Survival and Persistence of Nonpathogenic Escherichia coli and Attenuated Escherichia coli O157:H7 in Soils Amended with Animal Manure in a Greenhouse Environment

ABSTRACT Animal manure provides benefits to agriculture but may contain pathogens that contaminate ready-to-eat produce. U.S. Food and Drug Administration standards include 90- or 120-day intervals between application of manure and harvest of crop to minimize risks of pathogen contamination of fresh produce. Data on factors affecting survival of Escherichia coli in soils under greenhouse conditions are needed. Three separate studies were conducted to evaluate survival of nonpathogenic E. coli (gEc) and attenuated E. coli O157:H7 (attO157) inoculated at either low (4 log CFU/ml) or high (6 log CFU/ml) populations over 56 days. Studies involved two pot sizes (small, 398 cm3; large, 89 liters), three soil types (sandy loam, SL; clay loam, CL; silt loam, SIL), and four amendments (poultry litter, PL; dairy manure liquids, DML; horse manure, HM; unamended). Amendments were applied to the surface of the soil in either small or large containers. Study 1, conducted in regularly irrigated small containers, showed that populations of gEc and attO157 (2.84 to 2.88 log CFU/g) in PL-amended soils were significantly (P < 0.05) greater than those in DML-amended (0.29 to 0.32 log CFU/g [dry weight] [gdw]) or unamended (0.25 to 0.28 log CFU/gdw) soils; soil type did not affect E. coli survival. Results from study 2, in large pots with CL and SIL, showed that PL-amended soils supported significantly higher attO157 and gEc populations compared with HM-amended or unamended soils. Study 3 compared results from small and large containers that received high inoculum simultaneously. Overall, in both small and large containers, PL-amended soils supported higher gEc and attO157 populations compared with HM-amended and unamended soils. Populations of attO157 were significantly greater in small containers (1.83 log CFU/gdw) than in large containers (0.65 log CFU/gdw) at week 8, perhaps because small containers received more regular irrigation than large pots. Regular irrigation of small pots may have affected E. coli persistence in manure-amended soils. Overall, PL-amended soils in both small and large containers supported E. coli survival at higher populations compared with DML-, HM-, or unamended soils.

The environmental variables affecting Escherichia coli O157:H7 regrowth in dairy manure compost were investigated. Factors evaluated were moisture content, strain variation, growth medium of inoculum, level of background microflora and inoculum, different days of composting, and acclimation at room temperature. A mathematical model was applied to describe E. coli O157 regrowth potential in compost. Repopulation occurred in autoclaved compost with a moisture content as low as 20% (water activity of 0.986) in the presence of background microflora of 2.3 to 3.9 log CFU/g. The population of all three E. coli O157 strains increased from ca. 1 to 4.85 log CFU/g in autoclaved compost, with the highest increase in the spinach-outbreak strain. However, E. coli O157 regrowth was suppressed by background microflora at ca. 6.5 log CFU/g. By eliminating acclimation at room temperature and increasing the inoculum level to ca. 3 log CFU/g, E. coli O157:H7 could regrow in the presence of high levels of background microflora. E. coli O157:H7 regrowth in the autoclaved compost collected from the field study was evident at all sampling days, with the population increase ranging from 3.49 to 6.54 log CFU/g. The fate of E. coli O157:H7 in compost was well described by a Whiting and Cygnarowicz-Provost model, with R2 greater than 0.9. The level of background microflora was a significant factor for both growth and death parameters. Our results reveal that a small number of E. coli O157 cells can regrow in compost, and both background microflora and moisture content were major factors affecting E. coli O157:H7 growth.

The fate of Salmonella and Escherichia coli O157:H7 in swine or dairy manure amended into sandy loam or loam soil under field conditions was studied. Soil was amended with manure inoculated with a Salmonella or E. coli O157:H7 cocktail, then transferred to 0.22 μm pore size membrane walled vials. The vials were then placed on the surface or at 15 cm depth in the test plots. Pathogen numbers, soil moisture, rainfall, and temperature were measured throughout the three trials (20–47 weeks duration) representing spring or fall application. Survival curves were characterized by having an initial rapid decline in pathogen numbers followed by a slower inactivation phase with an occasional increase in culturable cells. The CT99.9 values (time to reach a 3 log CFU reduction) varied from 2 to 120 days, with the most rapid decrease being observed on the surface of sandy loam soil. The persistence of pathogens is primarily governed by variations in moisture and temperature, although season of application along with manure and soil type also contribute. To generate more accurate predictive pathogen models, there is a need for laboratory-based trials to mirror the dynamic variation in temperature and soil moisture encountered within the natural environment.

