Root exudation the ecological driver of hydrocarbon rhizoremediation

Rhizoremediation is a bioremediation technique whereby microbial degradation of organiccontaminants occurs in the rhizosphere. It is considered to be an effective and affordable “greentechnology” for remediating soils contaminated with petroleum hydrocarbons. Root exudation ofa wide variety of compounds (organic, amino and fatty acids, carbohydrates, vitamins, nucleotides,phenolic compounds, polysaccharides and proteins) provide better nutrient uptake for the rhizospheremicrobiome. It is thought to be one of the predominant drivers of microbial communities in therhizosphere and is therefore a potential key factor behind enhanced hydrocarbon biodegradation. Manyof the genes responsible for bacterial adaptation in contaminated soil and the plant rhizosphere arecarried by conjugative plasmids and transferred among bacteria. Because root exudates can stimulategene transfer, conjugation in the rhizosphere is higher than in bulk soil. A better understanding ofthese phenomena could thus inform the development of techniques to manipulate the rhizospheremicrobiome in ways that improve hydrocarbon bioremediation.

Background:: Nuts have been incorporated into guidelines for healthy eating since they contain considerable amounts of antioxidants and their effects are related to health benefits since they contribute to the prevention of nutritional deficiencies. The micronutrient characterization is based mainly on the determination of phenolics which is the most abundant class of bioactive compounds in nuts. Terpenes constitute another class of bioactive compounds that are present in nuts and show high volatility. The analysis of phenolic compounds and terpenes are very demanding tasks that require optimization of the chromatographic conditions to improve the separation of the components. Moreover, nuts are rich in unsaturated fatty acids and they are therefore considered as cardioprotective. Gas chromatography is the predominant instrumental analytical technique for the determination of derivatized fatty acids and terpenes in food matrices, while high performance liquid chromatography is currently the most popular technique for the determination of phenolic compounds Objective:: This review summarizes all the recent advances in the optimization of the chromatographic conditions for the determination of phenolic compounds, fatty acids and terpenes in nuts Conclusion:: The state-of-the art in the technology available is critically discussed, exploring new analytical approaches to reduce the time of analysis and improve the performance of the chromatographic systems in terms of precision, reproducibility, limits of detection and quantification and overall quality of the results

Background:: Luffa cylindrica is a plant that is widely distributed in Africa and Asia and it can be grown in regions with tropical or subtropical climates. Few patents dealt with Loofah biological properties, including some functional foods formulated from its leaves. Objective:: This study aimed to structurally and functionally characterize the bioactive compounds of L. cylindrica leaves grown in two different environments. Methods:: The extracts of L. cylindrica leaves collected from two Tunisian locations: Essouasi (LE), a semi-arid region and Medenine (LM) an arid region, were investigated for their phenolic compounds and fatty acids using HPLC/TOF-MS and GCMS techniques respectively. Furthermore, the antioxidant capacity was evaluated with DPPH, Chelating effect, Hydroxyl radical and Superoxide anion scavenging activities while the anticancer activity against HeLa cell lines was assessed using xCELLigence real time cell analyzer and lactate dehydrogenase cytotoxicity assay. Results:: The antiproliferative capacity of both extracts was time and dose-dependent with LE presenting the lowest HeLa cell index (CI = 0.035 ± 0.018, 250 μg/ml). LE also showed the best cytotoxic capacity (56.49 ± 0.8%) and antioxidant potential (IC50 = 54.41 ± 1.12 μg/ml for DPPH and 12.12 ± 0.07 μg/ml for chelating effet). 14 phenolic compounds were detected in LE with ferulic acid being the major compound (5128.5 ± 4.09 μg Phenols/g) while LM had only 6 phenolics. GCMS analysis showed the presence of omega-3 fatty acids in LE. Conclusions:: Our findings suggest that L. cylindrica leaves, especially when collected from semi-arid regions, are promising for formulating nutraceuticals of interest.

The present study aimed at the identification and quantitation of phenolic compounds, fatty acids, and further characteristic substances in the seeds of Geum urbanum L. and Geum rivale L. For this purpose, individual components of extracts recovered with MeOH, CH2Cl2, and by cold-pressing, respectively, were characterized by HPLC-DAD/ESI-MSn and GC/MS and compared with reference compounds. For both Geum species, phenolic compounds, such as flavonoids and gallic acid derivatives, and triterpenes, such as saponins and their aglycones, were detected. Surprisingly, both Geum species revealed the presence of derivatives of the triterpenoid aglycons asiatic acid and madecassic acid, which were characterized for the first time in the genus Geum. Furthermore, the fatty acids of both species were characterized by GC–MS after derivatization. Both species showed a promising fatty-acid profile in terms of nutritional properties because of high proportions of unsaturated fatty acids. Linoleic acid and linolenic acid were most abundant, among other compounds such as palmitic acid and stearic acid. In summary, the present study demonstrates the seeds of G. urbanum and G. rivale to be a valuable source of unsaturated fatty acids and bioactive phenolics, which might be exploited for nutritional and cosmetic products and for phytotherapeutic purposes.

The effect of compost on phytoremediation of diesel-contaminated soils was investigated using 130 small (200 g) containers in two screening tests. The experiments were conducted in a controlled environment using ryegrass from seed. Containers were destructively sampled at various times and analyzed for plant mass and total petroleum hydrocarbons. The results indicate that the presence of diesel reduces grass growth, and that compost helps reduced the impact of diesel on grass growth. The addition of compost helps increase diesel loss from the soils both with and without grass, though the addition of grass leads to lower diesel levels compared with controls. A second set of experiments indicates that the compost helps in phytoremediation of diesel-contaminated soil independent of the dilution effect that compost addition has. The results indicate that the compost addition allowed diesel loss down to 200 mg TPH/kg even though the compost would be expected to hold the diesel more tightly in the soil/compost mixture. The simplicity of the screening tests led to difficulties in controlling moisture content and germination rates. The conclusion of the research is that the tilling of compost into soils combined with grass seeding appears to be a valuable option for treating petroleum-contaminated soils.
