Quantitative modeling is increasingly utilized in the drug discovery and development process, from the initial stages of target selection, through clinical studies. The modeling can provide guidance on three major questions–is this the right target, what are the right compound properties, and what is the right dose for moving the best possible candidate forward. In this manuscript, we present a site-of-action modeling framework which we apply to monoclonal antibodies against soluble targets. We give a comprehensive overview of how we construct the model and how we parametrize it and include several examples of how to apply this framework for answering the questions postulated above. The utilities and limitations of this approach are discussed.

Effects of three diphenylether herbicides [2,4-dichlorophenyl-p-nitrophenyl ether (nitrofen); 2,4,6-trichlorophenyl-4′-nitrophenyl ether (hereinafter referred to as MC-1478); and 2,4′-dinitro-4-trifluoromethyl-diphenylether (hereinafter referred to as C-6989)] were measured on phosphorylation and electron transport in spinach(Spinacia oleraceaL.) chloroplasts, and mung bean(Phaseolus aureusL., var. Jumbo) and white potato tuber(Solarium tuberosumL.) mitochondria. All of the diphenylethers acted primarily as inhibitors of chloroplast noncyclic electron transport, and the coupled photophosphorylation. The compounds ranked in the following decreasing order of inhibitory effectiveness: MC-1478 ≥ C-6989 >> nitrofen. A site of action close to light reaction II was suggested. At high molar concentrations, marginal interference with cyclic electron transport or phosphorylation was obtained. In mitochondria, MC-1478 and nitrofen acted primarily as electron transport inhibitors with malate, NADH, and succinate as substrates. MC-1478 was a slightly stronger inhibitor than nitrofen. Only slight stimulation of ADP-limited oxygen uptake was obtained during the oxidation of NADH and succinate; whereas, strong inhibition of oxygen uptake was obtained with malate. C-6989 also weakly stimulated ADP-limited oxygen uptake with NADH and succinate but differed from the two chlorinated diphenylethers in that electron transport was not inhibited when ADP was present in excess. Interference with ATP generation could be one of the mechanisms through which the phytotoxicity of diphenylether herbicides is expressed.

Hypertension and obesity are two significant factors that contribute to the onset and exacerbation of a cascade of mechanisms including activation of the sympathetic and renin-angiotensin systems, oxidative stress, release of inflammatory mediators, increase of adipogenesis and thus promotion of systemic dysfunction that leads to clinical manifestations of cardiovascular diseases. Seaweeds, in addition to their use as food, are now unanimously acknowledged as an invaluable source of new natural products that may hold noteworthy leads for future drug discovery and development, including in the prevention and/or treatment of the cardiovascular risk factors. Several compounds including peptides, phlorotannins, polysaccharides, carotenoids, and sterols, isolated from brown, red and green macroalgae exhibit significant anti-hypertensive and anti-obesity properties. This review will provide a comprehensive overview of the recent advances on bioactive pure compounds isolated from different seaweed sources focusing on their potential use as drugs to treat or prevent hypertension and obesity. On the other hand, although it is obvious that macroalgae represent promising sources of antihypertensive and anti-obesity compounds, it is also clear that further efforts are required to fully understand their cellular mechanisms of action, to establish structure-inhibition relationships and mainly to evaluate them in pre-clinical and clinical trials.

ObjectiveTo determine whether atrophy relates to phenotypical variants of posterior cortical atrophy (PCA) recently proposed in clinical criteria (i.e., dorsal, ventral, dominant-parietal, and caudal) we assessed associations between latent atrophy factors and cognition.MethodsWe employed a data-driven Bayesian modeling framework based on latent Dirichlet allocation to identify latent atrophy factors in a multicenter cohort of 119 individuals with PCA (age 64 ± 7 years, 38% male, Mini-Mental State Examination 21 ± 5, 71% β-amyloid positive, 29% β-amyloid status unknown). The model uses standardized gray matter density images as input (adjusted for age, sex, intracranial volume, MRI scanner field strength, and whole-brain gray matter volume) and provides voxelwise probabilistic maps for a predetermined number of atrophy factors, allowing every individual to express each factor to a degree without a priori classification. Individual factor expressions were correlated to 4 PCA-specific cognitive domains (object perception, space perception, nonvisual/parietal functions, and primary visual processing) using general linear models.ResultsThe model revealed 4 distinct yet partially overlapping atrophy factors: right-dorsal, right-ventral, left-ventral, and limbic. We found that object perception and primary visual processing were associated with atrophy that predominantly reflects the right-ventral factor. Furthermore, space perception was associated with atrophy that predominantly represents the right-dorsal and right-ventral factors. However, individual participant profiles revealed that the large majority expressed multiple atrophy factors and had mixed clinical profiles with impairments across multiple domains, rather than displaying a discrete clinical–radiologic phenotype.ConclusionOur results indicate that specific brain behavior networks are vulnerable in PCA, but most individuals display a constellation of affected brain regions and symptoms, indicating that classification into 4 mutually exclusive variants is unlikely to be clinically useful.

