AbstractWe developed novel closed-form estimators of genetic and environmental correlation coefficients. We applied them to estimate over 4,000 genetic and environmental correlations between multiple phenotypes in a diverse sample from the Trans-Omics in Precision Medicine (TOPMed) program. We found substantial differences in heritabilities, genetic, and environmental correlations of multiple phenotypes and phenotype-pairs between Black, Hispanic/Latino and White populations as well as between sexes. Finally, we quantified genetic and environmental correlations between phenotypic domains, each characterized by multiple phenotypes. Altogether we provide a novel, in-depth framework for examining relations among complex human phenotypes and their determinants.

Background Ethnically endogamous populations can shed light on the genetics of type 2 diabetes. Such studies are lacking in India. We conducted this study to determine the genetic and environmental contributions of anthropometric traits to type 2 diabetes risk in the Sindhi families in central India. Methods We conducted a family study in Indian Sindhi families with at least one case of type 2 diabetes. Variance components methods were used to quantify the genetic association of 18 anthropometric traits with eight type 2 diabetes related traits. Univariate and bivariate polygenic models were used to determine the heritability, genetic and environmental correlation of anthropometric traits with type 2 diabetes related traits. Results We included 1,152 individuals from 112 phenotyped families. The ascertainment-bias corrected prevalence of type 2 diabetes was 35%. Waist circumference, hip circumference and the biceps, triceps, subscapular and medial calf skinfold thicknesses were polygenically and significantly associated with type 2 diabetes. The range of heritability of the anthropometric traits and type 2 diabetes related traits was 0.27–0.73 and 0.00–0.39, respectively. Heritability of type 2 diabetes as a discrete trait was 0.35. Heritability curves demonstrated a substantial local influence of type 2 diabetes related traits. Bivariate trait analyses showed that biceps and abdominal skinfold thickness and all waist-containing indexes were strongly genetically correlated with type 2 diabetes. Conclusions In this first study of Sindhi families, we found evidence for genetic and environmental concordance of anthropometric traits with type 2 diabetes. Future studies need to probe into the genetics of type 2 diabetes in this population.

AbstractThe present study aimed to determine the genetic and environmental etiology of the association between childhood negative emotionality (NE) and hyperactivity/inattention problems (HIP) using South Korean elementary school twins (mean age = 10.19 years, SD = 1.79 years). Telephone interviews were given to mothers of 919 twins (229 monozygotic males: 112 pairs and 5 individuals; 148 dizygotic males: 73 pairs and 2 individuals; 180 monozygotic females: 87 pairs and 6 individuals; 103 dizygotic females: 50 pairs and 3 individuals; 259 opposite-sex dizygotic twins: 127 pairs and 5 individuals) to assess their children’s NE and HIP. Consistent with prior studies, the phenotypic correlation between NE and the HIP was moderate (r = .29; 95% CI = .24, .34). Model-fitting analysis revealed that additive genetic and nonshared environmental influences on NE were .45 (95% CI [.34, .54]) and .55 (95% CI [.46, .66]), respectively, and that additive and nonadditive genetic, and nonshared environmental influences on HIP were .08 (95% CI [.03, .26]), .41 (95% CI [.21, .51]) and .51 (95% CI = .42, .61), respectively. In addition, the additive genetic correlation between NE and HIP was 1.0 (95% CI [.52, 1.00]), indicating that additive genetic factors are entirely shared between the two phenotypes. Nonadditive genetic influences were unique to HIP and not responsible for the NE-HIP association. Nonshared environmental correlation was significant but modest (re = .18, 95% CI [.06, .30]).

Bioelectrical impedance analysis (BIA)-derived phase angle (PhA) is a valuable parameter to assess physical health. However, the genetic and environmental aspects of PhA are not yet well understood. The present study aimed to estimate the heritability of PhA and investigate the relationships between PhA and anthropometric measurements. PhA and skeletal muscle mass index (SMI) were examined using multi-frequency BIA in 168 Japanese twin volunteers (54 males and 114 females; mean age = 61.0 ± 16.5 years). We estimated the narrow-sense heritability of these parameters and the genetic and environmental relationships between them using a genetic twin modeling. For the PhA, 51% (95% confidence interval: 0.33, 0.64) of the variance was explained by additive genetic effects, and 49% (95% confidence interval: 0.36, 0.67) was explained by unique environmental effects. The heritability of PhA was lower than the height, body weight, and body mass index. PhA shared almost no genetic variation with anthropometric measurements and SMI but shared an environmental variation (14%) with SMI. These findings suggest that the genes affecting PhA are different than those affecting anthropometric measurements and SMI. The correlation between PhA and SMI is caused by common environmental factors.

A foundational question in the social sciences concerns the interplay of underlying causes in the formation of people’s political beliefs and prejudices. What role, if any, do genes, environmental influences, or personality dispositions play? Social dominance orientation (SDO), an influential index of people’s general attitudes toward intergroup hierarchy, correlates robustly with political beliefs. SDO consists of the subdimensions SDO-dominance (SDO-D), which is the desire people have for some groups to be actively oppressed by others, and SDO-egalitarianism (SDO-E), a preference for intergroup inequality. Using a twin design (n = 1,987), we investigate whether the desire for intergroup dominance and inequality makes up a genetically grounded behavioral syndrome. Specifically, we investigate the heritability of SDO, in addition to whether it genetically correlates with support for political policies concerning the distribution of power and resources to different social groups. In addition to moderate heritability estimates for SDO-D and SDO-E (37% and 24%, respectively), we find that the genetic correlation between these subdimensions and political attitudes was overall high (mean genetic correlation 0.51), while the environmental correlation was very low (mean environmental correlation 0.08). This suggests that the relationship between political attitudes and SDO-D and SDO-E is grounded in common genetics, such that the desire for (versus opposition to) intergroup inequality and support for political attitudes that serve to enhance (versus attenuate) societal disparities form convergent strategies for navigating group-based dominance hierarchies.

Genetic parameters for milk and health related traits in low and high milk producing Holstein Friesian cows were estimated using farm collected information on the Jos, Plateau, Nigeria. Milk yield and health related records were extracted from database of the West Africa Milk Company Integrated Dairies Limited. An innovative statistical modeling in this study was the application of repeatability animal models to estimate the genetic parameters. The resulting dataset consisted of 5043 lactations from 608 dams and 39 sires. Genetic correlations were obtained by bivariate analyses of all pair-wise combinations between two traits. In high milk yield Holstein, heritabilities estimate recorded for milk yield (0.43), mastitis (0.26) and lameness (0.33) was high while NIS (0.56), PR (0.22), RR (0.67) and PR (0.22) recorded high estimate in low milk yield Holstein cows. Highest genetic correlation was observed between rectal temperature and vaginal temperature (0.91). Lameness had significant (p<0.05), positive and highest environmental correlation with mastitis (0.95). The heritability estimates of health related traits were low to high; therefore, genetic gain through direct selection alone would be slow, yet still positive and cumulative in the divergently classified Holstein Friesian cows on the Plateau.
