This study was designed to evaluate the effect of a daily dose of tramadol on selected biomarkers viz: haematological parameters, sperm count, kidney and liver damage in male albino rats. Twenty four wistar rats were divided randomly into two groups: control group and treated groups, the treated group were further divided into four groups and housed in cages. Clean drinking water was served to control (group 1), and 1.6 mg/kg bodyweight of tramadol was administered to group 2 (7 days treatment), group 3 (14 days treatment), group 4 (21 days treatment) and group 5 (21 days treatment +7 days withdrawal) in addition to a daily standard diet for all groups. Treatment of rats with tramadol caused significant decrease (P<0.05) in WBC, platelet and lymph. in group 2, on bicarbonate, AST and protein, it showed significant decrease (P<0.05) in group 3, and it showed significant decrease (P<0.05) in group 5 on Cl-, AST, ALT, bicarbonate, AST, PCV, Hb, RBC, WBC, platelet, lymphocytes and sperm count. The results indicates that tramadol has negative effects on the liver which may induce severe liver damage when used for a prolonged period, the results also shows that tramadol can cause anaemia as seen by the observed negative changes in the blood parameters evaluated. Therefore, administration should be with great caution and from a licensed pharmacist or doctor while self prescription or over the counter administration should be avoided considering the associated adverse effects.

The present study aimed to evaluate the inhibitory effect of Orobanche extract in ethylene glycol induced nephrolithiasis. Thirty male albino rats were divided into five groups each group contains 6 animals, group (1) control group, group (2) animals were supplied with 0.75% ethylene glycol in drinking water, group (3) animals were administrated Orobanche extract 3g/kg orally, group (4) animals were administrated Cystone 500 mg/kg in addition to 0.75% ethylene glycol, group (5) animals were administrated Orobanche extract 3g/kg orally in addition to 0.75% ethylene glycol the experiment continued for 28 days. Serum and the kidney homogenates were analyzed for various biochemical parameters and urine was examined microscopically for crystals. Orobanche treatment group and Cystone treatment group significantly decreased phosphorus, Calcium and Oxalate in kidney tissue of nephrolithiasis rats and significantly decreased kidney and liver marker in serum of nephrolithiasis rats. Conclusion this result revealed that Orobanche extract could be a potential candidate for phytotherapy against nephrolithiasis.

Background Aim. In case of high-dose acetaminophen intake, the active metabolite can not bind to the glutathione, thereby inducing cellular necrosis through binding to the cytosol proteins. This trial was performed to histologically and biochemically investigate whether leptin was protective against liver damage induced by paracetamol at toxic doses.Material and Method. In our trial, 30 female rats, divided into 5 groups, were used. IP leptin administration was performed after an hour in the group of rats, in which paracetamol poisoning was induced. The groups were as follows: Group 1: the control group, Group 2: 20 µg/kg leptin, Group 3: 2 g/kg paracetamol, Group 4: 2 g/kg paracetamol + 10 µg/kg leptin, and Group 5: 2 g/kg paracetamol + 20 µg/kg leptin.Results. The most significant increase was observed in the PARA 2 g/kg group, while the best improvement among the treatment groups occurred in the PARA 2 g/kg + LEP 10 µg/kg group (p< 0.05). While the most significant glutathione (GSH) reduction was observed in the PARA 2 g/kg group, the best improvement was in the PARA 2 g/kg + LEP 10 µg/kg group (p< 0.05).Conclusion. Liver damage occurring upon paracetamol poisoning manifests with hepatocyte breakdown occurring as a result of inflammation and oxidative stress. Leptin can prevent this damage thanks to its antioxidant and anti-inflammatory efficacy.

Background: We investigated whether the frequency of lower urinary tract symptoms (LUTS) increased in patients in whom double-J stents were applied. We also evaluated several medical therapy protocols to treat symptoms related with ureteral stents.Materials and Methods: A total of 108 patients, in whom unilateral double-j stent was applied during ureteral stone treatment, were included. Before the double-J stent was applied, all patients completed storage components of the “International Prostate Symptom Score” (IPSSs), quality of life components of the IPSS (IPSS-QOL) and “Overactive Bladder Questionnaire” (OABq) forms and scores were calculated. After the procedure, cases were randomized into 5 groups, an antiinflammatory was given to Group 1, spasmolytic to Group 2, anticholinergic to Group 3 and α-blocker to Group 4. No additional drug was given to Group 5 as this control group. During the fourth week of the procedure, IPSSs, IPSS-QOL and OABq forms were again completed and scores were compared with the previous ones.Results: When all the cases were evaluated, the IPSSs, IPSS-QOLand OABq scores of patients in whom the double-J stent was applied were statistically significantly higher the procedure. Compared to the control group, the cases where the double-J stent was applied showed a higher IPSSs, IPSS-QOL and OABq scores and none of the medical therapies could prevent this increase.Interpretation: The frequency of LUTS increased in cases where the ureteral stent was applied and discomfort continued as long as the stent stayed in the body.

