Aims: To evaluate the antibiotic sensitivity profile of bacteria isolated from swimming pools in Akure, Nigeria. Place and Duration of Study: Department of Microbiology, The Federal University of Technology, Akure Ondo state Nigeria between May and July, 2018. Methodology: Water samples were collected in the morning and evening periods including weekends, from ten (10) swimming pools in Akure, Ondo State, Nigeria. The temperature and the pH of the water samples were measured and recorded at the time of collection. The types and loads of bacteria at different times of each day were determined for each of the swimming pools. Characterization and Identification of the various bacterial isolates were based on Gram-staining techniques and biochemical tests. Antibacterial susceptibility profile of the isolates was evaluated using standard methods. Escherichia coli and Staphylococcus aureus being resistant to multiple antibiotics were subjected to plasmid analysis. Results: Bacteria isolated include; E. coli which had the highest occurrence rate (25%), S. aureus (18.75%), Shigella flexneri (14.50%), Klebsiella pneumoniae (14.50%), Proteus mirabilis (9.37%), Citrobacter freundii (6.25%) Pseudomonas aeruginosa (6.25%) Enterobacter faecalis (3.13%) and Salmonella bongori (3.13%). Ciprofloxacin, pefloxacin and tarivid recorded remarkable zones of inhibition against the isolates. The isolates were notably resistant towards chloramphenicol and septrin. The isolates were further examined for the presence of conjugative plasmids. The results showed that their resistance was chromosomal mediated. Conclusion: Despite the fact that these pools meet the World Health Organization minimum requirement, Microbiological examination of the swimming pools revealed that most of them are contaminated with various pathogenic microorganisms which are potentially harmful.

Packaged water, most especially sachet and bottled water, is one of the major sources of drinking water in Nigeria, especially for commuters, whose major way of quenching thirst is to buy from road side vendors and previous studies has shown that packaged water does not always measure up to standard. Aim: Investigations were done to determine adherence to physiochemical, microbiological standards. Antibiotic susceptibility profile and plasmid profiling of enumerated bacteria using disc diffusion method were carried out on vended packaged water samples. Location of Study: Eleven brands of sachet water and seven brands of bottled water samples vended in Akure, Ondo State were randomly selected. Methodology: Physiochemical analysis was carried out and the parameters were checked in line with standard organization of Nigeria (SON) specifications. Isolation of bacteria was carried out using standard procedures and isolates were identified by various biochemical tests.   Plasmid analysis and curing was conducted following standard protocols Results: Total bacterial counts of sachet water brands ranged from 0.2 × 102 to 4.5 × 102 CFU/ml while that of bottled water brands ranged from 0.1 × 102 to 4.2 × 102 CFU/ml. Bacillus subtilis, Corynebacterium diphtheriae, Enterobacter aerogenes, Enterococcus faecalis, Escherichia coli, Klebsiella pneumoniae, Salmonella typhi, Staphylococcus aureus, S. epidermidis and Shigella dysentariae.  Streptococcus pneumoniae was isolated from the samples. Conclusion: Most packaged water (Bottled water, sachet water) samples are improperly handled and produced, which could serve as a source for spread of antibiotic-resistant potential pathogens, a risk to public health. better surveillance should be done on packaged water sale and production, public health education is encouraged for safety of residents in the Metropolis.

The mean, standard plate count of water samples from the selected apartment, hotel, and municipal’s swimming pools were, 28407.14±28469.05, 8192.86±10556.36, and 3257.14±6250.17which above the WHO Guideline limit, thus signifying contamination.Conclusion: The study recommends improvement in the personal hygiene of swimmers, adequate cleansing of the pools, and enforcement of standards by the government.

The purpose of the study was to assess the water quality parameters of Ala River found in Akure, Ondo State, Southwest, Nigeria. The river is considered one of the most important irrigation and drinking water resources in Akure, Nigeria. Three locations were chosen spatially along the watercourse to reflect a consideration of all possible human activities that are capable of affecting the quality of the river water. The water samples were collected monthly for three consecutive months (February to April 2015) at the three sampling sites. The water samples collected were analyzed for physicochemical parameters which include pH, total dissolved solids, dissolved oxygen, biochemical oxygen demand, total hardness, phosphate,  temperature,  calcium, magnesium, chlorine, nitrate, iron and zinc using standard methods and their environmental effects on the river were investigated. There were variations in the quality of the sampled water when compared with the World Health Organization (WHO) standards for domestic and commercial water for the selected parameters. The traces of some hazardous physical and chemical impurities in the river were above the acceptable limits, and thereby pose a health risk to several rural communities who rely heavily on the river primarily as their source of domestic water. Therefore, the law should be enforced to discourage unnecessary waste dumping and discharging of another form of pollutants into surface water in Nigeria.

