activity confirmed Fagopyrum esculentum Moench. seed to be a good antioxidant and have lipid lowering potential.

Background & Introduction : Antioxidants are known to prevent oxidative stress-induced damage to the biomolecules and thus, delay the onset of cancers and many age-related diseases. Therefore, the development of novel and potent antioxidants is justified. Method: During this study, we synthesized symmetrical bis-Schiff bases of carbohydrazide 1-27, and evaluated their in vitro antioxidative activity and cytotoxic activity. Results: Among synthesized compounds, six compounds 20 (IC50 = 12.89 ± 0.02 µM), 16 (IC50 = 14.32 ± 0.43 µM), 17 (IC50 = 18.52 ± 0.83 µM), 19 (IC50 = 22.84 ± 0.62 µM), 24 (IC50 = 35.1 ± 0.82 µM) and 15 (IC50 = 40.03 ± 1.06 µM) showed an excellent 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, better than the standard butylatedhydroxyanisole (BHA) (IC50 = 44.6 ± 0.6 µM). Likewise, two compounds 16 (IC50 = 4.3 ± 1.3 µM) and 20 (IC50 = 6.6 ± 1.6 µM) showed oxidative burst scavenging activity better than the standard drug ibuprofen (IC50 = 11.2 ± 1.9 µM). Some synthesized compounds showed good to moderate toxicity against prostate cancer (PC-3) cell lines. Conclusion: This study has identified potent antioxidants and good cytotoxic agents with the potential to further investigate.

In the present study, protective effects of bitter melon (Momordica charantia) extract on lipid peroxidation induced by immobilization stress in rats have been assessed. Graded doses of extract (50, 100, and 150 mg/kg body weight) were administered orally to rats subjected to immobilization stress for two hours for seven consecutive days. Stress was applied by keeping the rats in a cage where no movement was possible. After seven days, rats were killed by decapitation after ether anesthesia. Blood and liver were collected to measure thiobarbituric acid reactive substances, reduced glutathione, and catalase. In vitro effects of M. charantia extract on lipid peroxidation in liver homogenate of normal, control, and rats pretreated with extract were carried out against cumene hydroperoxide-induced lipid peroxidation. Results reveal that in vivo M. charantia inhibited stress-induced lipid peroxidation by increasing the levels of reduced glutathione and activities of catalase. These results were further supported by in vitro results. In vitro inhibition of lipid peroxidation was indicated by low levels of thiobarbituric acid in the liver homogenate from pretreated rats and normal rats when incubated with both cumene hydroperoxide and extract. Inhibition was also noted in the homogenate where the rats were pretreated but the mixture contained no extract. Thus this plant provides protection by strengthening the antioxidants like reduced glutathione and catalase. Inclusion of this plant in the daily diet would be beneficial.

<p><strong>Objective: </strong>To determine the effect of <em>Tridham</em> (TD) and 1,2,3,4,6-penta-O-galloyl-β-d-glucose(PGG) on lipid peroxidation levels and mitochondrial antioxidants status in experimental mammary carcinoma.</p><p><strong>Methods</strong>:<strong> </strong><em>Elaecoarpus ganitrus </em>(fruits), <em>Terminalia chebula </em>(seed coats), <em>Prosopis cineraria </em>(leaves)<em>, </em>adult female albino rats of Sprague-Dawley strain weighing 170–190 g and 7,12-dimethylbenzeneanthracene (DMBA) were used for this study. Group I control rats, Group II rats mammary carcinoma induced with DMBA (25 mg in 1 ml olive oil) by gastric intubation. Group III, IV and V DMBA induced rats were treated with TD (400 mg/kg. b. wt/day), PGG (30 mg/kg. b. wt/day) and standard drug, Cyclophosphamide (30 mg/kg. b. wt/day), respectively for 48 d by gastric intubation. Group VI and VII rats served as TD and PGG treated controls, respectively for 48 d by gastric intubation. At the end of the experimental period, the rats were anaesthetized and sacrificed. Mammary glands were isolated and used for biochemical assays and histopathological evaluation.</p><p><strong>Results: </strong>In rats with cancer, the lipid peroxide levels (LPO) were significantly increased and mitochondrial antioxidant levels were decreased. Treatment with TD and PGG decreased LPO levels and increased mitochondrial antioxidant status in mammary carcinoma bearing rats. Histopathological analysis also confirmed the therapeutic effect of TD and PGG. No significant adverse effect was observed in sole drug treated group of rats.</p><p><strong>Conclusion: </strong>TD and PGG have definite therapeutic effect in experimental mammary carcinoma and inhibit growth of cancer cells by restoring mitochondrial antioxidant status and energy metabolism to normal states.</p>