Systemic sclerosis (SSc) is a life-threatening connective tissue disorder of unknown etiology characterized by widespread vascular injury and dysfunction, impaired angiogenesis, immune dysregulation and progressive fibrosis of the skin and internal organs. Over the past few years, a new trend of investigations is increasingly reporting aberrant epigenetic modifications in genes related to the pathogenesis of SSc, suggesting that, besides genetics, epigenetics may play a pivotal role in disease development and clinical manifestations. Like many other autoimmune diseases, SSc presents a striking female predominance, and even if the reason for this gender imbalance has yet to be completely understood, it appears that the X chromosome, which contains many gender and immune-related genes, could play a role in such gender-biased prevalence. Besides a short summary of the genetic background of SSc, in this review we provide a comprehensive overview of the most recent insights into the epigenetic modifications which underlie the pathophysiology of SSc. A particular focus is given to genetic variations in genes located on the X chromosome as well as to the main X-linked epigenetic modifications that can influence SSc susceptibility and clinical phenotype. On the basis of the most recent advances, there is realistic hope that integrating epigenetic data with genomic, transcriptomic, proteomic and metabolomic analyses may provide in the future a better picture of their functional implications in SSc, paving the right way for a better understanding of disease pathogenesis and the development of innovative therapeutic approaches.

The school inclusion involves much more than the right to a place in an institution of regular education. It encompasses the right of the student to rely on various pedagogical alternatives, according to their specific characteristics to help in the teachinglearning process and become subject in the construction of knowledge. Thinking about the difficulties of visually impaired students being included in the regular school and as data gathered through research, it was found restricted access to literary wo rks to the visually impaired. Thus students of the Curso Técnico em Publicidade from the Instituto Estadual de Educação Visconde de Cairu, Santa Rosa, Rio Grande do Sul, formed a study group in order to find alternatives to fill this gap. Came the idea of developing talking books from the tales and children's stories and thus was born Audiobooks Project. Throughout the project, notes, fiel d diaries were being made and were being collected materials so that each step was documented. This study discusses the development process Audiobooks Project, from its initial conception to distribution of materials to schools in the area. It is expected to provid e support for similar projects that are produced or that new pedagogical alternatives are designed, benefiting all special students and contributing to the construction of knowledge and effective citizenship.

Although neuroendocrine neoplasms are relatively uncommon entities, the appendix is a site with relatively frequent emergence of this pathology and its precise diagnosis and clinical management have been a challenge for practitioners. This paper aims to present and discuss a case report of neuroendocrine neoplasia in the appendix of a patient with acute abdomen who underwent appendectomy. A 46-year-old man referred to the Emergency Department complaining of abdominal pain in the right iliac fossa and signs of peritoneal irritation. After diagnosis of acute appendicitis, the patient underwent appendectomy. In a macroscopic analysis, an area of 1.9 cm long, hardened, irregular and yellowish shapes was noted in the distal third of the appendix. Histopathological analysis showed neoplasia consisting of invasive islands of monotonous rounded epithelioid cells, large areas of necrosis, high mitotic activity, neural and angiolymphatic invasion and extension to adipose tissue compatible with invasion of the mesoappendix. Circumferential resection was compromised, suggesting the persistence of neoplasia in the patient even after the surgical approach, which would probably recommend the need for surgical approach. Therefore, the importance of sending collected materials for anatomopathological analysis is emphasized, since it helps in the clinical evaluation, in the etiological diagnosis, guides the medical conduct in the evolution of the case, as well as assisting in family mourning in cases of mortality.Keywords: Neuroendocrine tumor, Appendix, Histopathological analysis.

The protection of intellectual property (IP) is a crucial area to support the development process of any country, as it is in this context that the biggest strategic disputes are taking place. In recent years Brazil has developed some actions to achieve greater efficiency in the public IP management system, but are we on the right track? The present study seeks to present answers regarding the performance of Brazil and to highlight the advances and challenges regarding the IP system. The methodological approach was structured based on a review in the literature, highlighting the scientific, economic and technological indicators on the development of IP and the main IP objects registered with the Brazilian national intellectual property body (INPI) in the period of 2013 to 2016, in the areas of patents, trademarks, industrial design, computer program, circuit topography, technology contracts and geographical indication.

Abstract An approach to support the appropriate definition, formulation and presentation of requirements obeying the needs of the employees is being presented. Classifiying the needs for information of the employees according to some characteristics, a guideline for how to promote the information needed by them can be developed. The goal is to ensure that everyone involved in the process — even the subcontractor — has the right information available in the right form at the right time. Defining requirements lists, splitting them into partial requirements lists for each area of the company, each process phase or each team, and then formulating these requirements and presenting them in an adequate way is the proceeding strived for. The result will be implemented into an information infrastructure to support the product development process continuously.

This book explores the history and the vicissitudes of one of Paris’s most extraordinary areas, the Marais. Centrally located on the Right Bank, this neighbourhood was from the Middle Ages through to the eighteenth century the most fashionable in the city, headquarters of the nobility who endowed it with resplendent architecture. The Court’s move to Versailles and the Revolution of 1789 led to the quartier’s decline, so that in the nineteenth century and the earlier part of the twentieth it was in parlous shape, its fine buildings run down and often severely overcrowded. It escaped wholesale destruction in the post-War frenzy of modernization largely thanks to André Malraux, who as Culture Minister fostered the restoration of the area. Malraux’s efforts were, however, not immune from criticism, sometimes seen as a form of socio-economic cleansing with concomitant fossilization, and thus emblematic of the problems faced by a city which has always been torn between the preservation of its past and the need to adapt to social and historical change. The book focuses particularly on literary, cinematic and other artistic reproductions of the quartier, of which it attempts to provide a comprehensive overview, and foregrounds particularly its importance as home to and base of two highly significant minorities – the Jewish and the gay communities.
