This study was designed to investigate the effect of penicillin on reproductive function in female Wistar rats. Fifteen female rats (120 – 160 g) were used for the estrous cycle and histopathological studies. Penicillin (17.14 mg/kg) was administered orally on daily basis for 21 and 50 days respectively for the estrous cycle and histological studies. Estrous cycle was carried out using the technique of Marcondes et al., histologies of the ovaries and uteri were also carried out. Data were analysed using descriptive statistics and student’s t-test at p=0.05. Treatment of rats for 21 days with penicillin (17.14 mg/kg) produced significant (p<0.05) increments in the estrous and metestrous phases as well as a significant (p<0.05) reduction in the proestrous phase of the estrous cycle relative to their respective controls. The histopathological study presented with a moderate endometrial congestion. It can therefore be concluded that penicillin probably has a pro-fertility effect with a moderate deleterious effect on the uteri at histological level in female Wistar rats.

Fifteen female rats (120 – 160 g) were used for the estrous cycle and histopathological studies. Artesunate (1.43 mg/kg) was administered orally on daily basis for 21 and 50 days respectively for the estrous cycle and histological studies. Estrous cycle was carried out using the technique of Marcondes et al., histologies of the ovaries and uteri were also carried out. Data were analysed using descriptive statistics and student’s t-test at p=0.05. Treatment of rats for 21 days with artesunate (1.43 mg/kg) produced significant (p<0.05) increments in the estrous and metestrous phases as well as a significant (p<0.05) reduction in the proestrous phase of the estrous cycle relative to their respective controls. The histopathological study presented with ovarian medullar that is severely congested (hemorrhagic) including expanded lumen of the endometrial glands. It can therefore be concluded that artesunate probably has pro-fertility effect with deleterious effect on the ovaries at histological level in female Wistar rats.

This study was designed to investigate the effect of methotrexate on reproductive function in female Wistar rats. Eighteen female rats (120 – 160 g) were used for the estrous cycle and histopathological studies. Methotrexate (0.071 mg/kg) was administered orally on daily basis for 21 and 50 days respectively for the estrous cycle and histological studies. Estrous cycle was carried out using the technique of Marcondes et al., histologies of the ovaries and uteri were also carried out. Data were analysed using descriptive statistics and student’s t-test at p=0.05. Treatment of rats for 21 days with methotrexate (0.071 mg/kg) produced significant (p<0.05) reductions in the proestrous and estrous phase of the estrous cycle as well as significant (p<0.05) increments in the metestrous and diestrous phases of the estrous cycle relative to their respective controls. The histopathological study revealed that treatment of rats with methotrexate (0.071 mg/kg) for 50 days presented with ovarian medullary part that is congested and oedematous as well as with multiple foci of hemorrhage within the endometria. It can therefore be concluded that that methotrexate probably has dominant pro-fertility effect, but also probably induced deleterious effects on the ovaries and uteri in female Wistar rats.

Clindamycin (8.57 mg/kg) was administered orally on daily basis for 21 and 50 days respectively for the estrous cycle and histological studies. Estrous cycle was carried out using the technique of Marcondes et al., histologies of the ovaries and uteri were also carried out. Data were analysed using descriptive statistics and student’s t-test at p=0.05. Treatment of rats for 21 days with clindamycin (8.57 mg/kg) produced significant (p<0.05) reductions in the proestrous and diestrous phases as well as significant (p<0.05) increments in the estrous and metestrous phases of the estrous cycle relative to their respective controls. The histopathological study presented with no deleterious effects on the ovarian and uterine tissues in the rats. It can therefore be concluded that clindamycin probably has pro-fertility effect without deleterious effects on the ovaries and uteri at histological level in female Wistar rats.

Estrous cycle was carried out using the technique of Marcondes et al., histologies of the ovaries and uteri were also carried out. Data were analysed using descriptive statistics and student’s t-test at p=0.05. Treatment of rats for 21 days with chloramphenicol (7.14 mg/kg) produced significant (p<0.05) reductions in the proestrous and diestrous phases as well as a significant (p<0.05) increment in the estrous phase of the estrous cycle relative to their respective controls. The histopathological study presented with no visible lesions on the ovaries and uteri, but few endometrial glands have expanded lumen. It can therefore be concluded that chloramphenicol probably has pro-fertility as well as exhibits non-deleterious effects on the reproductive tissues of female Wistar rats.

BACKGROUND: Leptin, which is the product of the obese gene, is believed to play important roles in pubertal development and reproductive function in females. In a study using adult male rats, it was found that leptin stimulated secretion of gonadotropin from the pituitary in a dose-related manner. However, there has been no such study in female rats. OBJECTIVE: To investigate the effects of leptin on the production of LH and FSH from the pituitary in female rats, using primary cultured pituitary cells. METHODS: In this study, we determined body weight, serum leptin concentration and serum estradiol (E(2)) concentration in female Wistar rats at 3, 5, 6, 7, 9 and 11 weeks of age, and cultured pituitary cells from 6-week-old female Wistar rats with leptin (0--10(-7) mol/l) and GnRH (0 or 10(-8) mol/l). Then basal and GnRH-stimulated extra- and intracellular LH and FSH were assayed by RIA. RESULTS: Serum leptin concentration increased with increases in body weight and E(2) concentration. The pubertal serum leptin concentration was about 10(-10) mol/l. At a lower or moderate concentration, leptin produced dose-related increases in both basal and GnRH-stimulated extra- and intracellular LH and FSH in pituitary cells. At a concentration of 10 mol/l, leptin significantly (P<0.05) stimulated both basal and GnRH-stimulated extra- and intracellular LH and FSH. However, at greater concentrations, these effects diminished. CONCLUSIONS: These results indicated that leptin induced pituitary cells to produce and secrete both LH and FSH, with or without GnRH. The concentration of leptin that induced the greatest production of gonadotropins by pituitary cells was 10(-10) mol/l, which was the same as the physiological pubertal concentration. Leptin may be involved in the onset of puberty. It is also conceivable that leptin may be a cause of ovulatory failure, not only in weight loss but also in weight gain.

