Abstract Daily water intake (DWI) is essential for survival in humans; however, accurate assessment of DWI from drinks and beverages (Wdrinks) or food moisture (Wfoods) is difficult as it depends on self-reported intakes that are prone to inaccuracy. Here, we established an objective method to assess DWI components using doubly labeled water (DLW). Deuterium and H218O were orally administered, and the dilution space and elimination rate of 2H and 18O were measured. DWI was calculated from the deuterium turnover corrected for metabolic water production and insensible water absorption from humidity. Wfoods was estimated using dietary record (Wfoods-DR) or calculated from the total energy expenditure assessed by DLW (Wfoods-DLW). The current results underscore Wfoods-DR underestimation using self-reported dietary assessments, which underestimates food intake. This study proposes novel methods for calculating each DWI component using DLW.

Adequate energy intake is critical for the healthy longevity of older adults, and the estimated energy requirement is determined by total energy expenditure (TEE). We aimed to identify the relationship between measured aerobic capacity and TEE, activity energy expenditure (AEE) or physical activity level (PAL) with the doubly labeled water (DLW) methods in the advanced older adults. A total of 12 physically independent older adults (10 males and 2 females) aged between 81 to 94 years participated in this study. Aerobic capacity was evaluated according to the lactate threshold (LT). TEE under free-living conditions was assessed using the DLW method, and self-reported physical activity was obtained through the Japanese version of the International Physical Activity Questionnaire (IPAQ). LT was significantly positively correlated with TEE, AEE, and PAL after adjustment for age and sex (ρ= 0.77 (P<0.01), 0.86 (p<0.01), and 0.86 (p<0.01), respectively). We found the LT as an aerobic capacity is positively and independently correlated with TEE, AEE or PAL. The present results suggest that maintaining aerobic capacity is an important factor for preventing frailty, although further research is needed to multisite studies and many samples.

Background: Diet is a modifiable behavior of interest in multiple sclerosis (MS); however, measures of diet in persons with MS have not been vetted for feasibility, acceptability, and validity. Methods: This cross-sectional study examined the Automated Self-Administered 24-H (ASA24) Dietary Assessment Tool in 30 persons with MS and 15 healthy control (HC) participants. Participants were prompted to complete six ASA24 recalls and undergo a standard doubly labeled water (DLW) protocol. Acceptability of ASA24 was assessed using an online questionnaire. Total energy expenditure (TEE) from DLW was compared with ASA24-reported energy intake for assessing validity. Results: All participants completed four or more ASA24 recalls, indicating feasibility of ASA24. Regarding acceptability, the hardest part of completing the ASA24 was remembering everything eaten the previous day. Pearson correlation coefficients between DLW TEE and ASA24 kcal/day were not significant among HC (r = 0.40; p = 0.14) or MS (r = 0.26; p = 0.16) participants. The absolute mean error between DLW TEE and ASA24 among HC participants was 694.96 ± 506.25 mean kcal/day and among MS participants was 585.37 ± 529.02 mean kcal/day; this represents a mean difference of 30 and 25%, respectively. Conclusion: This study established the feasibility and acceptability of ASA24 in persons with MS and provides a foundation regarding the need for further validation research examining appropriate outcomes for supporting dietary interventions.

Adequate nutritional therapy and research have been crucial for diabetes and obesity. Recent topics include Calorie restriction (CR) and Low Carbohydrate Diet (LCD). It is rather difficult to calculate energy intake in person, and also to calculate the energy of the meal. There are some methods for investigating these factors, such as the total energy expenditure (TEE), physical-activity-related energy expenditure (PAEE), metabolic equivalent (MET) values, and the doubly-labeled water (DLW) method. Multi factors would be involved in the study. Further investigation would be expected for the determination of an appropriate amount of energy intake and meal energy in the future.
