Telah dilakukan penelitian untuk menentukan kandungan total fenolik dan aktivitas antioksidan ekstrak metanol daun soyogik (Saurauia bracteosa DC) sebagai peredam radikal bebas asam linoleat. Kandungan total fenolik diukur dengan metode Folin-Ciocalteu sedangkan aktivitas antioksidan diukur dengan metode FTC (Ferric Thiocyanate) untuk mengetahui kemampuan penghambatan peroksida pada tahap pertama oksidasi lipid.  Hasil penelitian menunjukkan bahwa ekstrak metanol memiliki kandungan total fenolik sebesar 99,67 mg/kg dan kemampuan dalam menghambat oksidasi asam linoleat sebesar 49,464%.This research has been conducted to measure the total phenolic compounds and the antioxidant activity of methanol extract of soyogik leaves (Saurauia bracteosa DC) as a reducer of free radicals linoleic acid. The total phenolic content was measured using folin-ciocalteu method, meanwhile the antioxidant activity was measured using FTC (Ferric Thiocyanate) method and to determine the peroxide inhibitory capability in the first stages of lipid peroxidation. The results showed that the methanol extract had a total phenolic content of 99, 67 mg/kg and peroxide inhibition of 49, 464%

Indonesia traditional beverages such as minuman jahe, kunyit asam, beras kencur, and minuman temulawak are usually made with the addition of spices. They can be served in the form of  liquid or powder. This research was aimed to study the effect of  the beverage type, addition of spices, and the form of product on their antioxidant activity. The type of sugar used in this study was palm sugar juice. Total phenolic contents were measured to  indicate the level of antioxidants. Measurement of antioxidant  activity was conducted by analyzing malonaldehyde from linoleic  acid and capturing capacity of DPPH radical. The results showed  that Indonesian traditional beverage could inhibit the oxidation of linoleic acid and capture free radicals. Free radical capturing  capacity of curcuma beverage greater than other beverages, but  the malonaldehyde (MDA) inhibitor effect was not different. Total phenolic content of curcuma tended to be higher than the other traditional beverages. The addition of spices could raise the level  of total phenolic content in beverages and increase free radical  trapping capacity. Drying the beverages into powder could reduce the levels of total phenolic content and but did not reduce the  antioxidant activity. ABSTRAKMinuman tradisional Indonesia seperti minuman jahe, kunyit  asam, beras kencur, dan minuman temulawak biasanya dibuat  dengan tambahan berbagai rempah. Minuman tersebut dapat  disajikan dalam bentuk cair maupun serbuk. Penelitian ini  bertujuan untuk mengetahui pengaruh jenis minuman,  penambahan rempah tambahan, dan bentuk minuman terhadap aktivitas antioksidan minuman. Jenis gula yang ditambahkan  adalah gula nira. Jenis antioksidan yang diuji adalah total fenolik. Pengukuran aktivitas antioksidan dilakukan dengan uji  penghambatan malonaldehid dan kapasitas penangkapan radikal  DPPH. Hasil penelitian menunjukkan bahwa minuman  tradisional Indonesia dapat menghambat oksidasi asam linoleat dan menangkap radikal bebas. Kapasitas penangkapan radikal  bebas oleh minuman temulawak lebih besar dibandingkan  minuman yang lain tetapi kemampuan penghambatan  malonaldehid (MDA) oleh minumaan tradisional tidak berbeda.  Total fenolik minuman temulawak cenderung lebih besar  dibandingkan minuman tradisional yang lain. Penambahan  rempah-rempah dapat meningkatkan kadar total fenolik  minuman dan meningkatkan kapasitas penangkapan radikal  bebas. Pembuatan minuman serbuk pada penelitian ini dapat  menurunkan kadar total fenolik tetapi tidak menurunkan aktivitas antioksidan.

Extraction is an important step in separating bioactive compounds from the plant. The selection of extraction technique is also important in the standardization of herbal products, for if not observed it can remove the desired soluble constituents. One of Indonesia’s endemic plants which have been proven to have an antioxidant activity is Salam leaves (Syzygium polyanthum). This study aims to determine the effect of extraction method on total phenolic content and antioxidant activity of S. polyanthum leaves by counteracting free radicals mechanism using DPPH. The S. polyanthum leaves were extracted by maceration, soxletation, and infusa methods, and tested for the total phenolic content and the antioxidant activity. The results showed that the total phenolic content of each method extration is different. The total of the phenolic content extraction by maceration, soxhlet, and infusa method respectively were 338.62±21.3; 227.72±21.6; and 144.48±8.2 mgGAE/g. The best antioxidant activity was maceration method with IC50 17.53±0.11 µg/mL followed by soxhlet and infusa which were 18.73±0.31 and 40.26±0.18 µg/mL. The research conclusion is that the extraction method has an effect on the total phenol and antioxidant activity of S. polyanthum leaves.

