The objective of this study was to establish the effects of red spectrum of light (650nm, treated n=12) and normal spectrum of light (450nm control=12) on GnRH-I and GnIH mRNA expression, amplitude and frequency of luteinizing hormone (LH) and egg production from 42 to 52 weeks of age in white leghorn hens. Blood samples were collected at weekly interval from both the groups. At the 47th week of age blood samples from both the groups were collected at every 3 h for 36h to study the pulsatile secretion of LH surges. GnRH and GnIH mRNA expression pattern was studied between control and treated birds. Egg production and pause days were calculated between the two groups. LH concentration in the plasma was increased significantly (P<0.01) in hens exposed to red spectrum of light. Plasma LH concentration was higher (P<0.01) in treated birds with more number of LH surges. The amplitude and frequencies of LH were advanced in birds exposed to red spectrum of light during 36 h of sampling at 3h intervals. GnRH-I mRNA concentration was significantly (P<0.01) higher, whereas GnIH mRNA was significantly (P<0.01) lower in birds exposed to red spectrum of light compared to controls. It is hypothesized that exposure of birds to red spectrum of light enhanced (P<0.01) GnRH-I mRNA, along with LH required for ovulation and egg lay. During 77 days (42-52 weeks of age) of the experimental period, egg production was increased (p<0.01) with lower incidence of pause days in the treated group. It is concluded that low GnIH mRNA and higher levels of GnRH-I mRNA, LH, lower number of pause days enabled the birds to lay more eggs by stimulating GnRH through red spectrum of light.

The effects of a number of neuronally localized peptides have been ascertained on corticospinal and other unidentified neurons in the rat cerebral cortex. Motilin, somatostatin, and luteinizing hormone releasing hormone excited most of the corticospinal neurons on which they were tested. Cholecystokinin, Met-enkephalin, vasoactive intestinal peptide, and neurotensin also excited some corticospinal neurons. Many nonidentified neurons were excited by all of these peptides. Met-enkephalin had a depressant action on some (14%) corticospinal neurons. Leu-enkephalin depressed many identified and nonidentified neurons and had an excitatory action on a few neurons. Both excitatory and inhibitory actions of the enkephalins were antagonized by naloxone. Thyrotropin-releasing hormone had predominantly depressant actions on the spontaneous firing of corticospinal and nonidentified neurons but did excite some unidentified cortical neurons. Secretin had no effect on the firing of most of the neurons tested.

The aim of this study was to identify variations of vasoactive intestinal peptide (VIP) and VIP receptor-1 (VIPR-1) genes that might be associated with turkey reproductive traits. One hundred twenty turkey hens were recorded for age at first egg (AFE), first egg weight (FEW), egg number (EN), total egg weight (TEW), laying period (LP), and broodiness. The DNA was isolated from blood samples and subjected to PCR amplification of the meleagrine VIP and VIPR-1 genes. The SNPs were detected by single-strand conformation polymorphism and the variant DNA fragments were sequenced. One mutation in 3’-UTR of VIP (G5846A) and two SNPs in intron 2 of VIPR-1 (C17687T and A17690T) were found, all of them novel. The associations of the three detected SNPs with the reproductive traits of turkeys were evaluated. The detected polymorphisms were used for marker-trait association analyses. The results of association analysis showed that G5846A on 3’ UTR of VIP has a significant association with LP, EN, TEW, and AFE. The G allele of G5846A was the favourable SNP allele for LP, EN, and TEW traits. The AA genotype of A17690T on intron 2 of VIPR-1 was significantly associated with higher LP, EN, and TEW. AGAA haplotype showed association with higher EN and TEW. These results suggest that the SNPs in 3’-UTR of VIP and intron 2 of VIPR-1 genes may influence egg production traits in turkey hens.Keywords: broodiness, candidate gene, egg production, single nucleotide polymorphism, VIPR-1
