Background: trans-anethole is the main component of anise, and its flavor is characteristic of all the aniseed spirits. It is known that trans-anethole oil/ethanol/water solution could result in the undesirable emulsion effect called the “ouzo effect”. Moreover, trans-anethole can undergo chemical decay, producing unhealthy compounds, mainly cis-anethole and para-anisaldehyde. In this work, the trans-anethole stability was challenged simulating commercial products' long-term storage. Methods: Anethole standard solutions and two Italian aniseed spirits, namely Sambuca and Mistrà, were used to evaluate the trans-anethole response to light (both ultraviolet and visible) and its stability under thermal stress. Anethole decay was monitored by trans-anethole quantification using an HPLC method. Results: Light and high-temperature exposure affect the trans-anethole content of aniseed spirits during the storage, but sucrose revealed a role in the trans-anethole protection. Conclusions: as transport and storage are involved in the anethole decay, the Regulations on the minimum of the anethole that must be a guarantee in the aniseed spirits should be revised.

Knowledge of manure composition is important for farm nutrient management, either if importing manure onto a farm or transferring nutrients around the farm in ‘home produced’ manures. Many factors affect the nutrient content of the manure ready to spread onto the land (Smith & Frost, 2000): dietary input and quality, nutrient losses during housing and storage and additions of bedding material and/or water. There are many reports of average values for manures from conventionally raised livestock (e.g. Anon., 2000 for the U.K.). However, there are less data available for manures produced on organic holdings. It is probable that composition will differ from conventionally produced manures because of differences in diet and manure storage methods (composting and/or long-term storage). Therefore, we aimed to test this under U.K. conditions by analysing cattle manures from organic holdings for comparison with data on conventionally produced manures.

The objectives of this study were to understand how genotype, storage time, and storage conditions affect cooking time of beans and to indicate storage techniques that do not affect the cooking time. The grains were subjected to five different storage periods and six different storage conditions. The cooking time was estimated using the Mattson Cooker. The data were subjected to analysis of variance and a subsequent adjustment of simple linear regression for deployment of the interactions between the factors. Contrasts were used to determine the best levels of the factor storage condition. Genotype did not impact cooking time when the storage time and storage conditions were considered. Time and storage conditions affect the cooking time of beans in a dependent manner, but time of storage had the biggest influence. The best conditions for long-term storage of beans ensuring a smaller increase in cooking time is plastic storage at low temperatures. Thus, plastic freezer storage is a practical alternative for consumers.

The survival of five mycoplasma strains was studied in different storage media (Mycoplasma complet media without cryopreservative agent, Mycoplasma complete media with addition of horse serum, Mycoplasma complete media with addition of glycerol and lyophilized cultures without stabilizer) under different temperatures (+37°C, +4°C, -20°C, -85°C) during 24 months. Five Mycoplasma strains, Mycoplasma mycoides subsp mycoides (Mmm), Mycoplasma bovis (Mb), Mycoplasma agalactiae (Ma), Mycoplasma gallisepticum (Mg) and Mycoplasma synoviae (Ms) were isolated from various parts of the country. The initial titers of the strains determined by the agar plate count before storage were 42.4x10 7 UFC/ml (8.6 log UFC/ml) for Mmm strain; 32.4x10 8 UFC/ml (9.51 log UFC/ml) for M.bovis strain; 12.4x10 9 UFC/ml (10.09 log UFC/ml) for Ma strain; 2.4x10 9 UFC/ml (9.38 log UFC/ml) for Mg and 2.8x10 9 UFC/ml (9.45 log UFC/ml) for Ms strain. After 3 weeks of storage, no viable mycoplasmas were detected in all the conservation media at +37°C and after 3 months of storage at +4°C except for the lyophilized cultures in which an average viability rate of 17.81% was observed. Overall, the mycoplasma strains remained viable at freezing temperatures after 24 months regardless of the storage medium, but with decreasing titers, which was noticeable with mycoplasma complete media, and mycoplasma media with horse serum. Conversely, at -20°C the average viability rates after 24 months of storage were 84.36% (with glycerol) and 90.04% (lyophilized cultures). At -85°C after 24 months of storage, this was 87.98% (with glycerol) and 91.44% (lyophilized cultures). These findings suggest that, in the absence of the lysophylisation process, the addition of glycerol may be recommended for long-term storage of frozen mycoplasma isolates.

