  Objective: The present study was aimed to evaluate antihyperglycemic and antihyperlipidemic activities of methanolic extract of Aristolochia bracteolata (MEAB) against dexamethasone-induced diabetic rat model.Methods: Methanolic extract was prepared by soxhlet extraction and was evaluated for antihyperglycemic and antihyperlipidemic activity using dexamethasone-induced model. The MEAB was administered orally at a dose of 200 and 400 mg/kg body weight glibenclamide was used as standard drug. On 0th and 11th day, blood was collected by retro-orbit plexus.Results: In this model blood glucose levels were determined on 0th and 11th days and MEAB significantly reduced the blood glucose levels in diabetic rats. The effect of MEAB on serum lipid profile such as total cholesterol (TC), triglycerides (TGs), low-density lipoprotein (LDL), very LDL (VLDL), and high-density lipoprotein (HDL) was also measured on the 11th day in the diabetic rats. Significant reduction in TC, TGs, LDL, and VLDL levels and improvement in HDL level were observed in diabetic rats.Conclusion: From the results, it was found that the MEAB possess antihyperglycemic and antihyperlipidemic activities.

  Objective: The objective of the present study is to explore the antihyperglycemic and antihyperlipidemic activities of Jatropha gossypifolia methanolic extract (ME) in streptozotocin (STZ)-nicotinamide (NIC) induced diabetic model.Methods: Type II diabetes was induced by a single dose of NIC (110 mg/kg) and STZ (50 mg/kg b.w.) intraperitoneally. The diabetic animals were treated with ME (50 mg/kg and 100 mg/kg b.w.) of J. gossypifolia. At the end of experimental period, the effect of the ME on creatinine level, triglyceride (TG), total cholesterol (TC), high-density lipoprotein (HDL), low-density lipoprotein (LDL) and very LDL (VLDL) were analyzed. Liver function parameters such as glutamate oxaloacetate transaminase (GOT), glutamate pyruvate transaminase (GPT) were analyzed and liver glycogen content was estimated spectrophotometrically. After scarification of animals, the liver was collected and subjected to histopathology analysis. Glycogen content was estimated spectrophotometrically.Results: The ME treated diabetic rats showed a significant increase in HDL level and a decrease in creatinine, TG, TC, and VLDL levels. The treated group showed a significant decrease in liver function parameters such as GOT and GPT levels and significantly increased the liver glycogen content.Conclusion: These findings demonstrate that ME possess antihyperglycemic and antihyperlipidemic activity against STZ - NIC induced diabetic rats.

Abstract Background Curcuma amada is the most popular traditional medicine in India for the treatment of diabetes. The present study aimed to focus the antidiabetic and antioxidative activity of C. amada through the analysis of biochemical and genomic levels in a dose-dependent manner in streptozotocin-induced male adult rat. Method Streptozotocin-induced diabetic rats were administered orally with hydro-methanolic extract of C. amada at the dose of 10, 20, 40 and 80 mg/100 g body weight of rats for 28 days. The antidiabetic and antioxidative efficacy of the extract on glycemic, enzymatic, genomic and histological sensors along with toxicity study was investigated. Results The result showed a significant antidiabetic and antioxidative effect of the extract at dose-dependent manner. The significant recovery of fasting blood glucose level, serum insulin, activity of carbohydrate metabolic enzymes and antioxidative enzymes in extract-treated diabetic group as compared to untreated diabetic group were noted. After the extract treatment, the size of pancreatic islet and cell population densities were significantly increased. Activities of glutamate oxaloacetate transaminase and glutamate pyruvate transaminase in liver were significantly recovered along with the correction of Bax and Bcl-2 gene expression in hepatic tissue after the extract treatment in diabetic rats in respect to untreated diabetic group. Out of all the doses, the significant effects were noted at the dose of 20 mg/100 g body weight which has been considered as threshold dose in the concern. Conclusion It may be concluded that the significant and corrective effect in most of the sensors was noted at the minimum dose of 20 mg/100 g body weight of hydro-methanolic extract of C. amada without producing any toxicity.

Background: Diabetes mellitus (DM) is a metabolic disorder that has the phenotype of hyperglycemia. According to World Health Organization (WHO) there were 65.1 million diabetics in India in 2013, International Diabetes Federation estimates this to increase to 190 million by 2035. Although a number of drugs are available for treatment of DM, their cost and safety profile are major concern. Medicinal plants are used by clinicians for treatment of diabetes. Gymnema sylvestre (GS) extract has been reported to increase insulin levels in diabetic rats. This study was designed to compare the antihyperglycemic effect of Gymnema sylvestre with metformin.Methods: Diabetes was induced in Sprague-Dawley rats using streptozotocin 45mg/kg. Methanolic extract of Gymnema sylvestre 120mg/kg p.o. prepared using Soxhlet apparatus.Results: GS extract reduced blood glucose levels but not statistically significant. GS extract increased HDL and triglycerides, reduced both serum ALT and AST but no statistical significance seen. Metformin significantly increased serum urea, which was not seen in GS extract group. GS extract showed regenerative changes in pancreas, liver and kidney.Conclusions: The study investigation demonstrates that methanolic extract of GS possesses antihyperglycemic and hypolipidaemic activity and so it can be considered as a promising natural remedy in a prediabetic state and in mild hyperlipidaemia to prevent its progression. Increase in β cell regeneration activity could be a probable mechanism of action. However, further long term clinical studies are recommended to define its possible role in diabetes mellitus and hyperlipidaemia. Role of GS as a potential hepatoprotective agent also needs further evaluation.