Helicobacter pyloriis recognized by the World Health Organization to be the primary cause of peptic ulcers, chronic gastritis, and stomach cancer, though the source of human infection is not well understood. One of the problems in understanding the source of human contamination is the difficulty in isolating the organism from the environment. However, the combination of PCR results with those of culturing of 471 drinking water samples can provide a more accurate picture ofH. pyloridetection. In this method 78 presumptiveH. pyloricolonies out of 266 tap water samples were obtained in the preliminary detection on modified Columbia agar (MCUA) slant relying on urease positivity with a rate of 29.3%. However, only 11 out of them were confirmed by Gram staining and biochemical tests reducing the rate to 4.13% whereas only 3 (1.46%) from 205 reverse osmosis (RO) water samples. Furthermore, only 6 (54.5%) out of the 11 isolates from tap water and 1 (33.3%) of the 3 RO isolates were confirmed by 16SrRNA PCR. Thus PCR confirmation reduced the rate to 2.2%. In addition, only 4 (4%) of 100 tap water samples negative forH. pyloriby culture method wereH. pyloripositive by 16SrRNA. Water samples were collected from 24 districts of Basrah Governorate from February–December 2009. The direct recovery ofH. pylorifrom drinking water is both alarming and scientifically exciting in terms of the investigation of its epidemiology.

Biofilms formation by pathogens microbial Control considered important in medical research because it is the hazarded virulence factor leading to becoming difficult to treat because of its high resistance to antimicrobials. Glycopeptide antibiotic a (Vancomycin) and the commercial bacteriocin (Nisin A) were used to comparative with purification bacteriocin (MRSAcin) against MRSA biofilm. One hundred food samples were collected from Baghdad markets from July 2016 to September 2016, including (cheese, yogurt, raw milk, fried meat, grilled meat, and beef burger). All samples were cultures; S. aureus was confirmation by macroscopic culture and microscopic examination, in addition to biochemical tests. Methicillin resistance S. asureus (MRSA) were identification by antibiotic sensitivity test (AST), Vitek 2 system. The result shown the 60(60%) isolate were identified as S. aureus and 45(75%) gave positive result as MRSA isolate, M13 isolate was chosen as MRSA isolates highest biofilm formation for treatment with MRSAcin, Nisin A(bacteriocin) and Vancomycin (antibiotic) to compared the more antimicrobial have bacteriocidal effect. The sensitivity test uses to determine the effect of MRSAcin, Nisin A, and Vancomycin MIC on MRSA planktonic cell by (WDA). The new study shows the impacts of new kind Pure Bacteriocins (MRSAcin) from methicillin-resistant S. aureus (MRSA) highly effects then (Vancomycin and Nisin A) at different concentration. In a current study aimed to suggest new Bacteriocin is potent highly for the treatment of resistant bacteria biofilm infections in food preservatives

Pretendeu-se mostrar que o herbicida Tebuthiuron, selecionado entre outros usados na cultura de cana-de-açúcar, oferece risco de contaminação para a água subterrânea em função de suas características físico-químicas, principalmente, quando aplicado em áreas de recarga direta de aqüíferos consideradas de alta vulnerabilidade natural. Assim, efetuou-se monitoramento do referido herbicida no período compreendido entre 1995 e 1999, coletando amostras de água de poço semi-artesiano, com 53 metros de profundidade, localizado na microbacia do Córrego Espraiado, município de Ribeirão Preto/SP, Brasil, no qual tem sido freqüente o uso do produto em questão. Para efeito de testemunha da amostra de água foi considerado um poço semi-artesiano de profundidade semelhante, localizado cerca de três quilômetros de distância do poço objeto de monitoramento. Os resultados obtidos mostraram que o Tebuthiuron está presente em todas as amostras analisadas, embora em concentrações abaixo do nível crítico para padrão de potabilidade, conforme os limites mais restritivos estabelecidos pela Organização Mundial de Saúde (para os pesticidas em uso é de 0,1 µg/L por pesticida e de 0,5 µg/L para pesticidas totais). Mesmo com os baixos valores de concentração encontrados fica evidente que o Tebuthiuron está atingindo a água subterrânea podendo aumentar sua concentração pela existência de condições ambientais favoráveis à preservação da molécula em profundidade (tais como baixa temperatura, baixa atividade biológica e ausência de luz). Caso o nível crítico seja atingido, o risco de contaminação poderia ser minimizado por meio da substituição do Tebuthiuron por outro herbicida com menor mobilidade no perfil do solo. OCCURRENCE OF THE HERBICIDE TEBUTHIURON IN GROUNDWATER OF ESPRAIADO CATCHMENT RIBEIRAO PRETO - SP (BRASIL) Abstract The herbicide Tebuthiuron, selected among others utilized in sugar-cane crop, shows groundwater contamination risk due to its physicochemical properties, mainly, when applied in direct recharge areas of aquifers considered of high natural vulnerability. In this context, the monitoring of this herbicide in the period of 1995 through 1999 was realized, by collecting water samples of aquifers 53 m depht, located in Espraiado catchment, Ribeirao Preto/SP (Brazil) in which the product has been extensively used. For blank assays it was utilized water samples collected similarly 3 Km away from the monitoring area. The results obtained showed that Tebuthiuron is present in all analyzed samples. Although in lower concentrations of the critical level for drinkable water, following the restrictive limits established by World Health Organization (for pesticides in use is 0,1 µg/L by pesticide and of 0,5 µg/L for total pesticides). Even if the low concentration values found it is evident that the Tebuthiuron affects the groundwater, and could enhance its concentration by favorable ambiental conditions for molecule preservation in deep water (such as low temperature, low biological activity and absence of light). The present scenary reflects contamination risk in case of reaching the critical level, which could only be minimized by the substitution of Thebuthiuron for other herbicide with lower soil mobility.