Abstract Background: Pregabalin (PGB) used as analgesic in treatment of neurogenic pains of chronic diseases, is considered as one of the most abused anti-epileptic drugs worldwide and it has been proved that it induces addictive behaviors. The present histopathological study aimed to identify the effect of PGB administration on cerebral cortex and cerebellar cortex, in both acute and chronic toxicity. Seventy-two male and non-pregnant female adult albino rats’ 6- to 8-week-old divided into 3 main groups of 24 rats each were studied. Group 1 represented the control group and group 2 represented the acute toxicity group, in which rats were given a single dose of PGB (5000 mg/kg) orally by gavage and after 24 hours, rats were sacrificed and examined. Group 3 represented the chronic toxicity group; were given PGB 500 mg/kg orally by gavage for 12 weeks, after which rats were sacrificed and examined. Result: Cerebral cortex tissue of acute toxicity group displayed astrocytosis and dystrophic changes, while in chronic group showed degeneration, necrosis and cellular infiltrates. The cerebellum of chronic groups showed degeneration and shrunken of Purkinje cells. Conclusion: Acute and chronic intoxication with pregabalin adversely altered the structure of cerebral cortex and cerebellum.

In medical sciences, toxicity is an area wherein extensive studies have been carried to improve the diseases as well as to prevent. So, there is a high requirement for novel and improved alternative therapeutic strategies to manage diseases. The liver is the largest gland in the body, which executes several important mechanisms; it stores minerals and vitamins and releases them in periods of need. The main aim of this study was to give a closer insight into potent non- toxic compounds that is capable of modifying the responses. Animals were divided into five equal groups viz control (Group 1), administered with food and water ad libitum, (Group 2) administered with olive oil, (Group 3) administered with zingerone, (Group 4) administered with concanavalin A, (Group 5) administered with cyclosporine A followed by zingerone. Our results revealed significant changes in liver marker enzymes and liver histology of zingerone treated rats when compared to control rats.  A corollary, zingerone has no toxic effect on hepatocytes and was found to be safe at a dose of 10mg/kg b wt and also ameliorates hepatotoxicity.

Objective. To evaluate through FE-SEM the cleanliness and dentinal alterations promoted by different methods of dental sample preparation.Methods. Twenty-five human single-rooted teeth were used. The teeth were cleaned and autoclaved in wet medium and randomly divided into 5 groups (), according to the preparation methods employed—control group: no solutions applied; group 1: cement removal and irrigation with 5.25% % EDTA for 4 minutes each; group 2: 17%  % NaOCl (4 minutes ultrasonic bath); group 3: cement removal and 17%     buffer solution + distilled water (10 minutes ultrasonic); group 4: 17%   NaOCl (3 minutes ultrasonic bath). Specimens were analyzed by field emission scanning electron microscope (FE-SEM), at 1500x magnification. Data were submitted to qualitative analysis according to a scoring system and submitted to Kruskal-Wallis test.Results. In ascending order, as to bind parameters, (i) cleanliness: control, group 2, group 3, group 5, and group 4, (ii) dentinal alterations: group 1, group 5, group 2, group 3, and group 4.Conclusion. The proposed protocol was suitable for subsequent microbiological contamination, because it showed less dentinal morphological alterations with increased removal of organic waste.

Aim.The aim of this study was to evaluate the effect of different restoration techniques on the formation of internal microgaps between materials and dentin in class V restorations.Materials and Methods.Twenty-five extracted human premolars were prepared with standardized class V cavity outlines (3 mm × 2 mm × 2 mm). The cavities were randomly divided into 5 groups of 10 cavities each and restored according to manufacturer’s instructions: Group 1: preheating (55°C) conventional composite (Filtek Z250), Group 2: flowable composite (Filtek Flow), Group 3: Filtek Flow + Filtek Z250 light-cured separately, Group 4: Filtek Flow + Filtek Z250 light-cured simultaneously, and Group 5 (control): Filtek Z250 at room temperature (23°C). The specimens were then thermocycled and cross-sectioned through the center of the restoration. Subsequently, impressions were taken, and epoxy resin replicas were made. The internal adaptation of the materials to the axial wall was analyzed under SEM.Results.The preheated Filtek Z250 (Group 1) showed better internal adaptation than the room temperature groups(P<0.05). The combination of Filtek Flow with Filtek Z250 which was light-cured separately (Group 3) exhibited better internal adaptation than control group(P<0.05).Conclusion.Different restoration techniques exhibit different behavior regarding internal adaptation to dentin after photopolymerization.

