Four furanocoumarins: bergapten, xanthotoxin, isopimpinellin (linear furanocoumarins) and sphondin (angular furanocoumarin) were isolated for the first time from callus tissues of <em>Pastinaca sativa</em> L.(<em>Apiaceae</em>) cultured in vitro on solid medium. The compounds were identified using spectral methods. They are well-known secondary metabolites of the intact plant. This is the first report on the isolation of sphondin from in vitro plant cultures.

Multiple shoots of <em>Telekia speciosa </em>were cultivated on MS medium containing 4.44 µM BAP and 0.54 µM NAA, solidified with agar. After eight weeks of culture the shoots were harvested and extracted with methanol. From the methanol extract one pseudoguaianolide – 2,3-dihydroaromaticin and three thymol derivatives: 8-hydroxy-9,10-diisobutyryloxythymol, 10-isobutyryloxy-8,9-epoxythymyl isobutyrate and 10-(2-methylbutyryloxy)-8,9-epoxythymyl isobutyrate were isolated as major secondary metabolites. Moreover, the shoots produced megastigmane and monoterpene glucosides, which were isolated for the first time from the species. The content of 2,3-dihydroaromaticin in the shoot culture was similar to that found in the intact plant, whereas yields of the three thymol derivatives were higher from multiple shoots than from the plants grown in the open field.

Stevia rebaudiana, belongs to the family of Asteraceae, is a perennial and medicinal shrub. It’s leaves dry powder widely used as a natural sweetener which has no calories but reported to be 50– 300 times sweetener as sweet as sugar, that are said to be having insulin balancing properties without any side effects. This research was first time conducted in Bangladesh where a quantitative analysis were performed for determining the major constituents of in vitro propagated Stevia rebaudiana plant leaves dry powder that was prepared through oven dry after sun dry. The analysis revealed the powder as mild acidic (PH 5.345). Total chlorophyll contents were found to be 0.845mg% where chlorophyll–a and -b were 0.088 and 0.761 mg% respectively.  The major macro nutrients such as ash, crude fiber, total carbohydrate, total protein and fat were determined at the amount of 7.05, 10.5, 53.52, 13.13 and 3.55 gm% respectively where the micro nutrients like Iron (Fe), Sodium (Na), Potassium (K), Calcium (Ca), Magnesium (Mg), Phosphorus (P) and Chloride (Cl) were found at the amount of 34.2, 184.3, 2500, 534.43, 465.34, 304.7 and 49.6 mg% accordingly.  Screening for secondary metabolites showed the presence of alkaloids, flavonoids, saponins and tannins as the amount of 2.83, 17.20, 6.69 and 5.84 mg% respectively in dry powder. These findings also have been compared with the other reported values on Stevia from different countries of the world.   Int J Appl Sci Biotechnol, Vol 3(4): 721-726

AbstractHelichrysum plicatum species are used in Turkish folk medicine as lithagogue, diuretic, and nephritic. Research on the methanol (MeOH) extract of flowers of H. plicatum DC. subsp. plicatum resulted in the isolation of eight known compounds (1–8). The chemical structures of the compounds were determined as β-sitosterol (1), apigenin (2), nonacosanoic acid (3), astragalin (4), β-sitosterol-3-O-β-D-glucopyranoside (5), helichrysin A (6), helichrysin B (7), and isosalipurposide (8) by spectroscopic and chromatographic/spectrometric methods, including 1D and 2D nuclear magnetic resonance and liquid chromatography-tandem mass spectrometry. Nonacosanoic acid (3) was isolated for the first time from H. plicatum DC. subsp. plicatum. The MeOH extract and isolated compounds were evaluated for their in vitro human carbonic anhydrase I (hCAI) and II (hCAII), acetylcholinesterase (AChE), butyrylcholinesterase (BChE), and α-glycosidase inhibitory activities. The IC50 values of H. plicatum DC. subsp. plicatum MeOH extract for hCAI, hCAII, AChE, BChE, and α-glycosidase were found to be 77.87, 52.90, 115.50, 117.46, and 81.53 mg/mL, respectively. The compounds showed IC50 values of 1.43–4.47, 1.40–4.32, 1.69–2.90, 1.09–3.89, and 1.61–3.80 μM against hCAI, hCAII, AChE, BChE, and α-glycosidase, respectively. In summary, H. plicatum DC. subsp. plicatum secondary metabolites demonstrated strong inhibitory effects especially against hCAI and hCAII, whereas the MeOH extract showed a weak inhibitory effect on all enzymes.