Abstract Background The threat of methicillin-resistant Staphylococcus aureus (MRSA) exists globally and has been listed as a priority pathogen by the World Health Organization. One of the sources of MRSA emergence is livestock and its products, often raised in poor husbandry conditions. There are limited studies in Nepal to understand the prevalence of MRSA in dairy animals and its antimicrobial resistance (AMR) profile. A cross-sectional study was conducted in Chitwan, one of the major milk-producing districts of Nepal, from February 2018 to September 2019 to estimate the prevalence of MRSA in milk samples and its AMR profile. The collected milk samples (n = 460) were screened using the California Mastitis Test (CMT) and positive samples were subjected to microbiological analysis to isolate and identify S. aureus. Polymerase Chain Reaction (PCR) was used to identify the presence of the mecA gene and screen for MRSA. Results In total, 41.5% (191/460) of milk samples were positive in the CMT test. Out of 191 CMT positive milk samples, the biochemical tests showed that the prevalence of S. aureus was 15.2% (29/191). Among the 29 S. aureus isolates, 6.9% (2/29) were identified as MRSA based on the detection of a mecA gene. This indicates that that 1.05% (2/191) of mastitis milk samples had MRSA. The antibiotic sensitivity test showed that 75.9% (22/29) and 48.3% (14/29) S. aureus isolates were found to be sensitive to Cefazolin and Tetracycline respectively (48.3%), whereas 100% of the isolates were resistant to Ampicillin. In total 96.6% (28/29) of S. aureus isolates were multidrug-resistant (MDR). Conclusions This study revealed a high prevalence of S. aureus-mediated subclinical mastitis in dairy herds in Chitwan, Nepal, with a small proportion of it being MRSA carrying a mecA gene. This S. aureus, CoNS, and MRSA contaminated milk poses a public health risk due to the presence of a phenotype that is resistant to very commonly used antibiotics. It is suggested that dairy herds be screened for subclinical mastitis and treatments for the animals be based on antibiotic susceptibility tests to reduce the prevalence of AMR. Furthermore, future studies should focus on the Staphylococcus spp. to explore the antibiotic resistance genes in addition to the mecA gene to ensure public health.

Abstract Background Lack of sustainable access to clean drinking water continues to be an issue of paramount global importance, leading to millions of preventable deaths annually. Best practices for providing sustainable access to clean drinking water, however, remain unclear. Widespread installation of low-cost, in-home, point of use water filtration systems is a promising strategy. Methods We conducted a prospective, randomized, controlled trial whereby 16 villages were selected and randomly assigned to one of four treatment arms based on the installation location of Sawyer® PointONE™ filters (filter in both home and school; filter in home only; filter in school only; control group). Water samples and self-reported information on diarrhea were collected at multiple times throughout the study. Results Self-reported household prevalence of diarrhea decreased from 25.6 to 9.76% from installation to follow-up (at least 7 days, and up to 200 days post-filter installation). These declines were also observed in diarrhea with economic or educational consequences (diarrhea which led to medical treatment and/or missing school or work) with baseline prevalence of 9.64% declining to 1.57%. Decreases in diarrhea prevalence were observed across age groups. There was no evidence of a loss of efficacy of filters up to 200 days post-filter installation. Installation of filters in schools was not associated with decreases in diarrhea prevalence in school-aged children or family members. Unfiltered water samples both at schools and homes contained potential waterborne bacterial pathogens, dissolved heavy metals and metals associated with particulates. All dissolved metals were detected at levels below World Health Organization action guidelines. Conclusions This controlled trial provides strong evidence of the effectiveness of point-of-use, hollow fiber membrane filters at reducing diarrhea from bacterial sources up to 200 days post-installation when installed in homes. No statistically significant reduction in diarrhea was found when filters were installed in schools. Further research is needed in order to explore filter efficacy and utilization after 200 days post-installation. Trial registration ClinicalTrials.gov, NCT03972618.