Abstract BackgroundThe testis is the male reproductive gland or gonad having two vital functions - to produce both sperm and androgens, primarily testosterone.PurposeThe study aimed to investigate the effect of tramadol and boldenone injected alone or in combinatio for 2 months in rats on testicular function.MethodsGroup 1; normal control, Group 2; tramadol Hcl (TRAM) (20 mg/kg bwt.) (i.p). Group 3; boldenone undecylenate (BOLD) (5 mg/kg bwt) (i.m). Group 4; combination of TRAM (20 mg/kg bwt.) and BOLD (5 mg/kg), respectively for 2 months.ResultsTRAM and BOLD alone and in combination rats showed deteriorated testicular functions, lowered serum steroid levels (FSH, LH and testesterone), elevation in oxidative biomarkers (MDA & NO) and reduction in GSH and SOD, downregulation of StaR and HSD17B3 as well as assessment of testicular histopathological using H&E staining, PAS stain for histochemical assessment of polysaccharides and glycoproteins in the testes and Masson trichrome stain to assess the changes in the collagen fibers.ConclusionThe study illuminated the hazard of administration of these drugs for a long period as well as the prominent deleterious effects reported on concurrent use of both drugs.

The present study was performed in the Department of Medicine and in Veterinary clinic of Bangladesh Agricultural University (BAU), Mymensingh from January to June 2002. Myositis was produced experimentally by injecting oil of turpentine in the gluteal muscles of 4 goats of which 2 goats were given treatment with Diclofenac sodium @1mg/kg body weight (treatment group) for consecutive 3 days and remaining 2 goats were reared as control without giving treatment (control group). All the goats were observed for 7 days. Myositis was characterized by reduced appetite, increased body temperature (1-2°F), lameness, local swelling and cellular changes in blood level. The appetite in both groups reduced sharply from 1st day of myositis and improved from day 2 and became normal on 3rd day after treatment. Body temperature reduced to normal level from 2nd day after administration of diclofenac sodium but in control group, it came down in the same condition from 4th day onward. Moderate lameness was observed in all the goats of both groups. In treatment group lameness reduced completely in 100% goats after 2 consecutive days of treatment but in control group similar result was observed on day 6. Moderate local swelling was reduced to normal level in treatment group 2 days post treatment remain moderate in control group. Total leukocyte count (TLC) was increased significantly (p < 0.05) in both groups in myositic condition than in healthy condition and it came down to normal level in treatment group 3 days post treatment and in control group it remained higher. Similar changes were observed in Neutrophil count. The number of lymphocyte was reduced significantly (p < 0.05) in myositic condition than in healthy ones which again reached to normal level within 3 days of treatment. AbstractBackgroundKetoconazole (Keto), an antifungal drug and a common therapeutic option in the treatment of advanced prostate cancer, is known to cause reproductive dysfunctions. Like Keto, melatonin has antifungal and anticarcinogenic actions. Moreover, the hormone has been used to reverse the damaging effects of different toxicants on the reproductive system. Therefore, this study investigated the effects of Keto with/without melatonin on selected biomarkers in rats.MethodsForty rats of 10 animals per group were used in this study, which lasted for 6 weeks. The control group was administered with saline (0.1 mL/day), while group 2 was administered with Keto during the last 3 weeks of experiment; however, in groups 3 and 4, Keto was administered during the first 3 weeks; thereafter, they were administered with saline and melatonin, respectively, during the subsequent 3 weeks. Keto and melatonin were administered at 100 and 10 mg/kg b.w./day (p.o.), respectively.ResultsThe central effects of Keto are independent of the follicle stimulating hormone (FSH) and prolactin; however, relative to the control group, the drug significantly decreased the gonadotrophin releasing hormone (GNRH) and the luteinizing hormone (LH), substantiated by the corresponding significant decreases in sperm count and sperm morphology. Keto caused significant elevations in malondialdehyde (MDA) and lactate dehydrogenase (LDH) and a significant decrease in catalase (CAT) compared with the control group. Moreover, the drug triggered pro-inflammatory events. In group 3 (Keto recovery), MDA and uric acid levels were returned to the baseline (i.e. control), but not GNRH, LH, C-reactive protein (CRP), LDH, and CAT. Treatment with melatonin after Keto administration caused significant increases in FSH, LH, superoxide dismutase, total antioxidant capacity (TAC), sperm count, and sperm morphology but significant decreases in MDA and CRP, relative to groups 2 and 3.ConclusionsMelatonin ameliorates some biochemical alterations following ketoconazole administration.
