<p>Pengembangan standar berbasis bioteknologi pada sektor agroindustri dilakukan melalui peningkatan mutu laboratorium merupakan bagian infrastruktur standardisasi. Kondisi laboratorium bioteknologi di Indonesia yang diindikasikan di lapangan antara lain, metode uji, bahan acuan dan sebagian kompetensi SDM yang bekerja di laboratorium bioteknologi beragam. Penelitian ini dilakukan untuk memperkuat pengembangan metode uji, bahan acuan, dan standar SDM untuk  mendukung pengembangan lingkup akreditasi laboratorium bioteknologi di Indonesia. Penelitian dilaksanakan dengan menggunakan <em>desk study</em> dan FGD. Berdasarkan hasil penelitian, dapat disimpulkan bahwa beberapa acuan standar Internasional dapat dijadikan sebagai acuan dalam penembangan Standar Nasional Indonesia terkait deteksi GMO dan spesifik DNA  spesies antara lain, ISO 21569:2013 (<em>Qualitative nucleic acid based methods</em>), ISO 21570:2013<em> </em>(<em>Qua</em><em>nti</em><em>tative</em><em> </em><em>nucleic acid based methods</em>), ISO 21571:2013 (<em>Nucleic acid extraction</em>), dan ISO 21572:2013 (<em>Protein</em><em> based methods</em>), Selain itu, metode uji yang dikembangkan oleh <em>European Union Laboratory for GM Food and Feed </em>dan ENGL dapat digunakan sebagai rujukan dalam pengembangan SNI. Pengembangan bahan acuan pengujian GMO dan spesifik DNA dapat dirujuk dari <em>Institute Materials and Measurements</em> yang merupakan salah satu <em>Joint Research Centre EURL-GMFF</em> maupun LGC. InaCC di bawah Pusat Penelitian Biologi LIPI dapat dijadikan sebagai Bank RM sesuai skema yang dikonsepkan oleh KAN dilihat dari kesiapan dan kemampuannya. Beberapa rujukan standar SDM merujuk pada Peraturan bersama Kemendiknas dan Kepala BKN Nomor 02/V/PB/2010 dan Nomor 13 Tahun 2010 tanggal 6 Mei 2010 menjelaskan terkait dengan jabatan fungsional Pranata Laboratorium Pendidikan dan SKKNI bidang Jasa Pengujian Laboratorium merujuk poin MSL957014A mengenai <em>Perform Moleculer Biology Tests Procedures.</em></p>

VUCA world (Volatility, Uncertainty, Complexity, Ambiguity), according to experts of JRC (European Commission's Joint Research Centre), is now a special space that corrupts the integrity of science activity, limits thinking and vision of the situation by politicians. In fact, under such conditions, the only reliable resource for scientific organizations is a commercialization, and the payback is a condition of activity. These factors already pose a threat to global security. The mission orientation proposed by European Union experts is still seen as a rather weak alternative.

Studying the impacts of climate change requires looking at a multitude of variables across a broad range of sectors [1,2]. Information on the variables involved is often unevenly available or offers different uncertainties [3,4], and a lack of uniform terminology and methods further complicates the process of analysis, resulting in communication gaps when research enterprises span different sectors. For example, models designed by experts in one given discipline might assume conventions in language or oversimplify cross-disciplinary links in a way that is unfamiliar for scientists in another discipline. Geospatial Semantic Array Programming (GeoSemAP) offers the potential to move toward overcoming these challenges by promoting a uniform approach to data collection and sharing [5]. The Joint Research Centre of the European Commission has been exploring the use of geospatial semantics through a module in the PESETA II project (Projection of economic impacts of climate change in sectors of the European Union based on bottom-up analysis). <BR/>This manuscript has been accepted for publication in IEEE Earthzine 2014 Vol. 7 Issue 2, 2nd quarter theme: Geospatial Semantic Array Programming. This study aimed to assess standard and harsher nucleic acid extraction schemes for diagnostic helminth real-time PCR approaches from stool samples. A standard procedure for nucleic acid extraction from stool and a procedure including bead-beating as well as proteinase K digestion were compared with group-, genus-, and species-specific real-time PCR assays targeting helminths and nonhelminth pathogens in human stool samples. From 25 different in-house and commercial helminth real-time PCR assays applied to 77 stool samples comprising 67 historic samples and 10 external quality assessment scheme samples positively tested for helminths, higher numbers of positive test results were observed after bead-beating-based nucleic acid extraction for 5/25 (20%) real-time PCR assays irrespective of specificity issues. Lower cycle threshold values were observed for one real-time PCR assay after the standard extraction scheme, and for four assays after the bead-beating-based scheme. Agreement between real-time PCR results after both nucleic acid extraction strategies according to Cohen’s kappa ranged from poor to almost perfect for the different assays. Varying agreement was observed in eight nonhelminth real-time PCR assays applied to 67 historic stool samples. The study indicates highly variable effects of harsh nucleic acid extraction approaches depending on the real-time PCR assay used.

SARS-CoV-2 RT-PCR with pooled specimens has been implemented during the COVID-19 pandemic as a cost- and manpower-saving strategy for large-scale testing. However, there is a paucity of data on the efficiency of different nucleic acid extraction platforms on pooled specimens. This study compared a novel automated high-throughput liquid-based RNA extraction (LRE) platform (PHASIFYTM) with a widely used magnetic bead-based total nucleic acid extraction (MBTE) platform (NucliSENS® easyMAG®). A total of 60 pools of nasopharyngeal swab and 60 pools of posterior oropharyngeal saliva specimens, each consisting of 1 SARS-CoV-2 positive and 9 SARS-CoV-2 negative specimens, were included for the comparison. Real-time RT-PCR targeting the SARS-CoV-2 RdRp/Hel gene was performed, and GAPDH RT-PCR was used to detect RT-PCR inhibitors. No significant differences were observed in the Ct values and overall RT-PCR positive rates between LRE and MBTE platforms (92.5% (111/120] vs 90% (108/120]), but there was a slightly higher positive rate for LRE (88.3% (53/60]) than MBTE (81.7% (49/60]) among pooled saliva. The automated LRE method is comparable to a standard MBTE method for the detection of SAR-CoV-2 in pooled specimens, providing a suitable alternative automated extraction platform. Furthermore, LRE may be better suited for pooled saliva specimens due to more efficient removal of RT-PCR inhibitors.
