An efficient protocol was developed for plant regeneration, multiplication and rooting under in vitro condition in pointed gourd. Highest percent of shoot regeneration was 91.66%, when nodal explants were cultured on MS+1.0 mg/1 BAP. The maximum number of shoots (4.8) per explant was observed in MS + 1.0 mg/1 BAP + 0.2 mg/1 NAA from nodal segment. Among the two explants, nodal segment was found better for shoot regeneration and multiplication. The best response towards root induction was achieved on half MS medium supplemented with 0.4 mg/1 NAA. The regenerated healthy rooted plantlets were transferred to small plastic pot containing garden soil and compost in a ratio of 2:1. Immediately after transplantation the plantlets were covered with polythene bag to prevent desiccation. After acclimatization the plantlets were successfully transferred to the garden soil and the percentage of survivability in such condition was 90%.   DOI: http://dx.doi.org/10.3329/bjsir.v47i2.11457   Bangladesh J. Highest percent of shoot regeneration was 93.86 when nodal explants were cultured on MS+2.0 mg/1 BAP. The maximum number of shoots (4.00) per explant was observed in MS + 2.0 mg/1 BAP + 0.3 mg/1 NAA from nodal segment. Among the two explants, nodal segment was found better for shoot regeneration. Female genotypes responded better than the male genotypes for shoot induction and proliferation. Lower nodal segment performed the best shoot regeneration. The best response towards root induction was achieved on half MS medium supplemented with 0.5 mg/1 NAA. An in vitro multiple shoot regeneration protocol has been developed for Centella asiatica L. Urban from the nodal and shoot tips explants using MS medium with different concentrations and combinations of growth regulators. Multiple shoot regeneration was observed from both the explants on MS containing either BAP alone or in combination with IAA, NAA and Kn. Of the two explants, nodal segment showed comparatively better response towards multiple shoot regeneration. Maximum multiple shoots were found on MS supplemented with 1.0 mg/L BAP and 0.4 mg/L NAA. For root induction, well-developed shoots were excised and cultured on both MS and half strength of MS with various concentrations of IBA, IAA and NAA. Profuse healthy rooting was obtained on MS medium containing 0.2 mg/L IBA. An experiment was conducted to study the in vitro culture of pointed gourd. Cotyledon rescued from physiologically matured seeds (PMS) and immatured seeds (IMS) of pointed gourd were used as explants. Cotyledon excised from PMS responded very well in all culture conditions. Plant regenerated from cotyledon of PMS ranged from 38 to 96% in different hormonal formulations of culture media. Highest percentage of shoot regeneration was observed in MS + 1.0 mg/l BAP and lowest in MS + 2.5 mg/l BAP. No plant regeneration was observed in cotyledon from immatured seeds. The highest percentage of root induction (99%) was achieved in half MS medium supplemented with 0.5 mg/l NAA. The regenerated plantlets were successfully established in earthen pot. An efficient protocol was developed for in vitro plant regeneration and multiplication through callus culture in pointed gourd. Among the explants, highest percentage of cotyledon explants (92.00%) produced callus when this explant cultured in MS medium supplemented with NAA (0.1, 0.5, 1.0, 1.5, and 2.0 mg/l) and 2, 4-D (0.1, 0.5, 1.0, 1.5, and 2.0 mg/l). The highest number of shoots per explant was observed in MS + 0.5 mg/l BAP + 0.5 mg/l NAA followed by 1.0 mg/l BAP + 0.5 mg/l NAA when inter-node derived callus cultured in MS medium. Among the explants derived calli from leaf, inter-node and cotyledon in in vitro regeneration study, inter-node appeared as the most suitable explant for callusing and plant regeneration. The best response towards root induction was achieved on half MS medium supplemented with 0.5 mg/l NAA. The regenerated plantlets were successfully established in prepared earthen soil pot. The establishment of a micro-propagation protocol for banana (Musa sp.) cv. Sabri, was carried out using meristematic stem cuttings explant. MS medium supplemented with BAP singly or in combination with auxin, IAA and coconut water was used for this purpose. Highest percentage of shoot regeneration (90%) and maximum number of shoots (10) per explant were observed when cultured on MS + 4.0 mg/1 BAP + 2.0 mg/1 IAA + 13% (v/v) coconut water. Best response towards root induction was achieved on half MS medium supplemented with 0.5 mg/1 IBA. The regenerated healthy rooted plantlets were transferred to small plastic pot containing garden soil and compost in a ratio of 2:1. An efficient in vitro regeneration system was developed for Piper betle L. through direct and indirect organogenesis from nodal segment, leaf segment and petiole explants. Highest direct regeneration was recorded when nodal explants were cultured on MS with 1.0 mg/l BAP and 1.0 mg/l Kn where 80% explants produced multiple shoots and the average number of shoots per explants were 3.20. Remarkable results on callus induction and shoot initiation were observed when the explants cultured on MS + 2.0 mg/l BAP + 0.5 mg/l Kn + 1.0 mg/l IAA. It was observed that nodal explants were showed best response on shoot/explants 13.2 ± 4.5 after 8 weeks of callus culture on MS medium with 0.5 mg/l BAP. The best response towards root induction was observed on half strength of MS with 0.25 mg/l IBA. The well rooted plants were successfully acclimatized and transferred to soil.