The present study targets the protozoan parasite Trichomonas vaginalis that causes a healthy problem among women and rarely among men, by the application of natural product or secondary metabolites extracted from the microalgae Hapalosiphon aureus for the first time in Iraq. methanol extract was explained high activity in three concentration recording 100% of parasite death at 200 µg\ml of methanol extract in about two days while 150 and 100 µg\ml of extract reports activity against the parasite after fourand fivedays post treatment respectively. GC- Mass spectrum of the methanol extract has explain presence of the compound (2- deca - 3,d- dienyloxy) carbonyl benzeoic acid in about 13.28 % from the total composition of methanol extract of microalgae.

In August 2004, Glomerella acutata Guerber & Correll was detected on fruits from highbush blueberry (Vaccinium corymbosum L.) for the first time in Norway. Both the conidial (Colletotrichum acutatum J. H. Simmonds) and the ascigerous (G. acutata) stage developed on naturally infected blueberry fruits. Perithecia also readily formed on blueberries and strawberries inoculated with a culture from highbush blueberry, and on artificial, solid medium (both on strawberry leaf agar and potato dextrose agar). To our knowledge this is the first report worldwide of the teleomorph of Glomerella acutata on a naturally infected host. Accepted for publication 15 January 2007. Published 9 May 2007.

Solanum xanthocarpum is considered an important traditional medicinal herb because of its unique antioxidant, and anti-diabetic, anti-aging, and anti-inflammatory potential. Because of the over exploitation linked to its medicinal properties as well as destruction of its natural habitat, S. xanthocarpum is now becoming endangered and its supply is limited. Plant in vitro culture and elicitation are attractive alternative strategies to produce biomass and stimulate biosynthesis of medicinally important phytochemicals. Here, we investigated the potential influence of seven different monochromatic light treatments on biomass and secondary metabolites accumulation in callus culture of S. xanthocarpum as well as associated biological activities of the corresponding extracts. Among different light treatments, highest biomass accumulation was observed in white light-treated callus culture. Optimum accumulation of total flavonoid contents (TFC) and total phenolic contents (TPC) were observed in callus culture kept under continuous white and blue light respectively than control. Quantification of phytochemicals through HPLC revealed that optimum production of caffeic acid (0.57 ± 0.06 mg/g DW), methyl-caffeate (17.19 mg/g ± 1.79 DW), scopoletin (2.28 ± 0.13 mg/g DW), and esculetin (0.68 ± 0.07 mg/g DW) was observed under blue light callus cultures. Compared to the classic photoperiod condition, caffeic acid, methyl-caffeate, scopoletin, and esculetin were accumulated 1.7, 2.5, 1.1, and 1.09-folds higher, respectively. Moreover, high in vitro cell free antioxidant, anti-diabetic, anti-aging, and anti-inflammatory activities were closely associated with the production of these secondary metabolites. These results clearly showed the interest to apply multispectral light as elicitor of in vitro callus cultures S. xanthocarpum to promote the production of important phytochemicals, and allow us to propose this system as an alternative for the collection of this endangered species from the wild.

A new pterocarpan, aeschynocarpin (1), and the known pterocarpan 2-methoxymedicarpin (2) were isolated for the first time from Aeschynomene fascicularis (Fabaceae) and their structures elucidated by means of spectroscopic {UV/Vis, IR, and NMR (1H, 13C, COSY, HMQC, and HMBC)} and mass spectrometric (EI-MS and HRCIMS) techniques. Both compounds were tested in vitro for their cytotoxic and antiproliferative activities against a panel of cancer cell lines. This is the first report on the presence of pterocarpans in the genus Aeschynomene.
