Pollen morphology of 23 species belonging to Geranium have been studied in details, which represent eight sections of two subgenera i.e., G. sect. Dissecta, Geranium, and Tuberosa of subgen. Geranium, Divaricata, Lucida, Ruberta and Trilopha of subgen. Robertium. These plant species were collected from different phytogeographical regions of Iran. The palynological investigation was done using scanning electron microscopy (SEM) techniques. Different palyno-morphological features have been observed, and the closely related species were distinguished. We used different multivariate statistical methods to reveal the species relationships. Ward clustering analyses have been done to check out the relationship among the species. The shapes of pollen grains were monad, radially symmetric, isopolar, apertures were tricolporate, and of spheroid, prolate-spheroid or sub-prolate classes. Three pollen types were recognized on the basis of differences in exine sculpturing pattern: reticulate-clavate, striate-rugulate, reticulum cristatum with clavae. Observed differences were not of diagnostic importance in subgenera and sections level. The main objective of this study is to find distinguish pollen characters in the species of the genus Geranium and to elucidate their systematics importance.

AbstractPollen size and exine structure of the following five Onosma L. species from Slovakia and Italy were studied by light and scanning electron microscopy: Onosma echioides, O. tornensis, O. visianii, O. arenaria, and O. pseudoarenaria. Among the taxa studied, O. echioides has the smallest pollen grains in average (10.5–17.5 × 8.0–14.5 µm), and O. pseudoarenaria the largest grains in average (15.3–20.5 × 12.3–16.3 µm). Pollen grains of all 5 species are small sized, 3-syncolporate, subprolate, heteropolar, with ovate equatorial outlines and circular to rounded triangular polar outlines; the tectum is microechinate. A positive correlation is found between pollen size and chromosome number. The value of pollen characters for taxonomic purposes and the position of the taxa studied within the genus Onosma are discussed.

The paper presents pollen micromorphology by scanning electron microscopy of seven Anabasis L. species namely, A. haussknechtii Bge., A. aphylla L., A. calcarea (Charif & Allen) Bokhari & Wendelbo, A. eugeniae Iljin, A. eriopoda (Shrernk) Volkens, A. annua Bge. and A. setifera Moq. from Iran. Pollen grains in all studied species are peripolyporate, spherical and are of two basic types based on pores diameter inner of holes. The study showed that the sculpturing of exine provides valuable characters in species delimitation, sometimes even for closely related ones. Two new hybrids of Origanum, O. ×malyeri and O. ×sevcaniae, are described and illustrated in detail. The general morphology, pollen and chromosome features, and nuclear (ITS) and chloroplast (rpl32) genomes of collected specimens were studied. Origanum ×malyeri is a hybrid between O. boissieri and O. vulgare subsp. hirtum and O. ×sevcaniae is another hybrid between O. vulgare subsp. hirtum and O. vogelii. These two new hybrids have some morphological intermediate characters between their parents. The pollen morphology of O. ×sevcaniae and its parents were studied and documented in detail using light microscopy (LM) and scanning electron microscopy (SEM). O. ×sevcaniae pollen is small to medium (P=18.49–21.03 µm, E=22.20–26.63 µm), suboblate (73%) to oblate (22%), the range of P/E ratio is from 0.73 to 0.85. Pollen grains are hexacolpate with granular membranes. Pollen grains were identified as bireticulate exine ornamentation with scanning electron microscopy examinations. The somatic chromosome number of O. boissieri, O. vulgare subsp. hirtum, O. vogelii, and O. ×sevcaniae were counted as 2n = 30. Pollen characters and somatic chromosome numbers of O. ×malyeri could not be examined because it has only female flowers and has not any nutlets. Also, according to molecular results, O. ×malyeri and O. ×sevcaniae have some polymorphic nucleotide regions in their nuclear ITS genomes and which means they have two different DNA dataset belonging to their parents. Also, some individuals of O. ×sevcaniae parents have single nucleotide polymorphism, too. These individuals live together with their hybrids and hybridization is probably a continuing process in that hybrid swarms. These present results support previous reports for the speciation of Origanum members via hybridization.

The pollen morphology of 10 Allium L. taxa, 6 of which are endemic to Turkey, were investigated in detail by light and scanning electron microscopy. According to LM and SEM, the pollen grains of genera were monad, monosulcate percolate, heteropolar with bilateral symmetry, 25.30 to 53,85 µm long axis (LA) and 17.55 to 36.86 µm short axis (SA), the form was prolate (mean of LA/SA ratio 1.30 to 1.70 and in polar view boat-shaped. Three types of ornamentation were determined. Striate-rugulate-perforate type in Allium longisepalum, A. oreophilum, A. anacoleum, A. microspathum, A.shirnakiense, A. purpureoviride and A. armenum, Rugulate–perforate type in A. pervariense and A. gabardagense and Rugulate–reticulate-perforate type in A. arlgirdense. Sulcus membrane ornamentations were rugulate or psilate. The sulcus extends from the distal to proximal ends in A. anacoleum, A. arlgirdense and A. pervariense. The present study on some Turkish species of Allium showed that several morphological pollen characters may possess taxonomical value.

