The determination of mycotoxin and metabolite concentrations in human and animal urine is currently used for risk assessment and mycotoxin intake measurement. In this study, pig urine (n = 195) was collected at slaughterhouses in 2012 by the Swedish National Food Agency in three counties representing East, South and West regions of Sweden. Urinary concentrations of four mycotoxins, (deoxynivalenol (DON), zearalenone (ZEA), fumonisin B1 (FB1), and ochratoxin A (OTA)), and four key metabolites, (deepoxy-deoxynivalenol (DOM-1), aflatoxin M1 (AFM1, biomarker of AFB1), α-zearalenol (α-ZOL), and β-zearalenol (β-ZOL)) were identified and measured by UPLC-MS/MS. Statistically significant regional differences were detected for both total DON (DON + DOM-1) and total ZEA (ZEA + α-ZOL + β-ZOL) concentrations in pig urine from the three regions. These regional differences were in good agreement with the occurrence of Fusarium graminearum mycotoxins (DON + ZEA) in cereal grains harvested in 2011 in Sweden. There were no statistically significant differences in FB1, AFM1 and OTA urinary concentrations in pigs from the three regions. The overall incidence of positive samples was high for total ZEA (99–100%), total DON (96–100%) and OTA (85–95%), medium for FB1 (30–61%) and low for AFM1 (0–13%) in the three regions. Urinary mycotoxin biomarker concentrations were used to estimate mycotoxin intake and the level of mycotoxins in feeds consumed by the monitored pigs. The back-calculated levels of mycotoxins in feeds were low with the exception of seven samples that were higher the European limits.

The performances of four LC-MS/MS methodologies for determination of up to eight mycotoxin biomarkers in human urines were compared by involving three laboratories that analysed common urine samples spiked at two levels of each biomarker. Each laboratory received a calibration solution, spiked urines and the corresponding unspiked urine. The two spiking levels for each biomarker were chosen by considering the levels naturally occurring in human urines and the limits of quantification of the LC-MS/MS methodologies used by the participating laboratories. The results of each laboratory were evaluated for their z-score values. The percentage of satisfactory z-scores (| z | < 2) were: 100% for deoxynivalenol, de-epoxy deoxynivalenol, aflatoxin M1, β-zearalenol and zearalenone, 87% for α-zearalenol, 50% for ochratoxin A and 42% for fumonisin B1. Good method performances were obtained for most biomarkers at the levels tested in this study, as demonstrated by the overall percentage of satisfactory z-scores for all analytes (87%). Unsatisfactory/questionable z-scores (| z | ≯2) were obtained for fumonisin B1 (7/12 results), ochratoxin A (4/8 results) and ?-zearalenol (1/8 results). The percentage of satisfactory z-scores for fumonisin B1 and ochratoxin A increased from 42 to 83% for fumonisin B1 and from 50 to 62% for ochratoxin A when laboratories 1 and 2 used own calibrants. Factors that could explain the different results obtained for fumonisin B1 and ochratoxin A with provided and own calibration solutions could not be identified in this study and should be carefully investigated in future studies.

The major causal agents Fusarium graminearum (F. graminearum) and Fusarium asiaticum could produce multiple mycotoxins in infected wheat, which threatens the health of humans and animals. Specifically, deoxynivalenol (DON) and its derivatives 3- and 15-acetyldeoxynivalenol (3-ADON and 15-ADON) are commonly detected mycotoxins in cereal grains. However, the good chromatographic separation of 3-ADON and 15-ADON remains challenging. Here, an LC-MS/MS method for the chemotype determination of Fusarium strains was developed and validated. 3- and 15-ADON could be separated chromatographically in this study with sufficiently low limits of detection (LODs; 4 μg/kg) and limits of quantification (LOQs; 8 μg/kg). The satisfying intraday and interday reproducibility (both %RSDr and %RSDR were <20%) of this method indicated good stability. The recoveries of all analytes were in the range of 80–120%. In addition, three F. graminearum complex (FGC) strains, i.e., PH-1 (chemotype 15-ADON), F-1 (chemotype 3-ADON) and 5035 (chemotype 15-ADON), were selected to verify the accuracy of the method in differentiating phenotypes. The validation results showed that this LC-MS/MS method based on sample pretreatment is effective and suitable for the chromatographic separation of 3-ADON and 15-ADON in wheat.

