Different parts of eleven medicinal plants used in the traditional medicine in Puthan district were collected. Three different extracts, methanol, 50% aqueous methanoland 70% aqueous acetone extractswere prepared for each plant material. The methanol extractswere screened for the presence of different classes of phytochemicals. Total phenolic andflavonoid content, as well as DPPH freeradical scavenging activity, were evaluated. All the investigated extracts contain a high amount of phenolics and flavonoids.The highest amount of phenolicsand flavonoids were detected in the methanolextract of bark of Bauhinia variegate(355.35+3.69 mg GAE/g and 209.23 ±1.25 mg QE/g extract). Among the eight tested extracts, the highest radical scavenging activity was shown by methanol extract of bark of B. variegate (IC50 3.68 μg/ml). The extract having the highest phenolic and flavonoid content showed the lowest IC50 demonstrating the positive correlation between radical scavenging activity and total phenolic and flavonoid content.

Objective: The aim of this study is to evaluate the antioxidant potential, determination of total phenolic and flavonoid content in nine selected medicinal plants Spondias pinnata, Melia azedarach, Ageratina adenophora, Urtica dioica, Curcuma longa, Bauhinia variegata, Elaeocarpus angustifolius Blume, Achyranthes aspera, and Psidium guajava from Kavre district of Nepal using in vitro studies. Methods: Methanolic plant extracts were prepared by cold percolation method. The methanol extract of nine medicinal plants collected from Kavre district of Nepal, was screened for assessing bioactive phytoconstituents followed by antioxidant property, total phenolic, and flavonoid content. Different plants collected were powdered and extracted with methanol, concentrated by a rotatory evaporator and analyzed for the presence of phytochemicals. The antioxidant potential of the plant extracts was evaluated by 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay. Results: The phytochemical analysis of methanolic extracts of all nine medicinal plants displayed the presence of various secondary metabolites such as alkaloids, flavonoids, polyphenols, saponins, and quinones. The extract of S. pinnata showed the highest percentage of radical scavenging activity up to 87.94±1.88 with 50% inhibitory concentration (IC50) 17.51±1.27 μg/mL, followed by B. variegata, 80.63±1.06 with IC50 value 26.55±2.61 μg/mL. The standard, ascorbic acid has IC50 value of 20.13±1.17 μg/mL. Further, the ethyl acetate fraction of S. pinnata showed the maximum percentage of radical scavenging (85.92±1.37) with IC50 value of 46.95±1.17 μg/mL. Moreover, S. pinnata displayed the highest total phenolic content (TPC) 48.26±1.23 mg GAE/g (milligram gallic acid equivalent per gram) extract while the highest flavonoid content was displayed by Melia azedarach 41.07±1.53 mg QE/g (milligram quercetin equivalent per gram) extract measured by the Folin–Ciocalteu reagent method and aluminum chloride colorimetric method. Conclusions: The preliminary results of this study have put forward the extract of S. pinnata showed the highest percentage of radical scavenging activity and S. pinnata displayed the highest TPC while the highest flavonoid content was displayed by Melia azedarach methanolic extracts although the further studies are needed to assess its mechanism of action.