Outbreaks of enterohemorrhagic Escherichia coli O157:H7 infections associated with lettuce and other leaf crops have occurred with increasing frequency in recent years. Contaminated manure and polluted irrigation water are probable vehicles for the pathogen in many outbreaks. In this study, the occurrence and persistence of E. coli O157:H7 in soil fertilized with contaminated poultry or bovine manure composts or treated with contaminated irrigation water and on lettuce and parsley grown on these soils under natural environmental conditions was determined. Twenty-five plots, each 1.8 by 4.6 m, were used for each crop, with five treatments (one without compost, three with each of the three composts, and one without compost but treated with contaminated water) and five replication plots for each treatment. Three different types of compost, PM-5 (poultry manure compost), 338 (dairy manure compost), and NVIRO-4 (alkaline-stabilized dairy manure compost), and irrigation water were inoculated with an avirulent strain of E. coli O157:H7. Pathogen concentrations were 107 CFU/g of compost and 105 CFU/ml of water. Contaminated compost was applied to soil in the field as a strip at 4.5 metric tons per hectare on the day before lettuce and parsley seedlings were transplanted in late October 2002. Contaminated irrigation water was applied only once on the plants as a treatment in five plots for each crop at the rate of 2 liters per plot 3 weeks after the seedlings were transplanted. E. coli O157:H7 persisted for 154 to 217 days in soils amended with contaminated composts and was detected on lettuce and parsley for up to 77 and 177 days, respectively, after seedlings were planted. Very little difference was observed in E. coli O157:H7 persistence based on compost type alone. E. coli O157:H7 persisted longer (by >60 days) in soil covered with parsley plants than in soil from lettuce plots, which were bare after lettuce was harvested. In all cases, E. coli O157:H7 in soil, regardless of source or crop type, persisted for >5 months after application of contaminated compost or irrigation water.

Studies were conducted to determine the fate of Escherichia coli O157:H7 in dairy manure–based compost in a field setting. Two trials were performed involving duplicate compost heaps constructed at an outdoor fenced site. The compost heaps were composed of dairy manure, old hay, feed waste, a mixture of sawdust and calf feces, and fresh hay. Samples of the composting mixture were inoculated with stx-negative E. coli O157:H7 B6914 at initial concentrations of 107 and 105 CFU/g for trial 1 and trial 2, respectively. Individual sample bags were placed on the surface and at three locations (top, center, and bottom) within each heap. Although the compost heaps achieved temperatures of 50°C or above at all internal locations for at least 7 days, temperature stratification was observed. In trial 1, E. coli O157:H7 was detected by enrichment through 14 days within the heaps. When inoculated with 105 CFU/g in trial 2, E. coli O157:H7 was detected only through days 2, 2, and 5 at the top, center, and bottom locations, respectively. For both trials, the pathogen survived at the heap's surface for up to 4 months. The indicator commensal E. coli and coliforms were inactivated at a rate similar to that for E. coli O157:H7. Results indicate that composting, with periodic heap turning, can be a practical approach to inactivating E. coli O157:H7 in cattle wastes on the farm. Our data also suggest when compost heaps are not turned, E. coli O157:H7 may survive for months at the heap surface.

Studies were done to determine the fate of Escherichia coli O157:H7 in manure compost–amended soil and on carrots and green onions grown in an environmentally controlled growth chamber. Commercial dairy cattle manure compost was inoculated with a five-strain mixture of green fluorescent protein–labeled E. coli O157:H7 at 107 CFU g−1 and mixed with unsterilized Tifton sandy loam soil at a ratio of 1:5. Baby carrot or green onion seedlings were planted into the manure compost–amended soil in pots, and soil samples surrounding the plant, edible carrot roots and onion bulb samples, and soil immediately beneath the roots were assayed for E. coli O157:H7 in triplicate at weekly intervals for the first 4 weeks, and every 2 weeks for the remainder of the plant growth cycle (up to 3 months). E. coli O157:H7 cell numbers decreased within 64 days by 3 log CFU/g in soil and soil beneath the roots of green onions and by more than 2 log CFU/g on onions. E. coli O157:H7 survived better during the production of carrots, with a 2.3-log CFU/g reduction in soil and a 1.7-log CFU/g reduction on carrots within 84 days. These results indicate that the type of plant grown is an important factor influencing the survival of E. coli O157:H7 both on the vegetable and in the soil in which the vegetable is grown.

