The aim of this study was to evaluate the efficacy of a progesterone-based treatment on anoestrus in buffaloes. Primiparous acyclic buffaloes (n = 276), were divided into three classes according to their days in milk (DIM): from 50 to 90 (Class I; n = 86), from 91 to 150 (Class II; n = 102) and from 150 to 200 (Class III; n = 88). Animals were synchronized using P4 vaginal implants, followed by timed artificial insemination (TAI). They were then allowed to enter into a larger group of buffaloes for natural mating 15 days after AI was performed, and pregnancy status was monitored from then on at 15-day intervals. Finally, the temperature–humidity index (THI) was calculated. Statistical analysis was performed by ANOVA by means and both multiple and linear regression. The total pregnancy rate (PR) was 87.7%, with no differences among DIM classes (88.0, 92.4, and 80.0% in Classes I, II, and III, respectively). However, the PR at TAI tended to be higher (p = 0.07) in buffaloes in Class II. The follicle (FL) area in Class II buffaloes was larger (p < 0.01) than that of the other classes. No influence of the THI on the total PR was recorded. The pregnancy outcome at TAI was affected by the FL area (odds ratio = 2.237; p < 0.05) and body condition score (BCS) (odds ratio = 1.256; p < 0.05). In conclusion, treatment with vaginal P4 optimizes pregnancy rates in anoestrus buffaloes, particularly when the animals are in mid-lactation and show an optimal BCS. Furthermore, the THI does not seem to affect the efficiency of the progesterone treatment.

A Inseminação artificial em tempo fixo (IATF) está se difundindo no Brasil, pois contribui para o aumento da produtividade. O objetivo neste trabalho foi avaliar o uso do GnRH em protocolos de IATF de vacas criadas no pantanal Sul-mato-grossense. Participaram da pesquisa 531 vacas paridas da raça Nelore, que foram avaliadas de acordo com o escore de condição corporal (ECC), submetidas a três tipos de protocolos de IATF (lote 1, 2 e 3). Após 40 dias da inseminação artificial foi realizado diagnóstico de gestação. As matrizes que não receberam GnRH no lote 1 (estro aparente), lote 2 (estro parcial) e lote 3 (sem apresentação de estro) apresentaram, respectivamente 55,4%, 19,2% e 25,9% de prenhez, enquanto as que receberam o fármaco apresentaram 48,6%, 37,2% e 38,8%. Relacionado ao ECC, pode-se observar que a maior taxa de prenhez ocorreu em animais com ECC 3. Primíparas apresentaram melhor taxa de prenhez quando receberam GnRH independente do ECC. Concluiu-se que o uso de GnRH mostrou-se eficiente para melhoria da taxa de prenhez principalmente em primíparas, assim como nos animais que não apresentaram estro ou apresentaram estro parcial, e que matrizes com ECC 3 apresentaram melhores taxas de prenhez que vacas com escore inferior. Palavras-chave: bovino de corte; inseminação artificial em tempo fixo; produtividade; GnRH; Nelore.   Evaluation of cow reproductive performance in the region of Mato Grosso do Sul swamp submitted to TAI with GnRH application   ABSTRACT: The technique of timed artificial insemination (TAI) is spreading in Brazil, as it contributes to the increase of productivity. The objective of this work was to evaluate the use of GnRH in TAI protocols of cows raised in the swamp of Mato Grosso do Sul. 531 Nelore breed calved cows participate in the survey, which were evaluated according to the body condition score (BCS), and then performed three types of TAI protocols (batch 1, 2 and 3). After 40 days of the insemination, a pregnancy diagnosis was made. The matrices that did not receive GnRH in batch 1 (apparent oestrus), batch 2 (partial oestrus) and batch 3 (no oestrus presentation) presented respectively 55.4%, 19.2% and 25.9% of pregnancy, while those receiving the drug showed 48.6%, 37.2% and 38.8%. Related to BCS, it can be observed that the highest pregnancy rate occurred in animals with BCS 3. Primiparous had a better pregnancy rate when receiving GnRH regardless of BCS. It was concluded that the use of GnRH was efficient to improve the pregnancy rate mainly in primiparous as well as animals that showed no oestrus or partial oestrus, and matrices with BCS 3 had better pregnancy rates than cows with lower scores. Induction of ovulation for timed artificial insemination (TAI) with the Ovsynch protocol was evaluated in 49 anoestrous and lactating Bos taurus × Bos indicus cows. Palpation per rectum and transrectal ultrasonography were used on Days -30, -20, -10 and 0 (start of treatment) to confirm anoestrus but with the presence of follicles = 10 mm, and every other day during treatment to determine ovarian activity. Cows were randomly assigned to: (1) Ovsynch (n = 24; Day 0, 200 µg GnRH; Day 7, 150 µg PGF2a; Day 9, 200 µg GnRH + TAI 16 to 20 h later) and (2) control (n = 25; no treatment). Rates of ovulation for the first GnRH injection, detection of a corpus luteum (CL) at PGF2a injection, pregnancy and induction of cyclicity were greater (P < 0.05) with Ovsynch. There was no effect of body condition score (P ? 0.05). In conclusion, the Ovsynch protocol was not effective in obtaining acceptable pregnancy rate for TAI, but it was effective for induction of cyclicity in anoestrous and lactating Bos taurus × Bos indicus cows under tropical conditions.