This study aims to determine antioxidant activity and total phenolic content of mangosteen leaves in several different solvents. The antioxidant activity test of the extract was determined by the DPPH method and the total phenolic content was determined by the Folin-Ciocalteu method. The phytochemical test of mangosteen leaves showed the presence of phenolic compounds, saponins, triterpenoids and alkaloids. The extraction results increase with increasing water content in methanol, ethanol and acetone. The highest amount of extract was obtained using 50% acetone solvent (34.98 ± 0.17%). Extraction using 100% ethanol showed the highest total phenolic content (614.25 ± 5.58 mg GAE / g extract) and the lowest IC50 value (5.81 mg / L). The results of this study indicate that the use of 50% acetone solvent can optimize the amount of extract and 100% ethanol can optimize the content of total phenolic compounds and antioxidant activity of mangosteen leaves.

Teripang merupakan hewan jenis Echinodermata. Beberapa species teripang, misalnya H. scabra, H. fuscogilva dan T. ananas memiliki nilai jual yang tinggi. Meskipun tidak semua spesies memiliki nilai jual yang tinggi, teripang berpotensi untuk dikembangkan dalam bidang kesehatan, terutama dalam kemampuannya untuk meredam radikal bebas dan mencegah berbagai penyakit degeneratif. Tujuan dari penelitian ini yaitu untuk mengidentifikasi, menentukan aktivitas antioksidan, kandungan total fenol, serta kadar karotenoid pada ekstrak beberapa teripang yang berasal dari Perairan Karimunjawa Jepara. Hasil identifikasi dari ke empat spesies adalah Stichopus cf. quadrifasciatus, Pearsonothuria graeffei, Bohadschia vitiensis, dan Holothuria atra. Ekstraksi dilakukan dengan teknik maserasi dinding tubuh teripang menggunakan pelarut metanol. Uji aktivitas antioksidan dilakukan dengan menggunakan metode DPPH (1,1-difenil-pikrilhidrazil) sebagai agen radikal bebas. Penentuan total fenolik dilakukan menggunakan metode folin-ciocalteu, dan penentuan kadar karotenoid dilakukan dengan menggunakan spektrofotometer. Hasil menunjukkan bahwa aktivitas antioksidan Bohadschia vitiensis, Stichopus cf. quadrifasciatus, Pearsonothuria graeffei, dan Holothuria atra tergolong sangat lemah dengan nilai IC50 secara berturut-turut adalah sebesar 454,28 ± 14,4; 713,51 ± 9,1; 801,57 ± 8,4 dan 1535,47 ppm. Kandungan total fenol sebesar 21,08 ± 0,49; 14,325 ± 0,21; 14,033 ± 0,33 dan 10,67 ± 0,12 mg GAE/g sampel, dan kadar karotenoidnya sebesar 23,28 ± 0,07; 25,78 ± 0,56; 11,85 ± 0,04; dan 41,44 ± 0,008 µmol/g sampel.Kata kunci: Teripang, Antioksidan, DPPH, Fenolik, Karotenoid Sea cucumbers are sea benthos from phylum Echinoderms phylum. Some sea cucumbers such as H. scabra, H. fuscogilva, T. Ananas have a high economical price. Eventhough, some sea cucumbers have the potency of being a healthy food. It it scientifically proven that some sea cucumbers have the ability to reduce free radicals and prevent various degenerative diseases caused by the excessive free radicals. The aims of this study were to identified, determine antioxidant activity, total phenol content, and carotenoid levels in sea cucumber extracts originally from Karimunjawa waters. Some of the sea cucumbers obtained are Stichopus cf. quadrifasciatus, Pearsonothuria graeffei, Bohadschia vitiensis, and Holothuria atra. The extraction was done by macerating the body walls with methanol. The antioxidant activity test were tested using the DPPH method as free radicals, the total phenolic content tested using the folin-ciocalteu method, and determined the carotenoid levels spectrophotometrically. Based on the results, antioxidant activity of  B. vitiensis, S. cf. quadrifasciatus, P. graeffei, and H. atra were  classified as very weak with IC50 454.28 ± 14.4; 713.51 ± 9.1; 801.57 ± 8.4 dan 1535.47 ppm. Total phenolic content were 21.08 ± 0.49; 14.325 ± 0.21; 14.033 ± 0.33 dan 10.67 ± 0.12 mg GAE / g samples, while carotenoid levels were 23.28 ± 0.07; 25.78 ± 0.56; 11.85 ± 0.04; dan 41.44 ± 0.008 µmol / g sample.Keyword : Sea cucumber, Antioxidant, DPPH, Phenolic, Carotenoid