Abstract Background Myrianthus arboreus P. Beauv. (Cecropiaceae) is a medicinal plant used to treat female infertility. The aqueous extract of M. arboreus leaves was found to improve the fertility of healthy female Wistar rats. In the present study, we proposed evaluating the effects of such an extract on an animal model of infertility caused by polycystic ovary syndrome (PCOS), in order to bring scientific evidence to the curative action of this plant against female infertility. Methods Following a 21-day administration (gavage) of letrozole (1 mg/kg), animals with PCOS, indicated by overweight and an estrous cycle blocked in the diestrus phase, were co-treated with letrozole (1 mg/kg) and the aqueous extract of M. arboreus leaves at doses of 20, 110 and 200 mg/kg. The positive control received clomiphene citrate (1 mg/kg) and metformin (200 mg/kg). The negative control received distilled water. Each group of animals was made up of 10 female rats. Vaginal smear was examined 7 days before and during co-treatments. Co-treatments were orally administered for 30 consecutive days and 5 animals per group were sacrificed thereafter for biochemical and histological analyses. The 5 remaining animals in each group were crossbred with males of proven fertility for 5 consecutive days. The daily examination of vaginal smears allowed evaluating fertility index. Following parturition, gestation rate was calculated. Results The aqueous extract of M. arboreus leaves reversed letrozole effects by decreasing body weight, abdominal fat accumulation, and serum levels of LH and testosterone (p < 0.001). Ovarian dynamic was improved and the number of tertiary, Graafian follicles (p < 0.001) and corpus luteum increased while that of cystic (p < 0.001) and atretic follicles (p < 0.01) decreased. These effects were associated with increased serum levels of estradiol, decreased ovarian oxidative stress, the resumption of the estrous cycle, the hypertrophy of uterine epithelial cells and increased fertility index and gestation rate. Conclusions These results justify at least in part, the traditional use of M. arboreus against female infertility and suggest that this plant could be a promising alternative treatment to improve symptoms associated with different PCOS phenotypes.

ABSTRACT The mechanisms involved in the prolongation of the oestrous cycle following LH administration were studied in 4-day cyclic female Wistar rats. In females injected with LH on the morning of dioestrus I there was an increase in ovarian venous blood progesterone as compared with non-injected animals. In both LH-treated females, and those injected with progesterone on the morning of dioestrus I, a slowing up in follicular growth was observed from the afternoon of dioestrus I. The size of follicles greater than 400 urn present in LH or progesterone injected animals on the third day of cycle was similar to the size reached by the same range of follicles in non-injected animals on the second day of the cycle. Hence, the increase in endogenous ovarian progesterone elicited by LH was considered as the cause of the slowing up of follicular growth and therefore of the lengthening of the oestrous cycle duration in female rats injected with LH at the beginning of 4-day cycle.

Abstract Background Purslane is a widely distributed shrub used for the treatment of different ailments. The increasing reproductive complications associated with herbal treatments have led to the need to critically evaluate the safety and/or reproductive potentials of commonly used plant extracts. This study investigated the reproductive effect of methanolic extracts of Portulaca oleracea (MEPO) in adult female Wistar rats. Results Group C showed a significant decrease both in relative ovarian weight (p = 0.000), and relative uterine weight (p = 0.037), when compared with the control. There were no significant (p ˃ 0.05) changes in the levels of follicle-stimulating hormone, luteinizing hormone, progesterone, and estradiol. When compared to the control, groups B and C showed abnormal estrous cycle and cycle arrest especially at the metestrus phase with mild congestion of a few blood vessels in the ovary and uterus. Conclusions MEPO may possess some anti-fertility effect, as it disrupts the estrous cycle of adult female Wistar rats; although it has no major effect on the reproductive hormones, uterus, and ovarian histology of adult female Wistar rats. However, high dose consumption should be taken with precaution.

Cadmium (Cd) is an environmental toxicant and endocrine disruptor in humans and animals, and recent studies have illustrated that the uterus is exceedingly sensitive to Cd toxicity. The aim of the study was to investigate the influence of subchronic (90 days) oral Cd exposure in daily doses of 0.09-4.5 mg/kg b.w. on the balance of sex hormones by estimating estradiol (E2) and progesterone (P) concentrations in the uterus and plasma in comparison with the effects of 17β-E2. Additionally, the uterine weight, histopathological changes in the uterus and ovaries, the regularity of the estrous cycle, Cd bioaccumulation in uterine tissue, and selected biochemical parameters of oxidative stress were determined. A long period of observation (three and six months following the administration period) was used to assess whether the existing effects are reversible. The lowest dose of Cd caused effects similar to 17β-E2: an increase of E2 concentration in the uterus, endometrial epithelium thickness, and disturbed estrous cycle with estrus phase prolongation. The obtained results suggest that Cd causes nonlinear response. Higher doses of Cd caused a significant decrease in E2 concentration in the uterus and plasma, estrous cycle disturbances, endometrium atrophy, and structural damage in the ovaries. This dose additionally induces lipid peroxidation in the uterine tissues. It is noteworthy that a prolonged time of observation after terminating the exposure showed persistent changes in the concentration of E2 in uterine tissue, as well as alterations in estrous cycle phases, and an increase in lipid peroxidation in the uterus. Moreover, significant positive correlations between the plasma E2 concentration and endometrial epithelium thickness in all studied groups were found. In summary, subchronic oral Cd exposure of female rats may result in impaired fertility processes.