The present study provides novel information about the pollen of Salvia assurgens. Pollen grains were collected and described based on their observed characters by light microscope and scanning electron microscopy. The species is distinguished from other Mexican salvias by having small pollen grains (14.2 × 18.2 µm), thin primary muri (0.2 µm thick), elongated primary lumina (1.03 µm long) and secondary lumina with relatively few perforations (9, range 5–14). Pollen characters are similar to those of the majority of American salvias. Regarding Mexican sages, there has been little palynological research, and only 23 species of 32 examined have been quantitatively studied. Standardization is needed in different aspects of palynological studies, especially in relation to measurement protocols and data analysis, as well as the increased use of scanning electron microscopy (SEM), since the majority of differences among species are provided by SEM microscope observation.

AbstractThe Pollen morphology of 13 taxa 11 of which are endemics belonging to Paronychia Miller (Caryophyllaceae) viz., P. agryloba, P. angorensis, P. arabica subsp. euphratica, P. carica, P. cataonica, P. chinonea, P. condensata, P. davisii, P. dudleyi, P. galatica, P. kurdica subsp. kurdica, P. kurdica subsp. montis-munzur and P. mughlaei from Turkey has been investigated by light (LM) and scanning electron microscopy (SEM). LM observations show that pollen grains are usually radially symetrical, isopolar, pantoporate, polygonal (6-gonal) or polygonal-spheroidal. Tectum is psilate or punctate. Tectal surface sparsely-densely spinulose. The numbers of pores are between 6 and 12. On the basis of pollen sizes, P. davisii was the biggest pollen type (23.45 µm) and P. kurdica subsp. kurdica (16.2 µm) was the smallest pollen types. According to exine sculpturing, pollen size and spinule numbers per 1 µm2, three pollen types were distinguished.

Pollen morphology of 16 genera and 101 species from the Myrtaceae tribes Backhousieae, Melaleuceae, Metrosidereae, Osbornieae and Syzygieae was surveyed using scanning electron microscopy (SEM) and light microscopy (LM). The most common pollen type observed in these tribes was parasyncolpate with arcuate or angular colpi, and a rugulate exine pattern. There was little size variation in observed pollen, except for larger pollen in tribe Melaleuceae. All Metrosideros pollen grains had apocolpial islands, as well as all Callistemon species viewed by LM. Choricarpia of tribe Backhousieae had pollen with a distinctive exine pattern. Dicolporate pollen were observed in two tribes, Metrosidereae (Tepualia) and Syzygieae (Acmena), and may be of systematic value. The dicolporate grains of these two genera were also easily distinguishable from each other by using size and pollen side shape as diagnostic characters. Two pollen types were observed within the genus Melaleuca, and a number of pollen types were observed within the species-rich genus Syzygium.

The morphology of the microcirculation of uveal melanomas is a reliable market of tumor progression. Scanning electron microscopy of cast corrosion preparations can generate three-dimensional views of these vascular patterns, but this technique sacrifices the tumor parenchyma. Formalin-fixed wet tissue sections 100-150 microns thick from uveal melanomas were stained with the lectin Ulex europaeus agglutinin I (UEAI) and proliferating cell nuclear antigen (PCNA) to demonstrate simultaneously the tumor blood vessels and proliferating tumor cells. Indocarbocyanine (Cy3) was used as a fluorophore for UEAI and indodicarbocyanine (Cy5) was used for PCNA. Double labeled sections were examined with a laser scanning confocal microscope. Images of both stains were digitized at the same 5-microns intervals and each of the two images per interval was combined digitally to form one image. These combined images were visualized through voxel processing to study the relationship between melanoma cells expressing PCNA and various microcirculatory patterns. This technique produces images comparable to scanning electron microscopy of cast corrosion preparations while permitting simultaneous localization of melanoma cells expressing PCNA. The microcirculatory tree can be viewed from any perspective and the relationship between tumor cells and the tumor blood vessels can be studied concurrently in three dimensions. This technique is an alternative to cast corrosion preparations.

In order to evaluate the significance of the pollen morphology for generic and infrageneric taxonomy of the genus Eremanthus (Vernonieae, Asteraceae), and to provide additional data for its phylogenetic reconstruction, the pollen of 20 of the 23 species of the genus was examined using light and scanning electron microscopy. Acetolysed pollen grains were measured, described, and illustrated using light microscopy, while non-acetolysed pollen grains were analyzed using scanning electron microscopy. Pollen grains of these species are isopolar, oblate-spheroidal in most of the species, more rarely prolate spheroidal or suboblate, subtriangular amb, tricolporate and subechinolophate. The variation among quantitative characters does not correlate with the macromorphological subdivision of the genus or with the generic or specific limits.

Abstract Pollen grains of 10 species of Pedicularis occurring in Ukraine were investigated using light and scanning electron microscopy. Palynological data on 6 species are reported for the first time. General pollen morphology of the genus Pedicularis is presented. Three types of apertures (2-syncolpate, 3-colpate, and 3-syncolpate) and seven subtypes of sculpture (3-syncolpate pilate, 3-syncolpate microscabrate-tubeculate, 2-syncolpate microscabrate, 2-syncolpate microscabrate-tubeculate, 2-syncolpate microscabrate-tubeculate-perforate, 2-syncolpate microfoveolate, and 2-syncolpate microscabrate-perforate) are indentified. The subgenus Pedicularis is heterogeneous in its types of apertures and sculpture of the surface, which indicates the need of further taxonomic revision of the group.