Cattle feces and hides contribute to carcass contamination with Escherichia coli O157:H7, ultimately impacting beef safety. Primary objectives of our cross-sectional study were to evaluate associations among fecal, hide, and preevisceration carcass prevalence of E. coli O157:H7 and to assess factors affecting carcass contamination. Fecal, hide, and preevisceration carcass samples were collected from up to 32 cattle on each of 45 truckloads presented to a midwestern U.S. abattoir. Enrichment and selective culture were used to assess fecal, hide, and carcass prevalence, and direct plating was used to identify cattle shedding high levels of E. coli O157:H7 in feces. Fecal, hide, and carcass prevalence of E. coli O157:H7 within truckload were significantly correlated (P < 0.05) with each other. Enriched fecal sample prevalence was 13.8%, and high shedder prevalence was 3.3%; 38.5% of hides and 10.5% of carcasses were positive for E. coli O157:H7. We used logistic regression to assess animal- and truckload-level variables affecting the probability of carcasses testing positive for E. coli O157:H7. All truckload-level predictors significantly affected the probability of an E. coli O157:H7–positive carcass, including presence of a high shedder within the truckload (odds ratio [OR] = 4.0; confidence interval [CI], 1.6 to 10.1), high (>25%) within-truckload fecal prevalence (OR = 19.3; CI, 4.7 to 79.0), and high (>50%) within-truckload hide prevalence (OR = 7.7; CI, 3.1 to 19.6). The only significant animal-level predictor was having a positive hide (OR = 1.6; CI, 1.0 to 2.6). Our results suggest that preharvest interventions for reducing E. coli O157:H7 contamination of carcasses should focus on truckload (cohort)–level and hide mitigation strategies.

ABSTRACT Untreated biological soil amendments of animal origin (BSAAO), such as manure, are commonly used to fertilize soils for growing fruit and vegetable crops and can contain enteric bacterial foodborne pathogens. Little is known about the comparative longitudinal survival of pathogens in agricultural fields containing different types of BSAAO, and field data may be useful to determine intervals between manure application and harvest of produce intended for human consumption to minimize foodborne illness. This study generated 324 survival profiles from 12 different field trials at three different sites (UMES, PA, and BARC) in the Mid-Atlantic United States from 2011 to 2015 of inoculated nonpathogenic Escherichia coli (gEc) and attenuated O157 E. coli (attO157) in soils which were unamended (UN) or amended with untreated poultry litter (PL), horse manure (HM), or dairy manure solids (DMS) or liquids (DML). Site, season, inoculum level (low/high), amendment type, management (organic/conventional), and depth (surface/tilled) all significantly (P < 0.0001) influenced survival duration (dpi100mort). Spatiotemporal factors (site, year, and season) in which the field trial was conducted influenced survival durations of gEc and attO157 to a greater extent than weather effects (average daily temperature and rainfall). Initial soil moisture content was the individual factor that accounted for the greatest percentage of variability in survival duration. PL supported greater survival durations of gEc and attO157, followed by HM, UN, and DMS in amended soils. The majority of survival profiles for gEc and attO157 which survived for more than 90 days came from a specific year (i.e., 2013). The effect of management and depth on dpi100mort were dependent on the amendment type evaluated. IMPORTANCE Current language in the Food Safety Modernization Act Produce Safety Rule states no objection to a 90- or 120-day interval between application of untreated BSAAO and harvest of crops to minimize transfer of pathogens to produce intended for human consumption with the intent to limit potential cases of foodborne illness. This regional multiple season, multiple location field trial determined survival durations of Escherichia coli in soils amended with manure to determine whether this interval is appropriate. Spatiotemporal factors influence survival durations of E. coli more than amendment type, total amount of E. coli present, organic or conventional soil management, and depth of manure application. Overall, these data show poultry litter may support extended survival of E. coli compared to horse manure or dairy manure, but spatiotemporal factors like site and season may have more influence than manure type in supporting survival of E. coli beyond 90 days in amended soils in the Mid-Atlantic United States.