The potential risks to humans resulting from the usage of a pesticide must be carefully assessed before the product is registered. One of the components in the risk assessment is the determination of the amount of pesticide to which the applicator is exposed. Traditional methods estimated dermal exposure by measuring the amount of pesticide deposited on absorbent patches worn on the applicator's body. A more recent approach consists of measuring urinary metabolite levels. A review of data obtained in humans and in rats suggests that the urinary concentration of dimethyl thiophosphate is a good indicator of dermal exposure to azinphos-methyl.

ABSTRACT Oestrogens administered in lower doses than necessary to induce full cornification of the mouse vagina induce mucification. It was shown previously that the degree of mucification could be estimated by quantitative determination of sialic acids. A suitable parameter for oestrogen assay was the measurement of vaginal sialic acid concentration which exhibited a clear cut dose response curve. Eleven assays of various oestrogens were performed with this method. Their estimated relative potencies were in good agreement with other routine oestrogen assays. A statistically sufficient degree of precision was found. The sensitivity was of the same order, or slightly higher, than the Allen-Doisy test.

The choice of risk assessment procedure is one of the essential stages of efficient structuring of processes on prevention, localization and elimination of the consequences of accidents at main pipeline transport facilities. The authors analyzed themed publications and regulatory documents, governing procedures of risk assessment and forecasting of the consequences of possible accidents, and defined main problems in this area. Procedure for the risk assessment of accidents at main pipeline facilities was developed, the basis of which is the expert evaluation method. The procedure includes the determination of the main criteria for the assessment the probability of accident initiation and development and the evaluation of the severity of its consequences, an expert evaluation of criteria significance, their classification, and creation of a rating for hazardous pipeline sections. The application of the procedure application allows to specify the list of facilities that require high priority forecasting of accidents consequences, thus to optimize the distribution of resources and the overall increase of efficiency in planning while defining forces and special technical devices, necessary for containment and rectification of emergencies. Expert evaluation method application allows considering the specifics of certain enterprises, their technical and technological peculiarities, thereby increasing forecasting accuracy.

Abstract A completely automated analysis is described for the determination of serum calcium with glyoxal bis (2-hydroxyanil) solution (GBHA). The method is simple and precise, and the data obtained are in good agreement with results obtained by the manual GBHA procedure.

The hydrodynamic characteristics of the planing hulls in particular at the planing regime are completely different from the conventional hull forms and the determination of these characteristics is more complicated. In the present study, calm water hydrodynamic characteristics of planing hulls are investigated using a hybrid method. The hybrid method combines the dynamic trim and sinkage from the Zarnick approach with the Savitsky method in order to calculate the total resistance of the planing hull. Since the obtained dynamic trim and sinkage values by using the original Zarnick approach are not in good agreement with experimental data, an improvement is applied to the hybrid method using a reduction function proposed by Garme. The numerical results obtained by the hybrid and improved hybrid method are compared with each other and available experimental data. The results indicate that the improved hybrid method gives better results compared to the hybrid method, especially for the dynamic trim and resistance. Although the results have some discrepancies with experimental data in terms of resistance, trim and sinkage, the improved hybrid method becomes appealing particularly for the preliminary design stage of the planing hulls.

Human epididymis protein 4 (HE4) is an ovarian cancer marker. Various cut-off values of the marker in blood are recommended, depending on the method used for its determination. An alternative biosensor for HE4 determination in blood plasma has been developed. It consists of rabbit polyclonal antibody against HE4, covalently attached to a gold chip via cysteamine linker. The biosensor is used with the non-fluidic array SPRi technique. The linear range of the analytical signal response was found to be 2–120 pM, and the biosensor can be used for the determination of the HE4 marker in the plasma of both healthy subjects and ovarian cancer patients after suitable dilution with a PBS buffer. Precision (6–10%) and recovery (101.8–103.5%) were found to be acceptable, and the LOD was equal to 2 pM. The biosensor was validated by the parallel determination of a series of plasma samples from ovarian cancer patients using the Elecsys HE4 test and the developed biosensor, with a good agreement of the results (a Pearson coefficient of 0.989). An example of the diagnostic application of the developed biosensor is given—the influence of ovarian tumor resection on the level of HE4 in blood serum.