Abstract Previous experiments from our laboratory determined that postpartum beef cows with a functional corpus luteum (CL) at the start of an estrous synchronization protocol have greater pregnancy success to FTAI than those that did not have a CL. Therefore, it was hypothesized that incorporation a presynchronization program prior to FTAI in postpartum cows will improve pregnancy rates. The goal of this project was to compare the effectiveness of a presynchronization program prior to the 7-d CO-Synch+CIDR fixed timed artificial insemination (FTAI) protocol in postpartum cows. Postpartum cows (n = 310) at two locations were randomly assigned to receive either the presynchronization program, consisting of 2 PGF2α injections 14 d apart and 11 d prior to the start of the 7-d CO-Synch+CIDR protocol, or the 7-d CO-Synch + CIDR protocol. Following estrus synchronization, all cows were inseminated by trained technicians using semen from bulls of known fertility. Pregnancy status to FTAI was assessed using ultrasonographic diagnosis at 30 d post insemination. Data were analyzed using a MIXED procedure of SAS and examined for effects of inseminator, body condition score (BCS), and location and age. Statistical significance was determined at P &lt; 0.05 and a tendency at 0.05&lt; P &lt; 0.10. Age, BCS, and location did not affect (P &gt; 0.05) pregnancy rates. Cows exposed to the pre-synchronization program tended to have greater AI pregnancy rates compared to cows that did not receive the pre-synchronization program (57.7±5.3 vs. 46.9±5.3%, respectively; P = 0.0687). AI pregnancy rates were greater in cows with &gt;1 ng/mL P4 compared to cows with &lt; 1 ng/mL P4 at the start of FTAI (77.7±7 vs. 55.7±6.2%, respectively; P = 0.0204). Furthermore, AI pregnancy rates were decreased by inseminator C (26.1±7.4) compared to the other inseminators this breeding season (A-54.4±7.9; B- 62.3±5.3; D-57.8±7.2; and E -51.1±5.2; P = 0.0026). Based on these data, incorporating a pre-synchronization program to ensure the presence of a CL at the start of a FTAI protocol may improve pregnancy success in postpartum beef cows.

The aim of this study was to evaluate odds ratio (OR) of management factors on infertility of artificial insemination results. Three traditional farmer groups were purposively choosed, based on percentage of calf born of AI program. Group I, with calves born of 61.22%, Group II with calves born of 38,89 %, and group III with calves born of 30,0%. The management factors evaluated were 1. ownerships of cattle, 2. raise cattle in separate pen with other, 3. cattle raised in pen dirty pen, 4. not enough feed given 5. body condition score. The data was obtained by dichotomy questioner and observation then evaluated using odds ratio (OR), when OR more than 1 indicated the high risk of factor to infertility. The results of OR in Group I showed that ownership (1,18), separated pen (0,54), dirty pen (2,11), less feed (3,20) and body condition (0,22). The results in Group II showed that ownership (0,83) , separated pen (1,12), dirty pen (0,14), less feed (2,08) and body condition (2,00). The results of odds ratios in Group III showed that ownership (1,63), separated pen (3,83), dirty pen (0,88), less feed (1,00) and body condition (0,12). These data indicated that ownerships, dirty pen, less feed and body condition were factors which may confounding with other factors. However less OR of separate pen were, in Group I (0,54), Group II (1,12) and Group III (3,83) consistently with number of calves produced. It can be concluded that separate pen was a factor which influence calves produced in using artificial insemination.