Quercetin was isolated from ethyl acetate fraction of a methanol extract of the outer scale of onion by repeated sephadex LH-20 column chromatography. Methanol, 50% aqueous methanol and 70% aqueous acetone extracts of different parts of four medicinal plants, Ficus benghalensis, Elaeocarpus sphaericus, Ipomea carnea and Azeratum conyzoides were prepared and screened for total phenolic and flavonoid contents and free radical scavenging activity. The total phenolics contents were measured spectrophotometrically by using Folin-Ciocalteu reagent and total flavonoids content by using aluminum chloride colorimetric method. Free radical scavenging activity was evaluated using 2,2-diphenyl-1-picryl-hydrazyl (DPPH) assay. All the investigated medicinal plant extracts contained high amount of phenolics. The highest amount was detected in 70% aqueous acetone extract of E. sphaericus (298.8±9.03 mg GAE/g extract) and lowest amount in 50% aqueous methanol extract of F. benghalensis (6.7±0.73 mg GAE/g extract). The highest amount of flavonoid was found in methanol extract (78.2±2.72 mg quercetin/g extract) and the lowest amount was detected in 50% aqueous methanol extract (2.1±0.25 mg quercetin/g extract) of F. benghalensis. DPPH free radical scavenging assay was carried out only in 70% acetone extracts. The highest IC50 value was observed for E. sphaericus (34.0 μg/ml). Citrus fruits exhibit a high level of different phytoconstituents, of which the changes in the different parts of the fruit during ripening have not been thoroughly studied yet. Thus, in this study, we have investigated how different parts of pomelo fruit (Citrus grandis L.) are modified throughout the development of two consecutive growing seasons. In detail, the main phytochemical compounds, such as total phenolic content, total flavonoid content, antioxidant capacity, DPPH free radical scavenging activity, Ferric reducing antioxidant power (FRAP), and naringin and tannin content, were analyzed. A systematic metabolism of these compounds was found during the development of the fruit, but some pomelo tissues showed a fluctuating trend, suggesting a dependence on the different growing season. Focusing on the tissue distribution of these compounds, the fruit membrane contained the highest level of total phenolic and flavonoid content; fruit flavedo displayed the highest antioxidant capacities and FRAP activities, whereas maximum accumulation of naringin was noticed in fruit albedo. Instead, the highest DPPH free radical scavenging activity and tannin contents were found in the pomelo juice. Regarding the distribution of compounds, a possible bias pattern for the accumulation of those compounds has been noticed throughout the fruit development. From the GC-MS analysis, a total of 111 compounds were identified, where 91 compounds were common in both seasons. Overall, these results could be useful for the food processing industry as guidelines for excellent quality foods and for introducing health-beneficial products and components into our daily diets.

Antioxidant and free radical scavenging activity of methanol, ethanol, ethyl acetate and chloroform extracts of aerial parts of Lamium purpureum L. was determined by DPPH, ABTS, FRAP and TRP assays. Contents of flavonoids and phenols were also investigated. The total phenolic content in the extracts, determined using Folin?Ciocalteu assay, ranged between 8.57 to 128.00 mg GAE/g d.e. while concentrations of flavonoids in the extracts varied from 24.20 to 39.80 mg QuE/g d.e. The highest phenolic content was found in methanol extract (128.00 mg GAE/g d.e.). The highest content of total flavonoids was identified in the methanol extract (39.80 mg QuE/g d.e.) and the lowest was in the chloroform (24.30 mg QuE/g d.e.). DPPH scavenging of the extracts was determined and obtained IC50 values ranged from 0.12 to 3.12 mg/mL of solution. The values of ABTS radical scavenging activity ranged from 0.35 to 1.80 mg AA/g. The highest ABTS antiradical activity was registered for methanol extract. The FRAP value was found within the range 0.08 to 1.04 ?mol Fe/mg. The best radical scavenger was methanol (1.04 ?mol Fe/mg). In reducing power assay different extracts of L. purpureum showed increasing of activity with increased concentration, and all extracts possessed substantial dose dependent antioxidant activity. The best reducing capacity was obtained with methanol extract of L. purpureum (0.0132 mg AA/mL). The results in this study confirmed that L. purpureum possesses moderate antioxidant properties.

  Objectives: The goals of this research were to observe antioxidant properties from different parts of Simana lagi apple ([Malus domestica Borkh.] “Simana lagi”) using two antioxidant testing methods which were 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2’-azino-bis(3-ethylbenzthiazoline-6- sulfonic acid (ABTS) and correlation of total phenolic and flavonoid content with their inhibitory concentration 50 (IC50) of DPPH and IC50 of ABTS.Methods: Each sample was extracted by reflux using different polarity solvents. The extracts were evaporated using rotary evaporator. Antioxidant activities were tested using DPPH and ABTS assays, determination of total phenolic, and flavonoid content were carried out by ultraviolet-visible spectrophotometry and correlation with their IC50 of DPPH and IC50 of ABTS activities were analyzed by Pearson’s method.Results: The ethanolic leaves extract (LV3) of Simana lagi apple also exposed the highest total phenolic content (TPC) (13.88 g gallic acid equivalents/100 g), while the highest total flavonoid content was presented by ethyl acetate extract (LV2) (7.21 g QE/100 g). The lowest IC50 of DPPH scavenging activity 0.19 μg/ml, and the lowest IC50 of ABTS scavenging activity 0.15 μg/ml was given by ethanolic flesh extract (FL3) of Simana lagi apple. There were significantly negative correlation between TPC in all different parts extracts of Simana lagi apple with their IC50 of DPPH and IC50 of ABTS.Conclusions: All different parts extracts of Simana lagi apple were categorized as very strong antioxidant by DPPH and ABTS method (except n-hexane peels extract and n-hexane LV by DPPH method, and n-hexane FE by ABTS method). The major contributor in antioxidant activities of peels, flesh and leaves extracts of Simana lagi apple by DPPH and ABTS methods were phenolic compounds. Antioxidant activities of peels, flesh and leaves extracts of Simana lagi apple showed linear results by DPPH and ABTS methods.