In vitro study was carried out in an important medicinal plant Tinospora cordifolia (Willd.) Miers belonging to the family: Menispermaceae. Vegetative parts such as stem, leaf and nodal explants were excised from an elite in vivo grown mature plant and thereafter cultured on Murashige-Skoog (MS) medium supplemented with different hormonal concentrations for callus induction and organogenesis. Callus formation occurred from nodal segments, leaf and inter-node explants when planted on different combinations of hormones. Tinospora cordifolia showed response for in vitro shoot growth from the nodal segment. The best shoot growth was observed on MS medium supplemented with kinetin (1.5 mg/l). Similarly, the best result for root induction was obtained on MS medium supplemented with 6-benzylaminopurine (1.0 mg/l) and naphthaleneacetic acid (2.5 mg/l). Introduction: The present research work was undertaken with a view to developing a suitable protocol for in vitro plant regeneration of economically important plant (Glycine max) (Bangladesh Agricultural Research Institute BARI- 5) variety, via both direct and indirect organogenesis from in vitro grown seedlings. Methods: For micropropagation explants were cultured on MS and half strength Murashige and Skoog (MS) medium supplemented with various plant growth regulators (cytokinins and auxins). In the present study for inducting of callus, among 3 different hormone combinations, the suitable medium was 3.32 mg/L 2, 4-D containing MS medium and the callus was deep green in color. Different type of media like MS, 1/2 MS and MS with different (6-Benzyl Amino Purine) BAP concentration was used for seed germination of Glycine max. 100% of seed germination was observed in MS +1 mg/L BAP containing the medium. Results: In the present investigation, different concentration of cytokinins and auxins{BAP, 2, 4-D, and Naphthalene Acetic Acid (NAA)} were used individually or in combinations with MS medium to observe their effect on multiple shoot regeneration from the cotyledonary nodal segment. 100% shoot formation from cotyledonary nodal segment was recorded in 1.5 mg/L BAP and 0.15 mg/L BAP + 0.025 mg/L NAA containing MS medium, the best number of shoot was 10.9±2.0 found in MS + 1.5 mg/L BAP containing medium and highest length of shoot was 2 cm recorded in 1.5 mg/L BAP + 0.3 mg/L (different concentrations of Giberrellic acid) GA3 containing MS medium. In addition, for root induction in vitro raised well developed and elongated shoots were excised and cultured on MS and 1/2 MS medium supplemented with various concentration of Indole-3-Butyric acid (IBA). It was observed that MS medium containing 0.1 mg/L IBA and 1/2 MS medium containing 0.25 mg/L IBA was optimal for root induction. In which 100% shoots rooted well within 13 days of culture. The highest average number of roots per shoot was 6 recorded in MS +0.5 mg/L IBA containing the medium and highest average length of root was 8 cm recorded in 0.1 mg/L IBA containing MS medium. Conclusion: The most effective surface sterilization treatment for explants of Glycine max has been found in 0.1% HgCl2 solution for 15 minutes.

Investigation on in vitro multiple shoot regeneration in Bacopa monnieri (L.) Penn. using leaf and nodal explants was carried out on MS containing various concentrations and combinations of BAP, Kn, NAA and 2,4-D. Of the two explants, leaf showed the best response towards shoot regeneration and subsequent plant development on MS with 1.0 mg/l BAP and 0.25 mg/l Kn. In this combination, the mean number of shoots/explant was 10.6 ± 0.11 in leaf and 9.6 ± 0.29 in nodal explants. Maximum shoot length was recorded as 12.6 ± 0.21 and 11.20 ± 0.30 from leaf and nodal explants after six weeks of culture, respectively. Half strength of MS supplemented with 0.25 mg/l IBA was found to be the best medium for root formation. Ficus religiosa is an important industrial, medicinal, and ornamental plant, so in vitro regeneration is of high paramount in this valuable germplasm. Two efficient protocols were developed for indirect and direct shoot organogenesis through hypocotyl explants. In the first experiment, different concentrations of 2,4-dichlorophenoxyacetic acid (2,4-D) and indole butyric acid (IBA) (0.5, 1.0, and 1.5 mg·L−1) in combination with 6-benzyl amino purine (BAP) (ratio 10:1, respectively) were used for callus formation. Two types of callus were obtained from different concentrations of plant growth regulators (PGRs). Also, 2,4-D produced yellow-brownish and friable callus (Type I), whereas the green and compact callus (Type II) was achieved in IBA. The highest callus fresh weight (2.43 g) was observed in Murashige and Skoog (MS) medium containing 0.5 mg·L−1 2,4-D plus 0.05 mg·L−1 BAP. In the later experiments, various concentrations of thidiazuron (TDZ), 6-furfuryl amino purine (KN), and BAP (0.25, 0.5, 1.0, and 1.5 mg·L−1) in combination with IBA (ratio 10:1, respectively) were applied for shoot regeneration (direct and indirect organogenesis). In shoot regeneration from callus, the highest regeneration frequency (86.66%) and shoot number per callus (4.13) were achieved in MS medium supplemented with 1.5 mg·L−1 BAP plus 0.15 mg·L−1 IBA from type I calli. However, no regeneration was observed in type II calli. In direct shoot regeneration, the highest regeneration frequency (96.66%) and shoot number (6.26) were obtained in the medium mentioned previously. In root induction experiment, different concentrations of naphthalene acetic acid (NAA) and IBA alone or in combination were applied, and MS medium containing 2.0 mg·L−1 IBA along with 0.1 mg·L−1 NAA was the best hormonal balance for root induction. The rooted plantlets’ survival rate was more than 95% in the acclimatization stage. These results demonstrated that the direct regeneration method provides more shoot regeneration frequency and take a less time for shoot organogenesis than the indirect regeneration method. Based on our knowledge, this study is the first report of direct and indirect shoot organogenesis of F. religiosa via hypocotyl from in vitro–grown seedling.
