Gel microemulsion combines the advantages of the microemulsion, which can encapsulate, protect and deliver large quantities of active ingredients, and the gel, which is so appreciated in the cosmetic industry. This study aimed to develop and characterize new gel microemulsions suitable for topical cosmetic applications, using grape seed oil as the oily phase, which is often employed in pharmaceuticals, especially in cosmetics. The optimized microemulsion was formulated using Tween 80 and Plurol® Diisostearique CG as a surfactant mix and ethanol as a co-solvent. Three different water-soluble polymers were selected in order to increase the viscosity of the microemulsion: Carbopol® 980 NF, chitosan, and sodium hyaluronate salt. All used ingredients are safe, biocompatible and biodegradable. Curcumin was chosen as a model drug. The obtained systems were physico-chemically characterized by means of electrical conductivity, dynamic light scattering, polarized microscopy and rheometric measurements. Evaluation of the cytotoxicity was accomplished by MTT assay. In the final phase of the study, the release behavior of Curcumin from the optimized microemulsion and two gel microemulsions was evaluated. Additionally, mathematical models were applied to establish the kinetic release mechanism. The obtained gel microemulsions could be effective systems for incorporation and controlled release of the hydrophobic active ingredients.

Abstract Background Osteoarthritis (OA), though being treated via various methods and medicines, is still a major healthcare concern mostly due to the increase in diagnosis of these age-related diseases. The present study aimed at investigating the effects of oral and intra articular injection of grape seed oil on OA in male rat models. Methods and materials Seventy male rats were selected and their anterior cruciate ligament was cut to induce OA. They were divided into 7 groups (n = 10): C1, no treatment; C2, receiving 300 mg/day of Piascledine per os (PO); C3, 1 mg sodium hyaluronate intra-articularly in days 1, 7, 14; C4, 1 mg methyl-prednisolone acetate intra-articularly; E1, avocado and grape seed oil combination (2:1, 300 mg/day) PO; E2, 500 mg/day of grape seed oil PO; E3, 200 mg/day grape seed oil intra-articularly. After 10 weeks, the rats were anesthetized and evaluated radiologically and histopathologically. P value ≤ 0.05 was considered as statistically significant. Results All the groups made significant differences with C1 regarding all inspected radiological criteria (P ≤ 0.05). E1 and E3 showed significantly better effects on medial femoral condyle, medial tibial condyle, joint space width, total osteophyte, and OA scores (P ≤ 0.04). Joint surface, matrix, cell distribution, cell population viability, calcification, and subchondral bone in treatment groups had significantly better scores versus C1 (P ≤ 0.04). E1 and E3 had significantly superior results regarding joint surface, cell viability, and calcification (P ≤ 0.04). Conclusions Grape seed oil has protective effects, both in injectable form and PO in combination with avocado, on OA in rats. Further clinical trials are necessary.

AbstractGrape seed cakes (GSC) resulted from grape seed oil extraction represent a by-product rich in bioactive compounds such as polyphenols, polyunsaturated fatty acids, fibres, minerals, vitamins etc known for their beneficial anti-inflammatory, anti-microbial, anti-oxidative and immune-modulatory effects. In the present study, we have investigated the effects of dietary grape seed cakes bioactive compounds on several anti-inflammatory and antioxidative biomarkers in spleen of pigs during fattening phase. Twelve crossbred TOPIG hybrid fattening pigs were allocated to two experimental treatments: 1) commercial diet (control group) and 2) a diet including 5% grape seed cakes (GSC group) for 24 days. At the end of experimental period (day 24) pigs were sacrificed and spleen samples were collected and stored at –80°C until analysis. The results showed that GSC diet lowered the gene expression as well as the protein concentration of pro-inflammatory markers: interleukin 1 beta (IL-1β, -52.66%, p<0.05) and interferon gamma (IFN-γ, -42.13%, p<0.05) and had a tendency to decrease that of interleukin 6 (IL-6, -13.25%), tumour necrosis factor alpha (TNF-α, -9.06%) and interleukin 8 (IL-8, -11.08%) when compared to control diet confirming the anti-inflammatory properties of GSC’s active ingredients. The total splenic antioxidant capacity (TEAC) and gene expression of antioxidant enzymes (catalase-CAT and glutathione peroxidase-GPx) were higher in spleen of pigs fed GSC diet than in control group suggesting also the antioxidative potential of GSC. The results related to the molecular mechanism showed a higher expression of gene encoding for PPAR-γ and for Nrf2 in spleen of pigs receiving the GSC diet (Fc 1.81 and 1.58) suggesting that the anti-inflammatory and antioxidant effect of this diet is probably exerted by PPAR-γ and Nrf2 pathway. Further, other percent of dietary GSC inclusion need to be investigated in order to obtain a more complex response related to biomarkers which characterise the fattening/obesity process.

