Copper (Cu) is an important trace element with a two-sided effect on the growth performance of animals, which depends on the timing and dosage of Cu addition, etc. The purpose of this study was to determine the effects of oral copper sulfate (CuSO4, 350 ppm) on growth performance, cecal morphology, and its microflora of chickens (n = 60) after 30, 60, and 90 days. The results showed that after 90 days of copper exposure, the chickens lost weight, the cecum mucosa was detached, and vacuolation and inflammatory infiltration occurred at the base of the lamina propria. In addition, using the 16S rDNA sequencing method, we observed that copper exposure changed the richness and diversity of intestinal microorganisms. At the phylum level, Proteobacteria and Actinobacteria both significantly increased, while Bacteroidetes significantly decreased in the Cu group compared with control check (CK) group. At the genus level, the relative abundance of Rikenellaceae_RC9_gut_group decreased significantly, while Ruminococcaceae_UCG-014, Lachnoclostridium, and [Eubacterium]_coprostanoligenes_group increased significantly after copper exposure, and the change in microflora was most significant at 90 days. Moreover, the relevance of genus-level bacteria was altered. PICRUST analysis revealed potential metabolic changes associated with copper exposure, such as Staphylococcus aureus infection and metabolic disorders of nutrients. To sum up, these data show that subchronic copper exposure not only affects the growth and development of chickens but also causes the imbalance of intestinal microflora, which may further induce metabolic disorders in chickens.

This study aimed to investigate the efficacy of compound probiotics on broiler performance, intestinal microflora, immune function and serum biochemical indicators. Four hundred one-d-old female broiler chickens were randomly divided into 4 groups with 4 replicates in each group and 25 chickens per replication. The chickens in groupI(control group) were fed a basal diet, experimental groups II, III and IVwere fed the basal diet supplemented with 0.1%, 0.2% and 0.4% compound probiotics. The results of this work were as follows: (1) during the experimental period, compared with control group, the average daily in group III was significantly increased (P<0.05), the feed/gain and average daily feed intake were significantly lower (P<0.05); (2) at the age of 14 and 28 days, the number of cecal Lactic acid in group II and III were significantly higher than the control (P<0.05) and the content of E.coli has the opposite results. (3) the anti-body titer in group III was the highest, and significantly higher compared to the control (P<0.05), but there were no significantly differences between group IV and control group (P>0.05). (4) serum biochemical indexes of broilers in group III and IVwere significantly increased than that in control group (P<0.05). In conclusion, the supplementation of 0.2% compound probiotics could promote the growth performance, improve the cecal mico-ecosystem, enhance the anti-body titer and biochemical indexes in serum.

Objective: The objective of this work was to isolation, purification and characterization of solvent tolerant lipase from Bacillus sp. The objective of this work was to isolation, purification and characterization of solvent tolerant lipase from Bacillus sp. from kitchen grease for a variety of applications including organic synthetic reactions and preparation of enantiomerically pure pharmaceuticals.Methods: Lipase producing isolates were screened from kitchen grease on a selective medium rhodamine B olive oil agar, and tributyrin agar was used to screen the lipase and esterase producing an organism, respectively. The isolate identified using 16S rDNA sequencing method and enzyme activity was quantitatively assayed. Lipase production was characterized in different conditions.Results: The isolate showed highest lipase activity was which later was identified as Bacillus sp. using 16S rDNA sequencing method. The lipase was purified using ammonium sulfate precipitation. The isolate showed excellent tolerance to methanol, ethanol, acetonitrile, and moderate tolerance to butanol. The increased biomass concentration, maximum production, and activity were achieved at 37°C in 24 h incubation, then gradual reduction in production was observed. The maximum activity of lipase enzyme was obtained at pH between 6 and 9.Conclusion: The isolate produce solvent tolerance lipase enzyme and it can be a promising candidate of solvent tolerance lipase enzyme for variety of industrial applications.