Antioxidant fractions from Ophioglossum thermale were extracted with five different polar solvents using a Soxhlet type extractor. The total phenolic content of the extracts was determined by the Folin-Ciocalteu method. The ethyl acetate fraction of O. thermale was found to contain maximum phenolics. The dried fractions were screened for their antioxidant activity potential using in vitro model systems such as 1,1-diphenyl-2-picryl hydrazyl (DPPH), nitroblue tetrazolium (NBT) and lipid-peroxidation reduction at different concentrations. Results revealed that the EtOAc fraction exhibited the best performance in the DPPH assay, NBT assay and lipid peroxidation. All fractions showed more potent antioxidant capacity than green tea extract, a well-known antioxidant. Furthermore, the EtOAc fraction has the highest total phenolic content (475.65 mg of EGCG/g). In addition, the EtOAc fraction at 0.005% and 0.01% (g/100 ml) also significantly inhibited UVB irradiation-induced ROS generation in human dermal fibroblasts (HDFs). In a carrageenan-induced edema model, the EtOAc fraction showed an inhibitory effect (21.5%, p < 0.05) at 200 mg/kg (p.o.) after 300 min administration. Consequently, 3-O-methylquercetin (3MQ) was also isolated from the antioxidative EtOAc fraction. The data obtained using the above in vitro and in vivo tests suggest that the antioxidant activity of O. thermale and its anti-inflammatory effect on carrageenan-induced acute inflammation can be attributed to its ameliorating effect on oxidative damage, and thus it has great potential as a source for natural health products. To the best of our knowledge, this is the first report on the antioxidant activity of different polar extracts from O. thermale.

The total phenolic content and antioxidant activity of methanolic, ethanolic and aqueous extracts of myrtle ( Myrtus communis) leaves and berries were measured to find new potential sources of natural antioxidants. Total phenolic content was assessed by the Folin-Ciocalteau assay, while the antioxidant activity was evaluated by three methods: diphenyl-1-picrylhydrazyl (DPPH) radical scavenging capacity assay, the reducing antioxidant power assay and β-carotene linoleic acid assay. The total phenol content of myrtle extracts ranged between 9.0 and 35.6 mg GAE per g extract. For each solvent, leaf extracts contained significantly higher amount of total phenolic compounds than berry extracts. All of the extracts presented antioxidant capacity assessed by the three methods, but at different levels depending on the concentration, the extraction solvent and the part of the plant used. Generally, leaf extracts showed higher antioxidant activities than berry extracts, while the overall antioxidant strength was in the order methanol > water > ethanol in leaf extracts and methanol > ethanol >water in berry extracts. The phenolic content exhibited a positive correlation with the antioxidant activity: DPPH assay showed the highest correlation (r = 0.949), followed by the reducing power assay (r = 0.914) and the lowest for the β-carotene linoleic acid assay (r = 0.722).