The aim of this study was to evaluate the possible effect of two doses of hCG (100 and 300 IU) applied at two different times (7 and 14 d) after a fixed-time artificial insemination protocol (FTAI) upon some variables involved in the embryonic implantation rate in goats during the natural deep anestrous season (April, 25° north). The experimental units considered crossbred, multiparous, anovulatory goats (n = 69, Alpine, Saanen, Nubian x Criollo), with average body weight (43.6 ± 5.7 kg) and body condition score (1.86 ± 0.28 units) located in northern–semiarid Mexico (25° N, 103° W). Once the goat’s anestrus status was confirmed, goats were subjected to an estrus induction protocol. Upon estrus induction confirmation, goats (n = 61) were subjected to a FTAI procedure. Immediately after the FTAI, the goats were randomly distributed to five experimental groups: (1). G100-7 (n = 13) 100 IU, hCG 7 d post-FTAI, (2). G100-14 (n = 12) 100 IU hCG, 14 d post-FTAI, (3). G300-7 (n = 12) 300 IU, hCG, 7 d post-FTAI, (4). G300-14 (n = 12) 300 IU hCG 14 d post-FTAI, and (5). Control group, CONT (n = 12) 0.5 mL saline, 7 and 14 d post-FTAI. The response variables conception rate (39.36 ± 0.23), fertility rate (27.96%), prolificacy rate (1.1 ± 0.29 kids), ovulation rate (0.74 ± 0.20 corpus luteum) corpus luteum diameter (10.15 ± 0.59 mm), embryo number (1.58 ± 0.20), and embryo implantation rate (48.96%), did not differ between treatments. However, while the variables fecundity rate (67%), embryo efficiency index-1 (33.99 ± 0.20%), and embryo efficiency index-2 (27.94 ± 0.30%) were favored by the G300-14 treatment, the corpus luteum area was favored (p < 0.05) by both G300-7 (113.30 ± 0.19 mm2) and G300-14 (103.04 ± 0.17 mm2). Such reproductive strategy emerges as an interesting approach, not only to enhance the out-of-season reproductive outcomes, but also to boost one of the main rulers defining the global reproductive efficiency of a heard, namely, the embryo implantation efficiency.

The aim of the current study was to evaluate the relation of automatically determined body condition score (BCS) and inline biomarkers such as β-hydroxybutyrate (BHB), milk yield (MY), lactate dehydrogenase (LDH), and progesterone (mP4) with the pregnancy success of cows. The cows (n = 281) had 2.1 ± 0.1. lactations on average, were 151.6 ± 0.06 days postpartum, and were once tested with “Easy scan” ultrasound (IMV imaging, Scotland) at 30–35 d post-insemination. According to their reproductive status, cows were grouped into two groups: non-pregnant (n = 194 or 69.0% of cows) and pregnant (n = 87 or 31.0% of cows). Data concerning their BCS, mP4, MY, BHB, and LDH were collected each day from the day of insemination for 7 days. The BCS was collected with body condition score camera (DeLaval Inc., Tumba, Sweden); mP4, MY, BHB, and LDH were collected with the fully automated real-time analyzer Herd Navigator™ (Lattec I/S, Hillerød, Denmark) in combination with a DeLaval milking robot (DeLaval Inc., Tumba, Sweden). Of all the biomarkers, three differences between groups were significant. The body condition score (BCS) of the pregnant cows was higher (+0.49 score), the milk yield (MY) was lower (−4.36 kg), and milk progesterone in pregnant cows was (+6.11 ng/mL) higher compared to the group of non-pregnant cows (p < 0.001). The pregnancy status of the cows was associated with their BCS assessment (p < 0.001). We estimated that cows with BCS > 3.2 were 22 times more likely to have reproductive success than cows with BCS ≤ 3.2.

Abstract Introduction: This study aimed to evaluate the morphology of the genital tract from 98 Nellore cows by rectal palpation and combine them with the functional aspects for inclusion in a fixed-time artificial insemination (TAI) program. Methods: Were evaluated the ovaries, cervix and vulva, as well as the body condition score (BCS scale 1-5). Results: In relation to the ovaries, there were palpable structures found in 51 (17 CL and 34 FL) in the rights ovarian and 37 palpable structures (06 CL and 31 FL) in the left ovary. Asymmetry was found in 17.3% and uterine cervicitis in 20.4% of examined females. Changes were noted in vulva in 51.0% (40 to 10 papules and hyperemia) and the average of BCS was 3.15. On the basis of morphological aspects founded, 29 females with an average of BCS 2.7 and 01 with metritis were excluded and 07 (BCS-3, 5) inseminated immediately after the exam, with 61 included in the TAI program. These data support the conclusion that of all ovulations occurred, characterized by the presence of CL, most occurred in the right ovary (73.9%). The prevalence rate (51%) of vulvar aspects found, indicate a need for research of reproductive diseases in their flock. Conclusion: The BCS may impacts on the cyclicity and in the pregnancy rate of females included in the program.