Background: More than 700 plants are reported to be used for medicinal purposes in Nepal; however, many of them are not studied for their scientific evidences. The aims of the present study were the estimation of the total phenolic and flavonoid contents, and the evaluation of the free radical scavenging and α-amylase inhibitory activities of five selected medicinal plants from western Nepal: Aeschynanthus parviflorus Wall. (Gesneriaceae), Buddleja asiatica Lour. (Loganiaceae), Carica papaya L. (Caricaceae), Drepanostachyum falcatum (Nees) Keng f. (Gramineae) and Spondias pinnata (L. f.) Kurz (Anacardiaceae). Methods: The total phenolic content (TPC) and total flavonoid content (TFC) were measured using Folin-Ciocalteu’s phenol reagent and aluminium chloride methods, respectively. A 1,1–diphenyl–2–picrylhydrazyl (DPPH) free radical scavenging assay was used to evaluate the free radical scavenging activity and an α-amylase inhibitory assay was carried out to determine the in vitro antidiabetic activity. Results: The phytochemical screening of five hydroalcoholic plant extracts revealed the presence of various secondary metabolites, including alkaloids, flavonoids, reducing sugars, saponins, terpenoids and tannins. The amounts of total phenolics and flavonoids were found to be the highest in B. asiatica leaf extract, which also showed the most potent free radical scavenging activity. Extract of C. papaya fruits showed the highest α-amylase inhibitory activity, whereas the extracts of B. asiatica leaves and S. pinnata leaves exhibited moderate activity. Conclusions: Some of the medicinal plants selected in this study showed high TPC and TFC values and potent bioactivities. These results may provide the scientific evidences of the traditional uses of these plants. However, further detailed studies on bioactive compounds isolation and identification and evaluation of in vivo pharmacological activities should be performed in future.

Abstract Seaweed is one of the marine-biota that is widely known for its uses, one of them is in the food functional. Functional food is a program that promotes a healthy diet so that it can prevent a disease when consuming food. With functional food, daily eating patterns can be maintained and also the content of the food consumed daily has a good effect on the body. It is known that in seaweed, especially brown algae (S. crassifolium), it contains antioxidants and other compounds that are good for the health. In this study, phytochemical content and proximate composition was evaluated in S. crassifolium. Including total flavonoid content and total phenolic content, antioxidant capacity both of DPPH and ABTS assay used three type of solvents such as Ethanol, n-Hexana, and ethyl acetate for comparable the best solvents. Also, proximate analysis to determine the content of moisture content, crude fat content, crude protein content, and carbohydrates content. The results showed that n-Hexana and ethyl acetate solvents were significantly than ethanol. Which mean both of them had highest values of total flavonoid content (TFC), total phenolic content (TPC), and antioxidant activity of DPPH radical scavenging activity and ABTS radical scavenging activity. The results of chemical composition in proximate compounds showed that S. crassifolium was suitable to be candidate of functional food.

