Penelitian bertujuan untuk mengevaluasi efek pemberian infusa benalu teh (Scurrula sp.) terhadap toksisitas akut lethal dose 50 (LD50) dan mengkaji perubahan histopatologi hati, ginjal, dan limpa mencit. Sebanyak 30 ekor mencit jantan dan 30 ekor mencit betina dibagi menjadi enam kelompok dan lima ulangan. Mencit percobaan yang tidak diberi infusa benalu teh dikelompokkan sebagai kontrol, sedangkan yang diberi infusa benalu teh dosis 1, 5, 10, 15, dan 20 g/kg BB dikelompokkan sebagai perlakuan. Pemberian infusa benalu teh dilakukan satu kali pada awal penelitian secara oral kemudian mencit diamati gejala klinis, mortalitas, dan bobot badan. Pada akhir penelitian (hari ke-14), mencit percobaan dikorbankan untuk dievaluasi secara histopatologi. Hasil penelitian menunjukkan bahwa pemberian infusa benalu teh pada mencit sampai dengan dosis 20 g/kg BB tidak menimbulkan kematian ataupun gejala klinis spesifik serta pertumbuhan normal. Gambaran histopatologi menunjukkan bahwa pemberian infusa benalu teh pada semua mencit percobaan sampai dengan dosis 20 g/kg BB tidak ditemukan adanya degenerasi maupun nekrosis pada organ hati, ginjal, maupun limpa. Kesimpulan dari penelitian ini menunjukkan bahwa infusa benalu teh dikelompokkan dalam sediaan praktis tidak toksik dan tidak menimbulkan kelainan pada organ hati, ginjal, maupun limpa.

ABSTRACT The aim of this research was to asses the acute toxicity of ethanolic extract of malaka leaves using lethal dose 50 (LD50) on mice (Mus musculus). Twenty male mice weighing between 20-30 g were randomly divided into 4 groups (group K1-K4) of 5 mice each. All mice in group K1, K2, K3, and K4 were administered ethanolic extract of malaka leaves with the dose of 2, 4, 8, and 16 g/kg bw, respectively. Single dose of ethanolic extract of malaka leaves were given by oral gavage prior to clinical observation . The observation period was 14 days post administration, for sign of toxicity symptom, weight loss, and mortality. The result showed that no mortality was observed in the experimental animals during this study. Slight reduction of body weight was observed in group K2, K3, and K4, and no toxicity sign was found during fourteen days of observation. The LD50 of ethanolic extract of malaka leaves was higher than 16 g/kg body weight, thus, the substance was practically non toxic substance.

According to WHO (2010), the Trimeresurus albolabris snake is a venomous snake with a high number of bite cases in Indonesia. Information on the character and level of toxicity of snake venom in Indonesia, including Trimeresurus spp. snakes, is still minimal. This study aims to determine the LD50 value (lethal dose 50), clinical symptoms, and to determine the macroscopic appearance of the liver and kidneys of mice (Mus musculus) after being injected by a the white-lipped pit viper (Trimeresurus albolabris) venom. The LD50 value of Trimeresurus albolabris venom injected subcutaneously into mice was 5,09 mg/kgBW. Clinical symptoms that appear increased movement activity, increased respiratory rate, lethargy, decreased activity, and reduced grooming. Macroscopic appearance of the liver that looks abnormal is found (1) color changes in the form of pale, pale yellowish, and red spots (petechiae). (2) changes in surface structure found only one liver that is not flat. (3) changes in consistency in the form of brittle easily. In the kidney that looks abnormal, found (1) changes in color in the form of pale. (2) changes in surface structure were only found in one kidney in the form of nodules. (3) changes in consistency in the form of brittle easily and soft.

The potential hazard of 1080 baiting for predators to 14 species of non-target mammals in the pastoral areas of Western Australia and a further six from Western Australia's Fitzgerald River National Park, was assessed by comparing projected doses of 1080 (based on consumption of non-toxic bait by captive animals in the absence of alternative food) with the approximate lethal dose of 1080 for each species. These figures suggested that individuals from 12 species were potentially at risk from crackle baits, while only individuals from Dasyurus hallucatus, Ningaui spp., Sminthopsis crassicaudata, Planigale maculata, one population of Leggadina forresti and one population of Sminthopsis ooldea were potentially endangered by meat baits. Tests using the native mammals Zyzomys argurus and Pseudomys hermannsbergensis and laboratory mice (Mus musculus) and laboratory rats (Rattus norvegicus) showed that individuals of all species reduced their consumption of toxic bait relative to non-toxic bait, although this did not prevent three of five rats and one of three P. hermannsbergensis from being killed.

ZusammenfassungEs werden die Fälle einer disseminierten letalen Toxoplasmose bei einer Farbmaus (Mus musculus) und einem Roten Panda (Ailurus fulgens) vorgestellt. Es handelte sich um eine als Haustier gehaltene Farbmaus und einen Roten Panda aus einem sächsischen zoologischen Garten. Die pathologische Untersuchung ergab bei beiden Tieren eine systemische Toxoplasmeninfektion. Eine hochgradige nekrotisierende Hepatitis stellte in beiden Fällen den histologischen Hauptbefund dar. Parasitenzysten fanden sich massenhaft in der Leber, in mäßiger Zahl im Gehirn und in geringer Zahl in anderen Organen. Mittels PAS-Reaktion waren diese Zysten bei der Farbmaus kaum darstellbar, beim Roten Panda dagegen sehr deutlich. PCR bzw. Immunhistologie bestätigten die Diagnose.

SummaryThe role of Hageman factor in triggering intravascular coagulation has been studied in rabbits injected intravenously with Liquoid. Besides changes of coagulation parameters characteristic of consumption coagulopathy (e.g. decrease in platelet counts, fibrinogen levels, factor V activity), a pronounced drop in Hageman factor activity was observed after injection of Liquoid. Likewise, the partial thromboplastin time became prolonged.The activation of Hageman factor in vivo could be prevented by intravenous infusion of lysozyme. Twenty min after starting the lysozyme infusion, the partial thromboplastin time became prolonged from a mean of 29 sec to 108 sec. Animals infused with lysozyme and injected with a lethal dose of Liquoid did not develop a consumption coagulopathy. In the same manner, none of 10 animals treated with lysozyme developed the generalized Shwartzman reaction, whereas in the control group 19 out of 20 animals showed fibrin thrombi in the glomerular capillaries.From the present study it may be concluded that the intravascular coagulation process after intravenous injection of Liquoid is triggered by Hageman factor activation.
