Abstract Background Clinical outcomes vary among young people with the psychosis risk syndrome (PRS), with approximately 20% of individuals progressing to a psychotic disorder over 2–3 years and 30% achieving clinical remission. The identification of neurophysiological abnormalities associated with schizophrenia that predate and predict psychosis onset may enhance the accuracy of clinical outcome prediction in the PRS and help elucidate the pathogenic mechanisms of psychosis onset. Auditory P300 event-related potential (ERP) component amplitude reductions are well established in schizophrenia and reflect early attention-mediated information processing deficits. Recent studies employing auditory oddball tasks have shown that P300 amplitude deficits in PRS individuals are associated with later clinical outcomes, including both conversion to full-blown psychosis and remission from the at-risk state. The present study examined whether these effects extend to P300 in the visual modality using visual oddball task data collected as part of the North American Prodrome Longitudinal Study. Specifically, we evaluated whether visual P300 amplitudes are reduced in the PRS and predict future clinical outcomes. Methods 540 individuals meeting PRS criteria and 229 healthy individuals completed baseline EEG recording during a visual oddball task. Visual P300 subcomponents were measured in response to two stimulus types: (1) infrequent target stimuli, reflecting top-down allocation of attention (target P3b), and (2) infrequent non-target novel distractor stimuli, reflecting bottom-up orienting of attention (novelty P3a). P300 amplitudes of PRS participants who converted to psychosis (n=70) were compared with those of PRS non-converters who were followed clinically for 24 months and continued to be symptomatic (n=131) or fully remitted from the PRS (n=87). Results Group comparison effects did not differ by stimulus type. Visual P300 amplitudes were not significantly reduced in the PRS group relative to healthy individuals (p=.25). However, baseline target P3b and novelty P3a amplitudes were reduced in PRS individuals who later converted to psychosis relative to all PRS non-converters, including those who remitted (p=.006, d=.44) and those who remained symptomatic (p=.015, d=.37), as well as healthy individuals (p=.001, d=.44). Baseline P300 amplitudes were similar among healthy controls, PRS remitters, and PRS individuals who remained symptomatic (ps>.45). Moreover, visual P300 amplitudes differentiated future psychosis converters after accounting for PRS symptom severity. Finally, both target P3b and novelty P3a amplitudes predicted the time to psychosis onset in PRS participants (p=.03 and p=.02, respectively), such that more deficient P300 amplitudes were associated with shorter time to conversion. Discussion Baseline visual P300 amplitudes were reduced in future PRS converters relative to non-converters, with effect sizes comparable to those reported in previous auditory P300 studies of the PRS. Results implicate visual P300 as a neurophysiological vulnerability marker that predicts clinical outcomes among PRS individuals, including future transition to psychosis. Accordingly, together with prior auditory P300 studies, results suggest that P300 may have the potential to contribute to personalized early intervention in the PRS by distinguishing individuals with the greatest risk for psychotic illness, who require the most aggressive treatment, from those who may need minimal intervention.

Abstract The P300 component of the event-related potential was recorded during two blocks of an autobiographical oddball task. All participants performed honestly during the first block (Phone), i.e., the oddball stimuli were phone numbers. During the second block (Birthday), in which the oddball stimuli were participants' birthdays, a Truth group (N = 13) performed honestly and a Malinger group (N = 14) simulated amnesia. Amnesia simulation significantly reduced P300 amplitudes, both between groups and within the Malinger group (Phone vs. Birthday), possibly because of an increase in task difficulty in the Malinger condition. Analysis of scaled amplitudes also indicated a trend for a feigning-related alteration in P300 topography. Bootstrapping of peak-to-peak amplitudes detected significantly more (93%) Malinger individuals than bootstrapping of baseline-to-peak amplitudes (64%). Bootstrapping also provided evidence of a feigning-related amplitude difference between oddball stimuli (i.e., Phone > Birthday) in 71% of Malinger group individuals. In this comparison, the peak-to-peak measure also performed significantly better in intraindividual diagnostics.