Objective: The objective of the study was to in vitro evaluate of hypolipidemic effect of extracts of medicinal Dracaena cinnabari Balf. f. resin. Methods: About 800 g of dry powder of the resin of dracaena cinnabar was taken in a Soxhlet apparatus and subjected for sequential extraction of solvents from non-polar to polar end (hexane, benzene, diethyl ether, dichloromethane, chloroform, ethyl acetate, acetone, ethanol, methanol, and water); the extract samples were kept at 4°C for further assays. All the extracts were subjected to glucose uptake assay. Results: The ethanol extract showed significant (p<0.05) hypolipidemic effect by decreasing the activity of enzyme such as significant reduction in the pancreatic lipase enzyme, malic dehydrogenase enzyme, and glucose-6-phosphate dehydrogenase enzyme with IC50~13, ~13, and ~14, respectively. This results were similar to the standard drug atorvastatin with IC50~12, ~16, and ~17, respectively. Ethanol extract exhibited significant atherogenic index and percentage protection against hyperlipidemia. The potential biological activity of ethanol extract may be attributed to the highest polarity which needs further investigation.

Dennettia tripetala fruit is a popular Nigerian fruit from the family of plant known as Annonaceae. The whole fruit (flesh and seed) is usually consumed as snacks and it is oftentimes consumed with local gin (ufofop in Ibibio or kaikai in Igbo) or added to dishes as spice due to its peculiar strong pepperish taste and sweet aroma. The present study is aimed at evaluating the antioxidant potentials of ethanol extract of ripe and unripe D. tripetala (DT) fruit in vitro. The antioxidant activity of the ethanol extract of DT was evaluated spectrophotometrically using various in vitro models like 1, 1-diphenyl-2-picryl-hydrazyl (DPPH) and hydrogen peroxide scavenging activity; metal chelating activity and reducing power. Vitamin C was used as the standard antioxidant.Unripe and ripe DT fruits, as well as vitamin C showed a competitive inhibition of DPPH and H2O2 free radicals. As concentration of the extracts increased from 20 to100 µg/mL, the % scavenging activity for vitamin C increased from 87.86 ± 0.11 to 90.66 ± 0.07 and for ripe DT fruits from 15.15 ± 0.24 to 25.52 ± 0.23, while for unripe, fruits values increased from 12.09 ± 0.35 to 23.06 ± 0.12. The IC50 values was highest in unripe (549.23) followed by ripe (276.63) and lowest in vitamin C (12.92) indicating that vitamin C was the best scavenger of DPPH radical. Similar trend was obtained for H2O2 scavenging activity as well as reducing power. Unripe DT fruit extract was more potent at chelating metal ions (IC50 was 95.38), followed by the standard ascorbic acid with IC50 of 97.03 and was lowest in ripe DT fruit extract with IC50 value of 124.66. Unripe and ripe DT are potent antioxidants in nature and may be used to supplement our diets as rich sources of natural antioxidants for health protection.

Aim: The aim of this study was to investigate and compare the antioxidative properties of the mistletoe plant obtained from three different host species namely Psidium guajava, Vernonia amygdalina and Moringa olifera lam. Study Design: Experimental Design Place and Duration of Study: Department of Pharmaceutical Chemistry, Faculty of Pharmaceutical Sciences and Department of Biochemistry, College of Health Sciences, University of Jos, Nigeria. Methodology: Crude methanolic leaf extracts were studied for their antioxidative properties; Iron reducing and Iron-chelating activities, Nitric oxide (NO) radical and 2,2-diphenyl 1-picrylhydrazyl (DPPH) radical scavenging activities and the lipid peroxidation and thiobarbituric acid reaction (TBAR) methods. One way ANOVA was used for the result analysis with P<.05 for significant difference. Results: Mistletoes from Psidum guajava (MSPG) had significantly higher reducing property (0.16 – 0.20mg/mL); the chelating property of Mistletoes from Moringa olifera (MSMO) was significantly lower (45.7 – 58.9%); DPPH radical scavenging activity had no significant difference; and Nitric oxide scavenging activity was significantly higher in MSPG (72.1% in 75mg/mL) than the extracts from other hosts. MSPG had significantly higher TBAR inhibition using both FeSO4 (77.8% at 125µg/mL) and Sodium nitroprusside (61.6+1.0% at 125µg/mL) with an IC50 of 30.27µg/mL . Extract of Tapinanthus globiferus leaves from Psidium guajava had more antioxidative activities in the TBARs followed by Tapinanthus globiferus leaf extract from Vernonia amygdalina (MSVA). Conclusion: From the study, mistletoes from Psidium guajava had higher antioxidant activity compared to other hosts, which probably justifies its use for treatment of cancer in traditional medicinal practice.