The methanol extract of leaf of Picrasma javanica as well as its n-hexane, carbon tetrachloride,chloroform and aqueous soluble partitionates were subjected to screening for antioxidant and thrombolytic activities and brine shrimp lethality. The antioxidant potential was evaluated by DPPH, Folin-Ciocalteau reagent and phosphomolybdenum total antioxidant assays using butylatedhydroxytolune (BHT) and ascorbic acid as standards. All fractions showed moderate to strong antioxidant activity, of which the chloroform and aqueous soluble fractions demonstrated the highest activity with the IC50 value of 14.59 ± 0.73 ?g/ml and 18.6 ± 0.15 ?g/ml, respectively. The  total phenolic content of the chloroform and aqueous soluble fractions was 10.4 ± 0.64 and 5.15 ± 0.22 mg of GAE/g of extractive, respectively. Thus, a positive correlation was found between the total phenolic content and total antioxidant activity of P. javanica. The general toxicity was determined by brine shrimp lethality bioassay where the crude extract (LC50 1.04 ± 0.31 ?g/ml) and its n-hexane (LC50 1.28 ± 0.45?g/ml) soluble partitionate demonstrated the  presence of considerable bioactive principles. Mild to moderate thrombolytic activity was discerned by the methanol extract of leaf of P. javanica and its different fractions. During assay for thrombolytic activity, the carbon tetrachloride soluble materials revealed 34.165±1.57 % of clot lysis while standard streptokinase and water, used as positive and negative controls, demonstrated 66.77% and 3.791% lysis of clot, respectively. Chocolate is a product consumed worldwide and it stands out for presenting an important amount of phenolic compounds. In this study, the total phenolic content and antioxidant activity in the cerebral cortex, hippocampus, and cerebellum of male Wistar rats when consuming different types of chocolate, including milk, semisweet, dark, and soy, was evaluated. The total polyphenols concentration and antioxidant activity in vitro by the method of DPPH radical-scavenging test were evaluated in chocolate samples. Lipid peroxidation (TBARS), protein oxidation (carbonyl), sulfhydryl groups, and activity of SOD enzyme in cerebral cortex, hippocampus, and cerebellum of rats treated or not with hydrogen peroxide and/or chocolate were also evaluated. The dark chocolate demonstrated higher phenolic content and antioxidant activity, followed by semisweet, soy, and milk chocolates. The addition of chocolate in the diet of the rats reduced lipid peroxidation and protein oxidation caused by hydrogen peroxide. In the sulfhydryl assay, we observed that the levels of nonenzymatic defenses only increased with the chocolate treatments The SOD enzyme activity was modulated in the tissues treated with the chocolates. We observed in the samples of chocolate a significant polyphenol content and an important antioxidant activity; however, additional studies with different chocolates and other tissues are necessary to further such findings.

In the present study, we investigated the total phenolic content and in- vitro antioxidant activity of Sterculia oblongata. Total phenolic content (TPC) of the methanol extract was examined by Folin-Ciocalteu reagent and gallic acid as standard. The in-vitro antioxidant activity was performed by DPPH free radical scavenging activity and β-carotene bleaching assay. The result showed that the TPC of methanol extract was the highest (P<0.05) compared to other extracts. There is a positive and significantly very high Pearson’s correlation between antioxidant activity index (AAI) and TPC (r = 0.996). A negative and significantly high Pearson’s correlation was obtained between IC50 value and TPC (r = -0.885). The result of β-carotene bleaching assay showed that the rate of the decreased antioxidant activity of S.oblongata extract was lower than that of β-carotene/linoleic acid. The results suggest that the extract of S. oblongata can be used as a natural antioxidant. However, further investigation to isolate and identify the chemical compounds responsible for antioxidant was needed

During the lifetime, the human body forms a considerable amount of free radicals damaging DNA, cell membranes, and their components. A wider application of basil (Ocimum basilicum), an aromatic plant and one of the common gastronomic commodities in the human diet could help to prevent the formation of free radicals and to remove them from the human body. Therefore, determination of antioxidant activity and total content of phenolic substances in selected cultivars of basil (Ohře, Sweet green, Salad leaf, Purple opal, Thai) in a fresh and frozen state and the mixture with another plant (garlic, mint, rocket, spinach) in the fresh and refrigerated state was performed. The total content of phenolic substances in basil was established by the spectrophotometric Folin-Ciocalteu method. DPPH spectrophotometric method was used to analyze the antioxidant activity. The results have shown statistically significant differences between basil cultivars under the same growing conditions. The values of antioxidant activity in frozen samples ranged from 5.1 ±0.4 mg.g-1 AA FW to 11.71 ±0.18 mg.g-1 AA FW and the total phenolic content varied between 2.77 ±0.16 mg.g-1 GAE FW in TH and 8.93 ±0.13 mg.g-1 GAE FW. A statistically significant difference between fresh and frozen samples was established only in the “Ohře” cultivar. After the storage in cold temperatures, all mixtures showed a reduction in the antioxidant activity and total content of phenolic substances. The mixture of basil and mint performed the highest values of antioxidant activity and total content of phenolic substances and significantly differed from the other mixtures most often. The addition of basil and its mixtures to food and beverages can substantially increase their biological value and subsequently also the quality of human nutrition.
