Protein glycosylation pathways are present in all kingdoms of life and are metabolic pathways found in all the life kingdoms. Despite sharing commonalities in their synthesis, glycans attached to glycoproteins have species-specific structures generated by the presence of different sets of enzymes and acceptor substrates in each organism. In this review, we present a comparative analysis of the main glycosylation pathways shared by humans and the fungal pathogenCandida albicans:N-linked glycosylation,O-linked mannosylation and glycosylphosphatidylinositol-anchorage. The knowledge of similarities and divergences between these metabolic pathways could help find new pharmacological targets forC. albicansinfection.

Климат определенной местности нельзя рассматривать изолированно. Особенности климата отдельных регионов это преломление общих закономерностей в конкретной обстановке. Так, погодные условия высокогорных об ластей Кавказа зависят от высотной поясности, циркуляции атмосферы, географического положения, особенностей растительного покрова и других факторов. Результатами данного исследования являются обработка пятилетних дан ных (с 2011 2015 гг.) по температуре воздуха приземного слоя атмосферы и их сравнительный анализ. Данные получены с помощью автоматической метеостанции, расположенной на высоте 1700 м н. у. м. в Безенгийском ущелье. В ходе ис следований было выявлено, что среднегодовая температура приземного слоя атмосферы за 2014 г. была выше таковой в другие годы в 1,5 2,2 раза. The Climate of an area cannot be considered isolated. The climate of certain regions is the refraction of the General laws in a speci c situation. So weather conditions in high mountainous regions of the Caucasus depend on altitudinal zonation circulation of the atmosphere, geographical position, peculiarities of the vegetation cover and other factors. The results of this study are veyears treatment data on the air temperature in the atmospheric surface layer and their comparative analysis. These results obtained at the meteorological station located in Bezengi valley, Kabardino Balkar highmountain reserve.

The article presents theoretical and experimental studies of tribosystems operating under conditions of normal and abnormally low friction and wear when changing the external conditions of their operation. A comparative analysis of self-organization mechanisms of tribosystem operating under conditions of normal mechanochemical and abnormally low friction and wear is performed to determine the conditions of transition to such a mode. There was determined another possible way of converting tribosystems to conditions of abnormally low friction and wear by modifying the surface layer of bronze specimens using finishing treatment with the use of a mineral of amphiboles (jade) group.

The article considers issues of increasing wear resistance of cast iron products by methods of thermochemical treatment of their surface layer. Methods are proposed to solve the problem of wear from cast iron of grade EN-JS1060 due to development of technological processes of strengthening treatment. Alternative versions of strengthening technologies are represented by nitrocarburizing, liquid nitriding and carbonitration. Results of examination of control samples at increasing loads after 200 N before catastrophic wear are given. Comparative analysis of these results showed that samples after carbonitration and liquid nitriding had the greatest bearing capacity (1000 N). Carbonitration is selected as a most preferred option of the surface hardening treatment. Non-poisonous cyanoacid salts are used to realize it.

The enormous chemical diversity and strain variability of prokaryotic protein glycosylation makes a large-scale exploration exceptionally challenging. Therefore, despite the universal relevance of protein glycosylation across all domains of life, the understanding of their biological significance and the evolutionary forces shaping oligosaccharide structures remains highly limited.Here, we report on a newly established mass binning glycoproteomics approach that establishes the chemical identity of the carbohydrate components and performs untargeted exploration of prokaryotic oligosaccharides from large-scale proteomics data directly. We demonstrate our approach by exploring an enrichment culture of the globally relevant anaerobic ammonium-oxidizing bacterium Ca. Kuenenia stuttgartiensis. By doing so we resolved a remarkable array of oligosaccharides, produced by two entirely unrelated glycosylation machineries targeting the same surface-layer protein (SLP) simultaneously. More intriguingly, the investigated strain also accomplished modulation of highly specialized sugars, supposedly in response to its energy metabolism—the anaerobic oxidation of ammonium —which depends on the acquisition of substrates of opposite charge. Ultimately, we provide a systematic approach for the compositional exploration of prokaryotic protein glycosylation, and reveal for the first time a remarkable balance between maximising cellular protection through a complex array of oligosaccharides and adhering to the requirements of the ‘metabolic lifestyle’.

Many archaeal cell envelopes contain a protein coat or sheath composed of one or more surface exposed proteins. These surface layer (S-layer) proteins contribute structural integrity and protect the lipid membrane from environmental challenges. To explore the species diversity of these layers in the Methanosarcinaceae, the major S-layer protein inMethanosarcina barkeristrain Fusaro was identified using proteomics. The Mbar_A1758 gene product was present in multiple forms with apparent sizes of 130, 120, and 100 kDa, consistent with post-translational modifications including signal peptide excision and protein glycosylation. A protein with features related to the surface layer proteins found inMethanosarcina acetivoransC2A andMethanosarcina mazeiGoel was identified in theM. barkerigenome. These data reveal a distinct conserved protein signature with features and implied cell surface architecture in the Methanosarcinaceae that is absent in other archaea. Paralogous gene expression patterns in twoMethanosarcinaspecies revealed abundant expression of a single S-layer paralog in each strain. Respective promoter elements were identified and shown to be conserved in mRNA coding and upstream untranslated regions. PriorM. acetivoransgenome annotations assigned S-layer or surface layer associated roles of eighty genes: however, of 68 examined none was significantly expressed relative to the experimentally determined S-layer gene.