The introduction and active use of information transmission and storage systems in the Ministry of Defense (MoD) of Ukraine form the need to develop ways of guaranteed removal of data from media after their use or long-term storage. Such a task is an essential component of the functioning of any information security system. The article analyzes the problems of guaranteed destruction of information on magnetic media. An overview of approaches to the guaranteed destruction of information on magnetic media of different types is presented, and partial estimates of the effectiveness of their application are given by some generally accepted indicators of performance evaluation. The article also describes the classification of methods of destruction of information depending on the influence on its medium. The results of the analysis revealed the main problems of application of software methods and methods of demagnetization of the information carrier. The issue of guaranteed destruction of information from modern SSD devices, which are actively used in the formation of new systems of information accumulation and processing, became particularly relevant in the article. In today's conditions of development of the Armed Forces of Ukraine, methods of mechanical and thermal destruction are more commonly used today. In the medium term, the vector of the use of information elimination methods will change towards the methods of physical impact by the pulsed magnetic field and the software methods that allow to store the information storage device, but this today requires specialists to develop new ways of protecting information in order to avoid its leakage.

In three-year field experiments, the effect of genotype, flesh color, site conditions and storage on chlorogenic acid content (CAC) in tubers of potato cultivars with purple or red flesh was compared to yellow-fleshed cv. Agria. The results confirmed the significant effect of genotype on CAC. The highest CAC was characteristic on a three-year mean for the purple-fleshed cv. Vitelotte (769.5 mg/kg fresh weight (FW)), i.e. 1.19−2.6 times higher than in the other cultivars. In regard to the effect of flesh color, significantly higher mean CAC levels have been shown for the red-fleshed (2.8 times) and purple-fleshed (3.16 times) cultivars in comparison with cv. Agria (148 mg/kg FW). At the Uhříněves location with a warmer climate and frequent dry periods as compared to the second Valečov location, a higher CAC (1.18 times) was found. Cold storage (4°C, 6 months) resulted in a significant CAC increase varying from 33.2% in the Blaue St. Galler cultivar to 210.6% in the Vitelotte cultivar among all eight evaluated color-fleshed cultivars. On the other hand, the effect of storage on CAC was not evident in the yellow-fleshed Agria cultivar (inconclusive difference against CAC after harvest).

The work objective is to substantiate the technical bases for the production of gas hydrate blocks directly on-site for further transportation and storage in above ground storages, as an alternative to transporting and storing gas in underground gas storages. The theoretical bases have been considered and processes of gas hydrate blocks producing, physical-mechanical characteristics of artificial hydrates with the purpose of determining the conditions of their storage and prolonged storage were tested in laboratory conditions. The construction of above ground gas hydrate storage in the form of a building, partially deepened in a ground, which is separated from the environment by a wall of gravel cement elements made by the mixing method were substantiated. It is proved that such a constructive solution for a long-term storage of gas hydrate blocks is the most economical in terms of energy consumption to maintain internal negative temperature.

AbstractLarotrectinib, is an orally active novel small molecule approved for the treatment of solid tumors in pediatrics and adult patients. It acts by inhibiting tropomyosin receptor kinase. In this paper, we report the development and validation of a high-performance liquid chromatography (HPLC) method for the quantitation of larotrectinib in mice plasma as per the FDA regulatory guideline. Plasma samples processing was accomplished through simple protein precipitation using acetonitrile enriched with internal standard (IS, enasidenib). The chromatographic analysis was performed using a gradient mobile phase comprising 10 mM ammonium acetate and acetonitrile at a flow-rate of 0.8 mL/min on an X-Terra Phenyl column. The UV detection wave length was set at λmax 262 nm. Larotrectinib and the IS eluted at 3.85 and 6.60 min, respectively with a total run time of 8.0 min. The calibration curve was linear over a concentration range of 0.20–5.00 μg/mL (r2=≥0.992). The intra- and inter-day precision and accuracy results were within the acceptable limits. Results of stability studies indicated that larotrectinib was stable on bench-top, in auto-sampler, up to three freeze/thaw cycles and long-term storage at −80°C. The validated HPLC method was successfully applied to a pharmacokinetic study in mice.

Cereal and pasture hays, cut at the same site, were packaged into medium-sized round bales, large round bales and stacks. Halfthe number of each hay type and of each hay package were stored in the open for up to 15 months in a 500 mm rainfall area and half were transported to and stored for up to 15 months in a 1000 mm rainfall area. Densities of the pasture hay packages were within the manufacturers' specified ranges but those of the cereal hay packages were lower. Medium-sized round bales were less dense than large round bales and weathered more than large round bales and stacks after 10 and 15 months storage. Cereal hay packages deteriorated more than pasture hay packages with long-term storage. Medium-sized round bales lost more dry matter than large round bales after storage over the winter and more digestible dry matter over this period and after long-term storage. They also lost more crude protein than large round bales when stored over the first summer and over the following winter. It was economic to store all packages in the open over the first summer. It was uneconomic to store medium-sized round bales in the open over the following winter, while storage of large round bales and stacks was economic. For long-term storage, it was uneconomic to store any of the hay packages in the open, and storage under cover would have to be considered.