Abstract Background: Escherichia coli O157:H7 is one of the most potent zoonotic pathogens that causes mild diarrhea and leads to hemolytic uremic syndrome or death. This study was aimed to assess the prevalence and determinants of Escherichia coli O157:H7 related to diarrhea among under-five children with acute diarrhea. Methods: A cross-sectional study design was carried out in 2018 on 378 under-five-year children recruited randomly from Hospitals in Eastern Ethiopia. Stool specimens were collected and processed using enrichment, differential and selective medium. Among isolates, Escherichia coli O157:H7 was confirmed using latex test (Oxoid, Basingstoke, Hants, England). Factors associated with E. coli O157:H7 infection were identified using binary and multivariable logistic regression. Associations were reported by odds ratio with 95 % confidence interval. Results: Magnitude of E. coli O157:H7 related diarrhea was 15.3% (95%CI: 11.8-19.5). The E.coli infection was positively associated with rural residence (AOR;3.75, 95%CI:1.26-11.20), consumption of undercooked meat (AOR;3.95, 95%CI: 1.23-12.67), raw vegetables and/or fruit juice (AOR;3.37, 95%CI:1.32-8.62), presence of bloody diarrhea (AOR;4.42, 95% CI:1.78-10.94), number of under-five children in a household (AOR;7.16, 95%CI: 2.90-17.70), presence of person with diarrhea in a household (AOR;4.22, 95% CI: 1.84-12.69), owning domestic animal (AOR;3.87, 95% CI: 1.48-10.12) and uneducated mother (AOR;3.14, 95%CI: 1.05-9.42).Conclusion: The magnitude of E. coli O157:H7 related diarrhea among under-five children is relatively high in Eastern Ethiopia. The E. coli infection was associated with sanitation and hygiene in a household. Thus, education focused on food cooking and handling, child care and household sanitation associated with animal manure in rural resident children are helpful it.

In the last decade, Escherichia coli O157:H7 have emerged as important pathogens of the gastrointestinal tract of humans. Healthy cattle have been identified as the primary reservoir, however, the factors affecting heterogeneous E. coli O157:H7 fecal shedding are not fully understood. The aim of this study was to investigate the contribution of E. coli O157:H7 colonization of small and large intestinal sites to the heterogeneity of fecal shedding in cattle. There was a dose-dependant E. coli O157:H7 E318N colonization of duodenum, jejunum, ileum, cecum, ascending colon, spiral colon, descending colon, and the rectoanal junction in vitro with no difference in E. coli O157:H7 colonization of the rectoanal junction and other intestinal sites. There were 10–100 times greater E. coli O157:H7 colonization of intestinal sites from persistent shedding cattle compared with nonpersistent shedding cattle. Novel pathologies were associated with E. coli O157:H7 colonization sites in the small and large intestine. The first pathology, focal petechiae, was present throughout the intestinal tract of cattle that ceased shedding E. coli O157:H7 for 5–12 weeks or in the jejunum, ileum, cecum, and ascending colon of cattle shedding E. coli O157:H7 for 4–5 months. The second pathology, mucosal hemorrhages, was present in the same sites as the focal petechiae in cattle shedding for 5 months and these hemorrhages were in the final stages of repair. Several features of these hemorrhages support this conclusion including the brown appearance, low amount of classic E. coli O157:H7 induced A/E lesions, flattened epithelium, and blunted villi. Although mucosal hemorrhages were present in the jejunum, ileum, cecum, and ascending colon in cattle shedding for 4 months, many other pathologies were also present that were indicative of hemorrhagic enteritis as evidenced by the blood red appearance of hemorrhages, severe edema, and dark red erythema. Escherichia coli O157:H7 were associated with both pathologies suggesting it is the causative agent. The current study supports a relationship between the amount of E. coli O157:H7 colonization in intestinal sites and heterogeneous fecal shedding by cattle.