SummaryThe cationic detergent cetyltrimethylammonium bromide (CTAB), the anionic detergents sodium lauryl sulphate and sodium deoxycholate, and the nonionic detergents Tween 20, Tween 80 and Brij 35, and lecithin, starch, glycogen, chondroitin sulphate and polyethylene glycol, were tested for their ability to replace κ-casein in stabilizing αs- and β-caseins against precipitation by Ca2+. Of the materials tested, CTAB and Tween 20 stabilized both caseins and Brij 35 stabilized only αs-casein. Analytical ultracentrifugation of mixtures of CTAB with each casein and of Brij 35 with αs-casein indicated that both detergents acted by disrupting the casein aggregates and complexing with the monomers. Addition of CaCl2 did not aggregate the Brij 35–αs-casein complex. It is concluded that the basis of the stabilization phenomenon involves specific interactions between κ-casein and αs-casein or β-casein at mainly hydrophobic sites.

Capsules of different formulations were prepared by using a hydrophilic polymer, xanthan gum and a filler Ludipress. Metformin hydrochloride, which is an anti-diabetic agent, was used as a model drug here with the aim to formulate sustained release capsules. In the first 6 formulations, metformin hydrochloride and xanthan gum were used in different ratio. Later, Ludipress was added to the formulations in a percentage of 8% to 41%. The total procedure was carried out by physical mixing of the ingredients and filling in capsule shells of size ‘1’. As metformin hydrochloride is a highly water soluble drug, the dissolution test was done in 250 ml distilled water in a thermal shaker (Memmert) with a shaking speed of 50 rpm at 370C &plusmn 0.50C for 6 hours. After the dissolution, the data were treated with different kinetic models. The results found from the graphs and data show that the formulations follow the Higuchian release pattern as they showed correlation coefficients greater than 0.99 and the sustaining effect of the formulations was very high when the xanthan gum was used in a very high ratio with the drug. It was also investigated that the Ludipress extended the sustaining effect of the formulation to some extent. But after a certain period, Ludipress did not show any significant effect as the pores made by the xanthan gum network were already blocked. It is found here that when the metformin hydrochloride and the xanthan gum ratio was 1:1, showed a high percentage of drug release, i.e. 91.80% of drug was released after 6 hours. But With a xanthan gum and metformin hydrochloride ratio of 6:1, a very slow release of the drug was obtained. Only 66.68% of the drug was released after 6 hours. The percent loading in this case was 14%. Again, when Ludipress was used in high ratio, it was found to retard the release rate more prominently. Smart polymers are materials that respond to small external stimuli. These are also referred as stimuli responsive materials or intelligent materials. Smart polymers that can exhibit stimuli-sensitive properties are becoming important in many commercial applications. These polymers can change shape, strength and pore size based on external factors such as temperature, pH and stress. The stimuli include salt, UV irradiation, temperature, pH, magnetic or electric field, ionic factors etc. Smart polymers are very promising applicants in drug delivery, tissue engineering, cell culture, gene carriers, textile engineering, oil recovery, radioactive wastage and protein purification. The study is focused on the entire features of smart polymers and their most recent and relevant applications. Water soluble polymers with tunable lower critical solution temperature (LCST) are of increasing interest for biological applications such as cell patterning, smart drug release, DNA sequencing etc.