Plant based remedies are of much importance in healthcare due to low side effects. The effective constituents contained in seeds of a papaya have not been utilized efficiently in the production of medicines. The purpose of this study was driven towards determining the total phenolic content, total flavonoid content, antioxidant capacity, anti-inflammatory activity and antibacterial properties of the aqueous seeds extract of a Sri Lankan variety (Red Lady) of papaya (AESP). The AESP prepared according to the method of “Kasaya” in Ayurvedic medicine was used for the investigations according to standard procedures. Total phenolic content and the total flavonoid content of the AESP were 13.5±2.2 mg (pyrogallol equivalence)/g and 315.9±104.6 mg (quercetin equivalence)/g respectively. The 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity of AESP ranged from 12.4% to 81.2% for concentrations of 1.0-20.0 mg/ml; ascorbic acid gave 31.7% - 91.9% activity. The AESP displayed strong reducing power compared to ascorbic acid in the reducing power assay. The hydroxyl radical scavenging activity of the AESP ranged from 6.6% to 69.1% for concentrations ranging from 20.0-95.0 mg/ml; ascorbic acid activity was 37.7%-74.9%. The nitric oxide radical scavenging activity of AESP was 23.9%-62.7% for concentrations of 2.0-20.0 µg/ml; ascorbic acid gave 26.8%-63.5% activity. AESP concentrations ranging from 75.0-150.0 µg/ml exhibited human red blood cell (HRBC) membrane stabilization protection of 15.5%-22.7% compared to 50.8%-58.4% for aspirin. The AESP showed antibacterial activity against Bacillus subtilis and Staphylococcus aureus bacterial species. AESP possess promising antioxidant, anti-inflammatory and antibacterial activity. The aqueous decoction of the discarded seeds of papaya would facilitate remedies for many diseases in which radicals are implicated as well as assist against certain bacterial infections and also has an anti-inflammatory potential.

The phytochemical contents of Salvia virgata mericarps and the antioxidant and antimicrobial activities of its ethanol extract were studied for the first time. S. virgata mericarps were collected in the province of Trabzon, Turkey. Chemical analyses were performed using chromatographic methods. The total phenolic and flavonoid contents and antioxidant potential were measured using Folin-Ciocalteu, Al(NO3)3, and DPPH radical scavenging activity methods, respectively. The antimicrobial activity was evaluated using the microdilution method for all the tested bacterial and fungal strains, with the exception of Mycobacterium tuberculosis for which the resazurin microtiter plate method was applied. The mericarps were rich in glutamic acid (3934 mg/100 g), potassium (12578.8 ?g/g), calcium (12092.0 ?g/g), and dietary fibers (35.565 g/100 g). The total phenolic and flavonoid contents were 2.50 ?g GAE/mg extract and 0.34 ?g QE/mg extract, respectively. The most effective DPPH free radical scavenging activity determined for the highest applied concentration was 92.44%. The ethanol extract obtained from the mericarps was found to be as effective as the reference drug ampicillin (MIC value = 125 ?g/mL) against the nosocomial pathogen Acinetobacter baumannii. In conclusion, S. virgata mericarps provide good nutritional value with low amounts of carbohydrates and high dietary fibers, amino acids, minerals and total phenolic and flavonoid contents and medicinal properties.

Polyalthia longifolia (masquerade tree) is a plant which is believed to possess varied pharmacological and therapeutic values among different populations. The present report investigated the phytochemical composition, proximate, acute toxicity and antioxidant potential of P. longfolia root. All analyses were carried out using established methods; the antioxidant activity of the crude methanol extract and its fractions (n-hexane and ethyl acetate) were examined using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay while the total phenolic and flavonoid contents were assessed using the Folin-Ciocalteu and the aluminum chloride calibration methods respectively. The phytochemical analysis revealed the presence of alkaloids, carbohydrate, reducing sugars, tannins, saponins, flavonoids, phenolic compounds and protein in aqueous extract. The proximate analysis showed moisture content, total ash, alcohol extractive value, water extractive value, acid insoluble ash and water soluble ash at 8.80, 9.35, 3.28, 3.29, 2.27 and 7.29% respectively. The ethyl acetate fraction showed the highest antioxidant property compared to the n-hexane fraction and crude methanol extract in all assays conducted. Also, the methanol fraction was found to have the highest flavonoids and phenolic content among the extract and fractions. Oral administration of crude methanol extract of P. longifolia to Swiss mice was relatively non-toxic at a maximum dose of 5000 mg/kg. The root extract and fractions of P. longifolia indicated moderately high level of some phytochemicals with outstanding radical scavenging activity, and therefore substantiate its use as a conventional and comparatively non-toxic plant antioxidant.