The P300 event-related potential is an index of attentional resources related to target detection. Source localization and functional magnetic resonance imaging (fMRI) research has indicated that, among other regions, the prefrontal cortex contributes to the generation of the P300. Similar to fMRI, near infrared (NIR) spectroscopy measures change in blood oxygen levels, but offers several advantages including portability, low expense, and superior temporal resolution. No studies to date have examined the extent to which prefrontal cortex NIR spectroscopy measures are active during the P300 paradigm. To address this knowledge gap, participants completed a two-difficulty visual oddball task in which NIR spectroscopy and P300 data were collected in a counterbalanced order. Confirmatory results indicate that the P300 event-related potential is attenuated as a function of task difficulty. Similarly, NIR spectroscopy measures of oxygenated hemoglobin in the right medial prefrontal cortex are attenuated as a function of task difficulty. The results suggest that prefrontal cortex NIR spectroscopy measures are sensitive to task difficulty in a visual P300 oddball task.

Background: The other-race categorisation advantage (ORCA) is a well-established phenomenon, whereby other-race faces are categorised faster than own race faces. Objectives: This study investigated whether extraverts would demonstrate an ORCA-like effect toward unfamiliar other-race faces and familiar other-race faces in a modified oddball and choice reaction paradigm.  Methods: This event-related potential (ERP) study employed a repeated measures experimental design with one independent variable (racial familiarity) and three levels (familiar other-race/Malay faces, unfamiliar other-race/African faces, control group/furniture photos). In the oddball task, African faces and Malay faces were the target stimuli and furniture photos were the standard stimuli. Electroencephalography data (EEG) was collected during the oddball task, from which ERP components were derived. Results: The reaction time (RT) for African and Malay faces were not significantly different.  Significant effect of racial familiarity on P300 latencies at all electrode sites was not observed.  However, there was a significant effect of racial familiarity on P300 amplitudes at midline electrodes (Cz).  It was also observed that the P300 amplitude was larger for African faces than Malay faces at midline electrodes (Cz). Conclusion: An ORCA-like effect was not found in categorisation tasks involving faces from a familiar and an unfamiliar other-race, but a larger P300 amplitude was evoked by African faces. This dissociation between RT and P300 amplitude provided important theoretical implications with regard to models associated with ORCA. Specifically, the current findings lent support to the social cognition model and the Categorisation-Individuation Model (CIM).

The effects of self evaluation on the P300 event-related potential (ERP) were explored with 56 participants (16 men, 40 women; M age = 23.4 yr., SD = 1.2) across three conditions. The conditions included (1) a standard ERP auditory oddball discrimination between a random target (15% occurrence) and standard stimuli (85% occurrence), (2) the oddball task followed by the additional cognitive task of maintaining a mental count of the target tones, and (3) the oddball task followed by the additional cognitive task of self-evaluating whether they felt surprised by the current occurrence of the target tone. The added cognitive requirements for Conditions 2 and 3 required the subjects to maintain a cognitive readiness for the secondary stimulus-related task during their sensory discrimination response for the standard oddball task. During the self-evaluation condition, the P300 amplitude was significantly larger across all recording locations than the regular oddball condition and the cognitive count condition.

IntroductionThe effects of chronic antipsychotic administration on the human brain are debated. In particular, first-generation (FGAs) and second-generation antipsychotics (SGAs) seem to have different impacts on brain function and structure in subjects with schizophrenia. Few studies have investigated the effect of chronic administration of FGAs and SGAs on indices of brain function, such as event-related potentials (ERP) or neuropsychological performance.ObjectivesWithin the Italian Network for Research on Psychoses study, subjects stabilized on FGAs or SGAs were compared on P300, an ERP component, thought to reflect attention, working memory and context integration and on neurocognitive indices.MethodsERPs were recorded in 110 chronic, stabilized patients with Schizophrenia (28 used FGAs) during a standard auditory oddball task. P300 latency and amplitude were assessed at Pz channel. MATRICS Consensus Cognitive Battery (MCCB) was used for cognitive assessment.ResultsCompared with the SGAs group, patients on FGAs showed significant increased P300 latency (P = 0.003; Cohen's d = 0.67) and significant decreased P300 amplitudes (P = 0.023; Cohen's d = 0.38). The two groups did not differ on psychopathology and MCCB scores. Multiple linear regressions revealed that “FGAs vs. SGAs” (β = 0.298, P = 0.002) and MCCB neurocognitive composite T-score (β = –0.273, P = 0.004) were independent predictors of P300 latency, whereas only age (β = –0.220, P = 0.027) was an independent predictor of P300 amplitude.ConclusionsFGAs seem to affect the functional brain activity more than SGAs, particularly slowing cortical processing. Our results suggest that discrepant findings concerning P300 latency in schizophrenia might be related to the type of antipsychotic treatment used.
